Biliary secretion in elasmobranchs. I. Bile collection and composition.

Bile composition and secretion were studied in tow elasmobranch species, Squalus acanthias (spiny dogfish shark) and Raja erinacea (small skate), after ligation of the common duct and insertion of cannulas into the gallbladder lumen. Fish were then allowed to swim freely in large pools and bile was collected in balloons attached to the externalized cannulas. Both species secreted bile for periods of 4-7 days at a maximum rate of 1.77 +/- .89 ml/kg per 24 h in Squalus acanthias and 2.66 +/- .89 ml/kg per 24 h in Raja erinacea. Comparison of the composition of cannula and gallbladder bile indica

Adult chondrogenesis and spontaneous cartilage repair in the skate, Leucoraja erinacea.

Mammalian articular cartilage is an avascular tissue with poor capacity for spontaneous repair. Here, we show that embryonic development of cartilage in the skate (Leucoraja erinacea) mirrors that of mammals, with developing chondrocytes co-expressing genes encoding the transcription factors Sox5, Sox6 and Sox9. However, in skate, transcriptional features of developing cartilage persist into adulthood, both in peripheral chondrocytes and in cells of the fibrous perichondrium that ensheaths the skeleton. Using pulse-chase label retention experiments and multiplexed in situ hybridization, we ide

Sex may influence environmental diphenhydramine accumulation in Round Stingrays.

Despite the amount of treated wastewater discharged into the Southern California Bight, few studies have examined pharmaceutical compounds in local biota. The Round Stingray (Urobatis halleri) was selected as a representative elasmobranch species to perform an exploratory study on environmental pharmaceutical exposure. Archived liver samples of males and females from juvenile to adult size classes from several locations (n = 53) were examined for 18 pharmaceutical and illicit drug compounds using isotope-dilution LC-MS/MS. Very few compounds were detected in stingray livers, with diphenhydrami

Two classes of bipolar cell in the retina of the skate Raja erinacea.

We have used immunoreactions against serotonin and protein kinase C to visualize two distinct classes of bipolar cell in the all-rod retina of the skate, Raja erinacea. To enhance the immunoreaction in serotonin-accumulating bipolar cells, prior to fixation, some retinas were incubated in Ringer's solution containing serotonin and pargyline. We found the somata of serotonin-accumulating bipolar cells to be located slightly distal to the midline of the inner nuclear layer. With increasing eccentricity from the visual streak, the size of the perikarya increases, concomitant with a decline in den

When proglottids and scoleces conflict: phylogenetic relationships and a family-level classification of the Lecanicephalidea (Platyhelminthes: Cestoda).

This study presents the first comprehensive phylogenetic analysis of the interrelationships of the morphologically diverse elasmobranch-hosted tapeworm order Lecanicephalidea, based on molecular sequence data. With almost half of current generic diversity having been erected or resurrected within the last decade, an apparent conflict between scolex morphology and proglottid anatomy has hampered the assignment of many of these genera to families. Maximum likelihood and Bayesian analyses of two nuclear markers (D1-D3 of lsrDNA and complete ssrDNA) and two mitochondrial markers (partial rrnL and

DNA-based characterization of rays (Elasmobranchii: Batoidea) from Bangladesh using mitochondrial markers: Implications for conservation and management.

Rays are an iconic group of chondrichthyan fishes, with many species currently threatened with extinction. Although conservation laws exist in Bangladesh to protect their population, lack of comprehensive law enforcement strategies together with commercial exploitation and habitat destruction resulted in population decline of many species nonetheless. One significant challenge to this conservation effort is a rapid and authentic species identification strategy, as traditional morphological diagnosis is hindered by frequent misidentification, especially when species are morphologically similar

Zooplankton as a Bioindicator of Marine Contamination for Filter-Feeding Basking Sharks, Fin Whales and Devil Rays at Caprera Canyon (Mediterranean Sea).

The Canyon of Caprera (Mediterranean Sea) hosts high biodiversity, including threatened filter-feeding vertebrates such as the basking shark and fin whale. Due to their elusive nature, mobility and conservation status, direct contaminant assessment in these species is often challenging. To address this limitation, the present study focuses on zooplankton, an essential prey and bioindicator, to indirectly assess the levels of persistent organic pollutants and trace elements (TEs) in their feeding grounds. This approach provides a noninvasive method to estimate potential pollutant exposure in vu

Redescription and First Nucleotide Sequences of Rhinobatonchocotyle pacifica Oliva & Luque, 1995 (Monogenea: Hexabothriidae), a Parasite of Pseudobatos planiceps (Garman, 1880) (Rhinopristiformes: Rhinobatidae) from Peru.

INTRODUCTION: Specimens of Rhinobatonchocotyle pacifica Oliva & Luque, 1995 (Hexabothriidae) were collected from the gill filaments of Pacific guitarfish, Pseudobatos planiceps (Garman, 1880) (Rhinopristiformes: Rhinobatidae), from off the coast of Chorrillos, Lima, Peru. Since the original description of R. pacifica was incomplete, we herein redescribe R. pacifica based on re-examination of the holotype, paratypes, and newly collected voucher specimens. Nucleotide sequences (18S, 28S) for R. pacifica are presented for the first time, and a phylogenetic analysis conducted. MATERIALS AND METHO

The cutaneous cellular infiltrate to stingray envenomization contains increased TIA+ cells.

Stingrays result in approximately 2000 stings annually in the U.S.A., and thus are one of the most important venomous marine animals. After envenomization, there is immediate, intense pain with subsequent oedema, cyanosis followed by local erythema and petechiae. Progressive local necrosis and ulceration is variable, sometimes leading to gangrene. To characterize the inflammatory infiltrate at the site of a stingray injury, we examined tissue obtained approximately 4 days after stingray envenomization. Routine histology and immunohistochemical stains for lymphoid markers, including CD3, CD4, C

Molecular and morphological characterization, phylogenetic affinities and new records of Trypanorhyncha (Cestoda) from the Mexican Atlantic.

During a parasitological survey of elasmobranchs of the Atlantic Ocean on the Mexican coast, the digestive tract of some specimens of the southern stingray, Hypanus americanus (Hildebrand & Schroeder, 1928), and the Atlantic sharpnose shark, Rhizoprionodon terraenovae (Richardson, 1836), were analyzed for tapeworms. These elasmobranchs were obtained from commercial fishing in four localities. Five taxa of Trypanorhyncha were recovered: Kotorella pronosoma (Stossich, 1901) Euzet & Radujkovic, 1989, Oncomegas wageneri (Linton, 1890) Dollfus, 1929, Pterobothrium kingstoni Campbell & Beveridge, 19

Potamotrygon motoro stingray venom induces both neurogenic and inflammatory pain behavior in rodents.

Freshwater stingray accidents cause an immediate, intense, and unrelieved pain which is followed by edema, erythema and necrosis formation. Treatment for stingray envenomation is based on administration of analgesic, antipyretic and anti-inflammatory drugs. Concerning pain control, it is prescribed to immerse punctured limb on hot water to alleviate pain. There are no studies demonstrating specific targets on which stingray venom acts to promote pain. Therefore, the aim of this work was to investigate some mechanisms of Potamotrygon motoro venom (PmV) that contribute to nociception induction.

Single-cell and spatial transcriptomics inform mechanistic physiology in non-model animals.

Comparative physiology increasingly requires cellular resolution at the level of cell types, tissue microenvironments, and regulatory programs that associate genotype with function under environmental variation. Bulk transcriptomics and tissue-level assays have revealed pathways associated with stress responses, metabolic shifts, and developmental transitions; however, they average signals across heterogeneous cell populations and frequently obscure which cells contribute to physiological phenotypes. Single-cell RNA sequencing (scRNA-seq), single-nucleus RNA sequencing (snRNA-seq), and spatial

Identification of the midbrain locomotor region and its relation to descending locomotor pathways in the Atlantic stingray, Dasyatis sabina.

The midbrain locomotor region (MLR) in the Atlantic stingray, Dasyatis sabina, was identified and characterized. Stimulation (50-100 microA, 60 Hz) of the midbrain in decerebrated, paralyzed animals was used to elicit locomotion monitored as alternating activity in nerves innervating an antagonist pair of elevator and depressor muscles. Effective sites for evoking locomotion in the midbrain included parts of several nuclei: the caudal portion of the interstitial nucleus of the medial longitudinal fasciculus and the caudomedial parts of the cuneiform and subcuneiform nuclei. This region did not

Angiotensin-I converting enzyme inhibitory peptides from antihypertensive skate (Okamejei kenojei) skin gelatin hydrolysate in spontaneously hypertensive rats.

The aim of this study was to investigate antihypertensive effect of bioactive peptides from skate (Okamejei kenojei) skin gelatin. The Alcalase/protease gelatin hydrolysate below 1 kDa (SAP) exhibited the highest angiotensin-I converting enzyme (ACE) inhibition compared to other hydrolysates. SAP can decrease systolic blood pressure significantly in spontaneously hypertensive rats. SAP inhibited vasoconstriction via PPAR-γ expression, activation and phosphorylation of eNOS in lungs. Moreover, the expression levels of endothelin-1, RhoA, α-smooth muscle actin, cleaved caspase 3 and MAPK were de

Structural analysis of the evolution of steroid specificity in the mineralocorticoid and glucocorticoid receptors.

BACKGROUND: The glucocorticoid receptor (GR) and mineralocorticoid receptor (MR) evolved from a common ancestor. Still not completely understood is how specificity for glucocorticoids (e.g. cortisol) and mineralocorticoids (e.g. aldosterone) evolved in these receptors. RESULTS: Our analysis of several vertebrate GRs and MRs in the context of 3D structures of human GR and MR indicates that with the exception of skate GR, a cartilaginous fish, there is a deletion in all GRs, at the position corresponding to Ser-949 in human MR. This deletion occurs in a loop before helix 12, which contains the

Acanthobothrium oualimeriemae n. sp. (Eucestoda: Onchobothriidae) from the spiral valve of the skates Raja asterias and Raja radula (Rajiformes: Rajidae) off the Algerian coastline.

A new species of Acanthobothrium is described from the spiral intestine of Raja asterias Delaroche and Raja radula Delaroche in the Algerian Mediterranean Sea. Acanthobothrium oualimeriemae n. sp. was characterized using an integrative approach combining morphological and molecular (28S rDNA) data. The new species is assigned to morphological category 2 and can be distinguished from all known congeners by a unique combination of features, including total body length, number of proglottids, scolex configuration, dimensions of bothridial structures, length of the cephalic peduncle, length of the

Traumatic Pneumothorax Associated With Penetrating Neck Injury Caused by a Stingray: A Case Report.

We describe a patient with missed traumatic pneumothorax due to penetrating neck injury that was presumably caused by a stingray while diving. The barb can result in significant injuries to deep structures and organs despite a deceptively small entrance wound. This case underscores the importance of performing a thorough examination and having a high suspicion of distant injury in patients with penetrating neck injuries.

Reproductive biology of the Alaska skate Bathyraja parmifera, with comments on an intersexual individual.

A total of 1357 specimens of Alaska skate Bathyraja parmifera were collected in the eastern Bering Sea by fisheries observers and during scientific groundfish surveys from 2003 to 2005. Male and female gonads were examined for maturity stage and seasonal reproductive timing. Based on seasonal reproductive data, including the occurrence of egg cases, ovum size, ovum number, shell-gland width and gonado-somatic index, this species appears to reproduce continually throughout the year. Potential effects of maternal size upon the size and number of mature oocytes were also investigated, with total

Chondrogenesis and homology of the visceral skeleton in the little skate, Leucoraja erinacea (Chondrichthyes: Batoidea).

Chondrichthyan fishes possess visceral skeletons that differ considerably, morphologically, from those of their sister taxon, the osteichthyans. Here, we use histological techniques and whole-mount skeletal preparations to visualize and describe the sequence of visceral skeletal condensation and chondrogenesis in a chondrichthyan, the little skate (Leucoraja erinacea). We demonstrate that visceral skeletal condensation begins rostrally, with the mandibular arch, and progresses caudally with the hyoid arch and posterior branchial arches condensing soon after. We provide a detailed account of th
