First whole-genome sequence of Triatoma sanguisuga (Le Conte, 1855), vector of Chagas disease.

Triatoma sanguisuga is the most widespread triatomine bug species in the United States. The species vectors the human parasite Trypanosoma cruzi, which causes Chagas disease. Vector-borne Chagas disease is rarely diagnosed in the United States, but T. sanguisuga has been implicated in a handful of cases. Despite its public health importance, little is known about the genomics or population genetics of T. sanguisuga. Here, we used long-read sequencing to assemble the first whole-genome sequence for T. sanguisuga using DNA extracted from 1 adult specimen from Delaware. The final size of the geno

Quantitative imagery analysis of spot patterns for the three-haplogroup classification of Triatoma dimidiata (Latreille, 1811) (Hemiptera: Reduviidae), an important vector of Chagas disease.

BACKGROUND: Spots and coloring patterns evaluated quantitatively can be used to discriminate and identify possible cryptic species. Species included in the Triatoma dimidiata (Reduviidae: Triatominae) complex are major disease vectors of Chagas disease. Phylogenetic studies have defined three haplogroups for Mexico and part of Central America. We report here our evaluation of the possibility of correctly discriminating these three T. dimidiata haplogroups using the pattern of the dorsal spots. METHODS: Digital images of the dorsal region of individuals from the three haplogroups were used. Im

Population genetics of two chromatic morphs of the Chagas disease vector Rhodnius pallescens Barber, 1932 in Panamá.

Rhodnius pallescens is the principal vector of Chagas disease in Panama. Recently a dark chromatic morph has been discovered in the highlands of Veraguas Province. Limited genetic studies have been conducted with regards to the population structure and dispersal potential of Triatominae vectors, particularly in R. pallescens. Next generation sequencing methods such as RADseq and complete mitochondrial DNA (mtDNA) genome sequencing have great potential for examining vector biology across space and time. Here we utilize a RADseq method (3RAD), along with complete mtDNA sequencing, to examine the

Community participation and domiciliary occurrence of infected Meccus longipennis in two Mexican villages in Jalisco state.

The entomological features of Chagas disease in two western Mexican villages were analyzed through triatomines collection by the inhabitants and active research in the peridomicile. The inhabitant collections have the following comparable characteristics: 1) Meccus longipennis was the dominant species (> 91%), 2) around 43% of the insects were collected indoors, 3) about 70% of triatomines were adults, 4) cumulated rates of infestation of the dwellings reached 40-50%, 5) the triatomine infection rate by Trypanosoma cruzi was > 50%, and 6) the indoor triatomines frequently feed on humans (range

Agent of Chagas' disease from Honduran vector capable of developing in California insects: implications for cardiologists.

An exotic strain of Trypanosoma cruzi recovered from Triatoma dimidiata from Tegucigalpa, Honduras, was shown by isoenzyme studies to be closely related to the Miles' zymodeme 1 and laboratory reference strain Tehuantepec. It was injected into Swiss random-bred ICR mice. Clean Triatoma protracta nymphs and adults, which had been captured in Winters, California, fed on inoculated mice and were then examined over a 15-month period. Their feces contained multiplying epimastigote and infective trypomastigote forms of T. cruzi. This shows that exotic strains of T. cruzi can develop and survive for

[Chagas disease in Argentina: tools for schoolchildren to exercise vector surveillance and identify household risk factors].

The objective of this paper was to validate tools for schoolchildren to help control Chagas disease vectors in their own homes. Two types of tables were produced, one reporting on the presence of risk factors in residences and the other to record the presence of triatomines. The materials were tested in the field by 100 students from an endemic area in Argentina and validated by technical personnel. The results obtained by students concerning the percentage of households with risk factors and vector-positive residences were similar to those obtained by the technical personnel. This information

[Epidemiological surveillance of Chagas disease].

Chagas disease still constitutes an important medical problem in affected countries. In some, the extent of the disease is still unknown and control programs have not been implemented. In others the disease has been reduced due to regular control programs and other economic and social factors. Epidemiological surveillance with community participation to guard against disease transmission is now the basic challenge. Applied research and in-depth reformulation of health systems are required to establish efficient and sustainable Chagas disease surveillance programs, considering low density of pe

In vitro cytotoxicity of Rhodnius prolixus hemolytic factor and mellitin towards different trypanosomatids.

Trypanosoma cruzi strain Y and clone Dm28c and other trypanosomatids were exposed to two lytic agents, Rhodnius prolixus hemolytic factor (RHF) and mellitin, in vitro. In both cases, the result was a significant decrease in the number of parasites after a 30-min treatment at 37 degrees C. RHF and mellitin had distinct activities on different strains and species of trypanosomatids. These observations suggest that RHF may be an important factor in selecting resistant strains of trypanosomes for development in the vector's gut.

Palm trees and Chagas' disease in Panama.

An ecological survey of triatomines in the sylvan ecosystem of the Canal Zone and selected sites in Panama disclosed for the first time a close association of Rhodnius pullescens and Triatoma dimidiata, the two most important vector species of Chagas' disease in Panama, with a single species of a widely distributed palm tree, Scheelea zonensis. This association may explain why Chagas' disease is prevalent in certain rural communities in Central Panama and rare in others. An immense focus of zoonotic Trypanosoma cruzi infection exists in the forests of the Canal Zone with presence of large popu

Modelling American trypanosomiasis in an endemic zone: application to the initial spread of household infection in the Argentine Chaco.

The complex dynamics of Trypanosoma cruzi infection (Chagas disease) involves different actors and multiple transmission routes. Based on the information currently available, here, we propose a new and more comprehensive model to better understand the dynamics of the infection. This mathematical deterministic model was formulated considering: (i) the three clinical forms in humans: acute, chronic indeterminate and chronic with determinate pathology, (ii) the three main modes of transmission in the human population: vector-borne, congenital and transfusional, (iii) populations of triatomines an

Trypanosoma cruzi: specific detection of parasites by PCR in infected humans and vectors using a set of primers (BP1/BP2) targeted to a nuclear DNA sequence.

In the present work we evaluate Trypanosoma cruzi DNA detection by PCR using the nuclear oligonucleotides BP1/BP2 as primers. These primers are targeted to the 5' and 3' ends of the coding region for the flagellar protein F29. An amplification product of BP1/BP2 is a DNA band 692 bp long. Titration assays were performed to evaluate the minimum amount of parasite DNA that can be detected by this assay, resulting in 10 fg (equivalent to about 1/20 of the genome). The assay was also performed using T. cruzi DNA from different strains, clones, and human-derived isolates obtaining, in all cases, am

Influence of blood meal source on the biological parameters of Triatoma pallidipennis (Heteroptera: Reduviidae) from Mexico.

Chagas disease is one of the most important vector-borne diseases in Latin America, including Mexico. Triatoma pallidipennis (Stål) (Hemiptera: Reduviidae) is a Mexican triatomine vector commonly associated with different hosts. The influence of six blood meals (rabbits, rats, mice, dogs, cats and chickens) on six biological parameters of the biology of T. pallidipennis was evaluated. A significant difference was found in the period of egg-to-adult development between the five mammalian feeds (mean 195 days) and the chicken feed (221 days). The probability of survival was significantly lower i

[The national survey of seroprevalence for evaluation of the control of Chagas disease in Brazil (2001-2008)].

A survey for seroprevalence of Chagas disease was held in a representative sample of Brazilian individuals up to 5 years of age in all the rural areas of Brazil, with the single exception of Rio de Janeiro State. Blood on filter paper was collected from 104,954 children and screened in a single laboratory with two serological tests: indirect immunofluorescence and enzyme linked immunoassay. All samples with positive or indetermined results, as well as 10% of all the negative samples were submitted to a quality control reference laboratory, which performed both tests a second time, as well as t

Detection of Blastocrithidia spp. (Kinetoplastida: Trypanosomatidae) in Chagas disease vectors from Texas, USA.

Organisms highly similar to Blastocrithidia triatomae, a pathogenic parasite of Chagas disease triatomine bug vectors, were detected using polymerase chain reaction amplification and DNA sequence analysis of a segment of the small subunit rRNA gene in 3 of 203 triatomine specimens collected in Texas from June 2005 to October 2006. The parasite was identified in three species of triatomine bugs (Triatoma gerstaeckeri, T. indictiva, and T. neotomae) collected from three distinct geographic locations. Flagellated organisms indistinguishable from Trypanosoma cruzi were observed by direct microscop

In 1981, Kenyan authorities attempted eradication of tsetse (Glossina pallidipes) from the Lambwe valley, by sequential aerial spraying of endosulfan. Fly populations were reduced by over 99.9% in the main habitats, but recovered to their original levels within one year. In 1986, an Argentine research team attempted local elimination of domestic Triatominae (Triatoma infestans) in an area of Santiago del Estero. House infestation rates were reduced to an apparent zero, but recovered to pre-control levels in two years. In Sri Lanka, a combination of mosquito control with active case detection a

Melt With This Kiss: Paralyzing and Liquefying Venom of The Assassin Bug Pristhesancus plagipennis (Hemiptera: Reduviidae).

Assassin bugs (Hemiptera: Heteroptera: Reduviidae) are venomous insects, most of which prey on invertebrates. Assassin bug venom has features in common with venoms from other animals, such as paralyzing and lethal activity when injected, and a molecular composition that includes disulfide-rich peptide neurotoxins. Uniquely, this venom also has strong liquefying activity that has been hypothesized to facilitate feeding through the narrow channel of the proboscis-a structure inherited from sap- and phloem-feeding phytophagous hemipterans and adapted during the evolution of Heteroptera into a fan

Bionomics and Spatial Distribution of Triatomine Vectors of Trypanosoma cruzi in Texas and Other Southern States, USA.

Defining spatial and temporal occurrences of triatomine vectors of Trypanosoma cruzi, the agent of Chagas disease, in the US is critical for public health protection. Through a citizen science program and field collections from 2012 to 2016, we collected 3,215 triatomines, mainly from Texas. Using morphological and molecular approaches, we identified seven Triatoma species and report sex, length, and blood engorgement status. Many citizen-collected triatomines (92.9%) were encountered indoors, in peridomestic settings, or in dog kennels and represent spillover transmission risk of T. cruzi to

In July 2000, Heads of State of the 36th Session of the Organisation for African Unity signed a potentially important declaration on African trypanosomiasis, urging member states "to act collectively to rise to the challenge of eliminating the problem through concerted efforts in mobilising the necessary human, financial and material resources required to render Africa tsetse-free within the shortest time possible". To many, such an ambitious dream is received with some scepticism, recalling the doubts that surrounded a similar declaration signed in Brasilia in 1991, which paved the way for th
