Diagnostic Accuracy of Various Genetic Markers (tpi, gdh, and bg) for Prevalence and Genotyping of Giardia duodenalis in Human Samples: A Comparative Global Systematic Review and Meta-Analysis.

BACKGROUND: Giardia duodenalis is a widespread intestinal protozoan causing giardiasis in humans and animals. Precise detection and genotyping of this parasite are vital for understanding its epidemiology, implementing appropriate treatments, and mitigating transmission. Molecular techniques, particularly multilocus genotyping (MLG) targeting the beta-giardin (bg), triose phosphate isomerase (tpi), and glutamate dehydrogenase (gdh) genes, have improved diagnostic precision. However, variations in sensitivity and diagnostic performance among these genes can influence prevalence rates and genoty

Multilocus genotyping of Giardia duodenalis in Tibetan sheep and yaks in Qinghai, China.

Giardia duodenalis is a common gastrointestinal protozoon in mammals. Although many studies have been reported on the distribution of G. duodenalis genotypes in sheep and cattle raised under intensive farming, few studies are available on the distribution of G. duodenalis in Tibetan sheep and yaks, which are raised free ranging in a continental plateau climate. In this study, 495 fecal specimens from Tibetan sheep and 605 from yaks were collected from eight counties in Qinghai, China and analyzed for G. duodenalis by PCR targeting the β-giardin (bg), glutamate dehydrogenase (gdh), and trioseph

Synergistic Interaction of Light Alcohol Administration in the Presence of Mild Iron Overload in a Mouse Model of Liver Injury: Involvement of Triosephosphate Isomerase Nitration and Inactivation.

It is well known that iron overload promotes alcoholic liver injury, but the doses of iron or alcohol used in studies are usually able to induce liver injury independently. Little attention has been paid to the coexistence of low alcohol consumption and mild iron overload when either of them is insufficient to cause obvious liver damage, although this situation is very common among some people. We studied the interactive effects and the underlining mechanism of mild doses of iron and alcohol on liver injury in a mouse model. Forty eight male Kunming mice were randomly divided into four groups:

Triosephosphate isomerase deficiency: haemolytic anaemia, myopathy with altered mitochondria and mental retardation due to a new variant with accelerated enzyme catabolism and diminished specific activity.

A new triosephosphate isomerase (TPI) variant is described in an 8-year-old Turkish girl suffering from chronic haemolytic anaemia, myopathy and developmental retardation since early infancy. The enzyme activity profile revealed a generalized deficiency in erythrocytes, granulocytes, mononuclear blood cells, skeletal muscle tissue and cerebrospinal fluid. The concentration of enzyme substrate dihydroxyacetone phosphate was distinctly elevated. Biochemical examination showed accelerated enzyme deamidation, the first step in the normal catabolism of TPI during aging of the erythrocyte. The speci

Medical and Veterinary Importance of the Moonlighting Functions of Triosephosphate Isomerase.

Triosephosphate isomerase is the fifth enzyme in glycolysis and its canonical function is the reversible isomerization of glyceraldehyde-3-phosphate and dihydroxyacetone phosphate. Within the last decade multiple other functions, that may not necessarily always involve catalysis, have been described. These include variations in the degree of its expression in many types of cancer and participation in the regulation of the cell cycle. Triosephosphate isomerase may function as an auto-antigen and in the evasion of the immune response, as a factor of virulence of some organisms, and also as an im

Novel giardicidal compounds bearing proton pump inhibitor scaffold proceeding through triosephosphate isomerase inactivation.

Giardiasis is a worldwide parasitic disease that affects mainly children and immunosuppressed people. Side effects and the emergence of resistance over current used drugs make imperative looking for new antiparasitics through discovering of new biological targets and designing of novel drugs. Recently, it has determined that gastric proton-pump inhibitors (PPI) have anti-giardiasic activity. The glycolytic enzyme, triosephosphate isomerase (GlTIM), is one of its potential targets. Therefore, we employed the scaffold of PPI to design new compounds aimed to increase their antigiardial capacity b

Antibodies to triosephosphate isomerase in patients with neuropsychiatric lupus.

Sera from patients with neuropsychiatric lupus (NP lupus) were screened for antibodies to mouse choroid plexus cell line ECPC-4 by Western blotting. A 29-kDa protein band detected in NP lupus sera was identified as triosephosphate isomerase (TPI). Using Western blotting with TPI, TPI was confirmed as the reactive molecule in sera (6 of 14 samples) and in cerebrospinal fluids (1 of 2 samples) of patients with NP lupus. Enzyme-linked immunosorbent assay with TPI showed that the serum anti-TPI antibody index of 89.8 (SD, 70.1) in NP lupus group was significantly higher than in systemic lupus eryt

Integrating multi-omics reveals the mechanisms of elevated [CO2] enhances salt tolerance of tobacco plants with a photosynthetic regulatory network.

Abiotic stresses often occur concurrently with global changes, especially the combination of salt stress and elevated CO2 concentration ([CO2]) is a common phenomenon in salt-affected soils throughout the world. The synergistic regulatory mechanisms of tobacco (Nicotiana tabacum L.) plants in response to combined salt stress and elevated [CO2] are still poorly understood. This study aimed to reveal the potential mechanisms of elevated [CO2] modulating leaf photosynthesis of tobacco plants subjected to salt stress by integrating physiological, transcriptional, and metabolomics analyses. An optimized fast-repetition-rate fluorometry method reveals photochemical flux bottlenecks in C3 photosynthesis that limit carboxylation and growth.

Using the fast-repetition-rate fluorometry (FRRF) technique to monitor the variable yield of chlorophyll fluorescence emission (Fv = Fm - Fo), we develop a pulse sequence to measure flux bottlenecks that limit the transport of electrons and protons from their water oxidation source through the photosynthetic electron transport chain (PETC) into CO2 carboxylation and triose phosphate production within the Calvin-Benson-Bassham (CBB) cycle of living organisms. FRRF uses a single source of microsecond flashes to excite photochemistry and produce fluorescence, thus removing slower non-photochemica

Machine learning-driven discovery of antimicrobial peptides targeting the GAPDH-TPI protein-protein interaction in Schistosoma mansoni for novel antischistosomal therapeutics.

Schistosomiasis, caused by Schistosoma mansoni, remains a significant public health burden, particularly in endemic regions with limited access to effective treatment. The emergence of resistance to praziquantel necessitates the urgent discovery of novel therapeutic targets. This study explores the potential of antimicrobial peptides (AMPs) as inhibitors of the glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and triose phosphate isomerase (TPI) protein-protein interaction (PPI) in S. mansoni, a crucial glycolytic pathway component essential for parasite survival. A machine learning-driven app

Companion animals as a potential source of Giardia intestinalis infection in humans in the Czech Republic - A pilot study.

Giardia intestinalis is a common enteric single-celled parasite infecting both humans and animals. Its eight morphologically identical but genetically distinct groups called assemblages differ from each other in host range. While assemblages A and B infect a wide range of hosts, including humans, the other assemblages (C to H) limit their host preferences to particular animal groups only. In companion animals as Giardia hosts, genotyping data have previously shown various results depending on pet species, location, environmental or breeding conditions, and the study design. Frequency, diagnostic performance of coproantigen detection and genotyping of the Giardia among patients referred to a multi-level teaching hospital in northern India.

Giardiasis, a common gastrointestinal parasitic infection in tropics, is diagnosed on stool microscopy (gold standard); however, its sensitivity is low due to intermittent fecal shedding. Coproantigen detection (ELISA) is useful but requires further evaluation. We aimed to study: (a) detection of Giardia by stool microscopy and/or coproantigen, (b) diagnostic performance of fecal antigen detection and microscopy, and c) genotypic characterization of G. lamblia using PCR specific for triose phosphate isomerase (tpi) gene. Stool samples from 2992 patients were examined by microscopy from March 2

Multilocus genotyping of Giardia duodenalis in Malaysia.

Giardia duodenalis is considered the most common intestinal parasite in humans worldwide. In Malaysia, many studies have been conducted on the epidemiology of giardiasis. However, there is a scarcity of information on the genetic diversity and the dynamics of transmission of G. duodenalis. The present study was conducted to identify G. duodenalis assemblages and sub-assemblages based on multilocus analysis of the glutamate dehydrogenase (gdh), beta-giardin (bg) and triose phosphate isomerase (tpi) genes. Faecal specimens were collected from 484 Orang Asli children with a mean age of 7 years an

Occurrence and genetic characterization of Giardia duodenalis from captive nonhuman primates by multi-locus sequence analysis.

Giardia is the most common enteric protozoan that can be pathogenic to both humans and animals. Transmission can be direct through the faecal-oral route, or through ingestion of contaminated water or food. Genetic characterization of Giardia duodenalis isolates has demonstrated the existence of seven groups (assemblages A to G) which differ in their host distribution. Assemblages A and B are present in humans and other primates, dogs, cats, rodents, and other species of wild mammals, but the role of the different host animals in the epidemiology of human infection remains unclear. Genotyping and distribution of Giardia intestinalis assemblages in NSW, Australia.

Giardiasis is the most common enteric protozoan infection notifiable in New South Wales (NSW), Australia. Surveillance by NSW Health had shown a steady increase (prior to the COVID-19 pandemic) in the number of cases reported since 2012 and the reasons for this currently remain unknown. This study aimed to investigate the occurrence of Giardia intestinalis assemblages causing human infection in NSW. Individual faecal specimens were collected from participating hospitals and private laboratories, and the presence of Giardia and co-infections was confirmed by real-time multiplex-polymerase chain
