Streptococcus mutans outer membrane vesicles affect inflammasome activation and the glycolysis of macrophages.

Recent studies indicate that bacterial outer membrane vesicles (OMVs) play a significant role in bacterial virulence and pathogenicity. Streptococcus mutans (S. mutans), a principal pathogen in dental caries, secretes a substantial number of OMVs. However, the impact of S. mutans OMVs on oral health and their underlying pathogenic mechanisms remain poorly understood. Macrophages were the initial innate immune cells to respond to bacterial invaders and their products. Therefore, we purified S. mutans OMVs, which stimulated macrophages. Compared to controls, RT-PCR and ELISA analyses revealed th

Usnic acid brief exposure suppresses cariogenic properties and complexity of Streptococcus mutans biofilms.

Bacterial biofilms are highly structured surface associated architecture of micro-colonies, which are strongly bonded with the exopolymeric matrix of their own synthesis. These exopolymeric substances, mainly exopolysaccharides (EPS) initially assist the bacterial adhesion and finally form a bridge over the microcolonies to protect them from environmental assaults and antimicrobial exposure. Bacterial cells in dental biofilm metabolize dietary carbohydrates and produce organic acids. The blanket of exopolysaccharides over the bacterial communities hinders the buffering by saliva, contributing

Ribosomal-processing cysteine protease homolog modulates Streptococcus mutans glucan production and interkingdom interactions.

UNLABELLED: Glucan-dependent biofilm formation is a crucial process in the establishment of Streptococcus mutans as a cariogenic oral microbe. The process of glucan formation has been investigated in great detail, with glycosyltransferases GtfB, GtfC, and GtfD shown to be indispensable for the synthesis of glucans from sucrose. Glucan production can be visualized during biofilm formation through fluorescent labeling, and its abundance, as well as the effect of glucans on general biofilm architecture, is a common phenotype to study S. mutans virulence regulation. Here, we describe an entirely n

Zerumbone disrupts mixed biofilms of Candida albicans and Streptococcus mutans on acrylic resin.

The efficacy of Zerumbone (ZER) against mixed biofilms of fluconazole-resistant Candida albicans (ATCC 96901) and Streptococcus mutans (UA159) was evaluated. Biofilms were cultivated on acrylic resin specimens for 48 h, with alternating supplementation of glucose and sucrose. ZER's ability to inhibit biofilm formation (pre-treatment) and eradicate mature biofilms (post-treatment) was assessed. Control groups were treated with Chlorhexidine (CHX), Nystatin (NYS), Penicillin (ATB), and distilled water. The efficacy was measured by colony forming units (CFU/mm2) counts, biomass and biofilm's matr

Competitive dynamics and balance between Streptococcus mutans and commensal streptococci in oral microecology.

Dental caries, as a biofilm-related disease, is closely linked to dysbiosis in microbial ecology within dental biofilms. Beyond its impact on oral health, bacteria within the oral cavity pose systemic health risks by potentially entering the bloodstream, thereby increasing susceptibility to bacterial endocarditis, among other related diseases. Streptococcus mutans, a principal cariogenic bacterium, possesses virulence factors crucial to the pathogenesis of dental caries. Its ability to adhere to tooth surfaces, produce glucans for biofilm formation, and metabolize sugars into lactic acid contr

Exopolysaccharides metabolism and cariogenesis of Streptococcus mutans biofilm regulated by antisense vicK RNA.

BACKGROUND: Streptococcus mutans (S. mutans) is a pivotal cariogenic pathogen contributing to its multiple virulence factors, one of which is synthesizing exopolysaccharides (EPS). VicK, a sensor histidine kinase, plays a major role in regulating genes associated with EPS synthesis and adhesion. Here we first identified an antisense vicK RNA (ASvicK) bound with vicK into double-stranded RNA (dsRNA). OBJECTIVE: This study aims to investigate the effect and mechanism of ASvicK in the EPS metabolism and cariogenesis of S. mutans. METHODS: The phenotypes of biofilm were detected by scanning elec

Symbiotic relationship between Prevotella denticola and Streptococcus mutans enhances virulence of plaque biofilms.

OBJECTIVES: This study aimed to explore that whether interactions between Prevotella denticola and Streptococcus mutans could promote the establishment of hypervirulent biofilms on teeth surface and eventually influence the occurrence and development of caries. DESIGN: Based on single-species biofilms of either P. denticola or S. mutans, and dual-species biofilms of both bacteria, we compared the virulence properties associated with cariogenicity in vitro, including carbohydrate metabolism and acid productivity, synthesis of extracellular polysaccharides, biomass and architecture of biofilms,

Immunorecognition of Streptococcus mutans secreted proteins protects against caries by limiting tooth adhesion.

INTRODUCTION: Childhood caries, a prevalent chronic disease, affects 60-90 % of children in industrialized regions, leading to lesions in both primary and permanent teeth. This condition precipitates hospital admissions, emergency room visits, elevated treatment costs, and missed school days, thereby impeding the child's academic engagement and increasing the likelihood of caries into adulthood. Despite multiple identified risk factors, significant interpersonal variability remains unexplained. The immune system generates a unique antibody repertoire, essential for maintaining a balanced and h

Salivary Streptococcus mutans, Lactobacilli levels and buffer capacity in children with esophageal burns.

Children with esophageal burns due to toxic ingestion are medically compromised in many ways. They have problems like strictures in esophagus, dysphagia, gastroesophageal reflux and necessity to be fed with a special soft diet. Taking these distinctive features into consideration, we aimed to study these children with special focus on the dental caries risk. The experimental group consisted of 33 children between 2 and 14 years old (mean age 6.42 +/- 0.56) who had received stricture treatment in the Pediatric Surgery Department of our university. The control group consisted of 20 healthy child

[Studies on caries of Ishibashi rat--II. Streptococcus mutans population in oral cavity].

Previously, it was demonstrated that extensive carious lesions occurred in the Ishibashi rats (ISR) while they were bred with conventional diets. In this study, specimens from the oral cavity of the ISR were examined for the presence of Streptococcus mutans, and its occurrence and population were compared with those of the Wistar rats. The results were as follows: 1. S. mutans was first found in the mouth of the ISR when the first molars erupt. 2. The number of S. mutans (CFU on the gelatin agar) and its mean percentage of the total streptococci (CFU on the gelatin agar/CFU on the M-S agar) in

Backbone NMR resonance assignments for the C2 domain of the Streptococcus mutans adhesin P1.

Adhesin P1 (aka AgI/II) is an extracellular protein regulating adherence and detachment of Streptococcus mutans in the oral cavity and thus plays a pivotal role in biofilm development and maturation. P1's naturally occurring C-terminal truncation product, Antigen II (AgII), adopts both soluble, monomeric and insoluble, amyloidogenic forms during the bacterial life cycle. Monomeric AgII forms important quaternary interactions with P1's A3VP1 segment that is projected from the bacterial cell surface to promote cell adhesion, while the functional amyloid form of AgII promotes detachment of mature
