Glioblastomas have been historically difficult to treat with poor long-term survival. With novel strategies focused on targeting hypoxia-inducible factor (HIF) regulatory pathways, recent evidence has shown that Acriflavine (ACF) can effectively target glioma invasiveness and recurrence. However, local delivery of ACF and its combinatory effects with Temozolomide (TMZ) and radiation therapy (XRT) have not yet been optimized. In this study we test a novel polymeric matrix that can gradually release ACF at the tumor bed site in combination with systemic TMZ and XRT. In vitro cytotoxicity assays

PURPOSE: Glioma, especially glioblastoma (GBM), is the most aggressive malignant brain tumor and its standard therapy is often ineffective because of temozolomide (TMZ) resistance. Reversal of the TMZ resistance might improve the prognosis of glioma patients. We previously found that interferon-α (IFN-α) and anti-epileptic drug levetiracetam (LEV) could sensitize glioma to TMZ, respectively. In this study, we further investigated the efficiency of combining of LEV and IFN-α for improving the efficacy of TMZ.
METHODS: We evaluated whether LEV and IFN-α could increase TMZ efficacy using colony

INTRODUCTION: Diffuse intrinsic pontine glioma (DIPG) is the most common form of brainstem glioma. The present study was performed to assess if hypofractionated radiotherapy completed in < 3 weeks with temozolomide improves survival in DIPG.
MATERIAL AND METHODS: The present study is a phase II open label randomized trial. The study included newly diagnosed patients with DIPG. Patients in arm A received conventional fractionated RT of 60 Gy in 30 fractions over 6 weeks while patients in arm B received hypo-fractionated radiotherapy of 39 Gy in 13 fractions over 2.6 weeks along with concurrent

The blood-brain barrier (BBB) is major obstacle in drug or stem cell treatment in chronic stroke. We hypothesized that adding mannitol to temozolomide (TMZ) is a practically applicable method for resolving the low efficacy of intravenous mannitol therapy. In this study, we investigated whether BBB permeability could be increased by this combined treatment. First, we established a chronic ischemic stroke rat model and examined changes in leakage of Evans blue dye within a lesion site, and in expression of tight junction proteins (TJPs), by this combined treatment. Additionally, in an in vitro B

Chloride (Cl-), a primary anion in the extracellular fluid, plays an important role in a variety of physiological and pathological processes, such as cell apoptosis and proliferation. However, the information about Cl- in cancer cell apoptosis and chemoresistance is poorly understood. In the present study, we found that temozolomide (TMZ) treatment led to a decrease in intracellular concentration of Cl- ([Cl-]i) in both U87 and TMZ-resistant U87/R glioma cells. The decrease in [Cl-]i was more noticeable in U87 cells than in U87/R cells. Moreover, the expression of LRRC8A was reduced in U87/R c

Glioma is one of the most common primary tumors of the central nervous system in adults. Glioblastoma (GBM) is the most lethal type of glioma, whose 5-year survival is 9.8% at best. Glioma stem-like cells (GSCs) play an important role in recurrence and treatment resistance. MGMT is a DNA repair protein that removes DNA adducts and therefore attenuates treatment efficiency. It has been reported that interferon-α/β (IFN-α/β) downregulates the level of MGMT and sensitizes glioma cells to temozolomide. In the present study, we assessed whether IFN-α/β is able to sensitize GSCs to temozolomide by m

PURPOSE: Primary malignant gliomas of the optic nerves are rare tumors of adulthood, progressing rapidly to blindness and to death within several months, regardless of the type of treatment. Recently, treatments associating radiotherapy and temozolomide have been used in other types of glioblastomas, but their impact on optic nerve malignant gliomas is not known.
METHODS: This was a retrospective case series of 2 patients diagnosed with primary optic nerve and chiasm glioblastoma (GBM), treated with radiotherapy and concomitant temozolomide.
RESULTS: A 74-year-old man presented with visual l

We report here a rare case of a young male patient presenting with a Multiple Endocrine Neoplasia Type 1 - prolactin-secreting pituitary carcinoma, controlled long-term after temozolomide withdrawal. Initial presentation was pituitary apoplexy leading to surgery. Dopamine agonists and radiotherapy allowed control of prolactin secretion and pituitary remnant. Metastasis appeared 10 years after initial presentation, leading to the diagnosis of pituitary carcinoma. At that time, dopamine agonists were no more effective; temozolomide, an oral alkylating agent, was administered for 24 cycles, and a

OBJECT: Bioluminescence imaging (BLI) offers a rapid and accurate means for longitudinal study of tumor cell growth and response to therapy in rodent models. Because this technology has only recently come into use in the field of small animal imaging, applications in this area have been limited. In the current study we have applied BLI to the analysis of clinically relevant issues involving use of the DNA methylating agent temozolomide (TMZ) in a mouse model.
METHODS: An invasive glioblastoma multiforme xenograft was modified for BLI via transduction with a luciferase-encoding lentivirus. Sup

Between July 2003 and March 2004, nine patients with pretreated mycosis fungoides were enrolled into a phase II trial and were treated with temozolomide. The overall response rate was 33%. Further studies are needed to test the efficacy of temozolomide, alone or in combination in earlier stages of this disease.

BACKGROUND: Temozolomide resistance is a major barrier to effective glioma treatment and is frequently associated with enhanced DNA damage repair capacity. Although DPP9 has been linked to tumor development in several malignancies, whether it contributes to glioma therapeutic resistance has not been explored.
METHODS: Transcriptomic datasets, glioma specimens, and genetic gain- and loss-of-function approaches were employed to characterize DPP9 expression and function. RNA sequencing, co-immunoprecipitation, subcellular fractionation, and ubiquitination analyses were conducted to define the mo

Glioblastoma (GBM) represents the most aggressive primary malignant tumor of the adult central nervous system, with an extremely poor clinical prognosis. As the first-line chemotherapeutic agent for GBM, temozolomide (TMZ) has encountered a critical therapeutic bottleneck due to the development of drug resistance in tumor cells. GBM pathogenesis is jointly driven by multiple factors, including genetics, epigenetics, and the tumor microenvironment. O6-methylguanine-DNA methyltransferase (MGMT) -driven repair of DNA alkylation lesions is the central molecular mechanism underlying TMZ resistance

UNLABELLED: Glioblastoma multiforme (GBM) is a highly aggressive brain tumor with a poor prognosis. Temozolomide (TMZ) is the most widely used chemotherapeutic agent and can significantly improve patient survival rates. However, numerous patients develop TMZ resistance, leading to limited therapeutic benefits. Therefore, it is crucial to investigate the mechanisms of TMZ resistance in patients with GBM and identify the sensitizing targets of TMZ to improve its clinical efficacy. In this study, we demonstrated that acylphosphatase 2 (ACYP2) was involved in regulating the sensitivity of GBM to T

Glioblastoma is an aggressive primary brain tumor characterized by high recurrence rates and resistance to standard therapies, including temozolomide (TMZ). Emerging evidence suggests that microRNAs (miRNAs) play a critical role in regulating treatment response and tumor progression. In this study, we investigated the effects of TMZ and the JAK inhibitor ruxolitinib, alone and in combination, on miRNA expression profiles in glioblastoma cancer stem cells. miRNA expression levels were analyzed using quantitative real-time PCR, and differential expression was evaluated using the 2-ΔΔCt method. Temozolomide (TMZ) resistance in glioblastoma (GBM) is often linked to high MGMT protein levels, but MGMT-deficient tumors can still acquire resistance, implicating MGMT-independent repair pathways. Here, we identify an oxidative stress-responsive LMO7-ALKBH2 ubiquitination mechanism that regulates ALKBH2 stability and modulates alkylation damage tolerance in GBM. The E3 ligase LMO7 directly binds ALKBH2 and promotes K48-linked polyubiquitination and proteasomal degradation of ALKBH2, with Lys7 serving as a major ubiquitin acceptor site. Oxidative stress enhances LMO7-dependent ALKBH2 ubiquiti

Glioblastoma (GBM) is the most prevalent tumor in the central nervous system in adults. Lactotransferrin (LTF) is a molecule involved in the growth of various tumors. However, the underlying mechanism of LTF in GBM progression and chemotherapy resistance remains unclear. In this study, the clinical and diagnostic value of LTF were evaluated. In vitro and in vivo experiments were performed to explore the functional role of LTF in GBM. Immunoprecipitation and immunofluorescence assays were performed to clarify the effect of LTF on nuclear factor-κB (NF-κB) activation. LTF was overexpressed in GB

Glioblastoma (GBM) is the most aggressive and deadliest type of primary brain tumor, treated with temozolomide (TMZ) as first-line chemotherapy. However, temozolomide resistance remains a critical therapeutic hurdle in GBM, often resulting in treatment failure and tumor recurrence. Here, we aimed to identify an epigenetic target to overcome TMZ resistance in GBM. We established TMZ-resistant GBM cell lines, which exhibited increased expression of resistance markers such as E2F6, ABCG2, and phosphorylated STAT3, and decreased Bax expression. Through KDM inhibitor screening with these cells, we
