Hydrogen sulfide (H2S) is thought to signal through protein S-sulfuration (persulfidation; S-sulfhydration) in both mammalian systems and bacteria. We previously profiled proteome S-sulfuration in Staphylococcus aureus (S. aureus) and identified two thioredoxin-like proteins, designated TrxP and TrxQ, that were capable of reducing protein persulfides as a potential regulatory mechanism. In this study, we further characterize TrxP, TrxQ and the canonical thioredoxin, TrxA, by identifying candidate protein substrates in S. aureus cells using a mechanism-based profiling assay where we trap mixed

UNLABELLED: Microorganisms are important catalysts for the oxidation of reduced inorganic sulfur compounds. One environmentally important source of reduced sulfur is metal sulfide minerals that occur in economic mineral deposits and mine waste. Previous research found that Sulfuriferula spp. were abundant and active in long-term weathering experiments with simulated waste rock and tailings from the Duluth Complex, Northern Minnesota. We, therefore, isolated several strains of Sulfuriferula spp. from these long-term experiments and characterized their metabolic and genomic properties to provide

Hydrogen sulfide is a toxic compound that can affect various groups of water microorganisms. Photolithotrophic sulfur bacteria including Chromatiaceae and Chlorobiaceae are able to convert inorganic substrate (hydrogen sulfide and carbon dioxide) into organic matter deriving energy from photosynthesis. This process takes place in the absence of molecular oxygen and is referred to as anoxygenic photosynthesis, in which exogenous electron donors are needed. These donors may be reduced sulfur compounds such as hydrogen sulfide. This paper deals with the description of this metabolic process, repr

Sulfur amendment of zerovalent iron (ZVI) materials has been shown to improve the reactivity and selectivity of ZVI toward a select group of organohalide contaminants in groundwater, most notably trichloroethene (TCE). In previous studies, chemical or mechanochemical sulfidation methods were used; however, the potential of using sulfate-reducing bacteria (SRB) to enable sulfur amendment has not been closely examined. In this study, lab-synthesized nanoscale ZVI (nZVI) and Peerless iron particles (ZVIPLS) were treated in a sulfate-reducing monoculture (D. desulfuricans) and an enrichment cultur

Microbiologically induced concrete corrosion (MICC) poses a severe challenge to the long-term durability of infrastructure, particularly in sewer networks and marine environments, which is driven by microbial metabolic activities that attack cement hydrates (Ca(OH)2, C-S-H) mainly caused by biogenic sulfuric acid (from sulfur-oxidizing bacteria) or organic acids (from fungi), converting them into expansive gypsum and ettringite, and then cause cracking and spalling. This article reviews advances in mechanisms, key microorganisms, and protection strategies of MICC to enhance our understanding o

Dinitrogen (N2) fixation provides nitrogen sources supporting active chemoautotrophic processes in submarine hydrothermal systems. Here, the diazotrophic phylogenetic diversity and N2 fixation profile were investigated in depth along a sharp biogeochemical gradient from the hydrothermal vents to ambient seawater in a shallow submarine hydrothermal system (SHS) off Kueishantao Islet, Taiwan. Diazotrophic community compositions in the SHS covered all four known nifH clusters (I, II, III, and IV) and contained both characteristics of diazotrophic communities in deep-sea hydrothermal systems and t

The extraction of nickel, cobalt, and other metals from laterite ores via bioleaching with sulfur-oxidizing and ferric iron-reducing, autotrophic, acidophilic bacteria (e.g. Acidithiobacillus species) has been demonstrated under anaerobic as well as aerobic conditions in experiments in different laboratories. This study demonstrated the bioleaching of laterites from Brazil with the addition of elemental sulfur in 2-L stirred-tank bioreactors with pure and mixed cultures of Acidithiobacillus and Sulfobacillus species under aerobic conditions. In particular, a potential disturbance of mineral di

In order to study the influence of microorganisms on the mercury biogeochemistry, the metal content and the structure of microbial communities were determined in sediments from stations along the Adour Estuary. The comparison of the bacterial communities and their distribution in function of the environmental parameters by Canonical Correspondence Analysis (CCA) revealed the influence of metals on the bacterial communities structure. Sediments where the bacterial communities are mostly influenced by methylmercury were incubated in slurries with or without mercury, under oxic and anoxic conditi

Three anaerobic, dechlorinating consortia were enriched from different sites using methanol and tetrachloroethene (PCE) and maintained for approximately 3 years. These consortia were evaluated using chemical species analysis including distribution of dechlorination products, production of organic acids and methane, and using qualitative and quantitative PCR (qPCR), terminal restriction fragment length polymorphism (TRFLP), and denaturing gradient gel electrophoresis (DGGE) with primers specific to Dehalococcoides 16S rRNA gene sequences. TRFLP and analysis of organic acids revealed differing f

Ultraviolet cured-in-place pipe (UV-CIPP) has emerged as a prevalent trenchless rehabilitation method, applicable for repairing municipal drainage pipes eroded by sulfuric acid derived from sulfate reducing bacteria (SRB) activity, as well as marine outfall pipes degraded by hydrocarbon-degrading bacteria such as Alcanivorax. To assess UV-CIPP material durability, microbial and bending stress aging tests were conducted in simulated sulfuric acid and seawater environments, and the aging mechanisms and microplastics release were analyzed through comprehensive testing. Results indicated that micr

Sulfur is an essential element in determining the productivity and quality of agricultural products. It is also an element associated with tolerance to biotic and abiotic stress in plants. In agricultural practice, sulfur has broad use in the form of sulfate fertilizers and, to a lesser extent, as sulfite biostimulants. When used in the form of bulk elemental sulfur, or micro- or nano-sulfur, applied both to the soil and to the canopy, the element undergoes a series of changes in its oxidation state, produced by various intermediaries that apparently act as biostimulants and promoters of stres

Processes leading to the bioaccumulation of methylmercury (MeHg) in northern wetlands are largely unknown. We have studied various ecological niches within a remote, acidic forested lake ecosystem in the southwestern Adirondacks, NY, to discover that mats comprised of Sphagnum moss were a hot spot for mercury (Hg) and MeHg accumulation (190.5 and 18.6 ng g⁻¹ dw, respectively). Furthermore, significantly higher potential methylation rates were measured in Sphagnum mats as compared with other sites within Sunday Lake's ecosystem. Although MPN estimates showed a low biomass of sulfate-reducing ba

Genomic modification by sulfur in the form of phosphorothioate (PT) is widespread among prokaryotes, including human pathogens. Apart from its physiological functions, PT sulfur has redox and nucleophilic properties that suggest effects on bacterial fitness in stressful environments. Here we show that PTs are dynamic and labile DNA modifications that cause genomic instability during oxidative stress. In experiments involving isotopic labeling coupled with mass spectrometry, we observed sulfur replacement in PTs at a rate of ∼2% h-1 in unstressed Escherichia coli and Salmonella enterica. Wherea

A biosensor based on sulfur-oxidizing bacteria (SOB) for detection of toxic chemicals in water was developed. SOB are acidophilic microorganisms that get their energy through the oxidation of reduced sulfur compounds in the presence of oxygen to produce sulfuric acid. The reaction results in an increase in electrical conductivity (EC) and a decrease in pH. The bioassay is based on the inhibition of SOB in the presence of toxic chemicals by measuring changes in EC and pH. The effect of different physical factors such as HRT, inorganic sulfur (S°) particle size, and temperature on detection of C

Oral malodor, also known as bad breath or halitosis, is an extremely common problem. Bad breath can arise from many sources in the body, but most frequently is produced in the mouth by the action of gram-negative anaerobic bacteria on sulfur-containing proteinaceous substrates in the saliva, such as debris and plaque. The primary molecules responsible for oral malodor are volatile sulfur compounds (VSC), such as hydrogen sulfide and methylmercaptan. Increased malodor production is related to greater bacterial numbers, reducing conditions, availability of protein substrates, and a pH above neut

Hydrogen sulfide (H2S) production from waste activated sludge (WAS) is the main reason for odor emission during anaerobic fermentation system. CaO has been reported to effectively improve the resources recovery of WAS, but its potential effect on H2S production in anaerobic fermentation process remains unrecognized. In present study, it was found that the addition of 60 mg/g VSS CaO greatly inhibited H2S production and the maximum yield of H2S was 60.1 ± 1.8% lower than the control. Mechanism investigation demonstrated that CaO destroyed sludge structure and increased the release of intracellu

This study investigated the feasibility of co-treating H2S and CO2 in a biological trickling filter (BTF) inoculated with hydrogenotrophic methanogens (HMs) and nitrate-reducing, sulfur-oxidizing bacteria. This was accomplished by introducing a pure culture of Thiobacillus denitrificans in a BTF that was successfully upgrading a biogas mimic (60:40 CH4:CO2) to >97% methane using an enriched HM consortium. Nitrate was fed as the electron acceptor to oxidize H2S. The results revealed that a severe competition for hydrogen's electrons occurred between carbon dioxide and nitrate. Due to this compe

Microbial mercury (Hg) methylation transforms inorganic mercury to neurotoxic methylmercury (MeHg) mainly in aquatic anoxic environments. Sampling challenges in marine ecosystems, particularly in submarine canyons, leads to a lack of knowledge about the Hg methylating microbia in marine sediments. A previous study showed an enrichment of mercury species in sediments from the Capbreton Canyon where both geochemical parameters and microbial activities constrained the net MeHg production. In order to characterize Hg-methylating microbial communities from coastal to deeper sediments, we analysed t
