Closed-state inactivation and pore-blocker modulation mechanisms of human CaV2.2.

N-type voltage-gated calcium (CaV) channels mediate Ca2+ influx at presynaptic terminals in response to action potentials and play vital roles in synaptogenesis, release of neurotransmitters, and nociceptive transmission. Here, we elucidate a cryo-electron microscopy (cryo-EM) structure of the human CaV2.2 complex in apo, ziconotide-bound, and two CaV2.2-specific pore blockers-bound states. The second voltage-sensing domain (VSD) is captured in a resting-state conformation, trapped by a phosphatidylinositol 4,5-bisphosphate (PIP2) molecule, which is distinct from the other three VSDs of CaV2.2

Gap junction channels reconstituted in two closely apposed lipid bilayers.

Intercellular communication mediated by gap junction channels plays an important role in many cellular processes. In contrast to other channels, gap junction channels span two plasma membranes resulting in an intracellular location for both ends of the junctional pore and the regulatory sites for channel gating. This configuration presents unique challenges for detailed experimental studies of junctional channel physiology and ligand-activation in situ. Availability of an appropriate model system would significantly facilitate future studies of gap junction channel function and structure. Here

P2X7 Receptor Triggers Lysosomal Leakage Through Calcium Mobilization in a Mechanism Dependent on Pannexin-1 Hemichannels.

The P2X7 receptor is a critical purinergic receptor in immune cells. Its activation was associated with cathepsin release into macrophage cytosol, suggesting its involvement in lysosomal membrane permeabilization (LMP) and leakage. Nevertheless, the mechanisms by which P2X7 receptor activation induces LMP and leakage are unclear. This study investigated cellular mechanisms associated with endosomal and lysosomal leakage triggered by P2X7 receptor activation. We found that ATP at 500 μM and 5 mM (but not 50 μM) induced LMP in non-stimulated peritoneal macrophages. This effect was not observed i

Sodium channels, inherited epilepsy, and antiepileptic drugs.

Voltage-gated sodium channels initiate action potentials in brain neurons, mutations in sodium channels cause inherited forms of epilepsy, and sodium channel blockers-along with other classes of drugs-are used in therapy of epilepsy. A mammalian voltage-gated sodium channel is a complex containing a large, pore-forming α subunit and one or two smaller β subunits. Extensive structure-function studies have revealed many aspects of the molecular basis for sodium channel structure, and X-ray crystallography of ancestral bacterial sodium channels has given insight into their three-dimensional struc

Pancreatic β-cell KATP channels: Hypoglycaemia and hyperglycaemia.

The pancreatic β-cell ATP-sensitive K(+) channel (K(ATP) channel) plays a critical role in glucose homeostasis by linking glucose metabolism to electrical excitability and insulin secretion. Changes in the intracellular ratio of ATP/ADP mediate the metabolic regulation of channel activity. The β-cell K(ATP) channel is a hetero-octameric complex composed of two types of subunits: four inward-rectifying potassium channel pore-forming (Kir6.2) subunits and four high-affinity sulfonylurea receptor 1 (SUR1) subunits. Kir6.2 and SUR1 are encoded by the genes KCNJ11 and ABCC8, respectively. Mutations

A Kir6.2 mutation causing severe functional effects in vitro produces neonatal diabetes without the expected neurological complications.

AIMS/HYPOTHESIS: Heterozygous activating mutations in the pancreatic ATP-sensitive K+ channel cause permanent neonatal diabetes mellitus (PNDM). This results from a decrease in the ability of ATP to close the channel, which thereby suppresses insulin secretion. PNDM mutations that cause a severe reduction in ATP inhibition may produce additional symptoms such as developmental delay and epilepsy. We identified a heterozygous mutation (L164P) in the pore-forming (Kir6.2) subunit of the channel in three unrelated patients and examined its functional effects. METHODS: The patients (currently aged

Pannexin-1 channels contribute to seizure generation in human epileptic brain tissue and in a mouse model of epilepsy.

Epilepsies are characterized by recurrent seizures, which disrupt normal brain function. Alterations in neuronal excitability and excitation-inhibition balance have been shown to promote seizure generation, yet molecular determinants of such alterations remain to be identified. Pannexin channels are nonselective, large-pore channels mediating extracellular exchange of neuroactive molecules. Recent data suggest that these channels are activated under pathological conditions and regulate neuronal excitability. However, whether pannexin channels sustain or counteract chronic epilepsy in human pat

Structural insights into the mechanism of activation of the TRPV1 channel by a membrane-bound tarantula toxin.

Venom toxins are invaluable tools for exploring the structure and mechanisms of ion channels. Here, we solve the structure of double-knot toxin (DkTx), a tarantula toxin that activates the heat-activated TRPV1 channel. We also provide improved structures of TRPV1 with and without the toxin bound, and investigate the interactions of DkTx with the channel and membranes. We find that DkTx binds to the outer edge of the external pore of TRPV1 in a counterclockwise configuration, using a limited protein-protein interface and inserting hydrophobic residues into the bilayer. We also show that DkTx pa

Structural model of the outer vestibule and selectivity filter of the Shaker voltage-gated K+ channel.

A new generation of structural models were developed of the outer vestibule and ion-selective portion of the voltage-gated Shaker K+ channel. Some features of these models are similar to those that we have developed previously [Durrel S. R. and Guy H. R. (1992) Biophys. J. 62, 238-250; Guy H. R. A strongly interacting pair of residues on the contact surface of charybdotoxin and a Shaker K+ channel.

Charybdotoxin, a peptide neurotoxin of known molecular structure, blocks Shaker K+ channels by binding to a receptor at the outer opening of the ion conduction pathway. Analysis of variants of CTX at position 29 and of Shaker at position 449 shows that these two residues interact closely in the channel-toxin complex. The CTX mutation M29I leads to a slight strengthening of block when tested on Shaker-449T; the same CTX mutation weakens block 1700-fold when tested on Shaker-449F. The known position of CTX-29 on the toxin's interaction surface thus locates Shaker-449 within 5 A of the pore axis

Binding modes of two scorpion toxins to the voltage-gated potassium channel kv1.3 revealed from molecular dynamics.

Molecular dynamics (MD) simulations are used to examine the binding modes of two scorpion toxins, margatoxin (MgTx) and hongotoxin (HgTx), to the voltage gated K+ channel, Kv1.3. Using steered MD simulations, we insert either Lys28 or Lys35 of the toxins into the selectivity filter of the channel. The MgTx-Kv1.3 complex is stable when the side chain of Lys35 from the toxin occludes the channel filter, suggesting that Lys35 is the pore-blocking residue for Kv1.3. In this complex, Lys28 of the toxin forms one additional salt bridge with Asp449 just outside the filter of the channel. On the other

Covalent Organic Framework Mediated Polymerization for Multifunctional Applications.

Polymers constitute indispensable functional components across energy, environmental, and biomedical applications, yet precise control over their primary and higher-order structures remains a core challenge. Covalent organic frameworks (COFs), a class of crystalline porous materials featuring designable topologies, tunable chemical compositions, and permanent ordered pore channels, provide a molecular-level platform for monomer polymerization through their inherent pore confinement and interfacial synergistic effects. This review systematically summarizes two principal strategies for COF-media

Potassium benzoate-cyclodextrin metal-organic frameworks with multi-dimensional cavities: An adsorbent for removing aldehyde fishy odorants in aquatic products.

Fishy odorants substantially impair the flavor and quality of aquatic products, creating an urgent need for safe and effective deodorization methods. Herein, potassium benzoate-cyclodextrin metal-organic frameworks (PB-CD-MOFs) with multi-dimensional cavities was formulated to remove fishy odorants (2,4-decadienal, 4-heptenal, and nonanal). And the underlying adsorption mechanism of PB-CD-MOFs for fishy odorants was clarified by adsorption kinetics, isotherms, thermodynamics, and density functional theory (DFT) calculations. Based on the Single-crystal X-ray diffraction (SCXRD) analysis, PB-CD

IcmR-regulated membrane insertion and efflux by the Legionella pneumophila IcmQ protein.

Legionella pneumophila proliferates within alveolar macrophages as a central property of Legionnaires' disease. Intracellular growth involves formation of a replicative phagosome, which requires the bacterial Dot/Icm system, a multiprotein secretion apparatus that translocates proteins from the bacterium across the macrophage plasma membrane. Two components of this system, IcmR and IcmQ, are proposed to exhibit a chaperone/substrate relationship similar to that observed in other protein translocation systems. We report here that IcmQ inserts into lipid membranes and forms pores that allow the

Channel Dysfunction as the Basis for Comorbidities in Multiple Sclerosis and Depression.

Multiple sclerosis (MS) is a chronic inflammatory demyelinating disease of the central nervous system that is frequently accompanied by major depressive symptoms, which substantially worsen disease burden and quality of life. Increasing evidence suggests that this comorbidity is not solely a psychological reaction to neurological disability but may arise from shared neurobiological mechanisms. Ion channels play a central role in regulating neuronal excitability, synaptic transmission and immune cell function, and their dysregulation has emerged as a key feature of MS pathology. Demyelination a

Bisphenol A differently inhibits CaV3.1, Ca V3.2 and Ca V3.3 calcium channels.

Bisphenol A (BPA) is a widespread environmental contaminant detected in urine of 93 % of investigated US population. Recent epidemiological studies found correlation between BPA exposure and diseases including cardiovascular and neuronal disorders. BPA targets include hormone receptors and voltage-dependent ion channels. T-type calcium channels are important regulatory elements in both cardiovascular and neuronal system. Therefore, we investigated effects of BPA on T-type calcium channels. Calcium current flowing through recombinant T-type calcium channels expressed in HEK 293 cells was measur

Molecular Basis for Activation to Inhibition Switching in Kv7.2 Channel Modulators.

Kv7 voltage-gated potassium channels play a critical role in controlling electrical properties of excitable tissues. Neuronally-expressed Kv7 channels are involved in both common and rare neuropsychiatric disorders, ranging from epilepsy to depression and neurodegenerative diseases; thus, they represent attractive therapeutic targets. However, no Kv7 modulator is currently available for clinical use. Improved knowledge of the functional and structural determinants responsible for ligand-induced Kv7 channel modulation is likely to fill this gap. In the present work, we describe the cryo-electro

Emerging roles of mechanosensitive ion channels in acute lung injury/acute respiratory distress syndrome.

Acute lung injury/acute respiratory distress syndrome (ALI/ARDS) is a devastating respiratory disorder with high rates of mortality and morbidity, but the detailed underlying mechanisms of ALI/ARDS remain largely unknown. Mechanosensitive ion channels (MSCs), including epithelial sodium channel (ENaC), Piezo channels, transient receptor potential channels (TRPs), and two-pore domain potassium ion (K2P) channels, are highly expressed in lung tissues, and the activity of these MSCs can be modulated by mechanical forces (e.g., mechanical ventilation) and other stimuli (e.g., LPS, hyperoxia). A designer saxitoxin-engineered sodium channel pair as a general tool for subtype-selective inhibition.

We have used small molecule chemical design and protein engineering to fashion an orthogonal ligand-channel pair that enables acute and selective inhibition of a genetically encoded voltage-gated sodium channel (NaV). A modified ligand based on the nerve toxin, saxitoxin (STX), binds potently and reversibly to a NaV bearing two unique amino acid mutations in the outer mouth of the channel (Site 1). The same ligand is ineffective at blocking wild-type NaVs and does not disrupt action potential signals in neuronal cells or brain tissue at working concentrations. The versatility of this tool deri

P66 porins are present in both Lyme disease and relapsing fever spirochetes: a comparison of the biophysical properties of P66 porins from six Borrelia species.

The genus Borrelia is the cause of the two human diseases: Lyme disease (LD) and relapsing fever (RF). Both LD and RF Borrelia species are obligate parasites and are dependent on nutrients provided by their hosts. The first step of nutrient uptake across the outer membrane of these Gram-negative bacteria is accomplished by water-filled channels, so-called porins. The knowledge of the porin composition in the outer membranes of the different pathogenic Borrelia species is limited. Only one porin has been described in relapsing fever spirochetes to date, whereas four porins are known to be prese
