Effects of stimulation frequency and intensity in sacral neuromodulation on anorectal inputs to the somatosensory cortex in an experimental model.

INTRODUCTION: Although sacral neuromodulation (SNM) is an established treatment for faecal incontinence, stimulation parameters have been derived empirically and only one frequency (14 Hz) is employed clinically. The aim of this study was to test a range of stimulation frequencies to establish an optimal frequency of SNM for maximum augmentation of anal canal cortical evoked potentials (EPs) in an animal model. METHODS: In female Wistar rats, anal canal EPs were recorded over the primary somatosensory cortex using a flexible multielectrode array, and the effect of SNM was studied. SNM was app

Lack of activation of human secondary somatosensory cortex in Unverricht-Lundborg type of progressive myoclonus epilepsy.

Previous electroencephalographic and magnetoencephalographic studies have demonstrated giant early somatosensory cortical responses in patients with cortical myoclonus. We applied whole-scalp magnetoencephalography to study activation sequences of the somatosensory cortical network in 7 patients with Unverricht-Lundborg-type progressive myoclonus epilepsy diagnostically verified by DNA analysis. Responses to electric median nerve stimuli displayed 30-msec peaks at the contralateral primary somatosensory cortex that were four times stronger in patients than in control subjects. The amplitudes o

Facial nerve injury produces a latent somatosensory input through recruitment of the motor cortex in the rat.

Short-latency effects of unilateral facial nerve transection were studied on neuronal activation evoked in the primary motor cortex (MI) on both sides by vibrissa stimulation in adult rats. In the controls, unilateral trigeminal stimulation evoked activity in the whisker representation of both the contralateral somatosensory cortex (SI) and MI, but never in the ipsilateral MI. Unilateral transection of the facial motoric nerve facilitated evoked responses in the contralateral MI, and induced further neuronal activation (gross potentials and unit activity) in the MI ipsilateral to the stimulati

Environmental enrichment alters organizational features of the forepaw representation in the primary somatosensory cortex of adult rats.

The cortical forepaw area of young adult rats was mapped by recording the response properties of small clusters of neurons in layer IV of the primary somatosensory (SI) cortex. First we quantitatively analyzed the somatotopic organizational features of the cortical forepaw representation in terms of areal extent and topography, receptive field (RF) sensory modality, size, and location. We also assessed the influence of environmental enrichment, known to induce structural alterations in cortical connectivity, on the representational characteristics of the forepaw maps. Long-Evans rats were hous

A primate model for studying focal dystonia and repetitive strain injury: effects on the primary somatosensory cortex.

BACKGROUND AND PURPOSE: Job-related repetitive strain injuries (RSIs) are increasing, and current treatment strategies often fail to return injured people to work. This study documented the neural consequences of using two different movement strategies for active, repetitive hand closing and opening. METHODS: Two owl monkeys were trained for 20 weeks to repetitively close a handpiece against an 80-g force (3-400 trials per day, training at 80%-90% accuracy). One monkey used a highly articulated hand-squeezing strategy, and the other monkey used a proximal arm-pulling strategy. Changes in moto

Chaotic and phase-locked responses of the somatosensory cortex to a periodic medial lemniscus stimulation in the anesthetized rat.

Field potential responses of the somatosensory cortex to a periodic medial lemniscus (ML) fiber stimulation were investigated in anesthetized rats. Since the field potential responses of the cortex depend on the depth of anesthesia, two criteria were introduced to control the depth of anesthesia. One criterion is that spindle oscillations are caused by a single shock to ML fibers, and the other is that the dominant frequency of spontaneous field potential rhythm is in the frequency range of the delta wave. Phase-lockings and chaotic responses occurred depending on stimulus parameters under the

Immediate and chronic changes in responses of somatosensory cortex in adult flying-fox after digit amputation.

The somatosensory cortex of adult mammals has been shown to have a capacity to reorganize when inputs are removed by cutting afferent nerves or amputating a part of the body. The area of cortex that would normally respond to stimulation of the missing input can become responsive to inputs from other parts of the body surface. Although a few animals have been studied with repeat recording, no attempt has been made to follow the time-course of changes at cortical loci and the immediate effects of a small amputation have not been reported. We have followed the changes in response in the primary s

Connections of area 2 of somatosensory cortex with the anterior pulvinar and subdivisions of the ventroposterior complex in macaque monkeys.

The principal goal of the present study was to determine the thalamic connections of area 2 of postcentral somatosensory cortex of monkeys. The placement of injections of anatomical tracers (horseradish peroxidase, wheat germ agglutinin, or 3H-proline) was guided by extensive microelectrode maps of cortex in the region of the injection site. These maps identified the body parts represented in the cortex included in the injection site, and provided information about the physiological boundaries of area 2, which was related later to the cortical architecture. Most injections were placed in the r

Synchronal dual brain stimulation over the somatosensory cortex modulated social touch-induced analgesia depending on empathy.

Social touch, when provided as emotional support, can induce pain relief. Alpha inter-brain synchrony in the somatosensory cortex may be a neural factor contributing to the pain relief induced by social touch. This study investigated whether artificial manipulation of inter-brain communication in the pain-receiver and touch-giver could modulate touch-induced analgesia using transcranial alternating current stimulation. Sixteen same-sex pairs of participants were assigned the roles of pain-receiver and touch-giver during alpha-band transcranial alternating current stimulation applied to the som

Spared ulnar nerve injury results in increased layer III-VI excitability in the pig somatosensory cortex.

This study describes cortical recordings in a large animal nerve injury model. We investigated differences in primary somatosensory cortex (S1) hyperexcitability when stimulating injured and uninjured nerves and how different cortical layers contribute to S1 hyperexcitability after spared ulnar nerve injury. We used a multielectrode array to record single-neuron activity in the S1 of ten female Danish landrace pigs. Electrical stimulation of the injured and uninjured nerve evoked brain activity up to 3 h after injury. The peak amplitude and latency of early and late peristimulus time histogram

Altered brainstem-cortex activation and interaction in migraine patients: somatosensory evoked EEG responses with machine learning.

BACKGROUND: To gain a comprehensive understanding of the altered sensory processing in patients with migraine, in this study, we developed an electroencephalography (EEG) protocol for examining brainstem and cortical responses to sensory stimulation. Furthermore, machine learning techniques were employed to identify neural signatures from evoked brainstem-cortex activation and their interactions, facilitating the identification of the presence and subtype of migraine. METHODS: This study analysed 1,000-epoch-averaged somatosensory evoked responses from 342 participants, comprising 113 healthy

Home treatment against fatigue in multiple sclerosis by a personalized, bilateral whole-body somatosensory cortex stimulation.

BACKGROUND: Fatigue in multiple sclerosis (MS) is a highly invalidating symptom with no pharmacological efficacious therapies, which furthermore present frequent severe side effects. In two previous randomized controlled trials we observed the efficacy of a personalized neuromodulation treatment consisting of a personalized transcranial Direct Current Stimulation (tDCS) for 15 min per day for 5 days (Faremus). METHODS: By this medical-device phase II study, we aimed at assessing the feasibility, acceptance, safety and efficacy of Faremus treatment when applied at patients' home. We considered

Improved differentiation of tactile activations in human secondary somatosensory cortex and thalamus using cardiac-triggered fMRI.

Functional magnetic resonance imaging (fMRI) can reveal human brain activations with high precision. The accuracy may, however, be impaired by movement and deformation of brain tissue associated with cardiac pulsations. Here we corrected for such artifacts by time-locking the fMRI data acquisition to the cardiac cycle in ten subjects who received tactile stimuli to their lips, fingers, and toes. The imaged brain areas covered the parietal operculum and the thalamus, including the secondary somatosensory cortex (SII) bilaterally. Variance of the blood-oxygen-level-dependent signal decreased on

Muscarinic cholinergic receptor binding in rat hindlimb somatosensory cortex following partial deafferentation by sciatic nerve transection.

Peripheral nerve injury or amputation leads to extensive changes within the central representations of the mammalian body surface. The mechanisms responsible for post-traumatic reorganization of these maps in adults may also, at least partly, underlie a more general feature of the somatosensory system--the capacity for stimulus-dependent plasticity. Acetylcholine has been implicated in both of these processes. We studied the binding of the ligands [3H]QNB and [3H]pirenzepine in rat hindlimb somatosensory cortex from 1 to 14 days following sciatic nerve transection. Although the [3H]QNB binding

Brain-wide activity mapping reveals the somatosensory cortex as a sex-specific regulator of hedonic feeding.

Palatable food consumption engages reward circuitry and promotes neuroadaptations that contribute to overeating and obesity. While mesolimbic dopamine pathways are central to hedonic feeding models, the contribution of sensory cortical systems remains poorly understood. Here, we performed brain-wide activity mapping using Targeted Recombination in Active Populations (TRAP) and network analysis to define the distributed neural consequences of short-term high-fat diet (HFD) exposure in male and female mice. HFD increased caloric intake in both sexes, with females consuming significantly more tha

Changes in Intrinsic Activity of the Primary Somatosensory Cortex Causally Explain Differences in Emotion Perception in Autism.

Autism Spectrum Disorder (ASD) is characterized by certain difficulties in emotion-related processing. Recent research using electroencephalography (EEG) to measure somatosensory evoked potentials during emotion perception has shown reduced embodiment of emotional expressions in autistic compared to neurotypical individuals, independently from differences in visual processing. However, the underlying neural dynamics are not clear. In this study, we use Dynamic Causal Modeling (DCM) on EEG data to investigate whether reduced embodiment during emotion processing in ASD individuals is caused by c

Piezo1 and Piezo2 Ion Channels in Neuronal and Astrocytic Responses to MEA Implants in the Rat Somatosensory Cortex.

Intracortical microelectrode arrays (MEAs) are tools for recording and stimulating neural activity, with potential applications in prosthetic control and treatment of neurological disorders. However, when chronically implanted, the long-term functionality of MEAs is hindered by the foreign body response (FBR), characterized by gliosis, neuronal loss, and the formation of a glial scar encapsulating layer. This response begins immediately after implantation and is exacerbated by factors such as brain micromotion and the mechanical mismatch between stiff electrodes and soft brain tissue, leading

Regional brain morphology of the primary somatosensory cortex correlates with spicy food consumption and capsaicin sensitivity.

Objective: Habitual spicy food consumption leads to altered perception of capsaicin. Little is known about the neural morphological correlates of habitual spicy food intake and related trigeminal perceptions. In this study, we used voxel-based morphometry to identify brain regions where regional gray matter volume (GMV) correlates to spicy food consumption. Methods: Fifty-two participants were surveyed for their spicy food dietary habit by a composite score of spicy diet duration, frequency of spicy food consumption, and preferred degree of spiciness. Forty-two participants were further assess

Modulation of the rubber hand illusion by transcranial direct current stimulation over the contralateral somatosensory cortex.

In the rubber hand illusion (RHI), illusory bodily ownership is induced by synchronous touch of a participant's hidden hand and a visible surrogate. This paradigm allows investigating how the brain resolves conflicting multisensory evidence during perceptual inference. Previous studies suggest that the conflict between visual and proprioceptive information preceding the RHI is solved by attenuation of the somatosensory input. To investigate whether excitability-decreasing transcranial direct current stimulation (cathodal tDCS) over the primary somatosensory cortex may enhance the RHI, thirty h

Structural abnormalities in the primary somatosensory cortex and a normal behavioral profile in Contactin-5 deficient mice.

Contactin-5 (Cntn5) is an immunoglobulin cell adhesion molecule that is exclusively expressed in the central nervous system. In view of its association with neurodevelopmental disorders, particularly autism spectrum disorder (ASD), this study focused on Cntn5-positive areas in the forebrain and aimed to explore the morphological and behavioral phenotypes of the Cntn5 null mutant (Cntn5-/-) mouse in relation to these areas and ASD symptomatology. A newly generated antibody enabled us to elaborately describe the spatial expression pattern of Cntn5 in P7 wild type (Cntn5+/+) mice. The Cntn5 expre
