Distribution of functional opioid receptors in human dorsal root ganglion neurons.

Preclinical evidence has highlighted the importance of the μ-opioid peptide (MOP) receptor on primary afferents for both the analgesic actions of MOP receptor agonists, as well as the development of tolerance, if not opioid-induced hyperalgesia. There is also growing interest in targeting other opioid peptide receptor subtypes (δ-opioid peptide [DOP], κ-opioid peptide [KOP], and nociceptin/orphanin-FQ opioid peptide [NOP]) on primary afferents, as alternatives to MOP receptors, which may not be associated with as many deleterious side effects. Nevertheless, results from several recent studies

Lineage analysis of the olfactory epithelium using a replication-incompetent retrovirus.

We have analysed the lineage of olfactory receptor neurons using a replication-incompetent retrovirus injected beneath the olfactory epithelium of young rats. There are two major types of clusters of infected cells seen at 5--40 days after infection: (i) horizontal basal cells (HBCs); (ii) variable numbers of globose basal cells (GBCs), and immature and mature sensory neurons. Olfactory nerve lesion increased the frequency of the globose/sensory neuron clusters, as well as the number of cells/cluster, but did not change the number of HBC clusters or cells/cluster. No clusters contained sustent

Targeting of olfactory neurons.

Olfactory sensory neurons detect an enormous variety of small volatile molecules with extremely high sensitivity and specificity. The actual recognition and discrimination of odorous compounds is accomplished by specific receptor proteins located in the ciliary membrane of the sensory neurons. Axonal connections into the olfactory bulb, the first relay station for odor processing in the brain, are organized such that all neurons expressing the same odorant receptor converge their axons onto common glomeruli which are located at similar positions in all individuals from one species. For the est

Capsaicin-Sensitive Sensory Nerves and the TRPV1 Ion Channel in Cardiac Physiology and Pathologies.

Cardiovascular diseases, including coronary artery disease, ischemic heart diseases such as acute myocardial infarction and postischemic heart failure, heart failure of other etiologies, and cardiac arrhythmias, belong to the leading causes of death. Activation of capsaicin-sensitive sensory nerves by the transient receptor potential vanilloid 1 (TRPV1) capsaicin receptor and other receptors, as well as neuropeptide mediators released from them upon stimulation, play important physiological regulatory roles. Capsaicin-sensitive sensory nerves also contribute to the development and progression

Histamine induces peripheral and central hypersensitivity to bladder distension via the histamine H1 receptor and TRPV1.

Interstitial cystitis/bladder pain syndrome (IC/BPS) is a common chronic pelvic disorder with sensory symptoms of urinary urgency, frequency, and pain, indicating a key role for hypersensitivity of bladder-innervating sensory neurons. The inflammatory mast cell mediator histamine has long been implicated in IC/BPS, yet the direct interactions between histamine and bladder afferents remain unclear. In the present study, we show, using a mouse ex vivo bladder afferent preparation, that intravesical histamine enhanced the mechanosensitivity of subpopulations of afferents to bladder distension. Morphological Organization, Sensory Structures and Catecholaminergic Nervous System of Lindrilus flavocapitatus (Annelida: Protodrilidae).

Protodrilidae is a small family of almost exclusively interstitial annelids that lack parapodia and chaetae and possess a basiepithelial nervous system. This study presents a histological description of Lindrilus flavocapitatus (Uljanin, 1877), a protodrilid species last examined morphologically in the early 20th century, and provides detailed information on the organization of its nervous and sensory systems using histochemical detection of catecholamines (CAs), scanning electron microscopy (SEM), and alpha-tubulin immunolabelling. The epidermal ciliary structures on the head show a species-s

Glucagon-like peptide 1 and glucose-dependent insulinotropic polypeptide stimulate release of substance P from TRPV1- and TRPA1-expressing sensory nerves.

Glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP) are released from enteroendocrine cells (EECs) in response to nutrient ingestion and lower blood glucose levels by stimulation of insulin secretion and thus are defined as incretins. GLP-1 receptor (GLP-1R) expression has been identified on enteric neurons that include intrinsic afferent neurons, extrinsic spinal, and vagal sensory afferents but has not been shown to have an incretin effect through these nerves. GLP-1 and GIP enter the mesenteric lymphatic fluid (MLF) after a meal via the interstitial fluid

Implantation of neurotrophic factor-treated sensory nerve graft enhances survival and axonal regeneration of motoneurons after spinal root avulsion.

We previously showed that motor nerves are superior to sensory nerves in promoting axon regeneration after spinal root avulsion. It is, however, impractical to use motor nerves as grafts. One potential approach to enhancing axonal regeneration using sensory nerves is to deliver trophic factors to the graft. Here, we examined the regulation of receptors for brain-derived neurotrophic factor, glial cell line-derived neurotrophic factor, ciliary neurotrophic factor, and pleiotrophin after root avulsion in adult rats. We then tested their survival-promoting and neuroregenerative effects on spinal

Skin neurogenic inflammation.

The epidermis closely interacts with nerve endings, and both epidermis and nerves produce substances for mutual sustenance. Neuropeptides, like substance P (SP) and calcitonin gene-related protein (CGRP), are produced by sensory nerves in the dermis; they induce mast cells to release vasoactive amines that facilitate infiltration of neutrophils and T cells. Some receptors are more important than others in the generation of itch. The Mas-related G protein-coupled receptors (Mrgpr) family as well as transient receptor potential ankyrin 1 (TRPA1) and protease activated receptor 2(Par2) have impor

How Foraging Mode Sculpts Sensory Systems: Morphological Evidence From DiceCT and Histology in Sympatric Lizards.

The relationship between foraging modes and sensory system morphology is critical for understanding the ecological and evolutionary adaptations of lizards. This study investigates the nasal olfactory system (NOS) and vomeronasal system (VNS) of four sympatric lizards from the Turpan Basin, China, which exhibit distinct foraging strategies: the active foraging Eremias roborowskii (Lacertidae), the sit-and-wait foraging Phrynocephalus axillaris (Agamidae) and Tenuidactylus dadunensis (Gekkonidae), and the seasonally frugivorous Teratoscincus roborowskii (Sphaerodactylidae), which adopts active f

Butyrate Triggers Neuronal Signaling via Release of Colonic Serotonin in a Microfluidic Model of Gut-Brain Axis.

Host-microbiota interactions play a central role in regulating gastrointestinal and cardiometabolic physiology, yet the mechanistic pathways linking microbial metabolites to sensory neuronal activation remain poorly defined. In particular, the rapid signaling events through which short-chain fatty acids such as butyrate influence epithelial and neuronal function in the colon have not been fully resolved due to limitations of existing models. Here, we developed and validated a compartmentalized microfluidic tri-channel model that spatially organizes primary colonic epithelial cells, sensory neu

Ridge detection tactility deficits associated with carpal tunnel syndrome.

A ridge detection threshold task was administered to patients diagnosed as having carpal tunnel syndrome for studying performance in an occupationally relevant functional tactile inspection task. Thresholds were compared with a reference group of subjects not having carpal tunnel syndrome symptoms, performing the same task. The threshold detection task used the method of limits for studying the effects of carpal tunnel syndrome, rate of ridge height changes, ridge gradient, and direction of shearing against the skin on ridge detection thresholds for a repeated measures factorial experimental d

P2X(7) inhibition in stellate ganglia prevents the increased sympathoexcitatory reflex via sensory-sympathetic coupling induced by myocardial ischemic injury.

Purinergic signaling has been found to participate in the regulation of cardiovascular function. In this study, using a rat myocardial ischemic injury model, the sympathoexcitatory reflex mediated by P2X7 receptor via sensory-sympathetic coupling between cervical dorsal root ganglia (DRG) nerves and stellate ganglia (SG) nerves was explored. Our results showed that the systolic blood pressure, heart rate, serum cardiac enzymes concentrations, interleukin-6 (IL-6), and tumor necrosis factor-α (TNF-α) concentrations were increased, and the expression levels of P2X7 mRNA and protein in DRG and SG

EXPRESSION OF SEROTONIN RECEPTOR SUBTYPE 3A (5HT3A) ON RAT TRIGEMINAL SENSORY NEURONS.

Serotonin (5-hydroxytryptamine, 5HT) is a neurotransmitter and proinflammatory mediator found largely in the peripheral nervous system where it can initiate pain signaling. 5HT binds a variety of 5HT receptors on sensory nerve endings specialized in detecting noxious stimuli, termed nociceptors. A subset of sensory neurons involved in pain signaling express the transient receptor potential vanilloid 1 ion channel (TRPV1), a pain generator. 5HT can both directly activate sensory neurons and sensitize TRPV1 leading to enhanced nociceptor sensitivity (peripheral sensitization). Previous studies i

Enteric nervous system, serotonin, and the irritable bowel syndrome.

Intestinal motility, secretion, and blood flow are controlled and integrated by the enteric nervous system (ENS). The ENS is like a "brain-in-the-gut," with many of the neurophysiologic properties of the central nervous system. Serotonin is a neurotransmitter at synapses in the microcircuits of the ENS. Serotonin is also released from enterochromaffin cells and inflammatory/immune cells to act at serotonergic receptors on neurons of the ENS. Four important actions are (1) fast and (2) slow excitation of enteric neurons, (3) presynaptic inhibition of neurotransmitter release at synapses in ENS

Fernando de Castro and the discovery of the arterial chemoreceptors.

When de Castro entered the carotid body (CB) field, the organ was considered to be a small autonomic ganglion, a gland, a glomus or glomerulus, or a paraganglion. In his 1928 paper, de Castro concluded: "In sum, the Glomus caroticum is innervated by centripetal fibers, whose trophic centers are located in the sensory ganglia of the glossopharyngeal, and not by centrifugal [efferent] or secretomotor fibers as is the case for glands; these are precisely the facts which lead to suppose that the Glomus caroticum is a sensory organ." A few pages down, de Castro wrote: "The Glomus represents an orga

Acute corneal epithelial debridement unmasks the corneal stromal nerve responses to ocular stimulation in rats: implications for abnormal sensations of the eye.

It is widely accepted that the mechanisms for transducing sensory information reside in the nerve terminals. Occasionally, however, studies have appeared demonstrating that similar mechanisms may exist in the axon to which these terminals are connected. We examined this issue in the cornea, where nerve terminals in the epithelial cell layers are easily accessible for debridement, leaving the underlying stromal (axonal) nerves undisturbed. In isoflurane-anesthetized rats, we recorded extracellularly from single trigeminal ganglion neurons innervating the cornea that are excited by ocular drynes

Identification of perineal sensory neurons activated by innocuous heat.

C-fiber sensory neurons comprise nociceptors and smaller populations of cells detecting innocuous thermal and light tactile stimuli. Markers identify subpopulations of these cells, aiding our understanding of their physiological roles. The transient receptor potential vanilloid 1 (TRPV1) cation channel is characteristic of polymodal C-fiber nociceptors and is sensitive to noxious heat, irritant vanilloids, and protons. By using immunohistochemistry, in situ hybridization, and retrograde tracing, we anatomically characterize a small subpopulation of C-fiber cells that express high levels of TRP

The evolution of sensory placodes.

The vertebrate cranial sensory placodes are ectodermal embryonic patches that give rise to sensory receptor cells of the peripheral paired sense organs and to neurons in the cranial sensory ganglia. Their differentiation and the genetic pathways that underlay their development are now well understood. Their evolutionary history, however, has remained obscure. Recent molecular work, performed on close relatives of the vertebrates, demonstrated that some sensory placodes (namely the adenohypophysis, the olfactory, and accoustico-lateralis placodes) first evolved at the base of the chordate linea
