Tensilin-like stiffening protein from Holothuria leucospilota does not induce the stiffest state of catch connective tissue.

The dermis of sea cucumbers is a catch connective tissue or mutable connective tissue that exhibits large changes in mechanical properties. A stiffening protein, tensilin, has been isolated from the sea cucumber Cucumaria frondosa. We purified a similar protein, H-tensilin, from Holothuria leucospilota, which belongs to a different family to C. frondosa. H-tensilin appeared as a single band with an apparent molecular mass of 34 kDa on SDS-PAGE. No sugar chain was detected. Tryptic fragments of the protein had homology to known tensilin. H-tensilin aggregated isolated collagen fibrils in vitro

[Dangerous marine animals].

Sea-biological basic knowledge for divers is offered only in special lessons for advanced scuba divers. According to statistics, however, five per cent of the deadly diving accidents are caused by underwater organisms. This number could be reduced to a fraction, by correct behaviour during the dive and after an accident. The most frequent accidents with sea animals during water sports are not by unprovoked shark attacks, which cause six deaths world-wide per year on the average, but turn out with usually well camouflaged sea inhabitants, that do not attack humans, rather by their inadvertence

Anti-Inflammatory Effects of Bioactive Compounds from Seaweeds, Bryozoans, Jellyfish, Shellfish and Peanut Worms.

Inflammation is a defense mechanism of the body in response to harmful stimuli such as pathogens, damaged cells, toxic compounds or radiation. However, chronic inflammation plays an important role in the pathogenesis of a variety of diseases. Multiple anti-inflammatory drugs are currently available for the treatment of inflammation, but all exhibit less efficacy. This drives the search for new anti-inflammatory compounds focusing on natural resources. Marine organisms produce a broad spectrum of bioactive compounds with anti-inflammatory activities. Several are considered as lead compounds for

Transcriptomic characterization of the intestine in Stichopus monotuberculatus under gradient temperature stress and HSP gene family-mediated molecular adaptation.

The increasing frequency of extreme temperature events under climate change poses a growing threat to the stability of tropical sea cucumber aquaculture. To characterize the molecular responses of the tropical sea cucumber Stichopus monotuberculatus to acute temperature stress, juveniles were exposed for 96 h to 15 °C, 20 °C, 25 °C, 30 °C, and 35 °C, followed by transcriptomic profiling of the intestine. By transcriptomic analysis, 2258, 634, 1618, and 2980 differentially expressed genes (DEGs) were identified at 15, 20, 30, and 35 °C compared to control, respectively. More DEGs were generally

Holothurian fucosylated chondroitin sulfate.

Fucosylated chondroitin sulfate (FucCS) is a structurally distinct glycosaminoglycan found in sea cucumber species. It has the same backbone composition of alternating 4-linked glucuronic acid and 3-linked N-acetyl galactosamine residues within disaccharide repeating units as regularly found in mammalian chondroitin sulfates. However, FucCS has also sulfated fucosyl branching units 3-O-linked to the acid residues. The sulfation patterns of these branches vary accordingly with holothurian species and account for different biological actions and responses. FucCSs may exhibit anticoagulant, antit

Sulfated fuco-oligosaccharides from Acaudina leucoprocta: Structural characterization and anticoagulant activity.

Sulfated fucans from sea cucumbers are potent anticoagulants, but their structural complexity has hindered understanding of their structure-activity relationship at the oligosaccharide level. Herein, we report the preparation, structural characterization, and anticoagulant evaluation of structurally defined sulfated fuco-oligosaccharides from the sea cucumber, Acaudina leucoprocta. Crude polysaccharides were fractionated by anion-exchange chromatography to obtain a fucan sulfate fraction (WS-0). Mild acid hydrolysis of WS-0 generated an oligosaccharide mixture, which was purified by sequential

Marine sulfated glycans with serpin-unrelated anticoagulant properties.

Marine organisms are a rich source of sulfated polysaccharides with unique structures. Fucosylated chondroitin sulfate (FucCS) from the sea cucumber Ludwigothurea grisea and sulfated galactan from the red alga Botryocladia occidentalis are one of these unusual molecules. Besides their uncommon structures, they also exhibit high anticoagulant and antithrombotic effects. Earlier, it was considered that the anticoagulant activities of these two marine glycans were driven mainly by a catalytic serpin-dependent mechanism likewise the mammalian heparins. Its serpin-dependent anticoagulant action rel

Sulfation pattern of the fucose branch is important for the anticoagulant and antithrombotic activities of fucosylated chondroitin sulfates.

BACKGROUND: The aim is to compare the structures, anticoagulant and antithrombotic activities of two fucosylated chondroitin sulfates isolated from sea cucumbers Isostichopus badionotus (fCS-Ib) and Pearsonothuria graeffei (fCS-Pg), which were known to have different sulfation patterns on the fucose branches. METHODS: The structures of fCSs were identified using 2D NMR. Anticoagulant activities were measured by activated partial thromboplastin time (APTT) and thrombin time (TT), and inhibition of factors IIa, Xa and XIIa was assessed in vitro. Antithrombotic activity was determined ex vivo by

Cytotoxic activity of Holothuria tubulosa (Echinodermata) coelomocytes.

The immune system of marine invertebrates, in particular that of holothurians, still requires further study. Our research showed that coelomocyte cells contained in the coelomic fluid of the sea cucumber, Holothuria tubulosa, are able to lyse, in vitro, red blood cells in rabbits and sheep. A plaque-forming assay showed spherule cells to be the effector cells, able to release cytotoxic molecules after xenogenic cell contact. The coelomocyte lysate supernatant, analysed by polyacrylamide gel electrophoresis overlay technique, using rabbit and sheep erythrocytes, showed two different haemolytic

Marine-Food-Derived Ether Phospholipids Mitigated Alcoholic Liver Disease by Preferably Suppressing Lipid Peroxidation rather than Ferroptosis Defense.

Alcoholic liver disease (ALD) is one of the leading causes of liver-related mortality and poses a major global health burden. Ether phospholipids have emerged as novel candidates in protecting against alcoholic liver disease based on their unique structural and functional properties. In this study, we proposed that dietary ether phospholipids, particularly plasmenyl phosphatidylethanolamine (PlsEtn) and plasmanyl phosphatidylcholine (PakCho) derived from sea cucumber, demonstrate potent hepatoprotective activity against ALD by the inhibition of ferroptosis. In the murine model of ALD, PlsEtn a

Advances in Probiotic and Prebiotic Applications for Sustainable Aquaculture: Harnessing Their Roles in China's Marine Pasture Restoration and Health.

Aquaculture is the fastest-growing food-producing sector globally, yet intensive farming in China has led to severe ecological degradation of coastal marine pastures, including habitat loss, disease outbreaks, and dysbiosis of cultured species. Probiotics (beneficial live microorganisms) and prebiotics (non-digestible substrates that promote beneficial microbiota) offer eco-friendly alternatives to antibiotics and chemicals, aligning with China's "Blue Granary" and marine ecological restoration initiatives. Recent advances demonstrate that multi-strain probiotics (including Lactobacillus, Baci

Medical Gains of Chondroitin Sulfate Upon Fucosylation.

Chondroitin sulfate (CS) is a glycosaminoglycan (GAG) composed of alternating N-acetyl galactosamine and glucuronic acid units within disaccharide building blocks. CS is a key functional component in proteoglycans of cartilaginous tissues. Owing to its numerous biological roles, CS is widely explored in the pharmaceutical market as nutraceutical ingredient commonly utilized against arthritis, osteoarthrosis, and sometimes osteoporosis. Tissues like shark cartilage and bovine trachea are common sources of CS. Nonetheless, a new CS type has been introduced and investigated in the last few decade

Bioinspired Ionic Biogels for Conformal On-Skin Wearable Bioelectronics.

Conductive gels are promising materials for on-skin wearable bioelectronics owing to their softness, ionic conductivity, and tissue-like interfacial characteristics. However, conventional preformed conductive gels often cannot fully accommodate complex and dynamically deforming skin surfaces, thereby limiting their interfacial adhesion and signal-transmission performance. Inspired by the reversible mechanical switching behavior of sea cucumbers, we developed a thermoresponsive ionic biogel (IBG) composed of gelatin, water, and 1-ethyl-3-methylimidazolium ethyl sulfate. In this system, gelatin

A gene cluster encoding a nonribosomal peptide synthetase-like enzyme catalyzes γ-aromatic butenolides.

Sea cucumber-derived fungi have attracted much attention due to their capacity to produce an incredible variety of secondary metabolites. Genome-wide information on Aspergillus micronesiensis H39 obtained using third-generation sequencing technology (PacBio-SMRT) showed that the strain contains nonribosomal peptide synthetase (NRPS)-like gene clusters, which aroused our interest in mining its secondary metabolites. 11 known compounds (1-11), including two γ-aromatic butenolides (γ-AB) and five cytochalasans, were isolated from A. micronesiensis H39. The structures of the compounds were determi

Vitamin C Promoted Feeding and Growth Rate of Apostichopus japonicus by Increasing Its Digestive Capacity Through a 5-Hydroxytryptamine-Mediated Signaling Pathway.

To investigate the effects of dietary vitamin C (VC) supplementation at different concentrations (0, 5000, and 15,000 mg/kg) on the growth and feeding performance of sea cucumbers (Apostichopus japonicus), a 60-day feeding trial was conducted. The results demonstrated that supplementation with 5000 mg/kg VC significantly enhanced growth performance, evidenced by a higher specific growth rate and weight gain, along with a shorter feeding initiation time and increased tentacle feeding frequency. Furthermore, the activities of digestive enzymes-including amylase, lipase, and pepsin-were markedly

Implantable Bioresorbable Scaffold with Fucosylated Chondroitin Sulfate as a Promising Device for Delayed Stimulation of Hematopoiesis.

The aim of this study was to evaluate the prospects of using natural fucosylated chondroitin sulfate (FCS) from the sea cucumber Cucumaria japonica as the active component of an implantable biodegradable scaffold to stimulate hematopoiesis in mice with cyclophosphamide (CPh)-induced myelosuppression. The scaffolds were based on bioresorbable Fe-Mn-C and Fe-Mn-Pd alloys after equal-channel angular pressing (ECAP). The efficiency of the developed constructs with FCS was compared with the activity of the same scaffolds loaded with recombinant human granulocyte colony stimulating factor, as well a

Proteome analysis provides insights into sex differences in Holothuria Scabra.

Sex-determining mechanism is still ambiguous for sea cucumber Holothuria scabra which only manifests gonochorism in gonad. In this study, proteomic analysis was employed to delineate sex-related proteins and genes in gonads of H. scabra, subsequently validated through Quantitative real-time polymerase chain reaction (qRT-PCR). A total of 5,313 proteins were identified via proteome sequencing. Among these, 817 proteins exhibited expression in both the ovary and testis, with 445 proteins displaying up-regulation and 372 proteins showing down-regulation (ovary vs testis). Furthermore, 136 and 69

Different Temperature Regimes Affect Gonadal Development and Energy Budget in Apostichopus japonicus.

Apostichopus japonicus is a temperate marine echinoderm whose growth and reproduction are strongly influenced by temperature. We compared adult sea cucumbers exposed to stepwise warming (10-12, 12-14, and 14-16 °C across three successive 20-day stages) with animals maintained at 15 °C. Growth indices, gonadosomatic index (GSI), gonadal histology, and energy-budget components were evaluated in females and males. The 60 d experiment used three replicate tanks for each treatment-by-sex combination. Compared with the constant 15 °C treatment, the stepwise-warming regime showed a higher female GSI

First Report of Myocastor coypus Infected with Staphylococcus cohnii.

In December 2021, a wildlife conservation base in Hunan, China, reported illness and death among its captive Myocastor coypus population. A gram-positive pathogen was isolated from the affected animals. The 16S rDNA sequence of the isolated strain was approximately 1,500 bp long and exhibited 98.4% homogeneity with Staphylococcus cohnii found in sea cucumbers. Interestingly, the biochemical reactions of the isolated strain were consistent with the characteristics of S. cohnii. The LD100 for BALB/c mice was 1.2 × 107 CFU/g, while the MLD was 0.6 × 107 CFU/g. Inflammatory cell infiltration was o
