Substrate kinetics of the alternative oxidase of Neurospora crassa.

The kinetics of the succinate oxidation by cyanide-sensitive and cyanide-insensitive submitochondrial particles of Neurospora crassa cells suggest that both respiratory pathways use the same complex II. This is confirmed by comparing the kinetics of the reductase activities of the isolated succinate-ubiquinone oxidoreductase (complex II) of cyanide-sensitive and cyanide-insensitive cells respectively. No alternative-oxidase activity was found to be associated with the isolated complex II of cyanide-insensitive cells.

Synthesis of alternative membrane-bound redox carriers during aerobic growth of Escherichia coli in the presence of potassium cyanide.

Aerobic growth of Escherichia coli with an oxidizable substrate as carbon source in the presence of low concentrations of KCN leads to the synthesis and integration into the membrane of menaquinone and cytochromes b558, a1 and d in addition to the redox carriers normally present under aerobic growth conditions, namely ubiquinone and cytochromes b562, b556 and o. The results are discussed with reference to other phenotypic and genotypic modifications to the electron-transport chains of E. coli.

Involvement of Coenzyme Q10 in Various Neurodegenerative and Psychiatric Diseases.

Coenzyme Q10 (CoQ10), commonly known as ubiquinone, is a vitamin-like component generated in mitochondrial inner membranes. This molecule is detected broadly in different parts of the human body in various quantities. This molecule can be absorbed by the digestive system from various nutritional sources as supplements. CoQ10 exists in three states: in a of reduced form (ubiquinol), in a semiquinone radical form, and in oxidized ubiquinone form in different organs of the body, playing a crucial role in electron transportation and contributing to energy metabolism and oxygen utilization, especia

Myopathy induced by HMG-CoA reductase inhibitors in rabbits: a pathological, electrophysiological, and biochemical study.

A combination of electrophysiological, pathological, and biochemical studies were performed in myopathy induced by 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase inhibitors. Simvastatin (a lipophilic inhibitor) or pravastatin (a hydrophilic inhibitor) were administered by gavage to rabbits. In Group I (simvastatin-treated group, 50 mg/kg/day for 4 weeks), four rabbits showed muscle necrosis and high serum creatine kinase (CK) levels, and all six rabbits showed electrical myotonia. In Group II (pravastatin-treated group, 100 mg/kg/day for 4 weeks), no rabbit showed either condition.

Increased sensitivity to peroxidative agents as a possible pathogenic factor of melanocyte damage in vitiligo.

To examine the sensitivity of vitiligo melanocytes to external oxidative stress, we studied enzymatic and non-enzymatic anti-oxidants in cultured melanocytes of normal subjects (n = 20) and melanocytes from apparently normal skin of vitiligo patients (n = 10). The activity of superoxide dismutase and catalase and the intracellular concentrations of vitamin E and ubiquinone were evaluated in cultures at the fourth or fifth passage. In addition, cells were exposed to various concentrations of a peroxidizing agent, cumene hydroperoxide (CUH, 0.66-20 microM), for 1 and 24 h. Compared to normal mel

Molecular cloning of the rat NADH:ubiquinone oxidoreductase subunit and its up-regulation in the facial muscle after denervation: detected by means of differential display.

The differential display method was applied to identify gene expression which is especially up-regulated in the rat denervated skeletal muscle. Total RNA from normal and denervated facial muscles was isolated, amplified by PCR using certain primers, and separated by electrophoresis on polyacrylamide gel. PCR products which were apparently higher in the operated than in the control side were cut out from the gel and subsequently sequenced. One of the cDNA fragments obtained in the present study showed about 80% identity in nucleotide sequence and about 84% identity in amino acid sequence to one

NQR as a target for new antibiotics.

The rise in antimicrobial resistance has underscored the urgent need for identification of novel targets against antibiotic resistant bacteria, which pose enormous threats to public health. The respiratory enzyme NQR carries essential roles in pathogenic bacteria, producing an ion gradient across the plasma membrane that drives ATP generation by the oxidative phosphorylation system. The vital role of NQR in a multitude of pathogenic microbes for which drug development is a high priority, such as Vibrio cholerae, Chlamydia trachomatis, and Pseudomonas aeruginosa, makes it an ideal drug target m

Hypothalamic digoxin and isoprenoid pathway dysfunction relation to alcoholic addiction, alcoholic cirrhosis, and acquired hepatocerebral degeneration--relation to hemispheric chemical dominance.

The isoprenoid pathway produces three key metabolites--endogenous digoxin (modulate tryptophan/tyrosine transport), dolichol (important in N-glycosylation of proteins), and ubiquinone (free radical scavenger). It was considered pertinent to assess the pathway in alcoholic addiction, alcoholic cirrhosis, and acquired hepatocerebral degeneration. Since endogenous digoxin can regulate neurotransmitter transport, the pathway and the related cascade were also assessed in individuals with differing hemispheric dominance to find out the role of hemispheric dominance in its pathogenesis. Infrared and EPR studies on cyanide binding to the heme-copper binuclear center of cytochrome bo-type ubiquinol oxidase from Escherichia coli. Release of a CuB-cyano complex in the partially reduced state.

Cyanide-binding to the heme-copper binuclear center of bo-type ubiquinol oxidase from Escherichia coli was investigated with Fourier transform-infrared and EPR spectroscopies. Upon treatment of the air-oxidized CN-inhibited enzyme with excess sodium dithionite, a 12C-14N stretching vibration at 2146 cm-1 characteristic of the FeO3+ C=N CuB2+ bridging structure was quickly replaced with another stretching mode at 2034.5 cm-1 derived from the FeO2+ C=N moiety. The presence of ubiquinone-8 or ubiquinone-1 caused a gradual autoreduction of the metal center(s) of the air-oxidized CN-inhibited enzym

The microbial metabolism of Cl compounds. The stoicheiometry of respiration-driven proton translocation in Pseudomonas AM1 and in a mutant lacking cytochrome c.

This paper clarifies the role of cytochrome c in Pseudomonas AM1 by measuring the stoicheiometry of proton translocation driven by respiration of endogenous or added substrates in wild-type bacteria and in a mutant lacking cytochrome c (mutant PCT76). The maximum -->H(+)/O ratio (protons translocated out of the bacteria per atom of oxygen consumed during respiration) was about 4 and, except when respiration was markedly affected, this ratio was similar in mutant and wild-type bacteria. The -->H(+)/O ratios were unaltered when the usual oxidase (cytochrome a(3)) was inhibited by 300mum-KCN and

Chapter 12 Controlled expression of iron-sulfur cluster assembly components for respiratory chain complexes in mammalian cells.

Three of the respiratory chain complexes contain essential iron-sulfur (Fe/S) cluster prosthetic groups. Besides respiration, these ancient inorganic cofactors are also necessary for numerous other fundamental biochemical processes in virtually every known organism. Both the synthesis of Fe/S clusters and their delivery to apoproteins depend on the concerted function of specialized, often dedicated, proteins located in the mitochondria and cytosol of eukaryotes. Impaired function of the mitochondria-located Fe/S cluster (ISC) assembly machinery affects all cellular Fe/S proteins, including enz

A high molecular weight dihydro-orotate dehydrogenase of Neurospora crassa. Purification and properties of the enzyme.

Some of the unusual molecular and catalytic properties of a high molecular weight dihydro-orotate dehydrogenase (DHOD) from Neurospora crassa have been determined. Comparison of the properties of this enzyme with the properties of the soluble biosynthetic enzyme of prokaryotes has revealed several important differences. The fungal enzyme is located in a mitochondrial membrane in a position consistent with linkage with the respiratory chain through ubiquinone (Miller, R. W.: Arch. Biochem, Biophys. 146, 256-270 (1971)). Release of the enzyme from the membrane results in a solubilized protein co

Intragenic inversion of mtDNA: a new type of pathogenic mutation in a patient with mitochondrial myopathy.

We report an unusual molecular defect in the mitochondrially encoded ND1 subunit of NADH ubiquinone oxidoreductase (complex I) in a patient with mitochondrial myopathy and isolated complex I deficiency. The mutation is an inversion of seven nucleotides within the ND1 gene, which maintains the reading frame. The inversion, which alters three highly conserved amino acids in the polypeptide, was heteroplasmic in the patient's muscle but was not detectable in blood. This is the first report of a pathogenic inversion mutation in human mtDNA.

Studies on Hg(II)-induced H2O2 formation and oxidative stress in vivo and in vitro in rat kidney mitochondria.

Studies were undertaken to investigate the principal actions underlying mercury-induced oxidative stress in the kidney. Mitochondria from kidneys of rats treated with HgCl2 (1.5 mg/kg i.p.) demonstrated a 2-fold increase in hydrogen peroxide (H2O2) formation for up to 6 hr following Hg(II) treatment using succinate as the electron transport chain substrate. No increase in H2O2 formation was observed when NAD-linked substrates (malate/glutamate) were used, suggesting that Hg(II) affects H2O2 formation principally at the ubiquinone-cytochrome b region of the mitochondrial respiratory chain in vi

Mutation to ispA Produces Stable Small-Colony Variants of Pseudomonas aeruginosa That Have Enhanced Aminoglycoside Resistance.

Pseudomonas aeruginosa is a major pathogen in burn wound infections. We present one of the first reports of small-colony variant (SCV) emergence of P. aeruginosa, taken from a patient under aminoglycosides for a persistent burn wound infection. We confirm the causative role of a single ispA mutation in SCV emergence and increased aminoglycoside resistance. IspA is involved in the synthesis of ubiquinone, providing a possible link between electron transport and SCV formation in P. aeruginosa.

The induction of cyanide-resistant respiration in Hansenula anomala.

Cyanide-resistant respiration was induced in the yeast, Hansenula anomala in the presence of cyanide or antimycin A, which blocks the electron transport after ubiquinone. The de novo protein synthesis in cytosol and oxygen were deduced to be involved in this induction process. The period required for the induction varied during the growth stage, suggesting that involvement of additional physiological factor(s) in this induction process. The organism could multiply in the presence of antimycin A by developing cyanide-resistant respiration despite a decreased growth rate.

Effects of coenzymes Q2 and Q10 on the field potential of guinea pig olfactory cortex slices maintained in hypoxia.

The effects of exogenous CoQ2 and CoQ10 on field potentials were studied while brain slice preparations of guinea pig olfactory cortex were incubated in hypoxia. The potential amplitude, which diminished in hypoxia, recovered when the slices were superfused with solutions containing CoQ2 and CoQ10. Adenosine triphosphate (ATP) levels in the slices duplicated the experimental results of the potential amplitude. The effects of these ubiquinones on potentials measured in hypoxia were suppressed when the slices were incubated with a barbiturate solution.

[Interaction of paramagnetic probes with submitochondrial particle membranes].

Redox-transformations of some paramagnetic probes differing in chemical structure have been studied during their interaction with the membranes of submitochondrial particles. Dependence of the probe restoration kinetics on the conditions of functioning of the electron-transport chain, presence of oxygen, respiration substrate, electron transport inhibitors (antimycin, alpha tenoiltriftoracetate, potassium cyanide) in the reaction mixture is shown. Water-soluble as well as strongly hydrophobic spin probes are restored when the respiration substrate (succinate) is present; they interact with the
