Activatable Shiga toxin 2d (Stx2d) in STEC strains isolated from cattle and sheep at slaughter.

Shiga toxin producing Escherichia coli (STEC) harbouring the stx(2d-activatable) gene and expressing the mucus- and elastase-activatable phenotype have been associated with severe outcomes of human disease. However, there is limited data available on the occurrence of such strains in livestock reservoirs. In this study, we analyzed 11 STEC strains isolated from healthy cattle and sheep at slaughter that were originally detected to contain the stx(2c) allele, for the presence of the stx(2d-activatable) genotype. Ten of the eleven strains displayed the stx(2d-activatable) genotype as determine b

Prognosis and pathological characteristics of five children with non-Shiga toxin-mediated hemolytic uremic syndrome.

BACKGROUND: The three major signs of hemolytic uremic syndrome (HUS) are hemolytic anemia, thrombopenia and acute renal failure. HUS is classified into Shiga toxin-mediated HUS (Stx-HUS) and non-Shiga toxin-mediated HUS (nStx-HUS). The prognosis of nStx-HUS is reported to be less favorable than that of Stx-HUS. Although the association between the prognosis and pathological characteristics of HUS have been reported such that the prognosis was considered to be poor for thrombotic microangiopathy (TMA) with predominant arterial involvement (arterial TMA), good for TMA with predominant glomerular

Production of IgA monoclonal antibody against Shiga toxin binding subunits employing nasal-associated lymphoid tissue.

We established an IgA monoclonal antibody (mAb) against Shiga toxin 1 B subunits (Stx1B) from mouse nasal-associated lymphoid tissues (NALT) of BALB/c mice. We have developed an improved protocol in which cross-linked Stx1B is intranasally administered together with cholera toxin. Surface IgA-positive NALT lymphocytes from mice immunized in this manner were enriched and then fused with mouse myeloma cells to produce hybridoma cells. Hybridoma culture supernatants were examined to see if they contain IgA against Stx1B and if they can inhibit carbohydrate recognition by Stx1B. For the latter pur

Sequence of a variant Shiga-like toxin type-I operon of Escherichia coli O111:H-.

PCR amplification was used to screen faecal isolates of Escherichia coli from a 12-month-old boy with haemolytic uraemic syndrome for the presence of Shiga-like toxin (SLT)-encoding genes. One isolate, belonging to serotype O111:H-, was positive for SLT-I by this method. UV induction indicated that the strain was lysogenic for a lambdoid bacteriophage, but this did not encode the toxin. Southern hybridization analysis of chromosomal DNA revealed that the SLT-I gene was located on an 8.5-kb EcoRI fragment. SLT-I was further localized to within a 3.0-kb SphI-EcoRI fragment. A separate subclone c

Release of Shiga toxin by membrane vesicles in Shigella dysenteriae serotype 1 strains and in vitro effects of antimicrobials on toxin production and release.

Effects of various antimicrobials on in vitro Shiga toxin production and release by Shigella dysenteriae serotype 1 was investigated in this study with particular reference to the role of outer membrane vesicles in toxin release by the organism. Five antimicrobials, namely nalidixic acid, ciprofloxacin, norfloxacin, fosfomycin and mitomycin C, were chosen for the study and the toxin titre was measured by the reverse passive latex agglutination (RPLA) method using an available kit. Only mitomycin C was found to induce production of Shiga toxin in the bacteria and its release by outer membrane v

An Atypical Case of Hemolytic Uremic Syndrome Caused by Shiga Toxin Produced by Aeromonas spp.

Hemolytic uremic syndrome (HUS), traditionally recognized in pediatric populations, is characterized by renal insufficiency, hemolytic anemia, and thrombocytopenia, often linked to Shiga-like toxin (SLT) exposure. While typically associated with enteric pathogens like Escherichia coli (E. coli) and Shigella, Aeromonas spp. has also been identified as potential SLT producers, posing a new challenge. This study presents an exceptional case of HUS in a 77-year-old female, implicating Aeromonas hydrophila as the causative agent. The patient's clinical trajectory, marked by acute kidney injury post

The prevalence and genomic context of Shiga toxin 2a genes in E. coli found in cattle.

Shiga toxin-producing Escherichia coli (STEC) that cause severe disease predominantly carry the toxin gene variant stx2a. However, the role of Shiga toxin in the ruminant reservoirs of this zoonotic pathogen is poorly understood and strains that cause severe disease in humans (HUSEC) likely constitute a small and atypical subset of the overall STEC flora. The aim of this study was to investigate the presence of stx2a in samples from cattle and to isolate and characterize stx2a-positive E. coli. In nationwide surveys in Sweden and Norway samples were collected from individual cattle or from cat

Particulate Shiga Toxin 2 in Blood is Associated to the Development of Hemolytic Uremic Syndrome in Children.

Hemolytic uremic syndrome (HUS), the leading cause of acute renal failure in children (< 3 years), is mainly related to Shiga toxins (Stx)-producing Escherichia coli (STEC) infections. STEC are confined to the gut resulting in hemorrhagic colitis, whereas Stx are delivered in blood to target kidney and brain, with unclear mechanisms, triggering HUS in 5 to 15% of infected children. Stx were found on circulating cells, free in sera (soluble Stx) or in blood cell-derived microvesicles (particulate Stx), whereby the relationship between these forms of circulating toxins is unclear. Here, we have

Role of climate in the spread of shiga toxin-producing Escherichia coli infection among children.

Haemolytic-uraemic syndrome (HUS) is a rare disease mainly affecting children that develops as a complication of shiga toxin-producing Escherichia coli (STEC) infection. It is characterised by acute kidney injury, platelet consumption and mechanical destruction of red blood cells (haemolysis). In order to test the working hypothesis that the spread of the infection is influenced by specific climatic conditions, we analysed all of the identified cases of infection occurring between June 2010 and December 2013 in four provinces of Lombardy, Italy (Milano, Monza Brianza, Varese and Brescia), in w

Shiga toxins expressed by human pathogenic bacteria induce immune responses in host cells.

Shiga toxins are a family of genetically and structurally related toxins that are the primary virulence factors produced by the bacterial pathogens Shigella dysenteriae serotype 1 and certain Escherichia coli strains. The toxins are multifunctional proteins inducing protein biosynthesis inhibition, ribotoxic and ER stress responses, apoptosis, autophagy, and inflammatory cytokine and chemokine production. The regulated induction of inflammatory responses is key to minimizing damage upon injury or pathogen-mediated infections, requiring the concerted activation of multiple signaling pathways to

Effects of Shiga toxin 2 on lethality, fetuses, delivery, and puerperal behavior in pregnant mice.

Shiga toxin 2 (Stx2) is produced by enterohemorrhagic Escherichia coli (EHEC) and is known as the major virulence factor of EHEC. The aim of this study was to evaluate the effects of Stx2 on (i) maternal lethality, (ii) fetuses, (iii) delivery period, and (iv) maternal behavior after delivery. Timed pregnant ICR mice were injected intravenously with Stx2 on day 5 of pregnancy (early stage) or on day 15 (late stage). In early-stage experiments, the number of normal fetuses of mice injected with Stx2 was significantly lower than that of control mice. In late-stage experiments, mothers injected w

Depletion of liver and splenic macrophages reduces the lethality of Shiga toxin-2 in a mouse model.

The haemolytic uraemic syndrome (HUS) is a clinical syndrome consisting of haemolytic anaemia, thrombocytopenia, and acute renal insufficiency. HUS is the most frequent cause of acute renal failure in childhood. It has been previously suggested that the presence of Shiga toxin (Stx) is necessary but not sufficient for HUS development, and cytokines such as tumour necrosis factor-alpha (TNF-alpha) and IL-1beta appear to be necessary to develop the syndrome. Since the mononuclear phagocytic system (MPS) is the major source of these cytokines, macrophages might be one of the relevant targets for

Edema disease as a model for systemic disease induced by Shiga toxin-producing E. coli.

Edema disease (ED) is a naturally occurring disease of weaned pigs caused by host adapted strains of E. coli that produce Shiga toxin (STEC). We determined the temporal and quantitative relationships between intestinal colonization by STEC, levels of Shiga toxin (Stx2e) in the gut, in the blood, and clinical manifestations of ED. Bacterial colonization (10(8) CFU/cm ileum) was highest 4 days post inoculation (pi) in animals that did not develop clinical disease and 6 days pi in animals with clinical signs of ED. The mean time for the development of clinical signs of ED was 6 days pi (range 4-1

Shiga toxin-induced tumor necrosis factor alpha expression: requirement for toxin enzymatic activity and monocyte protein kinase C and protein tyrosine kinases.

Infections with Shiga toxin (Stx)-producing bacteria cause bloody diarrhea which may progress to life-threatening complications, including acute renal failure and neurological abnormalities. The precise mechanism of disease progression is unclear, although evidence suggests that the localized production of the host proinflammatory cytokines tumor necrosis factor alpha (TNF-alpha) and interleukin-1 may exacerbate toxin-mediated vascular damage. Purified Stxs have been demonstrated to elicit proinflammatory cytokine synthesis from human peripheral blood mononuclear cells and monocytic cell lines

Variants of Shiga-like toxin II constitute a major toxin component in Escherichia coli O157 strains from patients with haemolytic uraemic syndrome.

The prevalence and genotype of Shiga-like toxins (SLTs) in Escherichia coli (O)157 strains from patients in Germany with haemolytic uraemic syndrome (HUS) were investigated. This was done by PCR amplification of the B-subunit genes with two primer pairs--one complementary to slt-IB, and the other homologous to both slt-IIB and slt-IIvB sequences. To distinguish between slt-II and slt-IIv, the amplified DNA was digested with restriction endonucleases HaeIII and FokI. Of the 38 strains examined, 17 harboured sequences for slt-IIv; four contained only slt-IIv, three carried both slt-IIv and slt-I

Artificial Intelligence-Aided De Novo Design of High-Affinity Protein Binders Against Shiga Toxin 2.

Shiga toxin-producing Escherichia coli (STEC) is a major food-borne pathogen with limited treatment options. Targeting Shiga toxin (Stx) represents a promising therapeutic strategy. Using artificial intelligence (AI)-assisted de novo protein design combined with bacterial surface display screening, pull-down assays, and biolayer interferometry, we identified protein binders against the Stx2a B subunit (Stx2aB), among which Binder506 exhibited high affinity. Structural modeling predicted a three-α-helix fold, consistent with circular dichroism analysis, while differential scanning fluorimetry r

Epidemiology of Bloody Diarrhea, Shiga Toxin-producing Escherichia coli and Hemolytic Uremic Syndrome in Children.

BACKGROUND: Shiga Toxin-producing Escherichia coli -associated hemolytic uremic syndrome (STEC-HUS) remains a leading cause of acute kidney failure, worldwide. The disease often presents with acute bloody diarrhea and supportive care is the only therapeutic option; thus, early diagnosis and early management are of paramount importance. AIM: Herein, we share the results of a proactive diagnostic approach to acute bloody diarrhea in children as shown by a surveillance activity devoted to the early diagnosis of STEC infection. METHODS: Since 2010, in a well-defined area of Northern Italy (refer

Nigella sativa Seed Oil Antibacterial and Antibiofilm Activity against Escherichia coli O157H7 Shiga Toxin-2.

Shiga toxin type 2 (Stx2)-producing Escherichia coli O157H7 has become a global risk to public health. The study aimed to investigate the emergence of antimicrobial resistance patterns in the clinical isolates of E. coli. The isolates revealed a pattern of strong resistance to the majority of the common antibiotics. Most E. coli isolates were susceptible to gentamicin and were classified as MDR. The Stx2 determinant was screened in E. coli O157:H7 isolates through the PCR technique, and the Stx2 was purified by cation exchange and gel filtration chromatography. The purification yielded 5.2 mil

Shiga Toxin-Producing Escherichia coli in Aquaculture: A Decade (2015-2025)-Long Global Retrospective Outlook.

Shiga toxin-producing Escherichia coli (STEC) contamination and proliferation in aquaculture and aquatic systems is worrisome for global food safety, as well as veterinary and public health. Traditionally linked with terrestrial ruminant reservoirs, aquaculture matrices, including farmed finfish, shellfish, culture water, and benthic organisms, are increasingly acknowledged as potential conduits for the dissemination of STEC. Herein, the review documented data concerning the prevalence, genomic composition, and ecological dynamics of STEC across various aquaculture environments across differen
