Phenotypic and Genotypic Characterization of Resistance to Macrolides and Lincosamides in Streptococcus agalactiae Isolated from Pregnant Women in Misiones, Argentina.

The aim of this work was to determine the susceptibility, molecular profile, and clonal relationship in Streptococcus agalactiae (group B Streptococcus [GBS]) isolated from vaginal-rectal swab samples. We worked with 200 isolates collected from pregnant women between 35 and 37 weeks of gestation. The macrolide-lincosamide-streptogramin B (MLSB) resistance phenotypes were determined using the double-disc assay. Susceptibility to erythromycin (ERI) and clindamycin (CLI) was performed with the E-test. Resistance genes ermB and ermTR were detected by polymerase chain reaction. Clonal studies were

Distribution of Transferable Antibiotic Resistance Genes in Laboratory-Reared Edible Mealworms (Tenebrio molitor L.).

In the present study, the distribution of antibiotic resistance genes in laboratory-reared fresh mealworm larvae (Tenebrio molitor L.), their feeding substrates (carrots and wheatmeal), and frass was assessed. Microbial counts on selective media added with antibiotics highlighted the presence of lactic acid bacteria resistant to ampicillin and vancomycin and, more specifically, enterococci resistant to the latter antibiotic. Moreover, staphylococci resistant to gentamicin, erythromycin, tetracycline, and vancomycin were detected. Enterobacteriaceae resistant to ampicillin and gentamicin were a

The Rate of Inducible MLSB Resistance in the Methicillin-Resistant Staphylococci Isolated From Clinical Samples.

BACKGROUND: Staphylococci are one of the most common pathogens in nasocomial and community-acquired infections. Methicillin-resistant staphylococci are known to be resistant against all beta-lactam antibiotics. Therefore, non-beta-lactam antibiotics such as macrolide and lincosamides can be used. Resistance to those antibiotics may lead to therapeutic failure. The purpose of the present study was to determine the prevalence of macrolide-lincosamide-streptogramin B (MLSB ) resistance by using D-test in staphylococcal isolates from various clinical samples. METHODS: Seventy-one methicillin-resi

Staphylococcal tetracycline-MLSB resistance plasmid pSTE2 is the product of an RSA-mediated in vivo recombination.

OBJECTIVES: The complete nucleotide sequence of the 6913 bp plasmid pSTE2 from Staphylococcus lentus, which mediates inducible resistance to tetracyclines, macrolides and lincosamides, was determined. The plasmid was analysed for potential reading frames and structural features to gain insight into its development from potential ancestor plasmids. METHODS: Plasmid pSTE2 was transformed into Staphylococcus aureus RN4220. Suitable restriction fragments were cloned into E. coli plasmid vectors and sequenced. In vitro susceptibility testing was performed to confirm the resistance phenotype mediat

[Interpretive reading of the antibiogram in gram positive cocci].

Resistance to methicillin in Staphylococcus is related to the expression of the mecA gene, and involves resistance to all beta-lactams, with the exception of the new cephalosporins, ceftobiprole and ceftaroline. Breakpoints for interpretation of this mechanism differ in S. aureus and in coagulase-negative species. For macrolides-lincosamides-streptogramins B, (MLS(B)) the most frequent mechanism among resistant strains is expression of methylases (erm genes). Topoisomerase changes caused by point mutations and expression of the efflux pump NorA determine resistance to quinolones, but there are

Prevalence and Molecular Determinants of Antimicrobial Resistance in Clinical Isolates of Staphylococcus haemolyticus from India.

Aims: Although Staphylococcus haemolyticus is considered as a part of normal skin flora, infections associated with them are increasing. Irrespective of the low virulence profile it poses a severe threat to patients with indwelling devices due to its multidrug-resistant nature. The aim of this study was to determine antibiotic resistance patterns and to detect the genes responsible in clinical isolates of S. haemolyticus. Results: All the 356 S. haemolyticus isolates were susceptible to glycopeptides. 91.3% were resistant to cefoxitin, 85.4% to erythromycin, 57.3% to co-trimoxazole, 52.8% to c

Plausible Minimal Substrate for Erm Protein.

Erm proteins methylate a specific adenine residue (A2058, Escherichia coli coordinates) conferring macrolide-lincosamide-streptogramin B (MLSB) antibiotic resistance on a variety of microorganisms, ranging from antibiotic producers to pathogens. To identify the minimal motif required to be recognized and methylated by the Erm protein, various RNA substrates from 23S rRNA were constructed, and the substrate activity of these constructs was studied using three Erm proteins, namely, ErmB from Firmicutes and ErmE and ErmS from Actinobacteria The shortest motif of 15 nucleotides (nt) could be recog

Antimicrobial susceptibility of invasive Streptococcus pneumoniae isolates in Portugal over an 11-year period.

This national surveillance study presents the in vitro activities of the main antimicrobial agents against 1,331 S. pneumoniae isolates as tested by an agar dilution method according to the guidelines of the Clinical and Laboratory Standards Institute (formerly NCCLS). The strains were isolated in several regions of Portugal from cases of invasive disease over an 11-year period (1994 to 2004). This study shows that the percentage of penicillin-nonsusceptible strains increased from 12% in 1994 to 28.5% in 2000. Then the rate declined to 17.7% in 2003 but increased again to 23.2% in 2004. The Use of Ketolides in Treatment of Upper Respiratory Tract Infections.

Recent surveillance studies suggest that the incidence of resistance to macrolide antibiotics in common community-acquired respiratory tract pathogens, particularly Streptococcus pneumoniae and Streptococcus pyogenes, is increasing and limiting the usefulness of these drugs. The ketolides, of which telithromycin is the first to be available for clinical use (but not yet in the United States), represent a new class of antibacterials developed specifically to combat respiratory tract pathogens that have acquired resistance to macrolides. The ketolides possess innovative structural modifications,

The Complexity of erm-Mediated Resistance: Structural and Functional Perspectives From β-Hemolytic Streptococci.

Macrolide overuse and resulting resistance crippled the applicability of one of the most important and successful classes of antibiotics. The rising incidence of invasive infections caused by β-hemolytic group A and group B streptococci have been paralleled with marked increases in erythromycin and clindamycin resistance. As a result, resistant invasive group A and group B Streptococcus infections have been classified by the CDC as a concerning level threat since 2019. Acquisition of erythromycin resistance methylase (erm) genes is a major contributor of the MLSB [macrolide (i.e., erythromycin

Telithromycin.

Telithromycin is the first member of a new family of the macrolide-lincosamide-streptogramin-B (MLS(B)) class of antimicrobials, the ketolides. It has a good spectrum of activity against respiratory pathogens, including penicillin- and erythromycin-resistant pneumococci, as well as intracellular and atypical bacteria. Furthermore, it has a low potential to select for resistance or induce cross-resistance among other MLS(B) antimicrobials. At the recommended dosage of 800 mg orally once daily, telithromycin reaches maximal plasma concentrations of about 2 mg/L. It penetrates rapidly into bronch

[The prevalence of antiseptic resistance genes (qacA/B and smr) and antibiotic resistance in clinical Staphylococcus aureus strains].

Staphylococcus aureus is an organism of major medical importance, leading to skin and soft tissue infections, bacteremia, and endocarditis. S.aureus isolates are becoming increasingly resistant to numerous antimicrobial agents including antiseptics and disinfectants. Quaternary ammonium compounds are disinfectants that play an important role in the control of nosocomial infections. Presence of genes conferring resistance to quaternary ammonium compounds is widely distributed among clinical staphylococci isolated from certain areas of the world.
