Diabetes Enhances IL-17 Expression and Alters the Oral Microbiome to Increase Its Pathogenicity.

Diabetes is a risk factor for periodontitis, an inflammatory bone disorder and the greatest cause of tooth loss in adults. Diabetes has a significant impact on the gut microbiota; however, studies in the oral cavity have been inconclusive. By 16S rRNA sequencing, we show here that diabetes causes a shift in oral bacterial composition and, by transfer to germ-free mice, that the oral microbiota of diabetic mice is more pathogenic. Furthermore, treatment with IL-17 antibody decreases the pathogenicity of the oral microbiota in diabetic mice; when transferred to recipient germ-free mice, oral mic

Union between deciduous and permanent as intrusive luxation sequelae: A case report.

Traumatic dental injuries (TDI) are common in children due learning to walk and lack of balance that leads to falls. Luxation is the trauma that occurs most frequently in the deciduous, being that the intrusive and the avulsive are the ones that cause more damage to the permanent successors. The potential to cause disturbances to the developing permanent germ is high due to anatomical proximity and depends on age, direction of intrusion, severity and treatment. The consequences to the permanent range from hypocalcifications of enamel to retention of the permanent germ. In this case, the develo

Concomitant hypodontia and hyperodontia: an analysis of nine patients.

INTRODUCTION: Congenital absence of tooth germs and presence of supernumerary teeth (CHH) are anomalies which are classified as embryologically determined disorders. Both disorders can occur together (CHH), but relatively rarely. OBJECTIVE: The aim of the present study was to present and analyze nine cases collected during the past 24 years, where congenitally missing teeth occurred with supernumerary teeth in the same patient. MATERIALS AND METHODS: Dental records, radiographs and casts of nine patients, six boys and three girls (aged 9-29 years, mean age: 19 years) were examined. All the f

Congenital osteoclast deficiency in osteopetrotic (op/op) mice is cured by injections of macrophage colony-stimulating factor.

Osteopetrotic (op/op) mice have a severe deficiency of osteoclasts, monocytes, and peritoneal macrophages because of a defect in the production of functional macrophage colony-stimulating factor (M-CSF) resulting from a mutation within the M-CSF gene. In this study, we examined whether daily 5-microgram injections of purified recombinant human M-CSF (rhM-CSF) for 14 d would cure these deficiencies in the mutant mice. Monocytes in the peripheral blood of the op/op mice were significantly increased in number after subcutaneous injections of the factor two or three times a day. In contrast, osteo

Tooth replacement in the teleost fish Prionurus microlepidotus Lacépède.

Previous studies on tooth replacement in lower vertebrates have been plagued by a lack of common integrative approaches and methods making it impossible to furnish a phylogenetic synthesis. This study is based on serial sections of the jaw of Prionurus microlepidotus. Each Toothgerm was characterized by its developmental stage and its position in the jaw. The relationship between the developmental stage of toothgerm and position in the jaw has been studied and expressed in several graphical illustrations. The following conclusions have been made: (1) The initiation of toothgerms in P. microlep

A morphometric and biochemical study of the pre-eruptive development of hamster molars in vivo.

Surface area measurements of cross-sections of M1 and M2 at 1-11 days were related to biochemical parameters for cell proliferation ([3H]-thymidine incorporation) and mineralization (45Ca uptake) and with increase in dry weight. The pre-eruptive development of molar tooth germs was divided into 4 phases which partly reflect the life cycle stages of the ameloblast. In phase 1 (proliferation phase, duration in M2 about 5 days) mitotic activity and increase in total area were great; ameloblasts were undifferentiated or in a pre-secretory state. In phase 2 (differentiation phase, duration 2-3 days

Biomechanical stress regulates mammalian tooth replacement via the integrin β1-RUNX2-Wnt pathway.

Renewal of integumentary organs occurs cyclically throughout an organism's lifetime, but the mechanism that initiates each cycle remains largely unknown. In a miniature pig model of tooth development that resembles tooth development in humans, the permanent tooth did not begin transitioning from the resting to the initiation stage until the deciduous tooth began to erupt. This eruption released the accumulated mechanical stress inside the mandible. Mechanical stress prevented permanent tooth development by regulating expression and activity of the integrin β1-ERK1-RUNX2 axis in the surrounding

Experimental odontogenic tumors produced by ethylnitrosourea injections and mechanical injuries.

The present study was carried out to investigate odontogenic tumor induction in the rats by injections of carcinogen N-ethyl-N-nitrosourea (ENU) coupled with incisional wounds. The animals which received local injections of ENU in the region of incisor tooth germ of the right mandible every other day for 19 days after birth coupled with incisional wounding in the same region at 2 and 8 days, developed odontogenic carcinomas. However, the animals which were given local injections of ENU in the region of incisor tooth germ but did not receive incisional wounds, showed no pathologic changes. The

Laser-assisted management of ectopic eruption of permanent incisors.

Ectopic eruption originates from the malpositioning of a tooth bud ending in the eruption of the tooth in an improper direction. This anomaly is a consequence of ectopic development of the tooth germ. This condition gives rise to significant aesthetic and occlusal issues in the early mixed dentition stage. The most frequently influenced teeth are the maxillary first molars, accompanied by the maxillary cuspids. Ectopic eruption is seldom seen and might derive from traumatic injury. In some cases, hyperplastic and fibrotic gingival tissue may lead to ectopic eruption of a permanent incisor. Man

[Microscopic root canal treatment of fused mandibular molar with seven root canals: a case report].

Fused teeth are usually formed by the partial or complete fusion of two normal tooth germs during the development process and belong to dental developmental abnormalities. Fused teeth are relatively rare clinically, and those occurring in the posterior tooth area are even rarer. This article reports a case of fused teeth between the first permanent molar and the second permanent molar in the right mandible. This fused tooth had a complex root canal anatomical structure (seven root canals). The number and location of the root canals were analyzed by cone beam computed tomography, and root canal

A peculiar complication in Le Fort I osteotomy.

In two cases of routine Le Fort I osteotomy in secondary cleft palate surgery, a swelling noted in the antral mucosa was biopsied. In both cases, histological examination resulted in a diagnosis of odontogenic myxoma. After analysis of the radiographs, it was concluded that the biopsies were taken from displaced tooth germs. The close histological similarity between myxoma on the one hand and dental papilla tissue, a thickened tooth follicle and a thickened or polypoid antral mucosal lining on the other is discussed.

Intrusive luxation in primary teeth - Review of literature and report of a case.

Luxation injuries such as intrusion are commonly seen in the primary dentition. Intrusion drives the tooth deeper into the alveolar socket, which results in damage to the pulp and peridontium. Difficulty in gaining compliance from a very young child and the risk of damaging the permanent tooth germ makes the management of these injuries challenging. Careful clinical and radiographic examination along with regular follow-up is essential. A case of intrusive luxation to the maxillary central incisor in a 3-year-old patient is reported. Spontaneous reeruption was noted 4 months after injury, but

Revisiting the supernumerary: the epidemiological and molecular basis of extra teeth.

Supernumerary teeth are a common clinical and radiographic finding and may produce occlusal and dental problems. The aetiological basis of extra teeth is poorly understood in human populations; however, the mouse provides a useful model system to investigate the complex genetics of tooth development. This article describes recent advances in our understanding of the genetic basis of supernumerary teeth. We have reviewed biological evidence that provides insight into why supernumerary tooth formation may occur. Indeed, many of the molecular signalling pathways known to be involved in normal dev

Supernumerary teeth in anthropoid primates and models of tooth development.

Supernumerary teeth are usually regarded as de-novo structures that result from abnormal development by the splitting of a tooth germ. The notions of abnormal and de novo appear to be applicable in only one instance, the morphological dental twin. All other types of supernumerary deciduous teeth can be explained by either a progress zone or an interstitial budding model of tooth differentiation. The development of extra structures as well as the loss of normally-present teeth can be explained by differences in potential within the context of normal development.
