TGF-β Isoforms Affect the Planar and Subepithelial Fibrogenesis of Human Conjunctival Fibroblasts in Different Manners.

Three highly homologous isoforms of TGF-β, TGF-β-1~3, are involved in the regulation of various pathophysiological conditions such as wound healing processes in different manners, despite the fact that they bind to the same receptors during their activation. The purpose of the current investigation was to elucidate the contributions of TGF-β-1 ~3 to the pathology associated with conjunctiva. For this purpose, the biological effects of these TGF-β isoforms on the structural and functional properties of two-dimensional (2D) and three-dimensional (3D) cultured human conjunctival fibroblasts (Hcon

A high-throughput 3D conjunctival spheroid model for standardized in vitro testing.

New Approach Methodologies (NAMs) based on human cells are increasingly needed to improve the physiological relevance, reproducibility, and ethical acceptability of preclinical ocular surface research. We developed a reproducible and scalable three-dimensional in vitro conjunctival spheroid model using primary human conjunctival epithelial cells and conjunctival fibroblasts as a human-relevant, scaffold-free test system. Agarose-based microwell arrays were fabricated via a combination of custom-made high-resolution 3D printing and polydimethylsiloxane (PDMS) replica molding, enabling the forma

Bidirectional phenotypic transition in macrophages influences triple-negative breast cancer cell immune evasive patterns and response to chemotherapies in a co-culture spheroid model.

Tumor-associated macrophage (TAM) infiltration is a critical characteristic of triple-negative breast cancer (TNBC) related to drug resistance and poor prognosis. Integrating macrophages into TNBC spheroids is crucial to improve the accuracy of 3D in vitro models that mimic the complexity of the tumor microenvironment (TME) and assess treatment response. However, this remains challenging since the reciprocal effects of these two cell types on each other are not fully understood. In this study, we used the TNBC cell line, MDA-MB-231, and polarized M1-like or M2-like macrophages derived from THP

Beyond one-size-fits-all: cancer biology shapes nanoparticle behavior.

Nanoparticles (NPs) are a promising tool for cancer therapy, yet few have successfully reached clinical application. Current nanomedicine development pipelines are focused on optimizing the physical properties of NPs, overlooking the impact of cancer biology on their behavior. Here, we show that the polyethyleneimine-functionalized, silica-coated gold NPs (Au@mSi-PEI) exhibit distinct accumulation and penetration patterns in 3D spheroids derived from four spheroid models (representative of lung, colon, breast, and cervical cancer). We uncover an inverse relationship between NP uptake and penet

Organoid and Spheroid Tumor Models: Techniques and Applications.

Techniques to develop three-dimensional cell culture models are rapidly expanding to bridge the gap between conventional cell culture and animal models. Organoid and spheroid cultures have distinct and overlapping purposes and differ in cellular sources and protocol for establishment. Spheroids are of lower complexity structurally but are simple and popular models for drug screening. Organoids histologically and genetically resemble the original tumor from which they were derived. Ease of generation, ability for long-term culture and cryopreservation make organoids suitable for a wide range of

Embryonic development of pineal gland vesicles: a morphological and morphometrical study in chick embryos.

Pineal cell aggregates in 5, 10 and 15 day-old chick embryos have been studied. Cell aggregates were classified into rosettes or vesicles and spheroid and ellipsoid vesicles distinguished. The number of pineal vesicles per unit of surface (vesicle density) was determined in three pineal portions: apical, anterior and posterior. By day 5, only cellular rosettes were found, mainly in the apical portion. After 10 and 15 days, the presence of rosettes was occasional. The posterior wall showed only small spheroid vesicles, while in the apical and anterior areas ellipsoid vesicles were also observed

A physiologically relevantin vitro3D melanoma skin model for targeted therapy assessment.

Melanoma progression, regression, dormancy, and drug resistance involve a dynamic interplay between the tumor mass, dermal and epidermal cells, extracellular matrix (ECM), and the administered therapeutic agent. Understanding the mechanisms behind drug and melanoma interactions as well as the possible collateral effect on the surrounding healthy tissue could improve patient outcomes. To mechanistically unravel these complex interactions in melanoma, there is a need to develop realistic preclinicalin vitromodels; however, current melanoma models fail to replicate not only drug-cell, but also ce

Characterization and In Vivo Validation of a Three-Dimensional Multi-Cellular Culture Model to Study Heterotypic Interactions in Colorectal Cancer Cell Growth, Invasion and Metastasis.

Colorectal cancer (CRC) is the third cause of cancer-related mortality in industrialized countries. Local invasion and metastasis formation are events associated with poor prognosis for which today there are no effective therapeutic options. Invasion and metastasis are strongly modulated by cells of the tumor microenvironment (TME), in particular fibroblasts and endothelial cells. Unraveling interactions between tumor cells and cells of the TME may identify novel mechanisms and therapeutic targets to prevent or treat metastasis. We report here the development and in vivo validation of a 3D tum

Enhanced MSC spheroid adhesion on 3D-printed leaf-stacked scaffolds for functional tracheal regeneration.

Reconstruction of segmental tracheal defects using three-dimensional (3D)-printed scaffolds remains a formidable challenge. While polycaprolactone (PCL) is widely utilized for its mechanical integrity, its inherent hydrophobicity limits cellular adhesion and tissue integration. In this study, we developed a 3D-printed PCL tracheal scaffold featuring a Leaf-Stacked Structure (LSS) and evaluated a spatially organized Mesenchymal Stem Cell (MSC) delivery strategy for functional regeneration. MSC spheroids were employed to overcome the limitations of monolayer cells, as their 3D configuration crea

Development and Validation of a Scaffold-Free Human Multilineage Spheroid Model for Early Stage Cholangiopathies Driven by Cholangiocyte Senescence.

BACKGROUND AND AIMS: Cholangiopathies, including primary sclerosing cholangitis (PSC), primary biliary cholangitis (PBC), and post-COVID-19 cholangiopathy (PCC), involve chronic cholangiocyte injury, senescence, epithelial-stromal crosstalk, and progressive fibrosis. However, effective in vitro models to capture these interactions are limited. Here, we present a scaffold-free 3D multilineage spheroid model, composed of hepatocyte-like cells (HepG2), cholangiocytes (H69), and hepatic stellate cells (LX-2), designed to recapitulate early fibrogenic responses driven by senescent cholangiocytes.

Overcoming resistance in small-cell lung cancer.

INTRODUCTION: Small-cell lung cancer (SCLC) accounts for 15% of lung cancers and has a dismal prognosis due to early dissemination and acquired chemoresistance. The initial good response to chemotherapy is followed by refractory relapses within 1-2 years. Mechanisms leading to chemoresistance are not clear and progress is poor. AREAS COVERED: This article reviews the current evidence of the resistance of SCLCs at the cellular level including alteration of key proteins and the possible presence of cancer stem cells (CSCs). Without compelling evidence for cellular mechanisms and clinical failur

Photodynamic treatment of glioblastoma plus endothelial cell spheroid models: increased proliferative and migratory aggressiveness of surviving tumor cells due to iNOS/NO upregulation.

Photodynamic therapy (PDT) is a unique oxidative stress-based anti-tumor modality that has proven highly effective for various solid malignancies. Intrinsic and acquired resistance is a significant challenge for all cancer treatments, including PDT. We showed previously that several human cancer cell lines in 2D cultures can exploit nitric oxide (NO) from stress-upregulated inducible nitric oxide synthase (iNOS) to (i) resist photokilling sensitized by 5-aminolevulinic acid (ALA)-induced protoporphyrin IX, and (ii) promote growth and mobility aggressiveness of surviving tumor cells. The toxin biliatresone causes mouse extrahepatic cholangiocyte damage and fibrosis through decreased glutathione and SOX17.

UNLABELLED: Biliary atresia, the most common indication for pediatric liver transplantation, is a fibrotic disease of unknown etiology affecting the extrahepatic bile ducts of newborns. The recently described toxin biliatresone causes lumen obstruction in mouse cholangiocyte spheroids and represents a new model of biliary atresia. The goal of this study was to determine the cellular changes caused by biliatresone in mammalian cells that ultimately lead to biliary atresia and extrahepatic fibrosis. We treated mouse cholangiocytes in three-dimensional (3D) spheroid culture and neonatal extrahepa

Development of a Novel Fluorescent Recognition Guidewire System Toward Early Lung Cancer Biopsy.

Obtaining tissue biopsies from peripheral pulmonary lesions remains challenging due to the limited maneuverability of conventional bronchoscopic instruments in distal airways and the inability to distinguish malignant from benign tissue in real time. Here, we present a proof-of-concept guidewire-based biopsy system that integrates magnetic actuation for flexible navigation with fluorescence imaging for targeted lesion identification. The system comprises a magnetically driven biopsy tool, a miniaturized camera with an optical filter, a 488 nm laser fiber for fluorescence excitation, and a whit

The application of experimental models for the drug discovery for digestive tumors.

Drug development for digestive tumors depends on various preclinical models to evaluate their efficacy and safety. Traditional models, such as 2D cell lines and animal models, fail to recapitulate the complex pathology in the human body. Recently, novel models, such as 3D organoids, tumor spheroids, and organ-on-a-chip systems, have undergone rapid growth. These models can recapitulate tissue architecture and the microenvironment in a more faithful way, enhancing the translational relevance of in vitro experiments to clinical outcomes. Moreover, patient-derived xenografts and genetically engin

From inserts to 3D spheroids: MAC-T and BME-UV1 co-culture models for in vitro reconstruction of the bovine mammary epithelial architecture.

Two bovine mammary epithelial cell lines, MAC-T and BME-UV1, were co-cultured either in Transwell® inserts or in 3D culture systems to comparatively investigate their potential to mimic the structure and function of a mammary alveolus. Using either Matrigel® or animal-friendly alternatives, i.e. rat collagen type I, a mixture of rat collagen type I and mouse laminin, Vitrogel® or matrix-free ultra-low attachment surfaces, both co-culture mammosphere development and tight junction formation were assessed. On Transwell® inserts, co-culture failed to establish a functional epithelial barrier. In

Low-dose multi-walled carbon nanotubes are non-inflammatory alone but amplify lipopolysaccharide-induced cytokine responses in A549 lung epithelial spheroids.

BACKGROUND: Airborne pollutants comprise biological agents such as lipopolysaccharide (LPS) and engineered nanomaterials, including carbon nanotubes (CNTs), which are increasingly prevalent in industrial and consumer applications. Although the pulmonary toxicity of high-dose CNT exposure is established, the inflammatory consequences of low-dose, non-cytotoxic CNT exposure, particularly in combination with other aerosols, remain poorly understood. METHODS: We investigated cytotoxicity and proinflammatory responses to multi-walled CNTs in three-dimensional A549 human epithelial spheroids, compa

Engineering spatially organized hair peg-like structures in human skin constructs using keratinocyte-dermal papilla fibroblast spheroids and micropatterned dermal scaffolds.

Tissue-engineered skin substitutes (TESS) represent a key therapeutic option for extensive cutaneous injuries when autologous grafting is limited; however, current constructs lack skin appendages, particularly hair follicles, resulting in incomplete functional and aesthetic restoration. A central barrier to engineering follicular structures within TESS is the rapid loss of hair-inductive features in cultured human dermal papilla (DP) fibroblasts and the difficulty of recreating epithelial-mesenchymal interactions required for early folliculogenesis. Here, we report a bioengineering platform th

An advanced in vitro bladder cancer model integrating bladder cancer spheroids into a healthy human urothelium for preclinical therapeutic testing.

BACKGROUND: Despite therapy advances, better solutions for refractory bladder cancer remain an unmet need. Human cell-based models may aid in better treatment translation. We introduce 3D-UHU-TU, a bladder cancer microtissue model which incorporates spheroids derived from low- and high-grade human bladder cancer cell lines (RT112 and T24 respectively) into a healthy human urothelium. METHODS: After model characterisation with histopathology and immunofluorescence microscopy, we trialled both the conventional chemotherapeutic Mitomycin C (MMC), and a novel herpes simplex oncolytic virus (oHSV-

Spheroids with Artificial Mineral Organelles: Machine-Learning-Assisted Analysis and Control of Mechanical Properties, Fusion, and Ossification.

Controlling the balance between structural integration and functional maturation of microtissues remains a central challenge in bone tissue engineering, particularly in the design of composite cell-material constructs. Here, we demonstrate that intracellular loading of preosteoblast cells with submicron vaterite particles serves as a programmable material-based parameter to regulate spheroid behavior. Following controlled pre-loading and assembly into composite spheroids, we systematically quantified particle localization (confocal microscopy, PCC/Manders coefficients, Cellpose-based segmentat
