An update on proteases and protease inhibitors from trematodes.

Trematodes, a class of parasitic flatworms, are responsible for a variety of devastating diseases in humans and animals, with schistosomiasis and fascioliasis being prominent examples. Trematode proteolytic systems involved in the host-parasite interaction have emerged as key contributors to the success of trematodes in establishing and maintaining infections. This review concentrates on diverse proteases and protease inhibitors employed by trematodes and provides an update on recent advances in their molecular-level characterization, with a focus on function, structure, and therapeutic target

A case report of human gastrodiscoidiasis in Nepal.

Gastrodiscoidiasis is a snail-transmitted zoonotic infection caused by the digenean trematode Gastrodiscoides hominis, which is the only species in the genus Gastrodiscoides infecting humans. We report a case of human gastrodiscoidiasis in a 66 years-old Nepali man without history of travels outside of Nepal who was admitted in the hospital with pain in upper abdomen for 4 months with history of passage of black-tarry stools, anemia and eosinophilia. During upper endoscopy a living trematode worm was seen in the antrum of the stomach. After isolation, the worm was identified morphologically as

Diversity of trematode larvae in Indoplanorbis exustus from Phayao Lake, northern Thailand: Genetic evidence of Schistosoma spindale and a potential novel Schistosomatid associated with swimmer's itch.

Indoplanorbis exustus, a pulmonate snail, is an important component of freshwater ecosystems and serves as the primary intermediate host for the larval stages of several medically and veterinary significant trematodes. However, data on its infection status and associated trematode species in the Phayao Lake area of northern Thailand are currently lacking. This study aimed to investigate the prevalence of infection and identify the trematode species present in snails from this region. A total of 1290 snails were collected from Phayao Lake between November 2024 and February 2025 and examined usi

Potential Zoonotic Trematodes Recovered in Stray Cats from Kuwait Municipality, Kuwait.

Stray cats are a common feature roaming the streets and alleys of Kuwait; they could be a source of parasites, including trematodes, that affect humans. A survey was conducted to identify feline trematodes and throw the light on their public health significance in Kuwait. Out of 240 stray cats trapped from different localities of Kuwait from June 2011 to May 2012, 59 (24.6%) were found to be infected with 14 species of trematodes. The most common were trematodes of the genus Heterophyes, particularly H. heterophyes and H. dispar that were found in respectively 15.8% and 10.8% of the cats exami

Preliminary Survey of Larval Trematodes in Freshwater Snails of Phitsanulok Province in Lower Northern Thailand.

BACKGROUND: Freshwater snails serve as intermediate hosts for a variety of trematodes that cause illness in the human and animal populations. Several species of freshwater snails in Thailand have been found to have larval trematode infections. We aimed to investigate a freshwater snail in Phitsanulok Province and report on its current status of larval trematode infection. METHODS: Freshwater snails were collected from six localities (rice field and irrigation canal) by handpicking and using a count per unit of time sampling approach. The snails were identified by their external shell morpholo

Aestivation and its impact on the survival of snail intermediate hosts and trematode transmission in rice paddies.

In this study, we investigated the diversity and survival of aestivating snails in dry-season rice paddies, focusing on their role as intermediate hosts for trematodes. A total of 1,159 snails from various families were collected and analysed, revealing nine species, primarily from the Bithyniidae family, with Bithynia siamensis siamensis being the most abundant. Of the nine species collected, the most common species, B. s. siamensis, exhibited a relatively high survival rate of 81.5% throughout the three-month dry period post-harvest, whereas the remaining eight species had survival rates bel

Rare inventory of trematode diversity in a protected natural reserve.

In the face of ongoing habitat degradation and the biodiversity crisis, natural reserves are important refuges for wildlife. Since most free-living organisms serve as hosts to parasites, the diverse communities in protected areas can be expected to provide suitable habitats for a species-rich parasite fauna. However, to date, assessments of parasite diversity in protected nature reserves are rare. To expand our knowledge of parasite communities in natural habitats, we examined 1994 molluscs belonging to 15 species for trematode infections in a central European natural reserve. The parasitologi

Morphological and molecular data for Neocatatropis primorskii n. g. n. sp. and Notocotylus ikutai Sasaki & Nakao, 2021 (Trematoda: Notocotylidae).

Cercariae of two trematode species from the family Notocotylidae were found in the snails Radix auricularia and Boreoelona ussuriensis. Sexually mature flukes were obtained by experimentally infecting a duckling and a few chickens, respectively, with metacercariae from the found cercariae. Based on a combination of morphological and molecular (nuclear and mitochondrial markers) evidence, the trematodes grown in the duckling were identified as Notocotylus ikutai. The adult flukes obtained from the infected chickens were assigned to trematodes previously grouped into the genus Catatropis on the

DNA probes for the detection of Fasciola hepatica in snails.

Fasciola hepatica, also called the large liver fluke, is a trematode which can infect most mammals. Monitoring the infection rate of snails, which function as intermediate hosts and harbour larval stages of F. hepatica, is an important component of epidemiological studies on fascioliasis. For this purpose, DNA probes were generated which can be used for the detection of F. hepatica larvae in snails. Four highly repetitive DNA fragments were cloned in a plasmid vector and tested by Southern blot hybridization to the DNA of various trematodes for specificity and sensitivity. The probes Fhr-I, Fh

[Experimental study progress on tribendimidine, artemether and artesunate against Clonorchis sinensis and other trematodes].

Currently praziquantel is one of the major drugs used in treatment of schistosomiasis and other trematode infections. Recent experimental studies indicate that a new anthelmintic, tribendimidine, is used in the treatment of intestinal nematodes, also possesses effect against several species of trematodes including Clonorchis sinensis, Opisthorchis viverrini and Echinostoma caproni. Tribendimidine is even more effective against C. sinensis in rats that a single 300 mg/kg oral dose cures almost all of the animals treated, a lower cure dose than praziquantel (375-500 mg/kg). The anti-malarial dru

Trematode Fluke Procerovum varium as Cause of Ocular Inflammation in Children, South India.

Trematodes are recognized as a group of emerging parasites in tropical countries. We identified a trematode as a cause of ocular granulomas that developed in children who bathed in ponds or rivers in South India. DNA was isolated from patients' surgically excised granulomas and from the trematode cercariae (larvae) released by the snail Melanoides tuberculata in water in which the children bathed. Real-time and conventional PCRs were performed that targeted ribosomal DNA regions spanning the internal transcribed spacer 2 and 28S sequences of this trematode. The PCR-amplified products were subj

First record of Troglotrema acutum (Trematoda, Troglotrematidae) in European polecats Mustela putorius from Luxembourg.

We report the first record of the trematode Troglotrema acutum in skulls of European polecats (Mustela putorius) from Luxembourg. Thirty-one road-killed polecats from the northern and eastern parts of the country were examined. We found the trematode in three polecats from the administrative districts of Clervaux and Vianden in the northeast of the country. The parasites were detected in the frontal sinuses of one juvenile male as well as one adult of each sex. The animals were infected with six, nine and 13 adult trematodes, respectively, and we recovered 24 trematode eggs from one polecat. W

Heat sensitivity of first host and cercariae may restrict parasite transmission in a warming sea.

To predict global warming impacts on parasitism, we should describe the thermal tolerance of all players in host-parasite systems. Complex life-cycle parasites such as trematodes are of particular interest since they can drive complex ecological changes. This study evaluates the net response to temperature of the infective larval stage of Himasthla elongata, a parasite inhabiting the southwestern Baltic Sea. The thermal sensitivity of (i) the infected and uninfected first intermediate host (Littorina littorea) and (ii) the cercarial emergence, survival, self-propelling, encystment, and infecti

Hidden digenean diversity in two African protected areas: metabarcoding as a tool for parasite identification.

African protected areas support exceptional wildlife diversity, yet their parasite diversity remains poorly characterised. Documenting parasite biodiversity is increasingly urgent as global change alters species distributions and threatens to cause mass parasite extinctions. Digenean trematodes are thought to be particularly sensitive to environmental stressors due to their complex life cycles, involving gastropod intermediate hosts and a wide range of secondary and definitive hosts. This study used a metabarcoding approach targeting the nuclear 28S rDNA region with the aim of characterising t

Opisthorchis felineus and Metorchis bilis are the main agents of liver fluke infection of humans in Russia.

Liver fluke infections are gradually transforming from a local problem of individual geographic regions to a widespread problem. The observed expansion is likely to be connected with the ever-increasing intensity of traffic flow and migration of the infected carriers between cities, regions, and countries. Opisthorchis felineus, the trematode belonging to the family Opisthorchiidae, is a well known causative agent of the infection called opisthorchiasis. Metorchis bilis, also a member of the family Opisthorchiidae, causes metorchiasis, a disease very close to opisthorchiasis in symptomatology.
