Adverse muscle composition predicts all-cause mortality in the UK Biobank imaging study.

BACKGROUND: Adverse muscle composition (MC) as measured by magnetic resonance imaging has previously been linked to poor function, comorbidity, and increased hospitalization. The aim of this study was to investigate if adverse MC predicts all-cause mortality using data from UK Biobank. METHODS: There were 40 178 participants scanned using a 6 min magnetic resonance imaging protocol. Images were analysed for thigh fat-tissue free muscle volume and muscle fat infiltration (MFI) using AMRA® Researcher (AMRA Medical, Linköping, Sweden). For each participant, a sex, weight, and height invariant mu

Novel disease associations with schizophrenia genetic risk revealed in ~400,000 UK Biobank participants.

Schizophrenia is a serious mental disorder with considerable somatic and psychiatric morbidity. It is unclear whether comorbid health conditions predominantly arise due to shared genetic risk or consequent to having schizophrenia. To explore the contribution of genetic risk for schizophrenia, we analysed the effect of schizophrenia polygenic risk scores (PRS) on a broad range of health problems in 406 929 individuals with no schizophrenia diagnosis from the UK Biobank. Diagnoses were derived from linked health data including primary care, hospital inpatient records, and registers with informat

Genome-wide meta-analyses of stratified depression in Generation Scotland and UK Biobank.

Few replicable genetic associations for Major Depressive Disorder (MDD) have been identified. Recent studies of MDD have identified common risk variants by using a broader phenotype definition in very large samples, or by reducing phenotypic and ancestral heterogeneity. We sought to ascertain whether it is more informative to maximize the sample size using data from all available cases and controls, or to use a sex or recurrent stratified subset of affected individuals. To test this, we compared heritability estimates, genetic correlation with other traits, variance explained by MDD polygenic

Visual Function, Social Position, and Health and Life Chances: The UK Biobank Study.

IMPORTANCE: The adverse impact of visual impairment and blindness and correlations with socioeconomic position are known. Understanding of the effect of the substantially more common near-normal vision (mild impairment) and associations with social position as well as health and life chances is limited. OBJECTIVE: To investigate the association of visual health (across the full acuity spectrum) with social determinants of general health and the association between visual health and health and social outcomes. DESIGN, SETTING, AND PARTICIPANTS: A cross-sectional epidemiologic study was conduc

Heaviness, health and happiness: a cross-sectional study of 163066 UK Biobank participants.

BACKGROUND: Obesity is known to increase the risk of many diseases and reduce overall quality of life. This study examines the relationship with self-reported health (SRH) and happiness. METHODS: We conducted a cross-sectional study of the 163 066 UK Biobank participants who completed the happiness rating. The association between adiposity and SRH and happiness was examined using logistic regression. SRH was defined as good (excellent, good), or poor (fair, poor). Self-reported happiness was defined as happy (extremely, very, moderately) or unhappy (moderately, very, extremely). RESULTS: Poo

Mechanistic 5'UTR Variant Scoring Expands Rare Variant Discovery in the UK Biobank.

The 5' untranslated region (5'UTR) regulates protein output through upstream open reading frames (uORFs) and Kozak context, yet most deleteriousness scores rely heavily on evolutionary conservation of its nucleotide positions. Using 5ULTRA, a machine-learning classifier trained on 5'UTR regulatory biology, we annotated rare and low-frequency 5'UTR variants in 408,423 UK Biobank participants. We tested gene-level associations for all 59 quantitative blood-count and serum biochemistry traits. We identified 58 genome-wide significant gene-phenotype associations and CADD 38, including 24 shared, 3

Night-shift intensity, duration, cardiovascular susceptibility and incident cardiovascular disease: UK Biobank study.

OBJECTIVE: Night-shift work has been associated with adverse cardiovascular outcomes. It remains unclear whether the cardiovascular association of night-shift exposure differs according to underlying baseline cardiovascular susceptibility and whether short-term intensity and longer-term duration show similar patterns. METHODS: We analyzed 85,322 UK Biobank participants with occupational night-shift information. Baseline cardiovascular disease risk profile was classified as low, intermediate, or high using a composite score based on cardiovascular disease polygenic risk score tertiles, current

The choroid plexus, cognitive decline, and incident dementia: insights from the UK Biobank.

The choroid plexus (CP) plays a crucial role in cerebrospinal fluid secretion and the maintenance of brain homeostasis. Its structure and function have been implicated in the pathogenesis of dementia; however, the longitudinal associations of CP with dementia and structural brain biomarkers remain unclear. This prospective cohort study utilized data from the UK Biobank, including 45,306 participants (mean age, 64 years; 47.2% men) who underwent 3.0 T multiparametric brain MRI scans. CP volume and signal intensity were quantified by FreeSurfer software. Measures of grey or white matter macrostr

Population-Scale Plasma Proteomic Profiles Associated with Chronic Periodontitis in the UK Biobank.

Periodontitis is a chronic infectious disease characterized by the destruction of the tooth-supporting tissues, including the gingiva, periodontal ligament, and alveolar bone, which may ultimately lead to tooth loss. However, blood-based biomarkers reflecting systemic inflammation in periodontitis remain poorly defined. We analyzed plasma proteomic data from the UK Biobank using Olink Explore proteomics to identify systemic protein signatures distinguishing chronic periodontitis patients (n = 90) from healthy controls (n = 2234). Among 2151 proteins passing quality control, 29 proteins showed

Disentangling the relationship between glucose, insulin and brain health: A UK Biobank study.

BACKGROUND: Glycaemic traits are associated with poorer brain health and dementia risk. Recent advances in genetic instruments for specific glycaemic markers enable an in-depth investigation of the likely nature of associations and underlying mechanisms between diabetes-related mechanisms and brain health and dementia. METHODS: We used two-sample Mendelian randomisation (MR) in the UK Biobank (UKB) (maximum N = 357 883 White British, mean age 56.9 years, 54% female) applying inverse-variance weighted MR as our main estimator alongside MR-Egger, weighted median estimator (WME) and Mendelian Ra

Estimated Small, Dense LDL Cholesterol and Atherosclerotic Cardiovascular Risk in the UK Biobank.

BACKGROUND: A key step in primary prevention is the assessment of atherosclerotic cardiovascular disease (ASCVD) risk. Risk enhancer tests are additional tools used to further improve ASCVD risk assessment over conventional risk markers. Our objective was to determine whether estimated small, dense low-density lipoprotein cholesterol (E-sdLDL-C) can improve risk assessment and serve as a new risk enhancer test. METHODS: We used a prospective cohort analysis of participants in the UK Biobank study with a median (interquartile range) follow-up of 10 (6.7-12.3) years. We included 271 760 individ

Association of bilirubin with neurodegenerative disorders: A prospective cohort study from UK biobank.

BACKGROUND: Emerging evidence suggests complex interaction between bilirubin, a heme catabolism byproduct, and neurodegenerative disorders (NDDs), though existing studies report inconsistent associations. We aimed to investigate longitudinal relationship between serum bilirubin levels and NDD risk in a large population-based cohort. METHODS: Utilizing data from the UK Biobank, we investigated the association between baseline direct bilirubin (DBIL) and total bilirubin (TBIL) and the risk of common NDDs using Cox proportional hazards regression analysis. We further performed interaction and su

Sex-specific risk prediction models for aneurysmal subarachnoid hemorrhage-A UK Biobank study.

BACKGROUND: We recently developed and validated the SMA2SH2ERS risk prediction model for aneurysmal subarachnoid hemorrhage (ASAH) in the general population (c-statistic 0.62; 95% confidence interval [CI] 0.60-0.64). Given that women have higher ASAH incidence than men, and that predictors for ASAH have different effect sizes between sexes, we developed sex-specific risk prediction models. METHODS: Data from the prospective UK Biobank Study were used for model development. Participants with ASAH (per hospital-based ICD codes) before baseline or with missing predictor data were excluded. We de
