Acute cerebral focal ischaemia alters the adrenergic and NANC responses in the bisected rat vas deferens.

1. Disturbances of the autonomic nervous system are common in right hemisphere stroke patients, including a marked decline in male sexual functions. There is a lack of information on the influence of stroke on male secondary sex organs such as the vas deferens. 2. This study investigates the effect of right brain focal ischaemia on the adrenergic and purinergic responses in isolated epididymal and prostatic portions of rat vas deferens. 3. In both epididymal and prostatic portions the concentration-response curves to noradrenaline are flattened resulting in a reduction (up to 67 - 76%) of the

Investigation of the prejunctional alpha2-adrenoceptor mediated actions of MDMA in rat atrium and vas deferens.

1. We have investigated the effects of methylenedioxymethamphetamine (MDMA, 'ecstasy') on peripheral noradrenergic neurotransmission in the rat. 2. In rat atrial slices pre-incubated with [3H]-noradrenaline and in the presence of desipramine (1 micronM) to prevent effects of MDMA on basal outflow of tritium, MDMA (10 micronM) significantly inhibited the release of tritium evoked by short trains of six pulses at 100 Hz every 10 s for 3 min. This effect did not occur in the presence of the alpha2-adrenoceptor antagonist yohimbine (1 micronM). 3. In epididymal portions of rat vas deferens in the

Characterization of receptors for calcitonin gene-related peptide and adrenomedullin on the guinea-pig vas deferens.

1. The receptors which mediate the effects of calcitonin gene-related peptide (CGRP), amylin and adrenomedullin on the guinea-pig vas deferens have been investigated. 2. All three peptides cause concentration dependant inhibitions of the electrically stimulated twitch response (pD2s for CGRP, amylin and adrenomedullin of 7.90+/-0.11, 7.70+/-0.19 and 7.25+/-0.10 respectively). 3. CGRP8-37 (1 microM) and AC187 (10 microM) showed little antagonist activity against adrenomedullin. 4. Adrenomedullin22-52 by itself inhibited the electrically stimulated contractions of the vas deferens and also antag

Multiple receptors for calcitonin gene-related peptide and amylin on guinea-pig ileum and vas deferens.

1. The responses of the electrically stimulated guinea-pig ileum and vas deferens to human and rat calcitonin gene-related peptide (CGRP) and amylin were investigated. 2. The inhibition of contraction of the ileum produced by human alpha CGRP was antagonized by human alpha CGRP8-37 (apparent pA2 estimated at 7.15 +/- 0.23) > human alpha CGRP19-37 (apparent pA2 estimated as 6.67 +/- 0.33) > [Tyr0]-human alpha CGRP28-37. The amylin antagonist, AC187, was three fold less potent than CGRP8-37 in antagonizing human alpha CGRP. 3. Both human beta- and rat alpha CGRP inhibited contractions of the ile

Potassium channel inhibitors attenuate neuromodulatory effects of atrial natriuretic factor in the rabbit isolated vas deferens.

This study tested the hypothesis that neuromodulatory effects of atrial natriuretic factor (ANF) are mediated by an activation of potassium channels in the rabbit isolated vas deferens. The neuromodulatory effects of ANF were tested in the presence of the potassium channel inhibitors, tetraethylammonium, 4-aminopyridine, glibenclamide and charybdotoxin. The effects of the first three were ascertained by their prevention of neuromodulatory effects of a cromokalim enantiomer (BRL 38227), which opens ATP-sensitive potassium channels. The nonspecific potassium channel inhibitors, tetraethylammoniu

A possible crossed histamine-containing pathway adjacent to the sympathetic system of the rat vas deferens.

Studies using nerve degeneration techniques (ganglionectomy, interganglionic section, postganglionic axotomy, uni- or bilateral hypogastric nerve section and right pelvic ganglionectomy) and fluorometric determinations of histamine and norepinephrine have shown the presence of nervous pathways containing histamine adjacent to the sympathetic system of the rat vas deferens. The findings suggest that these pathways cross between the ganglionic clusters located at the angle formed by the seminal vesicle and the vas deferens. They are not structurally related to the central nervous system by way o

Incidental finding of duplication of vas deferens in inguinal hernia: a case report of rare anomaly.

Duplication of the vas deferens is a rare congenital anomaly, with an incidence of ~0.05% in the general population. It is typically discovered incidentally during surgeries involving the spermatic cord, such as inguinal hernia repair. This case report presents a 33-year-old male who underwent left inguinal hernioplasty, during which duplication of the vas deferens was incidentally identified. The patient had presented with left-sided inguinal swelling, painful testis, and a small left testis. Preoperative ultrasound suggested an inguinal hernia, but the vas deferens anomaly was only detected

Pharmacological characterisation of the CB1 receptor antagonist activity of cannabidiol in the rat vas deferens bioassay.

Cannabidiol is increasingly considered for treatment of a wide range of medical conditions. Binding studies suggest that cannabidiol binds to CB1 receptors. In the rat isolated vas deferens bioassay, a single electrical pulse causes a biphasic contraction from nerve-released ATP and noradrenaline. WIN 55,212-2 acts on prejunctional CB1 receptors to inhibit release of these transmitters. In this bioassay, we tested whether cannabidiol and SR141716 were acting as competitive antagonists of this receptor. Monophasic contractions mediated by ATP or noradrenaline in the presence of prazosin or NF44

Gubernaculum Sparing Laparoscopic Orchiopexy in Cryptorchidism with Ipsilateral Congenital Absence of the Vas Deferens: Unique Outcome.

Congenital absence of the vas deferens (CAVD) is an uncommon anomaly that occurs in up to 1% of the male population. It can be associated with various other anomalies, including cryptorchidism and renal anomalies, such as renal agenesis. We here present a case of cryptorchidism with ipsilateral congenital absence of the vas deferens and renal agenesis and used the Stephen-Fowler technique for Orchiopexy depending on gubernacular vessels. A 7-month-old boy was referred to our center with left grade 2 hydronephrosis, right renal agenesis, and right impalpable, undescended testis. Examination und

Gene expression in the efferent ducts, epididymis, and vas deferens during embryonic development of the mouse.

The tissues of the male reproductive tract are characterized by distinct morphologies, from highly coiled to un-coiled. Global gene expression profiles of efferent ducts, epididymis, and vas deferens were generated from embryonic day 14.5 to postnatal day 1 as tissue-specific morphologies emerge. Expression of homeobox genes, potential mediators of tissue-specific morphological development, was assessed. Twenty homeobox genes were identified as either tissue-enriched, developmentally regulated, or both. Additionally, ontology analysis demonstrated cell adhesion to be highly regulated along the

Contractile responses of prostatic and epididymal portions of isolated rabbit vas deferens to electrical field stimulation.

The prostatic and epididymal portions of rabbit vas deferens were different in the reactivity to electrical field stimulation (EFS), adenosine triphosphate (ATP) and noradrenaline (NA). The EFS produced biphasic contractile response; VIZ. the first rapid twitch like (phasic) and the second slow sustained (tonic) contraction. The ratio of the amplitude of the phasic contraction to that of the tonic contraction in response to the EFS was higher in the prostatic portion than that in the epididymal portion. The increase of stimulation frequency from 5 to 10 and 20 Hz more markedly augmented the to

The peritoneal cavity as a bioreactor for tissue engineering visceral organs: bladder, uterus and vas deferens.

Our objective was to produce avascular, myofibroblast-rich tissue capsules for use as autologous grafts for hollow, smooth muscle-walled visceral organs-bladder, uterus and vas deferens. To produce tissue for grafting, templates of the appropriate shape were implanted in the peritoneal cavities of rats or rabbits. After 2-3 weeks, the templates were removed, the encapsulating myofibroblast-rich tissue harvested and grafted to replace resected segments of bladder, vas deferens or uterus of the same animals in which the tissue was grown. Bladder grafts showed 100% patency after 14 months and had

Regional differences in the adenosine A(2) receptor-mediated modulation of contractions in rat vas deferens.

Adenosine receptors involved in modulation of contractions were characterized in the bisected rat vas deferens by combining pharmacological and immunohistochemical approaches. In both portions, noradrenaline-elicited contractions were enhanced by the adenosine A(1) receptor agonist N(6)-cyclopentyladenosine (CPA), and inhibited by the non-selective adenosine receptor agonist 5'-N-ethylcarboxamidoadenosine (NECA) in the presence of the adenosine A(1) receptor antagonist 1,3-dipropyl-8-cyclopentyl-l,3-dipropylxanthine (DPCPX). Cyclic AMP-mediated inhibition of noradrenaline-induced contraction and Ca2+ influx in guinea-pig vas deferens.

The relaxation effects of forskolin and methylxanthines on noradrenaline (NA)-induced contractions were investigated by measuring isotonic contraction and intracellular calcium concentration ([Ca2+]i) in the epididymal side of guinea-pig vas deferens. NA (100 microM) and high K+ (55 mM) induced a biphasic contraction; fast, transient (phasic) and slow, sustained (tonic) phases. Both phases in either NA or high K+ stimulation were abolished in Ca2+-free solution. Pretreatment with 10 microM nifedipine, an L-type Ca2+ channel blocker, reduced both phasic and tonic contractions induced by high K+

Prejunctional actions of some beta-adrenoreceptor antagonists in the vas deferens preparation of the guinea-pig.

1. The beta-adrenoceptor antagonists propranolol, pronethalol, MJ 1999 and Ciba 39089-Ba reduced responses to field stimulation of the guinea-pig isolated vas deferens preparation without significantly affecting responses to exogenously added noradrenaline.2. This prejunctional blocking action of the drugs cannot be correlated with their action as beta-adrenoceptor antagonists or non-specific depressants.3. The blockade produced was more pronounced at low (5-20 Hz) than at high (50 Hz) frequencies of stimulation.4. The blockade was slow in onset, and once established was poorly reversed by was

Bilateral transection of the vas deferens: an unusual trauma from a cross stab injury of the scrotum.

An unusual case of division of both vas deferens in a patient with a cross stab injury to the root of scrotum is described. The management of this case in the emergency setting is discussed. The case illustrates the need to explore every case of penetrating trauma to the scrotum, even in the absence of active bleeding, to avoid missing such injuries. These injuries, although not life-threatening, can have a profound impact on the patient's future fertility and this may only manifest several years after the injury.

Heterogeneity for mutations in the CFTR gene and clinical correlations in patients with congenital absence of the vas deferens.

Congenital absence of the vas deferens (CAVD) is a heterogeneous disorder, largely due to mutations in the cystic fibrosis (CFTR) gene. Patients with unilateral absence of the vas deferens (CUAVD) and patients with CAVD in association with renal agenesis appear to have a different aetiology to those with isolated CAVD. We have studied 134 Spanish CAVD patients [110 congenital bilateral absence of the vas deferens (CBAVD) and 24 CUAVD], 16 of whom (six CBAVD, 10 CUAVD) had additional renal anomalies. Forty-two different CFTR mutations were identified, seven of them being novel. Some 45% of the

Distribution of chromogranins A and B and secretogranin II (secretoneurin) in rat pelvic neurons and vas deferens.

The family of chromogranins/secretogranin peptides comprises three major subtypes: chromogranin A, chromogranin B and secretogranin II. We have characterized these proteins in rat vas deferens and pelvic ganglia by using two approaches. Firstly, extracts of rat vas deferens were subjected to molecular sieve chromatography followed by radioimmunoassay. The results indicate that, in the peripheral nerves of this organ, chromogranin B and secretogranin II are processed to small peptides, i.e. PE-11 and secretoneuron, respectively. Secondly, we investigated the localization of each of these peptid
