Abstract In transsulfuration reaction catalyzed by rat mercaptopyruvate sulfurtransferase (MST), a stable persulfide is formed at the catalytic site cysteine Cys(247) as a reaction intermediate. The outer sulfur atom is donated by the substrate, thiosulfate, or by mercaptopyruvate. MST serves as a thioredoxin-dependent antioxidant possessing self-regulated enzymatic activity. After oxidation of persulfurated MST by treatment with hydrogen peroxide, mass spectrometric analysis showed that the outer sulfur atom of the persulfide is oxidized to form Cys-thiosulfenate (Cys-Sγ-SO(-)), Cys-thiosulfi

Methionine sulfoxide reductase A (MsrA) protects proteins from oxidative damage through cysteine-based redox cycling, but how mammalian MsrA is structurally regulated during oxidation and reductive recycling has remained unclear. We report three crystal structures of mouse MsrA (mMsrA) that capture distinct redox states - reduced (3.0 Å), BME-trapped intermediate-like (BTIL; 1.6 Å), and oxidized (1.9 Å) - and reveal an unrecognized layer of regulation centered on Cys107. The BTIL structure visualizes catalytic Cys72 as a mixed disulfide with β-mercaptoethanol (BME) alongside a sulfenic acid at

Self-assembled liquid crystal polymer networks build directional transport channels via molecular alignment, enabling efficient and ordered lithium-ion migration, while single-ion conducting polymer electrolytes improve lithium-ion transference number by covalently anchoring anions, which alleviates concentration polarization and effectively suppresses lithium dendrite growth. Herein, a series of substrate-free Li salt-grafted cholesteric liquid crystal elastomers (CLCE) with tunable helical pitches and spiral orientation were fabricated, and quasi-solid electrolyte membranes were obtained thr

BACKGROUND: Glucosamine, a naturally occurring amino sugar abundant in cartilage, has long been utilized as a dietary supplement to alleviate osteoarthritis (OA) and joint pain. Beyond its structural role in maintaining joint integrity, glucosamine is metabolized through the hexosamine biosynthetic pathway (HBP) to generate UDP-N-acetylglucosamine (UDP-GlcNAc), the indispensable donor substrate for protein O-linked N-acetylglucosamine (O-GlcNAc) modification.
FINDINGS: Accumulating evidence indicates that glucosamine-driven modulation of HBP flux and the consequent alteration of O-GlcNAcylati

PURPOSE: We aimed to define intrauterine insemination (IUI) cycle characteristics associated with viable birth, identify thresholds below which IUI treatments are consistent with very poor prognosis and futile care, and develop a nomogram for individualized application.
METHODS: This retrospective cohort study evaluated couples using fresh partner ejaculate for IUI from January 2005 to September 2017. Variables included female age, semen characteristics, and ovarian stimulation type. Using cycle-level data, we evaluated the association of these characteristics with the probability of viable b

The transforming properties of the high-risk human papillomavirus (HPV) E7 oncoprotein are indispensable for driving the virus life cycle and pathogenesis. Besides inactivation of the retinoblastoma family of tumor suppressors as part of its oncogenic endeavors, E7-mediated perturbations of eminent cell cycle regulators, checkpoint proteins and proto-oncogenes are considered to be the tricks of its transformative traits. However, many such critical interactions are still unknown. In the present study, we have identified the anaphase-promoting complex/cyclosome (APC) co-activator, Cdh1, as a no

Pyruvate given in large doses may be neuroprotective in stroke, but it is not known to what degree the brain metabolizes pyruvate. Intravenous injection of [3-13C]pyruvate led to dose-dependent labelling of cerebral metabolites so that at 5 min after injection of 18 mmoles [3-13C]pyruvate/kg (2 g sodium pyruvate/kg), approximately 20% of brain glutamate and GABA were labelled, as could be detected by 13C nuclear magnetic resonance spectrometry ex vivo. Pyruvate, 9 mmoles/kg, was equivalent to glucose, 9 mmoles/kg, as a substrate for cerebral tricarboxylic acid (TCA) cycle activity. Inhibition

Photosynthesis is the main process by which biomass of living matter is created from inorganic carbon. However, the efficiency of photosynthesis is low. The main reason for the low efficiency of photosynthesis is the properties of the enzyme that fixes carbon dioxide - ribulose-1,5-bisphosphate carboxylase/oxidase (RuBisCO). There are reasons to believe that it is impossible to improve the properties of RuBisCO. It is apparently possible to significantly increase the efficiency of photosynthesis only by replacing RuBisCO with another enzyme. Among the known enzymes capable of fixing CO2, there

A reusable and self-calibrating Surface-Enhanced Raman Spectroscopy (SERS) substrate was developed for reliable detection and degradation of organic pollutants in complex environments. The substrate was fabricated by synthesizing Prussian blue (PB)-functionalized cotton (Cotton/PB), followed by Ag+ ion exchange with Fe3+ in the surface-bound PB and in situ reduction to form silver nanoparticles (Ag NPs). PB serves as an internal standard (IS) to correct SERS signal fluctuations and enables self-cleaning via photo-Fenton catalysis. The substrate exhibited high sensitivity, with detection limits

Recent studies highlight the emerging role of lipids as important messengers in malaria parasite biology. In an attempt to identify interacting proteins and regulators of these dynamic and versatile molecules, we hypothesised the involvement of phospholipid translocases and their substrates in the infection of the host erythrocyte by the malaria parasite Plasmodium spp. Here, using a data base searching approach of the Plasmodium Genomics Resources (www.plasmodb.org), we have identified a putative phospholipid (PL) scramblase in P. falciparum (PfPLSCR) that is conserved across the genus and in

The article discusses the removal of methylene blue (MB) dye, a common cationic dye used in the textile industry, from aqueous solutions through an adsorption process. The use of porous components as adsorbents are shown to facilitate complete separation after the process is completed. The substrate was synthesized by connecting zinc copper ferrite (ZnCuFe2O4), polyethyleneimine (PEI), and Graphene Oxide (GO) sheets to MCM-48, which is a mesoporous material. The surface of MCM-48 was modified using CPTMS, which created an O-Si-Cl bridge, thereby improving the adsorption rate. The substrate was

The human sodium-dependent multivitamin transporter (SMVT, SLC5A6) mediates cellular uptake of essential metabolic cofactors, including biotin, pantothenate, and lipoate. Its dysfunction is associated with neurological disorders, metabolic abnormalities, and cancer. However, the molecular mechanism underlying its multi-substrate transport has remained elusive. Here, we present cryo-electron microscopy structures of human SMVT in three conformational states: occluded, outward-open, and inward-open. These structural snapshots capture the complete transport cycle and reveal a conserved substrate-

Chiral binap/pica-Ru(II) complexes (binap=(S)- or (R)-2,2'-bis(diphenylphosphino)-1,1'-binaphthyl; pica=alpha-picolylamine) catalyze both asymmetric hydrogenation (AH) of ketones using H(2) and asymmetric transfer hydrogenation (ATH) using non-tertiary alcohols under basic conditions. The AH and ATH catalytic cycles are linked by the metal-ligand bifunctional mechanism. Asymmetric reduction of pinacolone is best achieved in ethanol containing the Ru catalyst and base under an H(2) atmosphere at ambient temperature, giving the chiral alcohol in 97-98 % ee. The reaction utilizes only H(2) as a h

Hepatocytes from different subacinar localizations differ not only in their enzyme equipment, but have also different functions in intermediary metabolism. Gluconeogenesis, amino acid breakdown, urea synthesis and oxidative metabolism occur predominantly periportal, whereas glycolysis, glutamine synthesis and drug metabolism are predominantly perivenous. This metabolic zonation is rather dynamic and is influenced by hormones, oxygen-tension, substrate concentrations, circadian rhythms and development. There are metabolic interactions between differently localized hepatocyte populations: the pr

Cyclin dependent-kinase 2 (CDK2) plays important functions during the mitotic cell cycle and also facilitates several key events during germ cell development. The majority of CDK2's known meiotic functions occur during prophase of the first meiotic division. Here, CDK2 is involved in the regulation of meiotic transcription, the pairing of homologous chromosomes, and the maturation of meiotic crossover sites. Despite that some of the CDK2 substrates are known, few of them display functions in meiosis. Here, we investigate potential meiotic CDK2 substrates using in silico and in vitro approaches

UNLABELLED: Coral gastrovascular cavities (GVCs) host diverse microorganisms and, as semi-closed compartments, are characterized by elevated nutrient concentrations and pronounced diel fluctuations in oxygen (O2) and pH. However, their broader chemical microenvironment and associated microbial activities remain poorly understood. Here, we provide direct evidence for active anaerobic metabolism in the GVC of Astraeosmilia curvata by measuring hydrogen (H2), nitric oxide (NO), and nitrous oxide (N2O) production, indicative of microbial fermentation and denitrification. Changes in GVC H2 concentr

This study expands the growing understanding that protein acetylation is a highly regulated molecular toggle of protein function in both host anti-viral defense and viral replication. We describe a pro-viral role for the human enzyme SIRT2, showing that its deacetylase activity supports HCMV replication. By integrating quantitative proteomics, flow cytometry cell cycle assays, microscopy, and functional virology assays, we investigate the temporality of SIRT2 functions and substrates. We identify a pro-viral role for the SIRT2 deacetylase activity via regulation of CDK2 K6 acetylation and the

Homogeneous preparations of liver glutathione reductase (GR, EC 1.6.4.2.), obtained from control rats and animals with toxic hepatitis, catalytic properties of this enzyme have been investigated. A number of the enzyme properties did not change under conditions of hepatitis. These included electrophoretic mobility (R(f) = 0.23 +/- 0.01), molecular mass (104 +/- 5.20 kDa), pH-optimum (7.4 +/- 0.37). There was similarity in pK values of functional groups. At the same time, decrease in affinity of enzyme to a substrate and coenzyme and occurrence substrate-linked inhibition was observed under con

The life cycle of many sessile marine invertebrates includes a dispersive planktonic larval stage whose ability to find a suitable habitat in which to settle and transform into benthic adults is crucial to maximize fitness. To facilitate this process, invertebrate larvae commonly respond to habitat-related chemical cues to guide the search for an appropriate environment. Furthermore, small-scale hydrodynamic conditions affect dispersal of chemical cues, as well as swimming behavior of invertebrate larvae and encounter with potential habitats. Shipworms within the family Teredinidae are depende

The urea cycle consists of six consecutive enzymatic reactions that convert waste nitrogen into urea. Deficiencies of any of these enzymes of the cycle result in urea cycle disorders (UCD), a group of inborn errors of hepatic metabolism that often result in life threatening hyperammonemia. Argininosuccinate lyase (ASL) is a cytosolic enzyme which catalyzes the fourth reaction in the cycle and the first degradative step, that is, the breakdown of argininosuccinic acid to arginine and fumarate. Deficiency of ASL results in an accumulation of argininosuccinic acid in tissues, and excretion of arg
