Bioactivity of Natural and Engineered Antimicrobial Peptides from Venom of the Scorpions Urodacus yaschenkoi and U. manicatus.

The spread of multidrug-resistant human pathogens has drawn attention towards antimicrobial peptides (AMPs), which are major players in the innate immune systems of many organisms, including vertebrates, invertebrates, plants and microbes. Scorpion venom is an abundant source of novel and potent AMPs. Here, we investigated natural and engineered AMPs from the scorpions Urodacus yaschenkoi and U. manicatus to determine their antimicrobial spectra as well as their hemolytic/cytotoxic activity. None of the AMPs were active against fungi, but many of them were active at low concentrations (0.25-30

Linking toxicity and predation in a venomous arthropod: the case of Tityus fuhrmanni (Scorpiones: Buthidae), a generalist predator scorpion.

BACKGROUND: Scorpions are arachnids that have a generalist diet, which use venom to subdue their prey. The study of their trophic ecology and capture behavior is still limited compared to other organisms, and aspects such as trophic specialization in this group have been little explored. METHODS: In order to determine the relationship between feeding behavior and venom toxicity in the scorpion species Tityus fuhrmanni, 33 specimens were offered prey with different morphologies and defense mechanisms: spiders, cockroaches and crickets. In each of the experiments we recorded the following aspec

The diversity of venom components of the scorpion species Paravaejovis schwenkmeyeri (Scorpiones: Vaejovidae) revealed by transcriptome and proteome analyses.

The recent publication of high-throughput transcriptomic and proteomic analyses of scorpion venom glands has increased our knowledge on the biodiversity of venom components. In this contribution, we report the transcriptome of the venom gland and the proteome of the venom for the scorpion species Paravaejovis schwenkmeyeri, a member of the family Vaejovidae. We report 138 annotated transcripts encoding putative peptides/proteins with sequence identity to known venom components available from different databases. A fingerprint analysis containing the molecular masses of 212 components of the wh

Venom composition of the scorpion Chactas exsul (Scorpiones: Chactidae) from the premontane tropical rainforest of Costa Rica.

Chactas exsul (Werner, 1939) is a specialized, saproxylic scorpion belonging to the diverse family Chactidae, inhabiting the humid tropical forests of Costa Rica and Panama. Characterized by robust pedipalps and a relatively small telson, C. exsul primarily relies on mechanical immobilization for prey capture, though it may utilize venom during challenging encounters. Despite its evolutionary importance, the venom of most of its members remains largely uncharacterized. In this study, we provide a comprehensive proteomic and biochemical analysis of the venom of C. exsul from the Caribbean lowla

Scorpion envenoming in Ecuador: district-level risk, updated scorpion diversity, and challenges for treatment and public health.

This study assessed scorpion envenoming risk in Ecuador at provincial and district levels between 2017 and 2021, estimated national incidence and mortality rates, and identified the likely implicated taxa. Data were obtained from the Ministry of Public Health and aggregated by province and district, with incidence and mortality rates calculated per 100,000 inhabitants using annual population estimates. District-level risk was visualized with choropleth maps, and scorpion diversity and distribution were compiled from updated national inventories. A total of 1514 scorpion stings were reported be

Comparison of the Scorpionism Caused by Centruroidesmargaritatus, Tityuspachyurus and Tityus n. sp. aff. metuendus Scorpion Venoms in Colombia.

Among other scorpion species, Colombia has two genera of the Buthidae family Centruroides and Tityus, considered to be dangerous to humans. This research shares scientific knowledge aiming to a better understanding about the pathophysiological effects of such venoms. The venom of the three species: Centruroides margaritarus, Tityus pachyurus, and T. n. sp. aff. metuendus with biomedical interest were studied. An initial pre-glycemic sample was taken from ICR mice. They were later intraperitoneally inoculated with doses of 35% and 70% of LD50 of total venom. Poisoning signs were observed during

Study on Distribution of Scorpions to Provide Prevention and Interventions in Combating Scorpionism in Poldokhtar County, Lorestan Province, Iran.

INTRODUCTION: Scorpions are arthropods of medical importance classified in the class Arachnida, inhabiting hot and dry environments. All scorpions have a venomous sting and several thousand people die each year from scorpion stings, but this mortality is due to the venom of about 25 species located in northern Africa, the Middle East, India, Mexico and parts of South America. Poldokhtar County belongs to one of the southern cities of Lorestan Province, providing suitable habitats for many different species of scorpions due to its specific climatic conditions. AIM: To examine the fauna of scor

The Role of ProBNP on Prognosis in Scorpion Stings.

INTRODUCTION: Scorpion stings are a major health problem with potentially fatal consequences. Children under the age of 10 y especially face a great risk. Predicting the prognosis is important in reducing mortality and morbidity because it enables the use of early treatment options. In this study, we examine the relationship between proBNP and prognosis in scorpion stings. METHODS: This is a retrospective analysis of patients aged ≤18 y who were admitted to the child emergency service with a scorpion sting. We examined the demographical data, clinical findings, laboratory records, treatments,

Scorpion venom phospholipases A2: A minireview.

Many venomous species, including snakes, bees and scorpions, contain a variety of secreted phospholipases A2 (sPLA2) that contribute to prey digestion and venom toxicity. Based on their primary structures, the different venom sPLA2 have been classified into four groups I, II, III and IX. While the structure of sPLA2 groups II and I has been well characterized, only one crystal structure of group III sPLA2 from bee venom was described. Scorpion venom sPLA2 belong to group III with a particular heterodimeric structure composed of a long enzymatic chain linked by a disulfide bridge to a Short one

Target-Driven Evolution of Scorpion Toxins.

It is long known that peptide neurotoxins derived from a diversity of venomous animals evolve by positive selection following gene duplication, yet a force that drives their adaptive evolution remains a mystery. By using maximum-likelihood models of codon substitution, we analyzed molecular adaptation in scorpion sodium channel toxins from a specific species and found ten positively selected sites, six of which are located at the core-domain of scorpion α-toxins, a region known to interact with two adjacent loops in the voltage-sensor domain (DIV) of sodium channels, as validated by our newly

Nephrotic syndrome after scorpion sting.

Scorpion venom is a water soluble, antigenic and heterogeneous mixture. The venom is composed of varying concentration of neurotoxin, cardiotoxin, nephrotoxin, haemolytic toxin, phosphodiesterase, phospholipases, hyaluronidases, glycosaminoglycans, histamine, serotonins, and tryptophan and cytokine releasers. The reported incidence of scorpion sting in India is 0.6 %. Scorpion sting resulting in acute renal failure has been reported in the past, but not the nephrotic syndrome. We report a patient of nephrotic syndrome after scorpion sting. The lacunae in the present knowledge linking scorpion

Epidemiological review of scorpion envenomation in iran.

This epidemiological review was carried out to display the magnitude and the geographic distribution of scorpion envenomation in Iran with focus on the southwestern region of Iran, particularly. The Iranian recognized scorpions belonging to two families, including Buthidae and Scorpionidae. Buthidae family consists of 14 genuses, 26 species, and 18 sub-species, while Scorpionidae family has three genuses and four species. The lack of basic knowledge, including the geographical distribution, clinical manifestations, and specific treatments related to scorpiofauna justifies such multidisciplinar

Emerging options for the management of scorpion stings.

Scorpion stings are common in many tropical countries. Although most scorpion stings cause only localized pain without life-threatening envenoming, about one third of stings cause systemic envenoming which can result in death. Children are particularly sensitive to scorpion envenoming. The severity of scorpion stings is related to the presence of neurotoxins in the venom that cause a sudden release of neurotransmitters from the autonomic nervous system, predominantly sympathetic. There is also a strong inflammatory response that worsens symptoms, including those of a respiratory nature. Severa
