Reconstitution of transport from the ER to the Golgi complex in yeast using microsomes and permeabilized yeast cells.

We have developed a highly efficient in vitro-transport assay that couples translocation across the ER membrane and transport to the Golgi complex using the secreted pheromone alpha-factor as a marker protein. Radiolabeled prepro-alpha-factor of high specific radioactivity is obtained by in vitro-translating this protein in a yeast lysate. Prepro-alpha-factor synthesized in vitro is then translocated directly into microsomes or the ER of permeabilized yeast cells. Conversion of the 26-kDa ER form of pro-alpha-factor to the high molecular weight Golgi form is dependent on the presence of ATP an

Distinct physiological strategies underpin antagonism of native Pichia yeasts against Botrytis cinerea.

Botrytis cinerea causes severe losses in viticulture and postharvest systems, requiring biological control agents adapted to the grape environment. Here, two native epiphytic yeasts, Pichia kluyveri B(F8) and Pichia kudriavzevii C(A4), were assessed for antagonistic activity against B. cinerea strains B05.10 and 992 using dual-culture and volatile-mediated in vitro assays, followed by evaluation of antagonistic activity on wine and table grape berries. Both yeasts markedly inhibited mycelial growth and significantly reduced disease severity on fruit. Additional physiological insight was obtain

Novel killer yeasts and toxins from the gardens of fungus-growing ants.

UNLABELLED: Killer toxins are proteinaceous antifungal molecules produced by yeasts, with activity against a wide range of human and plant pathogenic fungi. Fungus gardens of attine ants in Brazil were surveyed to determine the presence of killer toxin-producing yeasts and to define their antifungal activities and ecological importance. Our results indicate that 10 out of 59 yeast species isolated from fungal gardens are killer yeasts. Killer yeasts were less likely to inhibit the growth of yeasts isolated from the same environment but more effective at inhibiting yeasts isolated from other en

Screening of Brazilian Isolated Yeasts Reveals Multiple Potential Chassis for Industrial Biotechnology.

The future of sustainable industrial biotechnology relies on microbial hosts that can thrive where conventional strains fail. Although Saccharomyces cerevisiae remains the dominant industrial yeast, its metabolic constraints and stress sensitivity highlight the need for alternative microbial chassis. Environmental selection shapes microbial phenotypes, and ecologically diverse isolates represent an underexplored reservoir of industrially relevant traits. Here, we established the Brazilian Yeast Collection (BRYC), comprising 79 isolates spanning 18 genera and 28 named species collected across B

Improving Leishmania isolation in field conditions: Efficacy of caspofungin against yeasts contamination.

Cutaneous leishmaniasis (CL), a neglected disease prevalent in 88 countries, is commonly caused by Leishmania (L.) major in the Old World, posing significant public health concerns. Isolation from sand flies or infected mammals and in vitro cultivation of Leishmania parasites are critical for epidemiological studies, but these cultures are often compromised by bacterial and fungal contamination, especially when outsourced from vector digestive tracts. While the Leishmania parasite's natural resistance to antibiotics simplifies bacterial control, most antifungals also inhibit the parasites grow

Comparative Proteomics of Two Flor Yeasts in Sparkling Wine Fermentation: First Approach.

The traditional method is considered the highest-quality sparkling wine making technique. Its main characteristic is that the entire sparkling transformation takes place in the bottle, producing complex, refined wines with fine, persistent bubbles. Currently, the second fermentation in the bottle is initiated by a few commercially available strains of Saccharomyces cerevisiae. This lack of yeast diversity leads to a predominant uniformity in the sensory profiles of the final products and a lack of distinctive wines. The aim of the present study is to compare the proteomic profiles of the first

Virulence profile of pathogenic yeasts from snakes: Alternative ways for antifungal strategies.

Reptiles may act as reservoirs or spreaders of potential pathogenic microorganisms including Candida yeasts. While the epidemiology of yeast species has been thoroughly studied, the virulence profile of isolated species is not well investigated. Therefore, this study aimed to assess the haemolytic, phospholipase, lipase activities and biofilm formation of yeasts isolated from the cloacal swabs of venomous snakes from Marrakech, Morocco (Group I, n = 40) and from non-venomous snakes from Cocullo, Italy (Group II, n = 32). All the isolated yeasts from Group 1 showed low production of lipase (Lz

Machine learning identifies novel signatures of antifungal drug resistance in Saccharomycotina yeasts.

Antifungal drug resistance is a major challenge in fungal infection management. Numerous genomic changes are known to contribute to acquired drug resistance in clinical isolates of specific pathogens, but whether they broadly explain natural resistance across entire lineages is unknown. We leveraged genomic, ecological, and phenotypic trait data from naturally sampled strains from nearly all known species in subphylum Saccharomycotina to examine the evolution of resistance to eight antifungal drugs. The phylogenetic distribution of drug resistance varied by drug; fluconazole resistance was wid

Antifungal potential of Brazilian green and brown propolis against clinically relevant yeasts.

Brazilian biodiversity is rich in compounds with biological potential. Propolis is a natural product formed from bee saliva and plant exudates. Brazilian brown and green propolis exhibit antioxidant, antibacterial, antifungal, antiviral, and antiparasitic activities. These properties arise from their rich phenolic content; molecules such as artepellin-C, quercetin, and kaempferol are important phenolic biomolecules described in propolis. The objective of this study was to determine the antifungal activity and chemical parameters of brown and green propolis samples commercialized in Santa Rita

Why robust identification of rare yeasts is the need of the hour.

UNLABELLED: Infections caused by rare yeasts are surging, mainly due to the increase in immunocompromised or seriously ill patients. With the increasing clinical significance of rare yeasts, there is an urgent need for a rapid diagnostic method. The current process relies on the PCR of the ribosomal gene's internal transcribed spacer (ITS) region, followed by sequencing and BLAST search, which are labor- and time-intensive. Matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS) has revolutionized the rapid identification of yeasts at the species level. One

Snakes as sentinel of zoonotic yeasts and bio-indicators of environmental quality.

Reptiles in the wild or as pets may act as spreaders of bacteria, viruses, fungi and parasites. However, studies on the mycobiota of these animals are scanty. This study investigates the occurrence of yeasts from the cloacal swabs of snakes of different origins and the antifungal profile of the isolated strains. A total of 180 cloacal samples of snakes were collected from Morocco (Group I: n = 68) and Italy (Group II: n = 112). Yeast species were biochemically and molecularly identified. A total of 72 yeast strains belonging to 13 genera, 8 from snakes in Group I and five from snakes in Group

Saponaria officinalis saponins as a factor increasing permeability of Candida yeasts' biomembrane.

Saponins are a large group of compounds, produced mostly by plants as a side product of their metabolic activity. These compounds have attracted much attention over the years mostly because of their surface activity and antibacterial, anti-inflammatory and antifungal properties. On the other hand, most of the hitherto research has concerned the action of saponins against microbial cells as a whole. Therefore, knowing the possible interaction of saponins with biomembrane, we decided to check in-vitro the influence of saponin-rich extract of Saponaria officinalis on spheroplasts of two Candida s

Easy-to-use imaging-cytometry assay to analyze chitin patterns in yeasts.

BACKGROUND & AIM: Pathogenic fungi are a major threat to public health, and fungal infections are becoming increasingly common and treatment resistant. Chitin, a component of the fungal cell wall, modifies host immunity and contributes to antifungal resistance. Moreover, chitin content is regulated by chitin synthases and chitinases. However, the specific roles and mechanisms remain unclear. In this study, we developed a cytometric imaging assay to quantify chitin content and identify the distribution of chitin in the yeast cell wall. METHODS: The Candida albicans SC5314 and Nakaseomyces glab

Engineering yeasts to Co-utilize methanol or formate coupled with CO2 fixation.

The development of synthetic microorganisms that could use one-carbon compounds, such as carbon dioxide, methanol, or formate, has received considerable interest. In this study, we engineered Pichia pastoris and Saccharomyces cerevisiae to both synthetic methylotrophy and formatotrophy, enabling them to co-utilize methanol or formate with CO2 fixation through a synthetic C1-compound assimilation pathway (MFORG pathway). This pathway consisted of a methanol-formate oxidation module and the reductive glycine pathway. We first assembled the MFORG pathway in P. pastoris using endogenous enzymes, f

A Comprehensive Review of Identification Methods for Pathogenic Yeasts: Challenges and Approaches.

Given the increasing incidence of yeast infections and the presence of drug-resistant isolates, accurate identification of the pathogenic yeasts is essential for the management of yeast infections. In this review, we tried to introduce the routine and novel techniques applied for yeast identification. Laboratory identification methods of pathogenic yeast are classified into three categories; I. conventional methods, including microscopical and culture-base methods II. biochemical/physiological-processes methods III. molecular methods. While conventional and biochemical methods require more pre

Antifungal susceptibility profile of yeasts isolated from the oral cavity of cats.

BACKGROUND: Microorganisms living in the oral cavity play an important role in health and disease of the host. Cats are susceptible to oral infections, and it is documented that fungi in the oral cavity could impact these infections. Antifungal resistance has been increasing in recent years. OBJECTIVES: This study was designed to identify yeast isolates from the oral cavity of healthy cats and to evaluate their antifungal susceptibility pattern. METHODS: Oral specimens were collected from 60 cats and cultured at 37°C for 10 days. Yeasts were isolated and identified. Their antifungal suscepti

Extensive Diversity and Prevalent Fluconazole Resistance among Environmental Yeasts from Tropical China.

Yeasts play important roles in both the environment and in human welfare. While some environmental yeasts positively contribute to nutrient cycling and food production, a significant number of yeast species are opportunistic human pathogens, including several that are tolerant/resistant to commonly used antifungal drugs. At present, most of our understanding of environmental yeasts has come from a few terrestrial environments in selected geographic regions. Relatively little is known about yeast diversity in tropical environments and their potential impacts on human health. Here, we characteri

Non-Saccharomyces yeasts for beer production: Insights into safety aspects and considerations.

The application of non-Saccharomyces yeasts in beer as a natural tool for innovation, to create different aroma profiles and flavoured non-alcoholic beers, has attracted great interest from both researchers and commercial brewers. As a result, a higher diversity of non-Saccharomyces yeasts for beer production is expected on the market in the coming years. However, the safe use of non-Saccharomyces yeasts has not been broadly investigated and no guidance for the safety assessment of yeasts is published. The fundamentals of a safety assessment include an accurate taxonomic species identification

High-Titer Production of the Fungal Anhydrotetracycline, TAN-1612, in Engineered Yeasts.

Antibiotic resistance is a growing global health threat, demanding urgent responses. Tetracyclines, a widely used antibiotic class, are increasingly succumbing to antibiotic resistance; generating novel analogues is therefore a top priority for public health. Fungal tetracyclines provide structural and enzymatic diversity for novel tetracycline analogue production in tractable heterologous hosts, like yeasts, to combat antibiotic-resistant pathogens. Here, we successfully engineered Saccharomyces cerevisiae (baker's yeast) and Saccharomyces boulardii (probiotic yeast) to produce the nonantibio

Evaluation of overtime phenotypic variation of yeasts in chronic vulvovaginal candidosis cases.

Chronic vulvovaginal candidosis results either from reinfection or from the ability of Candida spp. to persist in the vulva and/or vagina. Persistence is usually associated with increased antifungal (mainly azoles) resistance rates, which can explain treatment failure, and/or increased expression of virulence factors by Candida spp. The aim of this study was to assess the mechanisms leading to Candida spp persistence, by studying sequential isolates from women with chronic vulvovaginal candidosis, focusing on strains genotypes, azole resistance, and ability to form biofilms along the period of
