THE ROLE OF TRANSIENT RECEPTOR POTENTIAL (TRPA1) CHANNEL IN PRURITUS.

Pruritus or itch is defined as the sensation that causes the desire to scratch, and it can be induced by mechanical, thermal and chemical stimuli. Persistent itch accompanying diseases of the skin and other organs can significantly impair the quality of life. There is a growing body of evidence implicating abnormal transient receptor potential (TRP) channel function, as a product of excessive or deficient channel activity, in pathological skin conditions such as pruritus and dermatitis. These data supports the notion that non-histaminergic itch mediators require the activation of TRPA1 channel

TRPA1s act as chemosensors but not as cold sensors or mechanosensors to trigger the swallowing reflex in rats.

We examined the role of TRPA1s in triggering the swallowing reflex. TRPA1s predominantly localized on thin nerve fibers and fibroblast-like cells in swallowing-related regions and on small to medium-sized superior laryngeal nerve-afferents in the nodose-petrosal-jugular ganglionic complex. Topical application of a TRPA1 agonist, allyl isothiocyanate (AITC), dose-dependently triggered swallowing reflexes. Prior topical application of a TRPA1 antagonist significantly attenuated the AITC-induced reflexes. Application of cold AITC (4 °C) very briefly reduced the on-site temperature to < 17 °C (tem

Systematic and quantitative mRNA expression analysis of TRP channel genes at the single trigeminal and dorsal root ganglion level in mouse.

BACKGROUND: Somatosensory nerve fibres arising from cell bodies within the trigeminal ganglia (TG) in the head and from a string of dorsal root ganglia (DRG) located lateral to the spinal cord convey endogenous and environmental stimuli to the central nervous system. Although several members of the transient receptor potential (TRP) superfamily of cation channels have been implicated in somatosensation, the expression levels of TRP channel genes in the individual sensory ganglia have never been systematically studied. RESULTS: Here, we used quantitative real-time PCR to analyse and compare mR

Recent developments in vascular endothelial cell transient receptor potential channels.

Among the 28 identified and unique mammalian TRP (transient receptor potential) channel isoforms, at least 19 are expressed in vascular endothelial cells. These channels appear to participate in a diverse range of vascular functions, including control of vascular tone, regulation of vascular permeability, mechanosensing, secretion, angiogenesis, endothelial cell proliferation, and endothelial cell apoptosis and death. Malfunction of these channels may result in disorders of the human cardiovascular system. All TRP channels, except for TRPM4 and TRPM5, are cation channels that allow Ca2+ influx

TRPA1 in Chemotherapy-Induced Peripheral Neuropathy: A Promising Target for Immunomodulation and Anti-Inflammatory Therapy.

Chemotherapy-induced peripheral neuropathy (CIPN) is a common and serious clinical condition that results when a chemotherapeutic agent damages the peripheral nervous system and is usually characterized by abnormal pain in the distal limbs and increased sensitivity to hot and cold stimuli. However, few clinical treatments for CIPN are available, so it is critical to identify effective treatment modalities. Transient receptor potential ankyrin 1 (TRPA1), an injury receptor, can be activated by oxidative stress, inflammatory mediators, and hypothermic conditions during the pathogenesis and progr

H2O2 enhances the spontaneous phasic contractions of isolated human-bladder strips via activation of TRPA1 channels on sensory nerves and the release of substance P and PGE2.

Several studies have indicated that reactive oxygen species (ROS) can lead to detrusor overactivity (DO), but the underlying mechanisms are not known. Hydrogen dioxide (H2O2) is used commonly to investigate the effects of ROS. In present study, we investigated the effects of H2O2 on phasic spontaneous bladder contractions (SBCs) of isolated human-bladder strips (iHBSs) and the underlying mechanisms. Samples of bladder tissue were obtained from 26 patients undergoing cystectomy owing to bladder cancer. SBCs of iHBSs were recorded in organ-bath experiments. H2O2 (1μM-10mM) concentration-dependen

Expression of the transient receptor potential channels TRPV1, TRPA1 and TRPM8 in mouse trigeminal primary afferent neurons innervating the dura.

BACKGROUND: Migraine and other headache disorders affect a large percentage of the population and cause debilitating pain. Activation and sensitization of the trigeminal primary afferent neurons innervating the dura and cerebral vessels is a crucial step in the "headache circuit". Many dural afferent neurons respond to algesic and inflammatory agents. Given the clear role of the transient receptor potential (TRP) family of channels in both sensing chemical stimulants and mediating inflammatory pain, we investigated the expression of TRP channels in dural afferent neurons. METHODS: We used two

Nuclear paraspeckle assembly transcript 1 promotes photophobia behavior in mice via miR-196a-5p/Trpm3 coupling.

BACKGROUND: The long noncoding RNA, NEAT1, is recognized as a key regulator of proinflammatory gene expression; Yet, its functional role in migraine remains unexplored, despite the central role of neuroinflammatory mechanisms in migraine pathophysiology. This study examines the implication of NEAT1 in the trigeminal ganglion activation, which underlies photophobia associated with migraine. METHODS: Light aversion behavior was induced by intranasal injection of the TRPA1 activator, umbellulone. Male mouse behavior was assessed by the total time the mouse stays in the light between the dark and

Role of TRPV1+ and TRPA1+ nociceptive neurons in delayed-onset muscle soreness: inhibition by hesperidin methyl chalcone.

OBJECTIVE: Delayed-onset muscle soreness (DOMS) is a type of pain caused by muscle injury provoked by eccentric, high intensity, or long-duration exercise. Hesperidin methyl chalcone (HMC) is a flavonoid with analgesic and anti-inflammatory actions. We investigated the effects of HMC against DOMS. METHODS: Mice received AMG9810 (100 nmol) or HC-030031 (10 μg) once intrathecally, or HMC twice (12 h plus 30 min before) intraperitoneally (1, 3, or 10 mg/kg) and were subjected to a single uninterrupted acute swimming session of 120 min to induce DOMS. Sham animals were subjected to swimming just

Cannabidiol activation of vagal afferent neurons requires TRPA1.

Vagal afferent neurons abundantly express excitatory transient receptor potential (TRP) channels, which strongly influence afferent signaling. Cannabinoids have been identified as direct agonists of TRP channels, including TRPA1 and TRPV1, suggesting that exogenous cannabinoids may influence vagal signaling via TRP channel activation. The diverse therapeutic effects of electrical vagus nerve stimulation also result from administration of the nonpsychotropic cannabinoid, cannabidiol (CBD); however, the direct effects of CBD on vagal afferent signaling remain unknown. We investigated actions of

Roles of TRPV1 and TRPA1 in Spontaneous Pain from Inflamed Masseter Muscle.

Craniofacial muscle pain, such as spontaneous pain and bite-evoked pain, are major symptoms in patients with temporomandibular disorders and infection. However, the underlying mechanisms of muscle pain, especially mechanisms of highly prevalent spontaneous pain, are poorly understood. Recently, we reported that transient receptor potential vanilloid 1 (TRPV1) contributes to spontaneous pain but only marginally contributes to bite-evoked pain during masseter inflammation. Here, we investigated the role of transient receptor potential ankyrin 1 (TRPA1) in spontaneous and bite-evoked pain during

Nociceptor neurons control pollution-mediated neutrophilic asthma.

The immune and sensory nervous systems, having evolved in parallel, communicate through shared receptors and transmitters to maintain homeostasis and respond to both external and internal disruptions. Although neural responses often confer protective benefits, they can also exacerbate inflammation during allergic reactions such as asthma. In our study, we modeled pollution-exacerbated asthma by exposing mice to ambient PM2.5 particles alongside ovalbumin. Compared to exposure to ovalbumin alone, this co-exposure significantly increased the numbers of neutrophils and γδ T cells in bronchoalveol

Transient receptor potential channels TRPA1 and TRPV1 are involved in chronic pain relief via Pulsed Radiofrequency in Rheumatoid Arthritis rat.

The relevance of transient receptor potential (TRP) channels ankyrin 1 (TRPA1, a sensor of noxious cold and mechanical stimuli) and vanilloid 1 (TRPV1, a nociceptor and heat sensor) in the pathogenesis of chronic pain associated with rheumatoid arthritis (RA) is profound. Multiple clinical studies have shown that pulsed radiofrequency (PRF) is effective in alleviating pain. However, the precise mechanism through which PRF alleviates pain remains unclear. In this study, we employed a collagen-induced arthritis (CIA) rat model to investigate the effects of intra-articular PRF or dorsal root gang

Transduction Mechanisms for Cold Temperature in Mouse Trigeminal and Vagal Ganglion Neurons Innervating Different Peripheral Organs.

AIM: To elucidate the molecular mechanism of cold sensing by visceral sensory endings, a side-by-side characterization of cold-sensitive (CS) neurons in adult mouse trigeminal (TG) and vagal ganglia (VG) was performed. METHODS: A combination of physiological, pharmacological, molecular, and genetic tools was employed on trigeminal and vagal neurons. RESULTS: CS neurons are more abundant in VG, and the majority co-express TRPA1. Cold-evoked responses are severely blunted in Trpa1 KO mice. In contrast, TRPM8 deletion or pharmacological TRPM8 blockade had little impact on VG cold sensitivity. I

FGF13 enhances the function of TRPV1 by stabilizing microtubules and regulates acute and chronic itch.

Itching is an aversive somatosensation that triggers the desire to scratch. Transient receptor potential (TRP) channel proteins are key players in acute and chronic itch. However, whether the modulatory effect of fibroblast growth factor 13 (FGF13) on acute and chronic itch is associated with TRP channel proteins is unclear. Here, we demonstrated that conditional knockout of Fgf13 in dorsal root ganglion neurons induced significant impairment in scratching behaviors in response to acute histamine-dependent and chronic dry skin itch models. Furthermore, FGF13 selectively regulated the function

Stephalagine, an aporphine alkaloid from Annona crassiflora fruit peel, induces antinociceptive effects by TRPA1 and TRPV1 channels modulation in mice.

Pain relief represents a critical unresolved medical need. Consequently, the search for new analgesic agents is intensively studied. Annona crassiflora, a native species of the Brazilian Savanna, represents a potential source for painful treatment. This study aimed to investigate the antinociceptive potential of A. crassiflora fruit peel, focusing on its major alkaloid, stephalagine, in animal models of pain evoked by the activation of transient receptor potential ankyrin 1 (TRPA1) and vanilloid 1 (TRPV1) channels. Male C57BL/6/J mice were submitted to formalin-, cinnamaldehyde-, and capsaicin

Exposure to formaldehyde at low temperatures aggravates allergic asthma involved in transient receptor potential ion channel.

Studies have indicated that formaldehyde and low temperature are considered to be the factors associated with several respiratory diseases. However, the effect of co-exposure to formaldehyde and low temperature on allergic asthma, and the potential mechanisms, are unknown. In this study, an allergic asthma mouse model was built and mice were exposed to 0.8 mg/m3 formaldehyde and/or subjected to low temperatures of 16 °C. The data showed that exposure to either low temperature or formaldehyde did not induce a significant aggravation on allergic asthma. However, simultaneous exposure to formalde

A TRP channel trio mediates acute noxious heat sensing.

Acute pain represents a crucial alarm signal to protect us from injury. Whereas the nociceptive neurons that convey pain signals were described more than a century ago, the molecular sensors that detect noxious thermal or mechanical insults have yet to be fully identified. Here we show that acute noxious heat sensing in mice depends on a triad of transient receptor potential (TRP) ion channels: TRPM3, TRPV1, and TRPA1. We found that robust somatosensory heat responsiveness at the cellular and behavioural levels is observed only if at least one of these TRP channels is functional. However, comb

Role of transient receptor potential channels on pathogenesis and treatment of psoriasis.

Psoriasis is a chronic inflammatory skin disease characterized primarily by hyperproliferation of keratinocytes, infiltration and activation of immune cells, including T lymphocytes and macrophages, as well as increased innervation by sensory neurons. Although several therapeutic options are available, the management of psoriasis remains unsatisfactory, with adverse effects and unmet clinical needs. In this context, channels from the Transient Receptor Potential (TRP) family, which are non-selective cation channels involved in various pathologies, have been identified as potential therapeutic
