Impact of chemical exposure on cancer mortality in a French cohort of uranium processing workers.

BACKGROUND: Nuclear workers may be exposed to a variety of chemical hazards, in addition to radiation. We examined the effect of chemical exposures on cancer mortality among French uranium processing workers at the AREVA NC Pierrelatte facility. METHODS: A cohort of 2,897 uranium processing workers employed for at least 6 months was followed from 1968 through 2006. Exposure to uranium and potentially carcinogenic chemicals was assessed with a plant-specific job-exposure matrix. Mortality hazard ratios (HRs) for cancers of the lung, lymphohematopoietic system, kidney and bladder, brain and cen

Renal effects of exposure to natural and depleted uranium: a review of the epidemiologic and experimental data.

Elevated levels of naturally occurring uranium in groundwater have been found in small geographic areas throughout the world. Relevant research was reviewed pertaining to natural and depleted uranium (DU) exposure and nephrotoxicity, including epidemiologic community-based and occupational studies, studies of Gulf War veterans exposed to DU, and experimental studies in animals. Occupational cohort studies do not provide evidence of an increased risk of kidney-related mortality among uranium-exposed workers. However, occupational and community-based studies of populations chronically exposed to

Uranium in drinking water: renal effects of long-term ingestion by an aboriginal community.

The authors conducted a study of an aboriginal community to determine if kidney func-tion had been affected by the chronic ingestion of uranium in drinking water from the community's drilled wells. Uranium concentrations in drinking water varied from < 1 to 845 ppb. This nonin-vasive study relied on the measurement of a combination of urinary indicators of kidney function and markers for cell toxicity. In all, 54 individuals (12-73 years old) participated in the study. Correlation of uranium excreted in urine with bio-indicators at p <or=.05 indicated interference with the kidney's reabsorptiv

Tissue studies of persons with intakes of the actinide elements: the U.S. Transuranium and Uranium Registries.

For more than three decades, the United States Transuranium and Uranium Registries (USTUR) have studied the biokinetics, dosimetry, and biological effects of plutonium, uranium, and americium through voluntary postmortem tissues from persons with known intakes. Radiochemical analyses of tissue obtained at autopsy have shown that plutonium and americium have different biokinetics and an appreciable deposition in the soft tissues of the body as well as the known depots in skeleton and liver. Studies of whole-body Thorotrast donors to the USTUR indicate that commonly accepted risk coefficients fo

Tooth enamel EPR spectroscopy study of radiation doses for uranium miners in Northern Kazakhstan.

PURPOSE: In order to estimate the effect of radiation exposure on the workers of a uranium enterprise, teeth samples were collected for EPR dosimetry of tooth enamel from workers of uranium mines living in Shantobe settlement (Akmola region, Northern Kazakhstan) and from residents of this settlement who had never worked in the mine as a control. METHODS: The accumulated radiation doses in enamel were estimated based on the magnitude of the radiation-induced EPR signal in the samples. Excess (additional) doses were obtained after subtracting the contribution of natural radiation at typical lev

Amidoxime-Based Near-Infrared Fluorescent Sensor for Highly Sensitive Uranium Detection in Living Systems.

The biological detection of uranium is crucial for nuclear emergency medical treatment, since uranium is the major nuclear fuel and its uptake would pose severe health risk to the workers and the public. But there is yet no clinical agent that can provide rapid and accurate evaluation of uranyl internal contamination level, which hampers the timely diagnosis and treatment. Herein, the fluorescent detection method was employed for in vivo uranyl monitoring owing to its advantages of noninvasiveness as well as high specificity and sensitivity. The fluorescent probe NRhB-AO was designed by conjug

Nicotinamide mononucleotide ameliorates impaired testicular spermatogenesis in uranium-exposed mice by modulating glycolytic pathways.

Natural uranium is a ubiquitous element in the environment, and human exposure to low levels of uranium is unavoidable. Several concerns have recently been raised about the reproductive effects of chronic exposure to low levels of uranium. Therefore, the aim of this study was to investigate the protective effect of nicotinamide mononucleotide (NMN) on uranium exposure-induced testicular sperm function in mice. To this end, a research model was established in which testicular damage and spermatogenic dysfunction were induced in adult male mice by intraperitoneal injection of 2 different doses o

Oxidative Addition of E-H (E=C, N) Bonds to Transient Uranium(II) Centers.

Two-electron oxidative addition is one of the most important elementary reactions for d-block transition metals but it is uncommon for f-block elements. Here, we report the first examples of intermolecular oxidative addition of E-H (E=C, N) bonds to uranium(II) centers. The transient U(II) species was formed in-situ by reducing a heterometallic cluster featuring U(IV)-Pd(0) bonds with potassium-graphite (KC8). Oxidative addition of C-H or N-H bonds to the U(II) centers was observed when this transient U(II) species was treated with benzene, carbazole or 1-adamantylamine, respectively. The U(II

Selective Sorption of Cerium Ions from Uranium-Containing Solutions by Remotely Activated Ion Exchangers.

This study investigated the effect of the remote activation of the ion exchangers Amberlite IR120 (H+ form) and AV-17-8 (OH- form) in aqueous media to increase the sorption activity of the interpolymer system "Amberlite IR120H:AV-17-8" (X:Y, molar ratio of ionic groups) towards cerium ions from uranium-containing solutions. The sorption properties of the above-mentioned interpolymer system with molar ratios X:Y of 6:0, 5:1, 4:2, 3:3, 2:4, 1:5, and 0:6 were studied using the methods of conductometry, gravimetry, and inductively coupled plasma-optical emission spectrometry. Uranium chelating ability of decorporation agents in serum evaluated by X-ray absorption spectroscopy.

Internal exposure to actinides such as uranium and plutonium has been reduced using chelating agents for decorporation because of their potential to induce both radiological and chemical toxicities. This study measures uranium chemical forms in serum in the presence and absence of chelating agents based on X-ray absorption spectroscopy (XAS). The chelating agents used were 1-hydroxyethane 1,1-bisphosphonate (EHBP), inositol hexaphosphate (IP6), deferoxamine B (DFO), and diethylenetriaminepentaacetate (DTPA). Percentages of uranium-chelating agents and uranium-bioligands (bioligands: inorganic

Multigenerational exposure to uranium changes morphometric parameters and global DNA methylation in rat sperm.

There is increasing evidence that environmental exposures early in fetal development influence phenotype and give rise to disease risk in the next generations. We previously found that lifelong exposure to uranium, an environmental contaminant, induced subtle testicular and hormonal defects; however, its impact on the reproductive system of multiple subsequent generations was unexplored. Herein, rats were exposed to a supra-environmental and non-nephrotoxic concentration of natural uranium (U, 40 mg·L-1 of drinking water) from postnatal life to adulthood (F0), during fetal life (F1), and only

Association of adverse birth outcomes with prenatal uranium exposure: A population-based cohort study.

Uranium (U) is a well-recognized hazardous heavy metal with embryotoxicity and fetotoxicity. However, little is known about its association with adverse birth outcomes. We aimed to investigate the potential correlation between prenatal U exposure and birth outcomes. Urine samples of 8500 women were collected before delivery from a birth cohort in Wuhan, China. Concentrations of urinary U and other metals were measured by inductively coupled plasma mass spectrometry. We used multivariable logistic regressions to evaluate the associations between urinary U concentrations and adverse birth outcom

Impulse oscillometry measurement of distal airways obstruction in depleted uranium-exposed Gulf War veterans.

INTRODUCTION: A cohort of Gulf War I veterans who sustained exposure to depleted uranium undergoes biennial surveillance for potential uranium-related health effects. We performed impulse oscillometry and hypothesized that veterans with higher uranium body burdens would have more obstructive abnormalities than those with lower burdens. METHODS: We compared pulmonary function of veterans in high versus low urine uranium groups by evaluating spirometry and oscillometry values. RESULTS: Overall mean spirometry and oscillometry resistance values fell within the normal ranges. There were no signi

Multi-electron reduction of sulfur and carbon disulfide using binuclear uranium(iii) borohydride complexes.

The first use of a dinuclear UIII/UIII complex in the activation of small molecules is reported. The octadentate Schiff-base pyrrole, anthracene-hinged 'Pacman' ligand LA combines two strongly reducing UIII centres and three borohydride ligands in [M(THF)4][{U(BH4)}2(μ-BH4)(LA)(THF)2] 1-M, (M = Li, Na, K). The two borohydride ligands bound to uranium outside the macrocyclic cleft are readily substituted by aryloxide ligands, resulting in a single, weakly-bound, encapsulated endo group 1 metal borohydride bridging the two UIII centres in [{U(OAr)}2(μ-MBH4)(LA)(THF)2] 2-M (OAr = OC6H2t Bu3-2,4,6

Grand rounds: nephrotoxicity in a young child exposed to uranium from contaminated well water.

CONTEXT: Private wells that tap groundwater are largely exempt from federal drinking-water regulations, and in most states well water is not subject to much of the mandatory testing required of public water systems. Families that rely on private wells are thus at risk of exposure to a variety of unmeasured contaminants. CASE PRESENTATION: A family of seven--two adults and five children--residing in rural northwestern Connecticut discovered elevated concentrations of uranium in their drinking water, with levels measured at 866 and 1,160 microg/L, values well above the U.S. Environmental Protec

Health effects and biological monitoring results of Gulf War veterans exposed to depleted uranium.

A small group of Gulf War veterans have retained fragments of depleted uranium (DU) shrapnel, the long-term health consequences of which are undetermined. We evaluated the clinical health effects of DU exposure in Gulf War veterans compared with nonexposed Gulf War veterans. History and follow-up medical examinations were performed on 29 exposed veterans and 38 nonexposed veterans. Outcome measures used were urinary uranium determinations, clinical laboratory values, and psychiatric and neurocognitive assessment. Gulf War veterans with retained DU metal shrapnel fragments were found to be stil

Effects of oxygen free radical scavengers on uranium-induced acute renal failure in rats.

Study was made to determine whether oxygen free radicals mediate uranium-induced acute renal failure (ARF). Superoxide dismutase (SOD), a superoxide anion scavenger, did not prevent uranium acetate (UA) (5 mg/kg, i.v.)-induced renal injury 48 h after injection. In contrast, dimethylthiourea (DMTU), a hydroxyl radical scavenger, significantly attenuated UA-induced rise in serum creatinine concentration (1.11 +/- 0.05 (DMTU) vs. 1.40 +/- 0.06 mg/dl (control), p < .05), and tubular necrosis. Dimethyl sulfoxide (DMSO), a hydroxyl radical scavenger, decreased UA-induced tubular damage. UA injection

Modulating tris-acid integrated proton-confined motifs for efficient uranium extraction from natural seawater.

Uranium predominantly exists as the anionic [UO2(CO3)3]4- complex in seawater; conventional uranium adsorbents cannot displace carbonate ligands from the uranyl core via competitive coordination, resulting in inherently low uranium extraction efficiency. Herein, we report a class of unprecedented porous adsorbents featuring tris-acid (triphosphonate/trisulfonate/tricarboxylate) integrated proton-confined motifs. In contrast to traditional sorbents, the incorporated tris-acid moieties suppress proton release through extensive hydrogen-bonding networks, enabling the formation of protonated urany

Spatial Confinement Engineering in a Bioinspired Capillary Electrode for Efficient Uranium Recovery From Seawater.

The electrochemical extraction of critical elements from complex aqueous environments is fundamentally limited by sluggish ion transport and low utilization efficiency in conventional porous electrodes, where reactions are confined to the surface. Inspired by the hierarchical network of blood capillaries, we designed an electrode incorporating carbon dots within amidoxime-functionalized g-C3N4 nanotubes (g-C3N4/CD-AO), forming an electroactive nanochannel network. The confined nanochannels enrich UO2 2+ via steric and coordination effects, while embedded carbon dots enable rapid charge transpo

Targeting the HIF-1α/PPARγ axis with curcumin carbon dots to combat uranium nephrotoxicity.

Uranium-induced nephrotoxicity involves complex mechanisms and lacks therapeutic interventions. This study identifies the HIF-1α/PPARγ axis as a key driver of renal lipid reprogramming and injury following uranium exposure. Using molecular docking, co-immunoprecipitation (Co-IP), ChIP-qPCR, and dual-luciferase reporter assays, we confirmed that HIF-1α directly regulates PPARγ expression and downstream lipotoxicity. The causal role of this axis was further validated using HIF-1α knockdown/knockout models and PPARγ pharmacological modulators (GW9662 and Rosi). Curcumin-derived carbon dots (CCDs)
