Aetiology and epidemiology of viral gastroenteritis outbreaks in Catalonia (Spain) in 2004-2005.

BACKGROUND: Acute infectious gastroenteritis causes substantial morbidity and economic loss. OBJECTIVE: The aetiology, epidemiology, and clinical features of acute viral gastroenteritis outbreaks reported during 1 year in Catalonia were investigated. STUDY DESIGN: This was a population-based study in which enzyme immunoassay and reverse transcriptase-polymerase chain reaction (RT-PCR) techniques were used to determine the presence of virus in stool specimens from outbreaks clinically and epidemiologically compatible with a viral aetiology and negative for bacteria, parasites and toxins. RES

Transmission dynamics of Norovirus GII and Enterovirus in Switzerland during the COVID-19 pandemic (2021-2022) as evidenced in wastewater.

Noroviruses and enteroviruses are major causes of endemic gastrointestinal disease associated with substantial disease burden. However, viral gastroenteritis is often diagnosed based on symptoms, with etiology infrequently tested or reported, so little information exists on community-level transmission dynamics. In this study, we demonstrate that norovirus (NoV) genogroup II and enterovirus (EV) viral loads in wastewater reveal transmission dynamics of these viruses. We report NoV and EV concentrations in wastewater from 363 samples between December 5 2020 and October 10 2022 (sampled every se

Sampling methods for recovery of human enteric viruses from environmental surfaces.

Acute gastroenteritis causes the second highest infectious disease burden worldwide. Human enteric viruses have been identified as leading causative agents of acute gastroenteritis as well as foodborne illnesses in the U.S. and are generally transmitted by fecal-oral contamination. There is growing evidence of transmission occurring via contaminated fomite including food contact surfaces. Additionally, human enteric viruses have been shown to remain infectious on fomites over prolonged periods of time. To better understand viral persistence, there is a need for more studies to investigate this

Norovirus inter-species transmission in an ex vivo model is dependent on virus internalization capacity.

Human noroviruses (HuNoV) are the most common cause of viral gastroenteritis worldwide, causing sporadic cases and outbreaks. Noroviruses that infect wild and domestic animals, including pigs and dogs, are genetically related to human noroviruses, increasing the potential for cross-species transmission. To investigate the potential of noroviruses to cross the species barrier, we used porcine, canine, and avian precision-cut intestinal slices. We show that human norovirus virus-like particles (VLPs) bind to pig and dog intestinal tissue and are taken up with a similar efficiency as the porcine

Efficacy of Neutral Electrolyzed Water for Inactivation of Human Norovirus.

Human norovirus (NoV) is the leading cause of acute gastroenteritis worldwide. Persistence on surfaces and resistance to many conventional disinfectants contribute to widespread transmission of norovirus. We examined the efficacy of neutral electrolyzed water (NEW; pH 7) for inactivation of human NoV GII.4 Sydney in suspension (ASTM method 1052-11) and on stainless steel surfaces (ASTM method 1053-11) with and without an additional soil load. The impact of the disinfectant on viral capsid was assessed using reverse transcriptase quantitative PCR (RT-qPCR; with an RNase pretreatment), SDS-PAGE,

[Environmental virology and sanitation in Brazil: a narrative review].

Sanitation services play a critical role in controlling transmission of numerous waterborne pathogens, especially viruses that cause acute gastroenteritis and hepatitis. The viral agents with the greatest public health impact are hepatitis A virus, rotaviruses and noroviruses, adenoviruses, and enteroviruses, contaminating many Brazilian aquatic ecosystems. Heavy circulation of viruses in the environment has been related to inadequate local sanitary conditions, including incomplete coverage of services or inefficacy of conventional technologies in eliminating or reducing the viral load in wate

Genetic diversity of norovirus associated with outbreaks in school children with acute gastroenteritis in Changzhou, China, 2018-2019.

Norovirus is one of the major causes of outbreaks and sporadic cases of acute gastroenteritis in school children. Obtaining local genotype diversity information regarding norovirus is important for developing and evaluating prevention strategies of the transmission of this virus in school children. Clinical specimens, obtained from the routine acute gastroenteritis surveillance network from 2018 to 2019, were primarily tested using commercial real-time PCR Kit. Samples with Ct value less than 25 were selected and used for complete genome sequencing and those with Ct value between 25 and 30 wer

Possible factors influencing the immunoglobulin G concentration in swine colostrum.

The immunoglobulin (Ig) G concentration in swine colostrum was determined by the single radial immunodiffusion method, using 157 samples collected from farm-raised sows in the Yamaguchi Prefecture of Japan during 1976 and 1977. The mean IgG value was 53.03 mg/ml, and the maximum and minimum values were 101.39 mg/ml and 11.74 mg/ml, respectively. The amount of IgG varied greatly among sows. To clarify the possible factors influencing the amount of IgG in colostrum, the following items were surveyed: season, district, breed, age of sows, number of parturitions, udder section from which samples w

Development and Application of a Triplex RT-qPCR Assay for Differentiating Major Lineages of Porcine Reproductive and Respiratory Syndrome Virus.

Porcine reproductive and respiratory syndrome (PRRS) represents a critical infectious disease caused by the PRRS virus (PRRSV), posing a substantial threat to the global swine industry. In China, there is currently an epidemic trend characterized by the coexistence of multiple evolving genotypes. Effective prevention and control measures are contingent upon the availability of rapid, precise, and sensitive pathogen detection technologies. Addressing the need for swift differentiation of the predominant circulating strains, including the classical strains (PRRSV-C), the highly pathogenic strain

Retinoic Acid Alleviates TGEV-Induced Ferroptosis by Activating the p62-NRF2-GPX4/HO-1 Pathway and Iron Metabolism in Intestinal Epithelial Cells.

Background: Transmissible gastroenteritis virus (TGEV) is a highly pathogenic porcine coronavirus that causes severe gastrointestinal damage in piglets. However, how TGEV affects host iron homeostasis, oxidative stress, and the ferroptosis process remains unclear. This study aimed to investigate the effects of TGEV infection on cellular iron metabolism, oxidative damage, and lipid peroxidation-mediated ferroptosis, as well as to evaluate the potential therapeutic role of retinoic acid (RA). Methods: Using an intestinal epithelial cell model of TGEV infection, we assessed key regulators of iron

Design of dual-effect molecules based on lysosome-targeting strategy: a mechanistic study on synergistic inhibition of ewing sarcoma and coronaviruses.

INTRODUCTION: Addressing clinical challenges such as the toxicity of conventional chemotherapy and viral drug resistance, this study proposes and validates an innovative "host organelle-targeting" strategy. By disrupting the function of host lysosomes-an organelle commonly exploited by both tumors and viruses-we aim to develop novel therapeutic agents with dual antitumor and antiviral activity. METHODS: We rationally designed and synthesized the target compound 3-chloro-4-(4-(diethylamino)butoxy)benzaldehyde, whose core design relies on the pH-responsive property of the 4-(diethylamino)butoxy

Molecular epidemiology and characterization of porcine adenoviruses in pigs with diarrhea in Thailand.

Pigs have been assumed as a source of human viral infections. Surveillance of viruses in animals is essential to evaluate the risk to human and animal health and to determine economic impact. A number of studies focused mainly on well- known enteritis viruses such as porcine epidemic diarrhea virus (PEDV), transmissible gastroenteritis virus (TGEV), and porcine group A rotavirus (PRVA), however, little data is available for porcine adenovirus (PAdV). In this study, the presence of PAdV was investigated in fecal samples collected from piglets with and without diarrhea from 31 commercial pig far

Identification and characterization of linear B cell epitopes on the nucleocapsid protein of porcine epidemic diarrhea virus using monoclonal antibodies.

The nucleocapsid (N) protein of porcine epidemic diarrhea virus (PEDV), the most important pathogen causing severe diarrhea in piglets, is a highly conserved structural protein. In this study, 5 monoclonal antibodies (McAbs) against the PEDV N-protein were prepared and identified. Three new epitopes, 56QIRWRMRRGERI67, 318GYAQIASLAPNVAALLFGGNVA VRE342 and 398HEEAIYDDV406, were firstly identified in the viral N-protein, by using McAbs 3F10, 6A11, and 1C9. The epitope 398HEEAIYDDV406 was deleted in SH strain (isolated by our lab) and different between CV777 and YZ strain (isolated by our lab). To

Isolation, Identification, and Comprehensive Genomic Characterization of a Bovine Rotavirus G10P[11] Strain in China.

Rotavirus is a principal infectious agent of diarrhea in both animals and humans, causing acute gastroenteritis with symptoms such as abdominal pain, fever, nausea, and vomiting. Bovine rotavirus (BRVA) not only causes illness in cattle but also poses a risk to human health through direct transmission or reassortment among different virus strains. Here, quantitative RT-PCR for the NSP5 gene was applied to pinpoint BRVA in six fecal specimens from calves. A positive sample was identified and further examined using MA104 cells to isolate the virus. The isolate was then identified through RT-PCR,

Characterization and cross-protection of experimental infections with SeCoV and two PEDV variants.

The aim of this study was to characterize the infection of weaned pigs with swine enteric coronavirus (SeCoV) - a chimeric virus most likely originated from a recombination event between porcine epidemic diarrhoea virus (PEDV) and transmissible gastroenteritis virus, or its mutant porcine respiratory coronavirus - and two PEDV G1b variants, including a recently described recombinant PEDV-SeCoV (rPEDV-SeCoV), as well as to determine the degree of cross-protection achieved against the rPEDV-SeCoV. For this purpose, forty-eight 4-week-old weaned pigs were randomly allocated into four groups of 12

Inhibition of SARS-CoV-2 by Highly Potent Broad-Spectrum Anti-Coronaviral Tylophorine-Based Derivatives.

Tylophorine-based compounds and natural cardiotonic steroids (cardenolides and bufadienolides) are two classes of transmissible gastroenteritis coronavirus inhibitors, targeting viral RNA and host cell factors, respectively. We tested both types of compounds against two types of coronaviruses, to compare and contrast their antiviral properties, and with view to their further therapeutic development. Examples of both types of compounds potently inhibited the replication of both feline infectious peritonitis virus and human coronavirus OC43 with EC50 values of up to 8 and 16 nM, respectively. St

The use of PCR genome mapping for the characterisation of TGEV strains.

Previous studies on different transmissible gastroenteritis virus (TGEV) strains, including porcine respiratory coronavirus (PRCV), have identified regions within the genome that are polymorphic as regards insertions and deletions. For example the 672 base deletion within the S gene and multiple deletions 5', within and 3' of the ORF-3a gene were detected in strains of PRCV. The presence of deletions may be associated with a change in the virulence, attenuation or tissue tropism of the isolate. The Nouzilly (188-SG) TGEV vaccine strain was attenuated by passage of a cell culture adapted virule

Cost-effectiveness of Ward Closure to Control Outbreaks of Norovirus Infection in United Kingdom National Health Service Hospitals.

BACKGROUND: Norovirus is the most common cause of outbreaks of acute gastroenteritis in National Health Service hospitals in the United Kingdom. Wards (units) are often closed to new admissions to stop the spread of the virus, but there is limited evidence describing the cost-effectiveness of ward closure. METHODS: An economic analysis based on the results from a large, prospective, active-surveillance study of gastroenteritis outbreaks in hospitals and from an epidemic simulation study compared alternative ward closure options evaluated at different time points since first infection, assumin
