The chemical composition of VLDL, LDL and HDL was studied in 82 patients with primary hyperlipoproteinaemia (37 type IIa, 7 type IIb, 3 type II, 25 type IV, 10 type V) and in ten metabolically normal individuals. Lipoprotein fractions were prepared by preparative ultracentrifugation. Each fraction was analysed for cholesterol, triglycerides, phospholipids and protein. Differences between patients and normal individuals, and between the individual types of hyperlipoproteinaemia were evident in the composition of all three lipoprotein fractions.

We have previously described a class of virus inhibitors which are produced spontaneously by many types of cells in culture and present in a number of physiological fluids. Extracellular vesicles (EVs) serve as key communication signals among embryos. They are secreted into the embryo culture medium and hold potential as biomarkers for assessing embryo quality. However, the limited volume of embryo-conditioned medium presents challenges for EV enrichment. This chapter details an effective method for purifying and characterizing EVs from in vitro embryo cultures by using the OptiPrep™ density gradient ultracentrifugation (ODG) method. Additionally, we also provide methods for preparing EV-free embryo culture media.

Deletion-mutant rabies viral (RV) vectors are powerful tools for neuroscience, allowing monosynaptic tracing of inputs to defined populations and rapid, high-level transgene expression in neurons targeted by multiple routes. High titers and high purity are critical for the successful use of RV vectors in vivo. Here we present a protocol for producing high-quality viral stocks that can be concentrated by ultracentrifugation for final titers in excess of 10(10) infectious units per milliliter.

A large spectrum of renal pathology is associated with hepatitis C virus (HCV). According to novel evidence, occult HCV infection (HCV-RNA in peripheral blood mononuclear cells or in serum after ultracentrifugation) could be involved in the pathogenesis of glomerular nephropathy among patients negative for conventional markers of HCV. Additional studies with appropriate size and technology are in progress to confirm the relationship between occult HCV and glomerular disease, which has multiple implications from the clinical standpoint.

The IgM globulin response has been measured following the onset of primary Q fever infections. A complement fixation test method was used to measure the IgM after separation by ultracentrifugation in a sucrose density gradient. Specific IgM was demonstrable between 2 and 10 wk after onset of symptoms. The test can be used to confirm a recent infection when only a 'convalescent' blood specimen has been collected and will be useful in the assessment of workers' compensation claims.

Soluble oligomers of the β-amyloid peptide, Aβ, are associated with the progression of Alzheimer's disease. Although many small molecules bind to these assemblies, the details of how these molecules interact with Aβ oligomers remain unknown. Huntington's disease (HD) results from expansions of polyglutamine stretches (polyQ) in the huntingtin protein (Htt) that promote protein aggregation, neurodegeneration, and death.
