[Review of the role of hyperhomocysteinemia and B-vitamin deficiency in neurological and psychiatric disorders--current evidence and preliminary recommendations].

Elevated concentration of total homocysteine (Hcy) in plasma (> 12 micromol/l) is a risk factor for several diseases of the central nervous system. Epidemiological studies have shown a dose-dependent relationship between concentrations of Hcy and the risk for neurodegenerative diseases. Hcy is a marker for B-vitamin deficiency (folate, B12, B6). Hyperhomocysteinemia (HHcy) causes hypomethylation which is an important mechanism that links Hcy to dementia. Supplementation with vitamins B aims at reducing the risk of neurodegenerative diseases. Current evidence suggests that Hcy-lowering treatmen

Vitamin B-12 deficiency induces anomalies of base substitution and methylation in the DNA of rat colonic epithelium.

Derangements of one-carbon metabolism can directly affect the integrity of the genome by producing inappropriate uracil insertion into DNA and by altering patterns of DNA methylation. Vitamin B-12, a one-carbon nutrient, serves as a cofactor in the synthesis of precursors of biological methylation and in nucleotide synthesis. We therefore examined whether vitamin B-12 deficiency can induce these molecular anomalies in the colonic mucosa of rats. Weanling male Sprague-Dawley rats (n = 30) were divided into 2 groups and fed either a vitamin B-12-deficient diet or a similar diet containing adequa

Anemia and deficiencies of folate and vitamin B-6 are common and vary with season in Chinese women of childbearing age.

Little is known about the micronutrient status of Chinese women of childbearing age. We assessed nonfasting plasma concentrations of folic acid, vitamin B-12, vitamin B-6 (as pyridoxal-5'-phosphate), hemoglobin (Hb), ferritin and transferrin receptor (TfR) in 563 nonpregnant textile workers aged 21-34 y from Anqing, China. All women had obtained permission to become pregnant and were participating in a prospective study of pregnancy outcomes. Mean (SD) plasma concentrations were 9.7 (4.1) nmol/L folic acid, 367 (128) pmol/L vitamin B-12, 40.2 (15.8) nmol/L vitamin B-6, 108 (12. 9) g/L Hb, 42.6

Optic neuropathy as a presenting feature of vitamin B-12 deficiency: A systematic review of literature and a case report.

INTRODUCTION: Vitamin B12 (VitB12) deficiency rarely manifests with visual symptoms. Optic nerve damage in VitB12 deficiency is thought to be via degeneration. However, optic neuritis, though infrequent, has been reported secondary to VitB12 deficiency. MATERIAL AND METHODS: We conducted a systematic review of all the reported cases of VitB12 deficiency with optic nerve involvement in Pubmed, Cochrane, and Google Scholar any date up to September 6, 2020. We have discussed the findings and compiled the available information on ophthalmological manifestations of VitB12 deficiency. We aim to pro

Involvement of 5-methyltetrahydrofolate in the amelioration of hyperhomocysteinemia caused by vitamin B(6) deficiency and L-methionine supplementation.

We attempted to clarify the reason why folate fortification ameliorates hyperhomocysteinemia induced by vitamin B(6) deficiency. Hyperhomocysteinemia caused by vitamin B(6) deficiency significantly decreased the rat liver 5-methyltetrahydrofolate level which was significantly improved by folate fortification. This result suggests that the amelioration of hyperhomocysteinemia in response to folate supplementation had enhanced the removal of homocysteine via methionine synthase.

Folic acid fortification ameliorates hyperhomocysteinemia caused by a vitamin B₆-deficient diet supplemented with L-methionine.

Vitamin B₆ (B₆) deficiency affects homocysteine metabolism, and this leads to hyperhomocysteinemia. In this study, we examined i) the effects of B₆-deficiency and graduated levels of dietary methionine on homocysteine metabolism, and ii) the effects of fortified folate on homocysteine metabolism. In experiment 1, Wistar male rats were fed a control or a B₆-deficient diet supplemented with L-methionine at a level of 3, 6, or 9 g/kg of diet for 5 weeks. The resulting plasma homocysteine levels in the B₆-deficient groups increased in relation to the increase in dietary methionine level. Next, in

Is vitamin B(6) deficiency an under-recognized risk in patients receiving haemodialysis? A systematic review: 2000-2010.

Vitamin B6 is a water-soluble vitamin, important for the normal functioning of multiple organ systems. In patients receiving haemodialysis, vitamin B6 deficiency has been reported. The impact of ongoing advances in renal medicine on vitamin B6 status has not been evaluated. The aims of this review were (i) to determine the current level of vitamin B(6) deficiency in the haemodialysis population; (ii) to determine the effect of current haemodialysis prescriptions on vitamin B(6) levels; and (iii) to consider the impact of recent medical advances in haemodialysis on vitamin B(6) levels. Effect of vitamin B-6 (pyridoxine) deficiency on lung elastin cross-linking in perinatal and weanling rat pups.

Weanling and perinatal rats were rendered vitamin B-6 (pyridoxine)-deficient. The rat pups were nursed from vitamin B-6-deficient or -sufficient dams and were killed at day 15 after parturition. The weanling rats were fed vitamin B-6-deficient or -sufficient diets and were killed after 5 weeks of treatment. Lung elastin from the groups of rats was then studied with respect to its content of lysine-derived cross-linking amino acids. Lung lysyl oxidase activity was also measured. B-6 deficiency decreased the number of lysine residues in elastin that were converted into the cross-linking amino ac

Vitamin B-12 (cobalamin) deficiency: a heretofore undescribed control mechanism for plasma corticosteroid-binding globulin concentration in man.

Six of eight patients with Addisonian pernicious anemia were found to have diminished corticosteroid-binding globulin (CBG) concentrations, which returned within 2 weeks to normal after the im administration of cobalamin. T4-binding globulin was found to be completely normal before and after cobalamin replacement. Other estrogen-responsive proteins, i.e. ceruloplasmin, alpha 1-trypsin inhibitor, haptoglobin, transferrin, and alpha 2-macroglobulin, also did not follow CBG concentration in a parallel fashion. The immunoglobulins similarly did not follow a course parallel to that of CBG. The CBG

The acute presentation of nitrous oxide-induced vitamin B-12 deficiency in a postoperative lumbar discectomy patient: illustrative case.

BACKGROUND: Vitamin B-12 deficiency can cause serious hematological and neurological complications. While most cases are chronic, acute presentations may occur. In autoimmune diseases, pernicious anemia is a common cause of vitamin B-12 deficiency. The authors present a case of acute vitamin B-12 deficiency following nitrous oxide anesthesia in a patient with Graves' disease, highlighting the impact of preexisting autoimmune conditions on perioperative management. OBSERVATIONS: A 44-year-old African American male with Graves' disease presented with symptoms of an L5-S1 disc herniation and und

Vitamin B Complex Deficiency After Roux-en-Y Gastric Bypass and Sleeve Gastrectomy-a Systematic Review and Meta-Analysis.

Bariatric surgery, although an effective method, still has complications, like nutritional deficiencies. Our aim was to summarize the evidence on the frequency of complex B vitamin deficiencies in studies comparing Roux-en-Y gastric bypass (RYGB) and sleeve gastrectomy (SG). We included 25 studies for qualitative synthesis and 21 studies for quantitative synthesis. Relevant data was extracted, including proportion of patients with deficiency and mean serum vitamin values in 3 different timeframes. B12 and folate were the most prevalent deficiencies. B12 deficiency was more common after RYGB an

Nutritional Supplementation with Chlorella pyrenoidosa Lowers Serum Methylmalonic Acid in Vegans and Vegetarians with a Suspected Vitamin B₁₂ Deficiency.

Since vitamin B12 occurs in substantial amounts only in foods derived from animals, vegetarians and particularly vegans are at risk of developing deficiencies of this essential vitamin. The chlorella used for this study is a commercially available whole-food supplement, which is believed to contain the physiologically active form of the vitamin. This exploratory open-label study was performed to determine if adding 9 g of Chlorella pyrenoidosa daily could help mitigate a vitamin B12 deficiency in vegetarians and vegans. Seventeen vegan or vegetarian adults (26-57 years of age) with a known vit

The effect of vitamin B-6 deficiency on host susceptibility to Moloney sarcoma virus-induced tumor growth in mice.

The effect of vitamin B-6 deficiency on in vivo host susceptibility to primary Moloney sarcoma virus (MSV)-induced tumor growth and to secondary challenge with MSB sarcoma cells was examined in mice. Female C57BL/6 mice, 6 weeks of age, were fed 20% casein diets with pyridoxine (PN) added at 1, 0.5, 0.1 or 0 mg/kg diet for 21 weeks. After 4 weeks of dietary treatment the mice were challenged with MSV. Vitamin B-6 deficiency resulted in an enhancement of tumor susceptibility as well as an increase in tumor size and regression time. The animals resistant to both MSV and MSV-transformed tumor cel
