Effect of thiamine deprivation on thiamine metabolism in mice.

Mice were made deficient by thiamin deprivation. Thiamin pyrophosphokinase and thiamin pyrophosphatase activities were measured, and their changes were compared to transketolase (TK), pyruvate and alpha-ketoglutarate dehydrogenase (DG) activities and thiamin pyrophosphate (TPP) level in the same tissues. Thiamin pyrophosphokinase activity was increased on day 10 of thiamin deficiency and remained unchanged within the next period of investigation. Thiamin pyrophosphatase activity decreased after 5 days and was enhanced in 10, 15 and 20 days of deficiency. This fact may be related to the regulat

Thiamine deficiency induced by polychlorinated biphenyls (PCB) and dichlorodiphenyltrichloroethane (DDT) administration to rats.

To clarify the effect of polychlorinated biphenyls (PCB), and dichlorodiphenyltrichloroethane (DDT) and other organochloride compounds on thiamine metabolism, rats were separated into six groups and placed on different admixed diets. Group 1 was fed a normal diet; group 2, a PCB-supplemented diet; group 3, a DDT-supplemented diet; group 4, a benzenehexachloride-supplemented (BHC) diet. After 50 days of these dietary regimens all animals were sacrificed and various studies were done on the excised tissues. In PCB-treated rats, thiamine levels in blood, liver and sciatic nerve decreased, transke

[Vitamin B 1 deficiency in chronic alcoholics and its clinical correlation].

50 chronic alcoholics reporting to the medical emergency ward of Basle University Hospital with alcohol-related illness were examined with respect to thiamine nutritional status by means of the transketolase activation test of erythrocytes (ETK). 46% of the chronic alcoholics, compared to only 2% of the control population (1152 healthy adults), had transketolase activation quotients indicating a strong probability of thiamine deficiency (alphaETK greater than 1.25). The most important symptoms associated with the biochemical parameters of thiamine deficiency were: anemia, pathologic liver func

Activities of thiamine-dependent enzymes in two experimental models of thiamine-deficiency encephalopathy: 1. The pyruvate dehydrogenase complex.

Chronic thiamine deprivation in the rat leads to selective neuropathological damage in brainstem structures whereas treatment with the central thiamine antagonist, pyrithiamine, results in more widespread damage. In order to further elucidate the neurochemical mechanisms responsible for this selective damage, the thiamine-dependent enzyme complex pyruvate dehydrogenase (PDHC) was measured in 10 brain structures in the rat during progression of thiamine deficiency produced by chronic deprivation or by pyrithiamine treatment. Feeding of a thiamine-deficient diet to adult rats resulted in 5-7 wee

Eighteen-month follow-up of infants fed evaporated milk formula.

In parts of Canada including Newfoundland and Labrador and among Aboriginal peoples, infants still consume evaporated milk (EM) formulas for cultural and economic reasons. At 3 and 6 months, full-term infants fed EM (n = 30) received low intakes of iron, thiamine, selenium and had higher weight velocity than breastfed (BF, n = 29) infants. EM infants had greater anemia, lowered transketolase activity (thiamine) and lowered glutathione peroxidase (selenium) activity (p < 0.05). To determine the later effect of early feeding deficit on nutritional status, we examined these same infants at 18 mon

Cell damage through pentose phosphate pathway in fetus fibroblast cells exposed to methyl mercury.

Methylmercury (MeHg) is a global pollutant that causes malformations. There has been no direct evidence for the effect of MeHg on pentose phosphate pathway (PPP). In embryonic development, PPP is much more active. This pathway produces ribose for DNA/RNA production. It is possible that one of teratogenicity mechanisms of MeHg is through PPP. The fetus fibroblast cells were incubated with different concentrations of MeHg (0.1-100 μm). A dose-response dependence was observed in MTT assay. Transketolase activity and DNA content were determined in cell exposed to MeHg. A defect at the level of DNA

The validity of biochemical assessment of thiamine, riboflavin and folacin nutriture.

Nutriture of thiamine, riboflavin and folacin was assessed by two tests: thiamine (109 subjects) by the TPP effect of erythrocyte transketolase activity and urinary excretion of thiamine (mug/g creatinine); riboflavin (81 subjects) by the activation coefficient of erythrocyte glutathione reductase and excretion of riboflavin in urine (mug/g creatinine); and folacin (91 subjects) by estimation of folacin in red blood cells and in serum (ng/ml). The following correlation coefficients (r) were obtained: transketolase activity vs thiamine excretion: -0.33; glutathione reductase vs riboflavin excre

Thiamine status in liver and brain of rats genetically selected for different sensitivity to hypnotic effect of alcohol.

BACKGROUND: The mechanisms of the different sensitivity or resistance of animals and humans to alcohol are still not completely understood. For further biochemical characterization of animals genetically selected for high-alcohol sensitivity (HAS) and low-alcohol sensitivity (LAS) with the hypnotic effect of alcohol, the thiamine status and thiamine metabolizing enzymes in these animals have been studied. METHODS: We investigated thiamine diphosphate and thiamine triphosphate levels as well as the activity of thiamine-dependent enzyme, transketolase, and thiamine-metabolizing enzymes, thiamin

Comparison of Secretome Profile of Pathogenic and Non-Pathogenic Entamoeba histolytica.

The obligatory intracellular protozoan parasite Entamoeba histolytica causes amebic dysentery and liver abscess. E. histolytica adheres to the host tissues in a contact-dependent manner. E. histolytica excretory-secretory products (ESP) might play critical roles during invasion. We comparatively analyzed the secretome profile of E. histolytica pathogenic HM-1:IMSS and non-pathogenic Rahman strains. The two ESP revealed similar but distinct spotting patterns. In both ESP, alcohol dehydrogenase, enolase 1, and transketolase, which control classical carbohydrate metabolism and other moonlighting

Effects of maternal thiamine deficiency on the development of thiamine-dependent enzymes in regions of the rat brain.

Previous studies suggest that developing rat brain is susceptible to reduced thiamine intake. In order to assess the metabolic basis for this susceptibility, activities of three thiamine-dependent enzymes (pyruvate dehydrogenase complex, ?-ketoglutarate dehydrogenase and transketolase) were measured in homogenates of brain tissue from the offspring of thiamine-deficient mothers. Control groups of animals were pair-fed to equal food consumption with the thiamine-deficient animals. The study revealed region-selective delays in the establishment of adult activities of thiamine-dependent enzymes a

Evidence of a thiamine deficiency in sheep fed maize silage.

Two groups of Merino sheep (with or without rumen fistulae) were used to study the effect of feeding maize-silage (compared to hay) on: thiamine concentrations in rumen fluid, tissues and organ; blood transketolase activity; microbial fermentation in the rumen. After sheep were transferred from a hay diet to maize-silage, thiamine concentrations in ruminal fluid fell to about 25% of control values on hay. Thiamine levels greatly decreased in kidney, heart, liver, rumen tissue and large intestine. Blood transketolase and TPP effect significantly decreased (P less than 0.01). Ruminal ammonia nit

[Wernicke-Korsakoff syndrome in anorexia (author's transl)].

A rare case of Wernicke-Korsakoff syndrome due to malnutrition (in a 35-year-old woman) is reported. It was probably the result of a psychiatric condition progressing to severe inanition. In the course of the malnutrition typical Wernicke-Korsakoff syndrome developed. Vitamin B1 deficiency was confirmed by determining erythrocyte transketolase. At the same time there was hypomagnesiaemia; substitution treatment led to complete recovery.

Assessment of thiamine status in humans.

Thiamine (vitamin B1) is an essential nutrient with established adverse health consequences associated with acute deficiency. Emerging evidence also suggests long-term neurocognitive deficits due to sub-clinical deficiency in early life, and there is a growing appreciation of a potential role for thiamine in Alzheimer's disease. Therefore, the assessment of thiamine biomarkers, particularly in large, population-based studies, is critical to grasp the true scope of this public health problem. The most reliable biomarkers of thiamine status are whole blood concentrations of thiamine diphosphate

Defective RNA ribose synthesis in fibroblasts from patients with thiamine-responsive megaloblastic anemia (TRMA).

Fibroblasts from patients with thiamine-responsive megaloblastic anemia (TRMA) syndrome with diabetes and deafness undergo apoptotic cell death in the absence of supplemental thiamine in their cultures. The basis of megaloblastosis in these patients has not been determined. Here we use the stable [1,2-13C2]glucose isotope-based dynamic metabolic profiling technique to demonstrate that defective high-affinity thiamine transport primarily affects the synthesis of nucleic acid ribose via the nonoxidative branch of the pentose cycle. RNA ribose isolated from TRMA fibroblasts in thiamine-depleted c

Thiamine deficiency and malaria in adults from southeast Asia.

BACKGROUND: Thiamine deficiency (beriberi) is common in some parts of southeast Asia. Acute thiamine deficiency can mimic many complications of malaria, such as encephalopathy and lactic acidosis. We examined the incidence of thiamine deficiency in adults admitted to hospital with malaria in Thailand. METHODS: For this prospective study, we recruited consecutive patients with malaria or other febrile illness who presented to Paholpolpayuhasena Hospital, Kanchanaburi, Thailand, between May and July, 1992. We used the activation coefficient (alpha) for transketolase activity in erythrocytes to

Correlation of enzymatic, metabolic, and behavioral deficits in thiamin deficiency and its reversal.

To clarify the enzymatic mechanisms of brain damage in thiamin deficiency, glucose oxidation, acetylcholine synthesis, and the activities of the three major thiamin pyrophosphate (TPP) dependent brain enzymes were compared in untreated controls, in symptomatic pyrithiamin-induced thiamin-deficient rats, and in animals in which the symptoms had been reversed by treatment with thiamin. Although brain slices from symptomatic animals produced 14CO2 and 14C-acetylcholine from [U-14C]glucose at rates similar to controls under resting conditions, their K+-induced-increase declined by 50 and 75%, resp

[Interstitial thiamine redistribution during the development of vitamin B1 deficiency in mice].

Investigation of diphosphothiamine content in the mouse tissues, associated with transketolase activity in the dynamic dependence on the body provision with vitamin B1, has proved the organ-specificity of deposition and elimination of diphosphothiamine under conditions of developing thiamine deficiency. The experimental data have evidenced that vitamin B1 exclusion from the food ration results in the diphosphothiamine redistribution between the tissues and in the provision with coenzymes of one tissue at the expense of the other.
