Serum levels of sICAM-1 and sE-selectin in patients with dengue virus infection.

The purpose of this study was to measure the serum level of sICAM-1 and sE-selectin as markers for endothelial damage in patients with dengue fever (DF) and dengue hemorrhagic fever (DHF). Twenty-nine patients with serologically-proven dengue virus infection (age 10.6 +/- 2.4 years, 55% male, DF = 13 and DHF = 16) were enrolled. Serum samples were collected from 25 healthy children (age 10.6 +/- 2.3 years, 40% male) as the control group. A follow-up was done at a mean interval of 15.9 +/- 1.6 days. The level of sICAM-1 at the toxic stage was significantly elevated compared to its level at the

Concanavalin-A-induced liver injury is severely impaired in mice deficient in P-selectin.

P-selectin (CD62P) is an adhesion molecule that mediates the initial attachment of leukocytes to activated platelets and endothelial cells in damaged tissues. We evaluated the role of P-selectin in concanavalin A (Con A)-induced hepatitis, a model characterized by CD4(+) T cell activation and infiltration of the liver. Con A injection induced transient P-selectin expression on hepatic venules and platelets. Mice lacking P-selectin showed impaired lymphocyte adhesion to liver venules and sinusoids, a striking reduction in intrasinusoidal occlusion, and decreased lymphocyte infiltration of liver

Transcriptional regulation of expression of carbohydrate ligands for cell adhesion molecules in the selectin family.

Cell adhesion mediated by selectins and their carbohydrate ligands is involved in the adhesion of cancer cells to endothelial cells during the course of hematogenous metastasis of cancer. In patients with leukemia, this adhesion is involved in the extravascular infiltration of leukemic cells. Extravasation and tissue infiltration of malignant cells in patients with adult T-cell leukemia is mediated by the interaction of selectins and their carbohydrate ligand sialyl Lewis X, which is strongly and constitutively expressed on the leukemic cells. Constitutive expression of Lewis X in these cells

Green fluorescent protein-based system for analysis of E-selectin-mediated adhesion.

Numerous cell-based or cell-free systems for study of selectin adhesion use radiolabeled tracers. However, in addition to handling problems associated with the use of radioisotopes, these assays have difficulty relating a number of counts to a number of adherent cells. Here, we describe an assay that uses the natural fluorescence of the green fluorescent protein (GFP) to measure binding of cells to E-selectin. We elaborated an adhesion system composed of a cell monolayer expressing E-selectin ligand to which monodispersed fluorescent Chinese hamster ovary (CHO) cells expressing E-selectin are

Relationship between cleaved L-selectin levels and the outcome of acute myeloid leukemia.

High plasma levels of the shed form of L-selectin (sL-selectin) are frequently detectable in acute myeloid leukemia (AML). sL-selectin can inhibit blast cell adhesion to vascular endothelium and may thereby influence the phenotype of AML. In this study, we have investigated the relationship between sL-selectin levels and clinical presentation or disease outcome in 100 patients with AML. Fifty-eight patients were found to have sL-selectin levels >/=3.12 microgram/mL (>/=3 SD above the mean of healthy controls: "increased"). Patients with extramedullary disease such as lymphadenopathies, splenom

P-selectin is up-regulated in vital organs during murine traumatic shock.

Murine traumatic shock is associated with increased adherence of neutrophils to the vascular endothelium resulting in neutrophil infiltration and tissue damage. We examined the effects of trauma on the expression of the adhesion molecule P-selectin in several vital organs (i.e., heart, lungs, liver, kidneys, and small intestine) 2 h after induction of Noble-Collip drum trauma in anesthetized rats. Total RNA was extracted from these organs and P-selectin mRNA was quantified by RNase protection assays. P-selectin mRNA was significantly increased over control nontraumatized, anesthetized rats in

[E-selectin expression in serum and tissue correlates with distant metastasis of colorectal cancer].

E-selectin is an adhesion molecule of endothelial cells that binds to cancer cells mediated by sialyl Lewis A (sLea) or sialyl Lewis X (sLex). It is suspected to be involved in hematogenous metastasis of tumors. Therefore, it is worth examining E-selectin expression in human colorectal cancer and its hepatic metastasis. In the present study, E-selectin was clearly revealed on the endothelial cells of small vessels adjacent to cancer nests in both primary and metastatic nests in immunohistochemistry. In these tissues, E-selectin was observed on the endothelial cells lining the lumen of small ve

L-selectin-deficient mice have impaired leukocyte recruitment into inflammatory sites.

L-selectin, a cell surface adhesion molecule that is expressed by most leukocytes, mediates leukocyte rolling along vascular endothelium at sites of inflammation. The contribution of L-selectin to leukocyte migration in models of chronic inflammation was assessed by using mice that lack cell surface L-selectin expression. Significant inhibition of neutrophil (56-62%), lymphocyte (70-75%), and monocyte (72-78%) migration into an inflamed peritoneum was observed 24 and 48 h after administration of thioglycollate, an inflammatory stimulus. L-selectin-deficient mice were also significantly impaire

Enforced hematopoietic cell E-selectin/L-selectin ligand expression enhances bone marrow stromal cells homing and amelioration of cerebral ischemia-reperfusion injury via induction of prostaglandin E2.

Ischemic stroke (IS) is a significant and potentially life-threatening disease with limited treatment options, often resulting in severe disability. Bone marrow stromal cells (BMSCs) transplantation has exhibited promising neuroprotection following cerebral ischemia-reperfusion injury (CIRI). However, the effectiveness is hindered by their low homing rate when administered through the vein. In this study, we aimed to enhance the homing ability of BMSCs through lentivirus transfection to express fucosyltransferase 7. This glycosylation engineered CD44 on BMSCs to express hematopoietic cell E-se

E-selectin and transferrin receptor functionalized synthetic miRNA nanoshuttles with therapeutic potential in stroke.

Current acute ischemic stroke treatment is limited to reperfusion therapies within the first hours after the onset, underscoring the urgent need for novel therapeutic strategies to mitigate the progression of the brain lesion. Here, we describe the development of bio-inspired synthetic nanoshuttles-lipid nanoparticles (LNPs)-designed to deliver miRNAs and target post-stroke inflammation, a key contributor to secondary brain damage. miRNAs isolated from human induced pluripotent stem cells (hiPSCs) were encapsulated into LNPs prepared by a microfluidic technique. We produced small sized, unifor

P2Y12-P-selectin-mediated platelet activation in a murine model of dengue-associated thrombocytopenia.

BACKGROUND: Dengue virus (DENV) infection frequently leads to thrombocytopenia, a hallmark of severe disease. However, the underlying mechanisms remain incompletely defined. Therefore, this study aimed to investigate the mechanisms underlying dengue-associated thrombocytopenia in a murine model, with a focus on defective thrombopoiesis and platelet activation. METHODS: Using a murine model that recapitulates key haematological and inflammatory features of human dengue, we investigated the impact of DENV infection on megakaryopoiesis, platelet activation, and thromboinflammatory responses. Cell deformability enhances adhesion of ATRA-differentiated HL-60 cells to P-selectin.

Acute promyelocytic leukemia (APL) is characterized by immature myeloid cells with unlimited proliferative capacity. All-trans retinoic acid (ATRA) can induce these cells to differentiate, but patients may develop differentiation syndrome (DS), a life-threatening complication linked to abnormal leukocyte recruitment. The mechanisms of DS remain poorly understood, despite insights gained from studies on APL cell lines. In this study, ATRA-differentiated HL-60 cells were perfused into a flow chamber simulating the physiological environment and functionalized with P-selectin to mimic the initial

Intrahepatic lipid content is independently associated with soluble E-selectin levels: The Maastricht study.

BACKGROUND: Evidence is accumulating that liver sinusoidal endothelial cells are involved in the pathogenesis of non-alcoholic fatty liver disease. Previous studies have suggested that the endothelial biomarker soluble E-selectin (sE-selectin) is to an important extent liver-derived. AIMS: To study the relationship of intrahepatic lipid (IHL) content with sE-selectin at the population level. METHODS: This study was conducted in participants of The Maastricht Study (n = 1,634), a population-based cohort study enriched with patients with type 2 diabetes. We assessed the cross-sectional associa

The presence of E-selectin in the myocardium indicates a sustained inflammatory reaction in patients with dilated cardiomyopathy.

BACKGROUND: It is still uncertain whether de-novo expression of E-selectin in endothelial cells may be considered an additional marker of chronic inflammation in heart failure (HF). METHODS: We studied 393 consecutive patients (313 M, 80 F) with HF secondary to dilated cardiomyopathy in whom the right ventricular endomyocardial biopsy was performed. For immunohistochemistry, HLA class I and II, E-selectin (ELAM-1), CD3 + lymphocytes and CD68 + macrophages were studied. Patients were divided into two groups: Group A, with ELAM-1 (+), and Group B with ELAM-1 (-) in the biopsy sections. RESULTS
