The utility of Ocimum basilicum, Perilla frutescens, and Mentha spicata essential oils to control larvae and engorged female Rhipicephalus sanguineus ticks on dogs.

The study evaluated the larvicidal and reproductive effects of essential oils from Ocimum basilicum, Perilla frutescens, and Mentha spicata on engorged female Rhipicephalus sanguineus ticks. Adult immersion and larval packet assays were used to evaluate their larvicidal efficacy and impact on tick reproductive abilities. For tick larvae, Perilla frutescens essential oil had the highest 50% lethal dose of 1.89 mg/mL, followed by Ocimum basilicum at 1.58 mg/mL, and Mentha spicata at 1.07 mg/mL. Regarding adult ticks, although Mentha spicata essential oil had the lowest reduced oviposition value,

A novel eukaryotic RdRP-dependent small RNA pathway represses antiviral immunity by controlling an ERK pathway component in the black-legged tick.

Small regulatory RNAs (sRNAs) are involved in antiviral defense and gene regulation. Although roles of RNA-dependent RNA Polymerases (RdRPs) in sRNA biology are extensively studied in nematodes, plants and fungi, understanding of RdRP homologs in other animals is still lacking. Here, we study sRNAs in the ISE6 cell line, which is derived from the black-legged tick, an important vector of human and animal pathogens. We find abundant classes of ~22nt sRNAs that require specific combinations of RdRPs and sRNA effector proteins (Argonautes or AGOs). RdRP1-dependent sRNAs possess 5'-monophosphates

The anal pore route is efficient to infect Amblyomma spp. ticks with Rickettsia rickettsii and allows the assessment of the role played by infection control targets.

Adult Amblyomma sculptum and Amblyomma aureolatum ticks are partially refractory to Rickettsia rickettsii when fed on infected hosts, hindering the functional characterization of potentially protective targets in the bacterial acquisition. In the current study, we used the anal pore route to infect adult A. sculptum and A. aureolatum ticks with R. rickettsii and to assess the effects of the knockdown of microplusin in infection control. The anal pore route was efficient to infect both species, resulting in a prevalence of around 100% of infected ticks. Higher loads of R. rickettsii were detect

Evaluation of the SELECT Tick Control System (TCS), a Host-Targeted Bait Box, to Reduce Exposure to Ixodes scapularis (Acari: Ixodidae) in a Lyme Disease Endemic Area of New Jersey.

We describe a 2-yr trial to evaluate the ability of SELECT Tick Control System (TCS) host-targeted bait boxes to reduce numbers of host-seeking Ixodes scapularis nymphs in a residential neighborhood. After four successive 9-wk deployments, nymphal and larval I. scapularis infestation prevalence and intensity were significantly reduced on target small mammals. In addition, these deployments resulted in 87.9% and 97.3% control of host-seeking nymphs in treatment sites at 1 yr and 2 yr postintervention, respectively. Installation of a protective metal cover around the SELECT TCS bait boxes elimin

Efficacy of amitraz applied to white-tailed deer by the '4-poster' topical treatment device in controlling free-living lone star ticks (Acari: Ixodidae).

White-tailed deer treated themselves with a commercial pour-on acaricide formulation containing 2% amitraz as they fed from an ARS-patented '4-poster' topical treatment device. Whole kernel corn attracted deer to a single device placed in each of two deer-fenced pastures. In the treatment pasture, the rollers of the treatment device were charged with the acaricide, whereas the rollers of the device in the other pasture remained untreated. Deer were allowed to use the '4-posters' during periods of tick activity beginning in early to midspring and lasting through late summer to early fall for th

Control of Hyalomma lusitanicum (Acari: Ixodidade) Ticks Infesting Oryctolagus cuniculus (Lagomorpha: Leporidae) Using the Entomopathogenic Fungus Beauveria bassiana (Hyocreales: Clavicipitaceae) in Field Conditions.

Entomopathogenic fungi are widely used to control arthropods not just in agricultural settings but also in Veterinary Medicine and Public Health. These products have been employed to control tick populations and tick-borne diseases. The effectiveness of these control measures not only depends on the fungi, but also on the tick species and environmental conditions. In Mesomediterranean areas, tick species are adapted to extreme climatic conditions and it is therefore especially important to develop suitable tick control strategies. The aim of this study was to evaluate the effectiveness of a ne

Genetic characterization of Rhipicephalus microplus (Acari: Ixodidae) in Africa reveals the presence of diverse haplotypes within a single clade: Implications for pathogens transmission and tick control in a predominantly pastoral production system.

The Asian blue tick, Rhipicephalus (Boophilus) microplus (Canestrini, 1888) is renowned as a notorious pest of veterinary importance globally. Its recent introduction to West Africa and subsequent eastward invasion and displacement of native tick fauna in many African countries is alarming, considering the prevalent unrestricted livestock migration in these regions. This study determined the genetic diversity of R. microplus in Africa based on the mitochondrial cytochrome oxidase subunit one (cox1) gene sequences in comparison with global sequences. Phylogenetic and genetic diversity analyses

Combination of essential oils from Ocimum americanum L., Ocimum gratissimum L., and Lippia multiflora Mold.: a potential solution for controlling the cattle tick Rhipicephalus microplus (Acari: Ixodidae).

The cattle tick Rhipicephalus microplus is one of the most significant ectoparasites affecting livestock in tropical and subtropical regions worldwide. This tick causes substantial economic losses in cattle production by reducing weight gain and milk yield and by transmitting pathogens responsible for diseases such as babesiosis and anaplasmosis. Conventional control strategies rely largely on synthetic acaricides; however, their use poses risks to human health and the environment and has contributed to the rapid emergence of resistant tick populations. Under these circumstances, the search fo

Efficacy of a low-dose fipronil bait in controlling blacklegged tick (Ixodes scapularis) larvae parasitizing white-footed mice (Peromyscus leucopus) under field conditions in the northeastern United States.

BACKGROUND: Lyme disease, caused primarily by Borrelia burgdorferi sensu stricto, remains the most common vector-borne disease in the United States despite control efforts. Peromyscus leucopus (white-footed mouse) is a key natural pathogen reservoir and Ixodes scapularis ticks, commonly infesting this rodent, serve as vectors for spirochetes causing human Lyme disease. Laboratory and simulated field studies demonstrated substantial efficacy of a 0.005% fipronil bait in systemically controlling I. scapularis parasitizing treated P. leucopus. To evaluate fipronil bait efficacy under natural cond

Efficacy of a novel topical combination of fipronil, amitraz and (S)-methoprene for treatment and control of induced infestations with four North American tick species (Dermacentor variabilis, Ixodes scapularis, Amblyomma americanum and Amblyomma maculatum) on dogs.

Five laboratory studies were conducted to confirm that a single topical dose of the novel combination of fipronil, amitraz and (S)-methoprene, CERTIFECT™ (Merial Limited, GA, USA), is efficacious for the rapid control of pre-existing infestations and the prevention of new infestations with Ixodes scapularis, Dermacentor variabilis, Amblyomma americanum and Amblyomma maculatum for at least 28 days on dogs. In each study, 8 male and 8 female purpose-bred, laboratory beagles were randomly assigned to one of two study groups (treated and untreated). Starting on the day before treatment, each dog w

Development of a low-dose fipronil deer feed: Bait-screening and range-finding to determine the optimal formulation to control blacklegged ticks (Ixodes scapularis) feeding on white-tailed deer (Odocoileus virginianus).

Lyme disease is the most prevalent vector-borne disease in the U.S., and acaricidal feeds administered to white-tailed deer (Odocoileus virginianus) have potential to disrupt blood feeding by the blacklegged tick, Ixodes scapularis. Two studies were conducted with the aim of determining an ideal formulation to deliver oral acaricides to white-tailed deer and finding the lowest fipronil dose level to effectively control I. scapularis. During formulation screening, various commercial attractants (baits) were presented to deer in large paddocks under semi-field conditions and motion-sensitive cam

Three-dose versus four-dose primary schedules for tick-borne encephalitis (TBE) vaccine FSME-immun for those aged 50 years or older: A single-centre, open-label, randomized controlled trial.

BACKGROUND: TBE vaccination failures among those past middle age have raised concern about immune response declining with age. We investigated immunogenicity of the TBE-vaccine FSME-Immun among those aged 50+ years using the standard three-dose primary series and alternative four-dose schedules. METHODS: In this single-centre, open-label, randomized controlled trial, 200 TBE-naive Swedish adults were given primary TBE vaccination with FSME-Immun. Those aged 50+ years (n = 150) were randomized to receive the standard three-dose (days 0-30-360) or one of two four-dose series (0-7-21-360; 0-30-9

Barriers to Effective Tick Management and Tick-Bite Prevention in the United States (Acari: Ixodidae).

Lyme and other tick-borne diseases are increasing in the United States. Development of tick control tools have focused primarily on the blacklegged tick, Ixodes scapularis Say. Application of acaricides or entomopathogenic fungal agents to kill host-seeking ticks or ticks on rodents can suppress I. scapularis abundance in residential landscapes, but evidence is lacking for impact on human tick bites or tick-borne disease. Similar studies remain limited for the lone star tick, Amblyomma americanum (L.). Other knowledge gaps include how well homeowners and pest control companies perform in the b
