Previously we reported the optimization of antiviral scaffolds containing benzimidazole and related heterocycles possessing activity against a variety of arenaviruses. These series of compounds were discovered through an HTS campaign of a 400,000 small molecule library using lentivirus-based pseudotypes incorporated with the Lassa virus envelope glycoprotein (LASV GP). This screening also uncovered an alternate series of very potent arenavirus inhibitors based upon an acylhydrazone scaffold. Subsequent SAR analysis of this chemical series involved various substitutions throughout the chemical

TP53 is the most frequently mutated gene in human cancers, with hundreds of mutations leading to loss of p53 tumor-suppressive functions. p53 mutations are closely associated with tumor progression, metastasis, and therapy resistance. In recent years, significant progress has been made in developing small-molecules targeting mutant (mut) p53 to restore its tumor suppressor activity. This review presents a comprehensive and updated overview on the recent advances in small-molecule mut p53 modulators reported between 2020 and 2025. This review distinguishes itself by focusing on the different mo

INTRODUCTION: Huntington's disease (HD) is a progressive neurodegenerative disorder caused by the accumulation of mutant huntingtin protein (mHTT) with expanded polyglutamine (polyQ) tracts. These aggregates contribute to neuronal toxicity and disease progression. Targeting aggregation, especially at the N-terminal domain (N17), may offer a therapeutic strategy. This study aims to identify potential small-molecule inhibitors that can bind to aggregation-prone regions of mHTT using computational methods.
METHODS: We characterized polyQ repeat regions and the N17 domain using CASTp to identify

Parkinson's disease (PD), a progressive neurodegenerative disorder, is primarily characterized by the accumulation of alpha-synuclein (α-syn) aggregates in the brain, leading to the neuronal dysfunction and degeneration. As a result, targeting α-syn aggregation is emerging as a promising therapeutic strategy for delaying or stopping disease progression. The present chapter tried to explore the progress made in the development of small molecule inhibitors in preventing or reversing the aggregation of α-syn. Overall, the chapter provides an overview of the mechanisms underlying α-syn misfolding

Virtual screening of large compound libraries against specific RNAs offers a promising and cost-effective approach for identifying novel lead compounds. Developing a reliable scoring function for RNA-ligand interactions is crucial for effective computational drug screening. However, current RNA-ligand scoring functions are primarily designed to predict native binding poses for given RNA-ligand pairs and have not been thoroughly evaluated for their virtual screening capabilities. Here, by leveraging more accurate descriptions of stacking, solvation, conformational flexibility, and other key eff

Increasing attention is being focused on skin health currently, especially the excessive deposition of melanin in the skin. Tyrosinase, the rate-limiting enzyme in melanin biosynthesis, is a crucial enzyme in melanin synthesis. However, existing tyrosinase inhibitors pose some degree of toxicity to humans. Therefore, the development of more efficient and low-toxicity tyrosinase inhibitors is urgently needed. This review briefly depicts the melanin biosynthesis process and the crystal structure and catalytic mechanism of tyrosinase. The latest research progress regarding small-molecule tyrosina

Human adenoviruses (HAdVs) present a significant health threat, particularly to immunocompromised individuals, where infections can progress to severe, life-threatening disease. Despite their clinical impact, no FDA-approved antiviral therapies specifically target HAdV infection. Current treatments, such as off-label cidofovir, are limited by nephrotoxicity, poor oral bioavailability, and resistance concerns, underscoring the urgent need for safer and more effective alternatives. This perspective critically evaluates recent advances in small-molecule anti-HAdV drug development, highlighting pr

Flow cytometry has great potential for screening in translational research areas due to its deep quantification of cellular features, ability to collect millions of cells in minutes, and consistently expanding suite of validated antibodies that detect cell identity and functions. However, cytometry remains under-utilized in discovery chemical biology due to the differences in expertise between chemistry groups developing chemical libraries and cell biologists developing single cell assays. This chapter is designed to bridge this gap by providing a detailed protocol aimed at both chemistry and

Shigella flexneri is a facultative intracellular pathogen that causes shigellosis, a human diarrheal disease characterized by the destruction of the colonic epithelium. Novel antimicrobial compounds to treat infections are urgently needed due to the proliferation of bacterial antibiotic resistance and lack of new effective antimicrobials in the market. Our approach to find compounds that block the Shigella virulence pathway has three potential advantages: (i) resistance development should be minimized due to the lack of growth selection pressure, (ii) no resistance due to environmental antibio

BACKGROUND: Malaria transmission depends on infected mosquitoes and can be controlled by transmission-blocking drugs. The recently discovered FREP1-mediated malaria transmission pathway is an excellent target to screen drugs for limiting transmission.
METHODS: To identify candidate small molecules, we used an ELISA-based approach to analyze extracts from a fungal library for inhibition of the FREP1-parasite interaction. We isolated and determined one active compound by chromatography and crystallography, respectively. We measured the effects of the bioactive compound on malaria transmission t

Natural product structure and fragment-based compound development inspire pseudo-natural product design through different combinations of a given natural product fragment set to compound classes expected to be chemically and biologically diverse. We describe the synthetic combination of the fragment-sized natural products quinine, quinidine, sinomenine, and griseofulvin with chromanone or indole-containing fragments to provide a 244-member pseudo-natural product collection. Cheminformatic analyses reveal that the resulting eight pseudo-natural product classes are chemically diverse and share b

Small-molecule fluorescent probes are powerful tools in chemical analysis and biological imaging. However, as the foundation of probe design, the meager existing set of core fluorophores have largely limited the diversity of current probes. Consequently, there is a high demand to discover fluorophores with new scaffolds and optimize the existing fluorophores. Here, we put forward a facile strategy of heterocyclic N-oxidation to address these challenges. The introduced N-O bond reconstructs the electron "push-pull" system of heterocyclic scaffolds and dramatically improves their photophysical p

The pre-mRNA splicing factor SF3B1 shows recurrent mutations among hematologic malignancies and some solid tumors. In 2007, the identification of two cytotoxic natural products, which showed splicing inhibition by binding to SF3b, prompted the development of small-molecule splicing modulators of SF3B1 as therapeutics for cancer. Recent studies suggested that spliceosome-mutant cells are preferentially sensitive to pharmacologic splicing modulation; therefore, exploring the clinical utility of splicing modulator therapies in patients with spliceosome-mutant hematologic malignancies who have fai

Obligate protozoan parasites of the kinetoplastids and apicomplexa infect human cells to complete their life cycles. Some of the members of these groups of parasites develop in at least two systems, the human host and the insect vector. Survival under the varied physiological conditions associated with the human host and in the arthropod vectors requires the parasites to modulate their metabolic complement in order to meet the prevailing conditions. One of the key features of these parasites essential for their survival and host infectivity is timely expression of various proteins. Even more i

This paper presents the effort of collecting and curating a data set of 15461 molecules tested against the malaria parasite, with robust activity and mode of action annotations. The set is compiled from in-house experimental data and the public ChEMBL database subsets. We illustrate the usefulness of the dataset by building QSAR models for antimalarial activity and QSPR models for modes of actions, as well as by the analysis of the chemical space with the Generative Topographic Mapping method. The GTM models perform well in prediction of both activity and mode of actions, on par with the class

A 2-amido benzo[ d]imidazole library has been constructed by solid-phase synthesis. The key step of this solid-phase synthesis involves the preparation of polymer-bound 2-amino benzo[ d]imidazole resin through desulfurative cyclization of thiourea resin using 2-chloro-1,3-dimethylimidazolinium chloride and N, N-diisopropylethylamine in dichloromethane (DCM), and the resin is then functionalized by acylation at the 2-amine position to afford 2-amidobenzo[ d]imidazole resin. In the case of 2-amidobenzo[ d]imidazole resin having a p-I or m-NO2, the resin was further functionalized by Suzuki/Sonog

Aquaporins (AQPs), a large family of membrane protein channels that facilitate transport of water and other small solutes, play important roles in physiological functions and human diseases. Up till now, 13 types of AQPs, numbered 0 through 12, have been identified in various mammalian tissues. Homologous genes for AQPs in amphibians, insects, and bacteria highlight the evolutionary conservation and, thus, the importance of these membrane channels. Many members of the AQP family are expressed in the eye. AQP1, which is a water-selective channel, is expressed in the anterior chamber (cornea, ci

Targeting epigenetic proteins is a rapidly growing area for medicinal chemistry and drug discovery. Recent years have seen an explosion of interest in developing small molecules binding to bromodomains, the readers of acetyl-lysine modifications. A plethora of co-crystal structures has motivated focused fragment-based design and optimization programs within both industry and academia. These efforts have yielded several compounds entering the clinic, and many more are increasingly being used as chemical probes to interrogate bromodomain biology. High selectivity of chemical probes is necessary
