Effect of starvation, malnutrition, and trauma on the gastrointestinal tract flora and bacterial translocation.

We have previously shown, in an animal model, that viable indigenous bacteria will cross the intact gastrointestinal (GI) mucosa and spread systemically, a process termed bacterial translocation, if the normal bacterial ecology of the gut was sufficiently disrupted to allow bacterial overgrowth or if the animals were severely immunosuppressed. Starvation or protein malnutrition disrupts the normal indigenous GI tract microflora and impairs host antibacterial defenses. Consequently, we tested the effect of the combination of starvation or protein malnutrition plus burn trauma in promoting bacte

Immunohistochemistry and morphometry of gastrin cells in the rat pyloric antrum during starvation.

The gastrin cells (G cells) in the rat pyloric antrum after 7, 14, 21 and 28 days of starvation were investigated by immunohistochemistry and electron microscopy. In the peroxidase anti-peroxidase method for light microscopy, gastrin immunoreactive cells during starvation markedly decreased in number and size. Quantitative electron microscopy revealed that during starvation the number of electron-lucent granules were greatly reduced, but the number of electron-dense granules increased; the number of intermediate granules were not remarkably changed in G cells. These results may suggest that th

Perinatal age determines the severity of retarded lung development induced by starvation.

Susceptibility of the lung to caloric restriction is age-dependent, with more permanent damage occurring during the phases of growth and differentiation. Because the guinea pig is born with more well-developed alveoli than are other rodents, the postnatal lung of this species may better resist alveolar hypoplasia than the prenatal lung. Control animals were raised from sows provided food ad libitum during and after normal gestations (66 to 68 days). Starvation groups received 50% rations of control food intakes during 1 of three 21-day periods: prenatal starvation, with sows rationed during th

Peripheral nerve changes in thiamine deficiency and starvation. An electron microscopic study.

Electron microscopic investigations on sciatic and plantar nerves of thiamine deficient and starved rats show a distally pronounced axonal degeneration. The changes are present in starved and thiamine deficient animals, but the deficient animals are more severely affected. The earliest alterations consist of an increase of mitochondria and a proliferation of vesicular elements of the endoplasmic reticulum. They are followed by loop formations of the axon membrane, clustering and disintegration of neurotubules and neurofilaments, axonal shrinkage and finally myelin disruption. The distal accent

From metabolic starvation to precision therapy: engineering amino acid-targeted tumor therapeutics.

Amino acid metabolism has emerged as a critical nexus linking tumor cell nutrient acquisition with immune microenvironmental regulation, providing a rich source of druggable targets for cancer therapy. Tumor cells engage in fierce metabolic competition with immune cells for key amino acids, including glutamine, arginine, tryptophan, and methionine, thereby establishing an immunosuppressive tumor microenvironment. Over the past decade, therapeutic strategies targeting this metabolic axis have evolved from preclinical proof-of-concept to active clinical development across multiple biotechnology

Hollow Mesoporous Silica Nanoparticles for Dual Chemo-starvation Therapy of Hepatocellular Carcinoma.

PURPOSE: This study aims at chemotherapy and starvation therapy of HCC via starvation and apoptosis. METHODS: Hollow mesoporous organosilica nanoparticles (HMONs) with the thioether-hybrid structure were developed using an organic/inorganic co-templating assembly approach. Hydrofluoric acid was used to remove the internal MSN core for yielding large radial mesopores for loading drug cargos. The morphology and structure of NPs were determined using TEM and SEM. HMONs were stepwise surface modified with glucose oxidase (GOx), oxygen (O2) and Doxorubicin (DOX), and cancer cell membrane (CCM) for

Nitrogen-inducible GLK1 modulates phosphate starvation response via the PHR1-dependent pathway.

Phosphorus (P) is a limiting nutrient for plant growth and productivity. Thus, a deep understanding of the molecular mechanisms of plants' response to phosphate starvation is significant when breeding crops with higher phosphorus-use efficiency. Here, we found that GARP-type transcription factor GLK1 acted as a positive regulator for phosphate-starvation response (PSR) via the PHR1-dependent pathway in Arabidopsis thaliana. GLK1 increased the transcription activity of PHR1 through the direct physical interaction and regulated the multiple responses to inorganic orthophosphate (Pi) starvation.

Scurvy, Starvation, and Flea Infestation - A Case Report From 21st Century Europe.

Scurvy is a disease caused by vitamin C deficiency, historically associated with long sea voyages, periods of famine and war. Currently, it is often misdiagnosed and underreported, as physicians tend to consider it a disease of the past. We present the case of a 79-year-old female who was admitted to the Emergency Department complaining of pruritus. The patient lived alone and in poor hygiene conditions. Diet was scarce. One week before admission she was in contact with flea-ridden stray dogs. Sometime later she noted several fleas and multiple pruritic small papules, crusts, and excoriations

Organotypic microfluidic breast cancer model reveals starvation-induced spatial-temporal metabolic adaptations.

BACKGROUND: Ductal carcinoma in situ (DCIS) is the earliest stage of breast cancer. During DCIS, tumor cells remain inside the mammary duct, growing under a microenvironment characterized by hypoxia, nutrient starvation, and waste product accumulation; this harsh microenvironment promotes genomic instability and eventually cell invasion. However, there is a lack of biomarkers to predict what patients will transition to a more invasive tumor or how DCIS cells manage to survive in this harsh microenvironment. METHODS: In this work, we have developed a microfluidic model that recapitulates the D

Concurrent metformin-associated lactic acidosis and euglycemic diabetic ketoacidosis from tirzepatide starvation ketosis.

SUMMARY: We describe a case of concurrent metformin-associated lactic acidosis (MALA) and euglycemic diabetic ketoacidosis (EDKA) in a 64-year-old woman with type 2 diabetes mellitus (T2DM) treated with metformin and tirzepatide who presented with shock, encephalopathy, and severe high anion gap metabolic acidosis after two weeks of markedly reduced oral intake following tirzepatide dose escalation. While incretin-based therapies, including dual glucose-dependent insulinotropic polypeptide/glucagon-like peptide-1 receptor agonists (GIP/GLP-1 RAs) such as tirzepatide, are highly effective for g

OsCK2 Phosphorylates OsSIZ1 to Regulate Phosphate Starvation Response and Defence Response in Rice.

Phosphorus is a crucial nutrient for the normal growth and development of rice. Enhancing the efficiency of phosphorus acquisition in plants is vital for optimising soil phosphorus utilisation. The SUMO E3 ligase OsSIZ1 is an important factor for regulating plant phosphorus acquisition and starvation response in rice. However, how its activity responds to Pi-supplied conditions remains unknown. In this study, we discovered that the mutations in OsCK2β3, which encodes a regulatory subunit of CK2, influenced the Pi starvation response. The global level of SUMOylation was found to decrease in Osc

A Simple Restriction, a Complex Collapse: Multisystem Wernicke Encephalopathy Triggered by Self-Starvation.

Wernicke encephalopathy (WE) is an acute neuropsychiatric emergency caused by thiamine deficiency, most commonly associated with chronic alcoholism but increasingly recognized in non-alcoholic contexts. This report describes a 23-year-old woman who developed WE secondary to prolonged self-imposed starvation driven by distorted body image. She had deliberately restricted food intake for several weeks, believing she was overweight, which led to severe malnutrition, weight loss, and profound weakness. On admission, she presented with bilateral ptosis, nystagmus, and lower limb weakness but withou

Enhancing Leukemia Treatment: The Role of Combined Therapies Based on Amino Acid Starvation.

Cancer cells demand amino acids beyond their usage as "building blocks" for protein synthesis. As a result, targeting amino acid acquisition and utilization has emerged as a pivotal strategy in cancer treatment. In the setting of leukemia therapy, compelling examples of targeting amino acid metabolism exist at both pre-clinical and clinical stages. This review focuses on summarizing novel insights into the metabolism of glutamine, asparagine, arginine, and tryptophan in leukemias, and providing a comprehensive discussion of perturbing their metabolism to improve the therapeutic outcomes. Cascade-Catalyzed Nanogel for Amplifying Starvation Therapy by Nitric Oxide-Mediated Hypoxia Alleviation.

Glucose oxidase (Gox)-mediated starvation therapy offers a prospective advantage for malignancy treatment by interrupting the glucose supply to neoplastic cells. However, the negative charge of the Gox surface hinders its enrichment in tumor tissues. Furthermore, Gox-mediated starvation therapy infiltrates large amounts of hydrogen peroxide (H2O2) to surround normal tissues and exacerbate intracellular hypoxia. In this study, a cascade-catalyzed nanogel (A-NE) was developed to boost the antitumor effects of starvation therapy by glucose consumption and cascade reactive release of nitric oxide

Acidity-Responsive Nanoreactors Destructed "Warburg Effect" for Toxic-Acidosis and Starvation Synergistic Therapy.

"Warburg Effect" shows that most tumor cells rely on aerobic glycolysis for energy supply, leading to malignant energy deprivation and an "internal alkaline external acid" tumor microenvironment. Destructing the "Warburg Effect" is an effective approach to inhibit tumor progression. Herein, an acidity-responsive nanoreactor (Au@CaP-Flu@HA) is fabricated for toxic acidosis and starvation synergistic therapy. In the nanoreactor, the fluvastatin (Flu) could reduce lactate efflux by inhibiting the lactate-proton transporter (monocarboxylate transporters, MCT4), resulting in intracellular lactate a

Morphological change and differential proteomics analysis of gill in Mytilus coruscus under starvation.

Mytilus coruscus is a dominant shellfish in the Yangtze estuary and its adjacent sea area. Food deprivation often occurs during their growth due to fluctuations in algal abundance caused by seasonal freshwater flushing and high-density aquaculture mode. To investigate the coping strategies of M. coruscus to starvation stress, electron microscopy and differential proteomic analysis were performed on the critical feeding organ gill of the mussels after 9 days of starvation. The electron microscopy results showed that the cilia of the mussel gills were dissolved, and the gaps between gill filamen
