Ultrasound frequency of sonication applied in microbiological diagnostics has a major impact on viability of bacteria causing PJI.

OBJECTIVES: Sonication of explanted prosthesis constitutes an element of microbiological diagnostics. The aim of performing this procedure is to remove biofilm and to increase sensitivity of diagnostics. Ultrasound used in medical purposes are low-frequency and low-intensity. With this wide range of frequency which can be used in sonication process it is necessary to find the golden mean between biofilm dislodging and planktonic bacteria sparing. MATERIALS AND METHODS: The aim of this study was to determine the least harming low-intensity ultrasound frequency (35 kHz, 40 kHz or 53 kHz) used d

Toward Inter-Sonication Monitoring of Skull and Brain Temperatures in Transcranial MRgFUS Using a 3D Stack-of-Spirals Sequence.

PURPOSE: To develop an MRI approach for simultaneous skull and brain temperature monitoring between sonications in transcranial MR-guided focused ultrasound (MRgFUS) surgeries. METHODS: A 3D stack-of-spirals dual-echo ultra-short TE (UTE) sequence incorporating fat suppression, spiral deblurring, trajectory measurement, and B1 correction was implemented. A variable TE strategy enabled UTEs as short as 0.05 ms for skull T1 mapping, while a fixed late-TE (LTE) was used for brain temperature measurement. The approach was evaluated in laboratory and clinical settings through phantom and human stu

Combination of advanced nano-Fenton process and sonication for destruction of diclofenac and variables optimization using response surface method.

Diclofenac (DCF) as a non-steroidal pharmaceutical has been detected in aquatic samples more than other compounds due to its high consumption and limited biodegradability. In this study, ultrasound waves were applied along with an advanced nano-Fenton process (US/ANF) to remove DCF, and subsequently, the synergistic effect was determined. Before that, the efficiency of the US and ANF processes was separately studied. The central composite design was used as one of the most applicable responses surface method techniques to determine the main and interactive effect of the factors influencing DCF

Clinical evaluation of dithiothreitol in comparison with sonication for biofilm dislodgement in the microbiological diagnosis of periprosthetic joint infection.

Sonication of explanted devices is well investigated method and was shown to improve the microbiological diagnosis of impant-associated infections by physical removal of bacterial biofilms. Recently, novel approach with chemical agents have been investigated for biofilm dislodgement such as dithiothreitol (DTT). We compared the biofilm dislodgement efficacy of chemical method (dithiothreitol, DTT) compared to the sonication procedure in the diagnosis of prosthetic joint infections (PJI). In a prospective cohort, 187 patients undergoing hip and knee prostheses explantation were included, of who

SONIC: Supersizing motion tracking for natural humanoid whole-body control.

Despite the rise of billion-parameter foundation models trained across thousands of graphical processing units (GPUs), similar scaling gains have not been shown for humanoid control. Current neural controllers for humanoids remain modest in size, target a limited set of behaviors, and are trained on a handful of GPUs. We show that scaling model capacity, data, and compute yields a generalist humanoid controller capable of natural, robust whole-body movements. We position motion tracking as a scalable task for humanoid control, leveraging dense supervision from diverse motion-capture data to ac

AMBRA1 enhances Sonic Hedgehog signaling during cerebellar development and in medulloblastoma.

AMBRA1 (autophagy and beclin 1 regulator 1) is primarily recognized as a tumor suppressor. However, its role as a tumor promoter has garnered increasing attention. Here, leveraging clinical data of an international multi-omic medulloblastoma (MB) cohort, we identified that elevated AMBRA1 protein levels, independently of its mRNA expression, correlate with poor prognosis in the Sonic Hedgehog subgroup (MBSHH). Mechanistically, AMBRA1 enhances SHH signaling by stabilizing GLI1, the pathway's final effector, via inhibition of its βTrCP-mediated degradation. Additionally, AMBRA1 protein stability

Sonic Hedgehog signaling in spinal cord injury: mechanisms and therapeutic implications.

Spinal cord injury is a serious neurological condition that results in severe dysfunction below the level of injury, often leading to long-term disability and impaired quality of life. Despite significant advances in medical care, effective treatment options remain limited. Recent research has highlighted the role of endogenous signaling pathways, including Sonic Hedgehog, in the natural healing response following SCI. Sonic Hedgehog (Shh) signaling pathway plays a vital role in spinal cord development and post-injury regeneration by regulating neuroprotection, axon regeneration, synaptic remo

The role of sonic hedgehog signaling in the oropharyngeal epithelium during jaw development.

Sonic hedgehog (Shh) is expressed in the oropharyngeal epithelium, including the frontonasal ectodermal zone (FEZ), which is defined as the boundary between Shh and Fgf8 expression domains in the frontonasal epithelium. To investigate the role of SHH signaling from the oropharyngeal epithelium, we generated mice in which Shh expression is specifically deleted in the oropharyngeal epithelium (Isl1-Cre; Shhf/f). In the mutant mouse, Shh expression was excised in the oropharyngeal epithelium as well as FEZ and ventral forebrain, consistent with the expression pattern of Isl1. Isl1-Cre; Shhf/f mic

Sonic Hedgehog Determines Early Retinal Development and Adjusts Eyeball Architecture.

The eye primordium of vertebrates initially forms exactly at the side of the head. Later, the eyeball architecture is tuned to see ahead with better visual acuity, but its molecular basis is unknown. The position of both eyes in the face alters in patients with holoprosencephaly due to Sonic hedgehog (Shh) mutations that disturb the development of the ventral midline of the neural tube. However, patient phenotypes vary extensively, and microforms without a brain anomaly relate instead to alternation of gene expression of the Shh signaling center in the facial primordia. We identified novel mis

Pharmacological modulation of Sonic Hedgehog signaling pathways in Angiogenesis: A mechanistic perspective.

The Sonic hedgehog (SHh) signaling pathway is an imperative operating network that helps in regulates the critical events during the development processes like multicellular embryo growth and patterning. Disruptions in SHh pathway regulation can have severe consequences, including congenital disabilities, stem cell renewal, tissue regeneration, and cancer/tumor growth. Activation of the SHh signal occurs when SHh binds to the receptor complex of Patch (Ptc)-mediated Smoothened (Smo) (Ptc-smo), initiating downstream signaling. This review explores how pharmacological modulation of the SHh pathw

PTCHD1 Binds Cholesterol but Not Sonic Hedgehog, Suggesting a Distinct Cellular Function.

Deleterious mutations in the X-linked Patched domain-containing 1 (PTCHD1) gene may account for up to 1% of autism cases. Despite this, the PTCHD1 protein remains poorly understood. Structural similarities to Patched family proteins point to a role in sterol transport, but this hypothesis has not been verified experimentally. Additionally, PTCHD1 has been suggested to be involved in Hedgehog signalling, but thus far, the experimental results have been conflicting. To enable a variety of biochemical and structural experiments, we developed a method for expressing PTCHD1 in Spodoptera frugiperda

Sonic Hedgehog acts as a macrophage chemoattractant during regeneration of the gastric epithelium.

Sonic Hedgehog (Shh), secreted from gastric parietal cells, contributes to the regeneration of the epithelium. The recruitment of macrophages plays a central role in the regenerative process. The mechanism that regulates macrophage recruitment in response to gastric injury is largely unknown. Here we tested the hypothesis that Shh stimulates macrophage chemotaxis to the injured epithelium and contributes to gastric regeneration. A mouse model expressing a myeloid cell-specific deletion of Smoothened (LysMcre/+;Smof/f) was generated using transgenic mice bearing loxP sites flanking the Smo gene

Gas1 Regulates Patterning of the Murine and Human Dentitions through Sonic Hedgehog.

The mammalian dentition is a serially homogeneous structure that exhibits wide numerical and morphological variation among multiple different species. Patterning of the dentition is achieved through complex reiterative molecular signaling interactions that occur throughout the process of odontogenesis. The secreted signaling molecule Sonic hedgehog (Shh) plays a key role in this process, and the Shh coreceptor growth arrest-specific 1 (Gas1) is expressed in odontogenic mesenchyme and epithelium during multiple stages of tooth development. We show that mice engineered with Gas1 loss-of-function

Cardioprotective effect of sonic hedgehog ligand in pig models of ischemia reperfusion.

Sonic hedgehog (SHH) signaling pathway is involved in embryonic tissue patterning and development. Our previous work identified, in small rodent model of ischemia reperfusion, SHH as a specific efficient tool to reduce infarct size and subsequent arrhythmias by preventing ventricular repolarization abnormalities. The goal of the present study was to provide a proof of concept of the cardioprotective effect of SHH ligand in a porcine model of acute ischemia. Methods: The antiarrhythmic effect of SHH, either by a recombinant peptide (N-SHH) or shed membrane microparticles harboring SHH ligand (M

Dysregulation of sonic hedgehog pathway and pericytes in the brain after lentiviral infection.

BACKGROUND: Impairment of the blood-brain barrier (BBB) has been associated with cognitive decline in many CNS diseases, including HIV-associated neurocognitive disorders (HAND). Recent research suggests an important role for the Sonic hedgehog (Shh) signaling pathway in the maintenance of BBB integrity under both physiological and pathological conditions. METHODS: In the present study, we sought to examine the expression of Shh and its downstream effectors in relation to brain pericytes and BBB integrity in HIV-infected humans and rhesus macaques infected with simian immunodeficiency virus (

Sonic hedgehog-mediated epithelial-mesenchymal transition in renal tubulointerstitial fibrosis.

The sonic hedgehog (SHH) signaling pathway plays a critical role in embryonic development, tissue regeneration and organogenesis. The activation of SHH signaling produces profibrogenic effects in various tissues, such as the liver and the biliary ducts. However, the role of SHH signaling in renal fibrogenesis remains to be elucidated. For this purpose, in the present study, we evaluated the hypothesis that activated SHH signaling promotes the acquisition of a myofibroblastic phenotype through the epithelial-mesenchymal transition (EMT), resulting in renal interstitial fibrosis (RIF).
