Tooth root growth impairment after mantle radiation in long-term survivors of Hodgkin's disease.

The tooth root growth impairment that resulted from 35 to 37 Gy mantle port radiation in 47 long-term survivors of childhood Hodgkin's disease was quantified and related to specific age groups and categories of teeth. Root measurements of the mandibular permanent canines, first and second premolars, and first and second molars were made from sequential panoramic radiographs taken at the time of radiation therapy and after the closure of root apexes. The severity of root growth impairment was greatest in patients who received radiation during the early stages of odontogenesis. With later stages

Root formation in ectopically transplanted teeth of the frog, Rana pipiens I. Tooth morphogenesis.

In order to determine the intrinsic (genetic) and extrinsic (functional) parts of the naturally ankylosed tooth, a qualitative study of the development and fate of tooth buds transplanted individually or within the premaxilla was carried out. These teeth were transplanted as autografts during crown formation. They were placed in either the anterior chamber of the eye or in a dorsal subcutaneous site. Most of the 163 transplants survived and were examined at intervals from 0 to 180 days by gross, histologic and microradiographic methods. Tooth buds transplanted with or without the premaxilla we

Sensory Nerves Regulate Odontoblast Differentiation via the SPP1/ITGA4 Axis During Tooth Root Development.

AIM: The stem/progenitor cell is crucial for organogenesis. Sensory nerves, as key components of the stem cell niche, secrete various factors to modulate stem/progenitor cell fate decision. Here, we utilised tooth root development as a model to explore the role of sensory nerves in this regulatory process and to elucidate the underlying mechanism. METHODOLOGY: Spatiotemporal dynamics of nerve innervation were characterised during tooth root development. We treated mouse dental papilla cells (mDPCs) with trigeminal ganglion-conditioned medium (TG-CM) and employed a subrenal co-culture of TG-to

Notch signaling inhibition protects against root resorption in experimental immature tooth movement in rats.

INTRODUCTION: This study aimed to build an experimental immature tooth movement model and verify less resorption of incompletely developed roots than those fully developed during the same orthodontic treatment, followed by investigating the cellular and molecular mechanism. METHODS: The development of Wistar rat tooth was investigated using in vivo microcomputed tomography and hematoxylin and eosin staining to decide the optimal ages of rats for immature tooth and mature tooth groups. The rats in the immature tooth and mature tooth groups were divided into experimental, sham control, and blan

Geomorphologic study of human tooth root surfaces following simulated avulsion by scanning electron microscopy.

BACKGROUND/AIMS: Tooth avulsion is the most severe traumatic tooth injury. Immediate replantation after avulsion provides the highest success rate, whereas delayed replantation with dehydration for >1 h always results in ankylosis, replacement resorption, and eventual tooth loss. The aim of this study was to investigate the geomorphologic change of ultrastructure on tooth root surfaces due to dehydration. MATERIAL AND METHODS: Twenty-four sound human premolars and fully developed third molars without periodontitis were selected for the experiment. Roots were separated into 6-7 pieces with an

BoneCeramic graft regenerates alveolar defects but slows orthodontic tooth movement with less root resorption.

INTRODUCTION: BoneCeramic (Straumann, Basel, Switzerland) can regenerate bone in alveolar defects after tooth extraction, but it is unknown whether it is feasible to move a tooth through BoneCeramic grafting sites. The objective of this study was to investigate 3-dimensional real-time root resorption and bone responses in grafted sites during orthodontic tooth movement. METHODS: Sixty 5-week-old rats were randomly assigned to 3 groups to receive BoneCeramic, natural bovine cancellous bone particles (Bio-Oss; Geistlich Pharma, Wolhusen, Switzerland), or no graft, after the extraction of the ma

Effect of odanacatib on root resorption and alveolar bone metabolism during orthodontic tooth movement.

The aim of this study was to investigate the effect of local administration of odanacatib (ODN) on orthodontic root resorption and the status of alveolar bone metabolism in rat molars. All specimens were scanned using microcomputed tomography and then the raw images were reconstructed. The total volume of the root resorption craters of the 60 g-NS (normal saline) group was higher than in the 60 g-ODN group and the control group. In the 60 g-NS group, the bone volume fraction values of alveolar bone were significantly decreased compared with the other 2 groups. There were no significant differe

Strontium ranelate improved tooth anchorage and reduced root resorption in orthodontic treatment of rats.

The anchorage mechanisms currently used in orthodontic treatment have various disadvantages. The objective of this study was to determine the applicability of the osteoporosis medication strontium ranelate in pharmacologically induced orthodontic tooth anchorage. In 48 male Wistar rats, a constant orthodontic force of 0.25 N was reciprocally applied to the upper first molar and the incisors by means of a Sentalloy(®) closed coil spring for two to four weeks. 50% of the animals received strontium ranelate at a daily oral dosage of 900 mg per kilogramme of body weight. Bioavailability was determ

Continuous versus interrupted continuous orthodontic force related to early tooth movement and root resorption.

The aim of the present clinical investigation was to assess the effects of continuous and interrupted continuous forces of the same magnitude (50 cN = 50 g) on orthodontic tooth movement and related adverse tissue reactions, i.e., root resorption. Thirty-two maxillary first premolars in 16 patients, 8 boys and 8 girls (mean age 13.9 years), were moved buccally by means of a fixed orthodontic appliance with a sectional arch. The patients were divided into two groups of 8, for experimental periods of 4 and 7 weeks. The continuous force was checked and reactivated weekly to 50 cN. The interrupted

Interactive effects of periodontitis and orthodontic tooth movement on dental root resorption, tooth movement velocity and alveolar bone loss in a rat model.

BACKGROUND: Many adult orthodontic patients suffer from chronic periodontitis with recurrent episodes of active periodontal inflammation. As their number is steadily increasing, orthodontists are more and more frequently challenged by respective treatment considerations. However, little is currently known regarding interactive effects on undesired dental root resorption (DRR), tooth movement velocity, periodontal bone loss and the underlying cellular and tissue reactions. MATERIAL AND METHODS: A total of 63 male Fischer344 rats were used in three consecutive experiments employing 21 animals e

Mutations in LRP5 and BMP4 are associated with mesiodens, tooth agenesis, root malformation, and oral exostoses.

WNT/β-catenin and BMP signaling pathways play important roles in the process of tooth development. Dysregulation of WNT/β-catenin and BMP signaling is implicated in a number of human malformations, including dental anomalies. Whole exome and Sanger sequencing identified seven patients with LRP5 mutations (p.Asn1121Asp, p.Asp856Asn, p.Val1433Met, and p.Val1245Met) and six patients with BMP4 mutations (p.Asn150Lys, p.Gly168Arg, p.Arg269Gln, and p.Ala42Glu). All patients were affected with isolated dental anomalies (dental anomalies with no other structural defects), including mesiodens, tooth ag

Imaging of growth factors on a human tooth root canal by surface-enhanced Raman spectroscopy.

Endodontic treatment of immature permanent teeth with necrotic pulp poses several clinical challenges and is one of the most demanding interventions in endodontics. Recently, with new discoveries in the field of tissue engineering, novel treatment protocols have been established. The most promising treatment modality is revascularization, whose integral part is the exposure of collagen matrix and embedded growth factors. However, optimization of the treatment protocol requires a development of analytical procedures able to analyze growth factors directly on the sample surface. In this work, me
