Genetic evidence for a functional association between Parkinson's disease proteins leucine-rich repeat kinase 2 and α-synuclein during axonal transport.

Mutations in α-synuclein (α-syn) and LRRK2 cause familial Parkinson's disease (fPD), yet how these proteins functionally interact remain ambiguous. We previously showed that α-syn undergoes bi-directional transport within axons and influences mitochondrial health, while other studies suggested that LRRK2-G2019S disrupts the axonal transport of autophagic vesicles and mitochondria. Here we tested the hypothesis that α-syn and LRRK2 are functionally linked during axonal transport. Expression of human LRRK2-WT in Drosophila larval nerves caused modest CSP-containing axonal blockages whereas no de

Sodium ion uptake into isolated plasma membrane vesicles: indirect effects of other ions.

Vesicles derived from plasma membrane of corneal endothelium were agitated to their minimum size distribution. When isotonic salt solutions surrounding the vesicles were changed there were alterations to the vesicle size distribution: the modal point of the logarithmic distribution did not change but the log variance did, indicating that substantial fission and fusion of vesicles occurred depending upon the nature of the surrounding solute. Orientation and total membrane area was conserved in the transformed population of vesicles. Although the ions added to the external isotonic salt solution

Dysfunction of the sarcoplasmic reticulum in polymyositis.

The transport of calcium in vesicles of sarcoplasmic reticulum isolated from muscle specimens from 6 patients with early, active polymyositis and from 11 controls was examined. The time courses of calcium uptake and calcium-dependent ATPase activity were measured simultaneously. Calcium uptake by sarcoplasmic reticulum vesicles from patients with polymyositis was 50% less than that by vesicles from controls (P less than 0.001). In contrast, no difference in calcium-dependent ATPase activity was noted between vesicles from patients with polymyositis and controls. The demonstrated defect may be

Anion transport in biologically relevant lipid mixtures.

A potent squaramide-based anion transporter was used to evaluate how vesicles prepared with tailored lipid compositions, which mimic organelle membranes, can impact transmembrane transport. Using HPTS and ISE assays, we show that the lipid environment and POPE flip-flop dynamics significantly influence transport efficiency.

Transport of the cholera toxin B-subunit from recycling endosomes to the Golgi requires clathrin and AP-1.

The retrograde pathway is defined by the transport of proteins and lipids from the plasma membrane through endosomes to the Golgi complex, and is essential for a variety of cellular activities. Recycling endosomes are important sorting stations for some retrograde cargo. SMAP2, a GTPase-activating protein (GAP) for Arf1 with a putative clathrin-binding domain, has previously been shown to participate in the retrograde transport of the cholera toxin B-subunit (CTxB) from recycling endosomes. Here, we found that clathrin, a vesicle coat protein, and clathrin adaptor protein complex 1 (AP-1) were

Staphylococcal toxic shock toxin specifically binds to cultured human epithelial cells and is rapidly internalized.

Staphylococcal toxic shock syndrome toxin (TST) was labeled with 125I under mild conditions without apparent destruction of the molecule. [125I]TST bound specifically to human epithelial (Chang) cells in culture; the binding was inhibited by a 100-fold excess of unlabeled toxin. Scatchard analysis of the binding data indicated about 10(4) receptor sites per cell and a dissociation constant (Kd) of 4 X 10(-9) M. When cells pretreated with TST at 4 degrees C were swiftly transferred to 37 degrees C, the amount of surface-bound toxin rapidly declined, as determined by release of noninternalized l

Modification of Mecp2 dosage alters axonal transport through the Huntingtin/Hap1 pathway.

Mecp2 deficiency or overexpression causes a wide spectrum of neurological diseases in humans among which Rett Syndrome is the prototype. Pathogenic mechanisms are thought to involve transcriptional deregulation of target genes such as Bdnf together with defects in the general transcriptional program of affected cells. Here we found that two master genes, Huntingtin (Htt) and huntingtin-associated protein (Hap1), involved in the control of Bdnf axonal transport, are altered in the brain of Mecp2-deficient mice. We also revealed an in vivo defect of Bdnf transport throughout the cortico striatal

Immunofluorescence-Based Study of Cargo Transport Mediated by Cytoskeleton-Associated Molecular Complexes.

The cytoskeleton plays a fundamental role in orchestrating the intracellular cargo transport, providing a dynamic scaffold that ensures the appropriate sorting, movement, and distribution of membrane-bound compartments. Through the coordinated activity of microtubules and actin filaments, cargo vesicles are driven along defined routes to ensure efficient intracellular trafficking. Among the molecular machineries that interact with the microtubule network, the intraflagellar transport (IFT) system has emerged as a key regulator of vesicular dynamics. Initially identified for its essential role

Japanese encephalitis virus is transported across the cerebral blood vessels by endocytosis in mouse brain.

Japanese encephalitis (JE) virus principally infects neuron systems of animals and causes severe encephalitis. The mechanism by which the virus enters the central nervous system (CNS) from the circulatory system remains elusive. In this study, electron-microscopic techniques have been used to determine these sequential events in the suckling mouse brain. The results indicate that (1) endocytosis is employed when JE virus is transported across the cerebral blood vessels (CBV) and breaches the blood-brain barrier (BBB). (2) Uncoated vesicles, which may be caveolae, and coated vesicles are involv

The Role of Extracellular Vesicles in Metabolic Diseases.

Extracellular vesicles represent a group of structures with the capacity to communicate with different cells and organs. This complex network of interactions can regulate multiple physiological processes in the organism. Very importantly, these processes can be altered during the appearance of different diseases including cancer, metabolic diseases, etc. In addition, these extracellular vesicles can transport different cargoes, altering the initiation of the disease, driving the progression, or even accelerating the pathogenesis. Then, we have explored the implication of these structures in di

Failure of epithelial tube maintenance causes hydrocephalus and renal cysts in Dlg5-/- mice.

Epithelial tubes represent fundamental building blocks of metazoan organisms; however, the mechanisms responsible for their formation and maintenance are not well understood. Here, we show that the evolutionarily conserved coiled-coil MAGUK protein Dlg5 is required for epithelial tube maintenance in mammalian brain and kidneys. We demonstrate that Dlg5(-/-) mice develop fully penetrant hydrocephalus and kidney cysts caused by a deficiency in membrane delivery of cadherin-catenin adhesion complexes and loss of cell polarity. Dlg5 travels with cadherin-containing vesicles and binds to syntaxin 4

Huntingtin-mediated axonal transport requires arginine methylation by PRMT6.

The huntingtin (HTT) protein transports various organelles, including vesicles containing neurotrophic factors, from embryonic development throughout life. To better understand how HTT mediates axonal transport and why this function is disrupted in Huntington's disease (HD), we study vesicle-associated HTT and find that it is dimethylated at a highly conserved arginine residue (R118) by the protein arginine methyltransferase 6 (PRMT6). Without R118 methylation, HTT associates less with vesicles, anterograde trafficking is diminished, and neuronal death ensues-very similar to what occurs in HD.

Alteration of renal amino acid transport system in cadmium-intoxicated rats.

Effects of cadmium intoxication on renal transport systems for various amino acids were studied. Subcutaneous injections of CdCl2, at a dose of 2 mg Cd/kg.day for 2 weeks, resulted in polyuria, proteinuria, glycosuria, phosphaturia, and aminoaciduria, as observed in chronic cadmium-intoxicated humans and experimental animals. The nature of aminoaciduria was nonspecific, including iminoacid as well as almost all species of neutral, acidic, and basic amino acids. In renal cortical brush border membrane vesicles isolated from cadmium-intoxicated rats, Na(+)-dependent transport of L-proline, L-ala

Investigation and intervention of autophagy to guide cancer treatment with nanogels.

Cancer cells use autophagy to resist poor survival environmental conditions such as low PH, poor nutrients as well as chemical therapy. Nanogels have been used as efficient chemical drug carriers for cancer treatment. However, the effect of nanogels on autophagy is still unknown. Here, we used Rab proteins as the marker of multiple trafficking vesicles in endocytosis and LC3 as the marker of autophagy to investigate the intracellular trafficking network of Rhodamine B (Rho)-labeled nanogels. The nanogels were internalized by the cells through multiple protein dependent endocytosis and micropin

Mesothelial intercellular junctions and pathways.

Freeze-etch preparations of mesothelial cells taken from the peritoneum of mouse reveal the presence of vesicles invaginating the apical and the basal cell surfaces. These vesicles are scarcely seen within the cytoplasm. Long tortuous tubular profiles extend for considerable distance within the cytoplasm and are frequently associated with the vesicles. The possible nature and role of the vesicles and the tubules in transport phenomena across the mesothelial barrier, are discussed in relation to the pore theory advanced by physiologists and the "stomata" concept observed by early German and con

[Preparation of inside-out vesicles from red blood cell membranes and measurement for its Ca2+ uptake].

Sealed, inside-out red cell membrane vesicles (IOV) from rats were prepared by a modified method of Steck (1974). The preparation contained 70% of IOV. In the presence of ATP and Mg2+, these vesicles actively took up Ca2+ with a high efficiency and reproducibility. The intraassay coefficient of variance (C.V.) was 3.6% and the interassay C.V. was 13.58%. The active Ca2+ transport reached the maximal level in 10 minutes of incubation, and linearly correlated with the IOV concentration. The Ca2+ uptake rate in Wistar rats was 9.39 +/- 1.28 nmol/mg IOV protein/min. IOV rapidly lost Ca2+ when A231

Short-term studies of taurocholate uptake by ileal brush border membrane vesicles: anion effects.

Experiments allowing Na+-dependent short-term uptake measurements by ileal brush border vesicles were described. Glucose uptake was compared with taurocholate uptake in the presence of NaCl, NaSCN and Na2SO4. In contrast to the observation made with glucose, taurocholate transport was the same for the three electrolytes, indicating electroneutral taurocholate transport.

VE-Cadherin and Vesicles Differentially Regulate Lymphatic Vascular Permeability to Solutes of Various Sizes.

Lymphatic vascular permeability prevents lymph leakage that is associated with lymphedema, lymphatic malformations, obesity, and inflammation. However, the molecular control of lymphatic permeability remains poorly understood. Recent studies have suggested that adherens junctions and vesicle transport may be involved in regulating lymphatic vessel permeability. To determine the contribution of each transport pathway, we utilized an ex vivo permeability assay to directly measure the solute flux of various molecular weight solutes across a range of pressures in intact murine collecting lymphatic

Trans-Golgi network and subapical compartment of HepG2 cells display different properties in sorting and exiting of sphingolipids.

In HepG2 cells, the subapical compartment (SAC) is involved in the biogenesis of membrane polarity. By contrast, direct apical transport originating from the trans-Golgi network (TGN), which may contribute to polarity establishment, has been poorly defined in these cells. Thus, although newly synthesized sphingolipids can be directly transported from the TGN to the apical membrane, numerous apical resident proteins are traveling via the transcytotic route. Here, we developed an in vitro transport assay and compared the molecular sorting of 6-[N-(7-nitrobenz-2-oxa-1,3 diazol-4-yl)amino] hexanoy
