Active Zone Scaffold Protein Ratios Tune Functional Diversity across Brain Synapses.

High-throughput electron microscopy has started to reveal synaptic connectivity maps of single circuits and whole brain regions, for example, in the Drosophila olfactory system. However, efficacy, timing, and frequency tuning of synaptic vesicle release are also highly diversified across brain synapses. These features critically depend on the nanometer-scale coupling distance between voltage-gated Ca2+ channels (VGCCs) and the synaptic vesicle release machinery. Combining light super resolution microscopy with in vivo electrophysiology, we show here that two orthogonal scaffold proteins (ELKS

Changes in Properties of Auditory Nerve Synapses following Conductive Hearing Loss.

UNLABELLED: Auditory activity plays an important role in the development of the auditory system. Decreased activity can result from conductive hearing loss (CHL) associated with otitis media, which may lead to long-term perceptual deficits. The effects of CHL have been mainly studied at later stages of the auditory pathway, but early stages remain less examined. However, changes in early stages could be important because they would affect how information about sounds is conveyed to higher-order areas for further processing and localization. We examined the effects of CHL at auditory nerve syna

VGLUT1 synapses and P-boutons on regenerating motoneurons after nerve crush.

Stretch-sensitive Ia afferent monosynaptic connections with motoneurons form the stretch reflex circuit. After nerve transection, Ia afferent synapses and stretch reflexes are permanently lost, even after regeneration and reinnervation of muscle by motor and sensory afferents is completed in the periphery. This loss greatly affects full recovery of motor function. However, after nerve crush, reflex muscle forces during stretch do recover after muscle reinnervation and reportedly exceed 140% baseline values. This difference might be explained by structural preservation after crush of Ia afferen

Opioids potentiate electrical transmission at mixed synapses on the Mauthner cell.

Opioid receptors were shown to modulate a variety of cellular processes in the vertebrate central nervous system, including synaptic transmission. While the effects of opioid receptors on chemically mediated transmission have been extensively investigated, little is known of their actions on gap junction-mediated electrical synapses. Here we report that pharmacological activation of mu-opioid receptors led to a long-term enhancement of electrical (and glutamatergic) transmission at identifiable mixed synapses on the goldfish Mauthner cells. The effect also required activation of both dopamine

Differential effects of GABA in modulating nociceptive vs. non-nociceptive synapses.

GABA (γ-amino-butyric acid) -mediated signaling is normally associated with synaptic inhibition due to ionotropic GABA receptors that gate an inward Cl(-) current, hyperpolarizing the membrane potential. However, there are also situations where ionotropic GABA receptors trigger a Cl(-) efflux that results in depolarization. The well-characterized central nervous system of the medicinal leech was used to study the functional significance of opposing effects of GABA at the synaptic circuit level. Specifically, we focused on synapses made by the nociceptive N cell and the non-nociceptive P (press

Stress-induced priming of glutamate synapses unmasks associative short-term plasticity.

Exposure to a stressor sensitizes or 'primes' the hypothalamic-pituitary-adrenal axis to a subsequent novel stressor. The synaptic mechanisms underlying this priming, however, are not known. We found that exposing a rat to a single stressor primed glutamate synapses in the paraventricular nucleus of the hypothalamus and allowed them to undergo a short-term potentiation (STP) following a burst of high-frequency afferent activity. This transient potentiation requires a corticotrophin-releasing hormone-dependent depression of postsynaptic NMDA receptors (NMDARs). The long-term depression of NMDAR

Dynamically reconfigurable silicon array of spiking neurons with conductance-based synapses.

A mixed-signal very large scale integration (VLSI) chip for large scale emulation of spiking neural networks is presented. The chip contains 2400 silicon neurons with fully programmable and reconfigurable synaptic connectivity. Each neuron implements a discrete-time model of a single-compartment cell. The model allows for analog membrane dynamics and an arbitrary number of synaptic connections, each with tunable conductance and reversal potential. The array of silicon neurons functions as an address-event (AE) transceiver, with incoming and outgoing spikes communicated over an asynchronous eve

Pharmacologically distinct presynaptic calcium channels in cerebellar excitatory and inhibitory synapses.

We have used whole-cell patch clamp recordings and pharmacological blockers of Ca channels to compare the pharmacology of Ca channels that mediate synaptic transmission at the three types of synapses innervating Purkinje cells in rat cerebellar slices. Both parallel fiber and climbing fiber excitatory synapses were sensitive to the P-type Ca channel blocker, omega-AgaIVA and the P/Q/N-type channel blocker, omega-conotoxin MVIIC. Transmission at inhibitory interneuronal synapses was not suppressed by these toxins, or by the N-type (omega-conotoxins GVIA and MVIIA) or L-type (nimodipine) channel

MINFLUX nanoscopy to study the NK cell immune synapse.

Antibody dependent cellular cytotoxicity (ADCC) is an effector function performed by natural killer (NK) cells to target and clear viral infections and cancer. ADCC is a critical feature of several antibody and cellular therapeutics as well as vaccination strategies. Using microscopy to understand the details of molecular events driving ADCC is essential to improving such therapeutics but has been limited by technologies that cannot practically provide the spatial resolution necessary to study protein function at the single molecule level in cells. In this chapter, we describe a model system u

Topological acoustic synapse for high-dimensional neuromorphic computing.

The human brain performs complex, high-dimensional (HD) computations, such as causal reasoning, counterfactual thinking, and abstraction, with ~1011 neurons while consuming ~20 watts of power. Neuromorphic computing seeks similar efficiency, but current devices face bottlenecks in bandwidth, energy, wiring, footprint, and reliability that limit scalability. Here, we introduce the topological acoustic synapse (TAS), an acoustic-wave neuromorphic device that circumvents these limits by mapping information in multivariate state spaces. A single TAS generates and manipulates numerous computing cha

WONOEP appraisal: Biomarkers and treatment strategies beyond the synapse.

Epilepsy is a heterogeneous neurological disorder affecting more than 70 million people worldwide, posing significant challenges for clinicians due to its complex etiology, diverse manifestations, variable treatment responses, and the inability to predict seizures or disease onset reliably. Despite advances in antiseizure medications, approximately 30% of patients remain treatment-resistant, highlighting the urgent need for therapies with antiepileptogenic or disease-modifying effects. To optimize and individualize strategies to predict and treat seizures and epilepsy, efforts to identify biom

The Impact of Zinc on T Cell Motility and the Immunological Synapse.

Zinc is an essential trace element with a critical role in regulating immune functions. Patients with autoimmune diseases or chronic lymphatic leukemia often exhibit lower serum zinc levels. As T cells are key mediators of adaptive immunity, disturbances in zinc homeostasis can strongly affect their function. Effective T cell activity depends on directed migration to inflamed tissues, requiring coordinated cytoskeletal reorganization. This process involves the formation of a leading edge and a trailing edge (uropod) and is regulated by the ezrin-radixin-moesin (ERM) complex and its interaction

The role of Grn in the development of mouse cochlear nucleus synapses.

Progranulin (encoded by Grn) is a multifunctional glycoprotein implicated in neuronal survival and synapse function, yet its role in central auditory circuit development remains unclear. In this study, we investigated the role of Grn in mouse cochlear nucleus (CN) synapse development. Grn-/- and Grn+/+ mice were used in this study. Grn (Progranulin) expression was examined by reverse transcription PCR and immunofluorescence. Neuronal density and synaptic puncta expression within the CN were analyzed using immunofluorescence labeling and quantitative image analysis. Auditory function was assess

Dysregulation of complement at the synapse in P301S mice and human tauopathies.

Synapse loss is the best correlate of cognitive decline in neurodegenerative diseases (NDDs). In Alzheimer's disease complement dysregulation, triggered by amyloid-β accumulation, plays a major role in synapse loss, but its contribution in other NDDs where amyloid is absent, most notably tauopathies, is elusive. Aggregation of tau is a prominent co-pathology in many NDDs and is characteristic of classical tauopathies in which amyloid pathology is lacking. Here we explore the effect of tau accumulation on complement dysregulation and its contribution to neuronal damage and synapse loss in a mou

Astrocytic synapse engulfment is differentially controlled by APOE genotype.

APOE gene variants encoding the apolipoprotein E (ApoE) protein are strong genetic modifiers of risk of Alzheimer's disease (AD) with the APOE ε4 allele (APOE4) associated with substantially increased disease risk, APOE ε2 allele (APOE2) associated with decreased risk and APOE ε3 allele (APOE3) considered neutral. Recently the Christchurch variant of APOE3 (APOE3CH) has been shown to protect people from familial AD. Despite this strong evidence for APOE mediating AD risk, the exact biological mechanisms through which APOE influences pathogenesis remain unknown. Our previous work implicates APO

Astrocyte-Synapse Structural Plasticity in Neurodegenerative and Neuropsychiatric Diseases.

Synaptic dysfunction is a common feature across a broad spectrum of brain diseases, spanning from psychopathologies such as posttraumatic stress disorder and substance use disorders to neurodegenerative diseases such as Alzheimer's disease and Parkinson's disease. While neuroscience research aiming to understand the mechanisms underlying synaptic dysfunction has traditionally focused on the neuronal elements of the synapse, recent research increasingly acknowledges the contribution of astrocytes as a third element controlling synaptic transmission. This also sparked interest to investigate the

Glial phagocytosis for synapse and toxic proteins in neurodegenerative diseases.

Glia, as resident immune and supportive cells of the central nervous system, play a critical role in maintaining brain homeostasis. One of their key homeostatic functions is phagocytic capacity in pruning synapses and removing cellular debris/protein aggregates, a process vital for synaptic plasticity and brain maintenance. However, these phagocytic functions are often dysregulated with aging and in neurodegenerative diseases (NDs), such as Alzheimer's disease, Parkinson's disease, Huntington's disease, amyotrophic lateral sclerosis, and frontotemporal dementia. This review aims to examine the

Passing the torch: The ascendance of the glutamatergic synapse in the pathophysiology of schizophrenia.

For nearly fifty years, the dopamine hypothesis has dominated our understanding of the pathophysiology of schizophrenia and provided the lone target for drug development. However, with the exception of clozapine, the dopamine D2 receptor antagonizing anti-psychotic drugs have little impact on the negative symptoms and cognitive deficits, aspects of the disorder that robustly predict outcome. Pathologic studies reveal cortical atrophy and wide-spread loss of glutamatergic synaptic spines, unexplained by dopaminergic malfunction. Recent genome-wide association studies indicate that at least thir
