Soil-transmitted helminth infections and associated risk factors in three Orang Asli tribes in Peninsular Malaysia.

Currently, information on prevalence of soil-transmitted helminth (STH) infections among different tribes of Orang Asli (aboriginal) is scarce in Malaysia. The present study is a cross-sectional study aimed at determining the factors associated with the prevalence of STH infections among the Proto-Malay, Negrito and Senoi tribes. Faecal samples were collected from 500 participants and socioeconomic data was collected via pre-tested questionnaire. All samples were processed using formalin-ether sedimentation and Wheatley's trichrome staining. Trichuris trichiura (57%) was the most common STH se

Global progress towards eliminating gastrointestinal helminth infections.

PURPOSE OF REVIEW: To highlight the gastrointestinal helminths in the context of renewed global commitment to control or eliminate neglected tropical diseases in the coming decade. RECENT FINDINGS: Two key documents, namely the 2012 London Declaration for Neglected Tropical Diseases and the 2013 World Health Assembly resolution, emphasize the importance of mass drug administration (MDA) for controlling several key neglected tropical diseases. These documents, together with the Global Burden of Disease Study 2010, establish the major gastrointestinal helminth infections, including the soil-tra

[Therapeutic possibilities in diseases caused by intestinal parasites].

Compatible and efficient drugs for the therapy, caused by protozoal- or helminthic-infections, are available today. Many of them are not registered in Austria. Metronidazole, Mebendazole and Praziquantel are particularly effective. Most protozoal-infections can be treated with metronidazole. Mebendazole can be used against numerous nematode-infections (Ascariasis, Trichuriasis, Oxyuriasis, Ancylostomiasis), Niclosamide against cestode-infections (Taeniasis, Hymenolepiasis, Diphyllobothriasis), whereas, Praziquantel is applied against trematode-infections.

Polypyridylruthenium(II) complexes exert in vitro and in vivo nematocidal activity and show significant inhibition of parasite acetylcholinesterases.

Over 4.5 billion people are at risk of infection with soil transmitted helminths and there are concerns about the development of resistance to the handful of frontline nematocides in endemic populations. We investigated the anti-nematode efficacy of a series of polypyridylruthenium(II) complexes and showed they were active against L3 and adult stages of Trichuris muris, the rodent homologue of the causative agent of human trichuriasis, T. trichiura. One of the compounds, Rubb12-mono, which was among the most potent in its ability to kill L3 (IC50 = 3.1 ± 0.4 μM) and adult (IC50 = 5.2 ± 0.3 μM)

Immune Profile of Honduran Schoolchildren with Intestinal Parasites: The Skewed Response against Geohelminths.

Soil-transmitted helminth infections typically induce a type-2 immune response (Th2), but no immunoepidemiological studies have been undertaken in Honduras, an endemic country where the main control strategy is children's annual deworming. We aimed to characterize the immune profile of Honduran schoolchildren harbouring these parasitoses. Demographic and epidemiological data were obtained through a survey; nutritional status was assessed through anthropometry; intestinal parasites were diagnosed by formol-ether and Kato-Katz; and blood samples were collected to determine immunological markers

Nutritional factors, parasite infection and allergy in rural and suburban Vietnamese school children.

Urban areas often have more allergy than rural areas. Dietary patterns and parasite infection have been suggested as possible related factors. This study evaluated the prevalence of allergy in school children in one rural and suburban area of Vietnam where parasite infection is common. A total of 195 children aged 9 to 13 years old completed a self-administered allergy questionnaire and provided blood and stool samples for analysis. Nutritional status, dietary intake and parasite infection were determined in all participants. Allergy was more common in girls (10.7% vs. 7.6%), suburban children

Efficacy of albendazole and its combinations with ivermectin or diethylcarbamazine (DEC) in the treatment of Trichuris trichiura infections in Sri Lanka.

The efficacy of the drugs currently available for treatment of infection with Trichuris trichiura is low compared with that of the drugs used against roundworm and hookworm. Single-dose combinations of albendazole with ivermectin or of albendazole with diethylcarbamazine (DEC) have recently been seen to produce raid and sustained reductions in Wuchereria bancrofti microfilaraemia. This observation prompted the present study, on the efficacy of these combinations against trichuriasis. The drug regimens tested were albendazole (400 mg) alone, albendazole (400 mg) with ivermectin (200 micrograms/

Survey for soil-transmitted helminths in Asahan Regency, North Sumatra, Indonesia.

The survey on soil-transmitted helminthiasis was carried out in the three villages of Asahan Regency, North Sumatra, Indonesia, between October 1981 and December 1982. The prevalence rates of geohelminthiasis proved to be extremely high (average 97%) in the three villages and more than 70% harboured three or more helminths, especially Ascaris lumbricoides, Trichuris trichiura and hookworm. Necator americanus and Ancylostoma duodenale were detected in 61.1% and 52.2% of 835 fecal specimens, respectively. The effects of two anthelmintics, Combantrin and Trivexan, were essentially the same agains

Trichuris trichiura egg extract proteome reveals potential diagnostic targets and immunomodulators.

Embryonated eggs are the infectious developmental stage of Trichuris trichiura and are the primary stimulus for the immune system of the definitive host. The intestinal-dwelling T. trichiura affects an estimated 465 million people worldwide with an estimated global burden of disease of 640 000 DALYs (Disability Adjusted Life Years). In Latin America and the Caribbean, trichuriasis is the most prevalent soil transmitted helminthiasis in the region (12.3%; 95% CI). The adverse health consequences impair childhood school performance and reduce school attendance resulting in lower future wage-earn

Flubendazole versus mebendazole in intestinal helminthic infections.

In a double-blind study the vermicidal effect of flubendazole, a new benzimidazole derivative, was evaluated and compared to mebendazole. Both drugs were administered in a single dose of 600 mg. While in the treatment of Ascaris and hookworm infestations flubendazole and mebendazole showed a similar efficacy, mebendazole seemed to be slightly superior in the treatment of trichuriasis. Both drugs were well tolerated and no side effects were observed even in patients with a heavy worm load.

[Treatment of intestinal nematode infections with albendazole. Preliminary results (author's transl)].

A randomized double-blind study, in order to evaluate effectiveness and tolerance of albendazole, was carried out in 186 african subjects presenting intestinal nematode infections in Mali and Senegal. The drug, well tolerated, given as a single dose of 0.4 g (adolescents and adults) or 0.2 g (children) is curative in more than 90% for ascariasis. For ancylostomiasis and trichuriasis, the effectiveness is less satisfactory in children, probably due to insufficient dosage in this age group.

Structural insights into Trichuris muris eggs through 3D modeling, Cryo-SEM, and TEM of samples prepared with HPF-FS.

Trichuriasis - a disease caused by Trichuris trichiura - affects underserved communities. Infection occurs by the ingestion of embryonated eggs, a resilient structure against environmental fluctuations - an essential feature for the survival and transmission of trichurids. This study aims to enhance our comprehension of the trichurid eggs by providing a characterization of the ultrastructure of eggshell and first-stage (L1) larvae of T. muris, a key experimental model for trichuriasis. We employed the following microscopy techniques: light, fluorescence, confocal, Cryo-SEM, and Transmission el

Efficacy and safety of ivermectin-albendazole combination versus ivermectin or albendazole monotherapy in soil-transmitted helminthiasis: A systematic review and meta-analysis.

BACKGROUND: Soil-transmitted helminthiasis remains a daunting challenge to global health, exerting its greatest toll on resource-limited regions of the world. A dual drug approach using the co-administration of ivermectin and albendazole has shown promising results in comparison to the traditional monotherapy strategy. In light of this, a systematic review and meta-analysis of randomized controlled trials was conducted. METHODS: Several electronic databases including PubMed, Cochrane Central, Google Scholar, and Embase were explored to search for relevant studies from inception to September 2

Comparative trials using albendazole and mebendazole in the treatment of soil-transmitted helminths in schoolchildren on Penang, Malaysia.

Trials using albendazole and mebendazole, as single 400 mg dose treatments, against soil-transmitted helminths, were carried out in 7-9 and 10-12 years-old schoolchildren living in urban and rural environments in Penang, Malaysia. Both drugs were equally effective in treating trichuriasis and ascariasis in both age groups and environments. However, mebendazole is not so effective in the treatment for hookworms when compared to albendazole. It is suggested that albendazole should be considered the drug of choice for mass chemotherapy for Penang.

Public health significance of intestinal parasitic infections. WHO Expert Committee.

Intestinal parasitic infections are distributed virtually throughout the world, with high prevalence rates in many regions. Amoebiasis, ascariasis, hookworm infection and trichuriasis are among the ten most common infections in the world. Other parasitic infections such as abdominal angiostrongyliasis, intestinal capillariasis, and strongyloidiasis are of local or regional public health concern. The prevention and control of these infections are now more feasible than ever before owing to the discovery of safe and efficacious drugs, the improvement and simplification of some diagnostic procedu

Adverse metabolic effects of antiparasitic drugs.

Several drugs are available for mass campaigns against enteric helminths. Mebendazole and pyrantel pamoate are quite effective against ascariasis and reasonably effective against the hookworms. Only mebendazole is effective against trichuriasis. Two drugs have recently been introduced for mass therapy of schistosomiasis, but they remain experimental, pending further evaluation. Other drugs are mentioned, and possible adverse metabolic effects are discussed. The fundamental question remaining is whether the eradication of certain parasites will improve the nutritional status of the infected pop

The Use of MALDI-TOF MS as a Diagnostic Tool for Adult Trichuris Species.

Trichuriasis is considered a neglected tropical disease, being the second most common helminthiasis in humans. Detection of Trichuris in routine diagnosis is usually done by microscopic detection of eggs in fecal samples. Other molecular analyses are more reliable and could be used, but these analyses are not routinely available in clinical microbiology laboratories. The use of matrix-assisted laser desorption/ionization-time of flight (MALDI-TOF) mass spectrometry (MS) is increasing since the last decades due to its recent evidence as a potential role for reliable identification of microorgan
