Role of phenolic acids with different functional groups in the regulation of starch digestion in simulated dietary intake patterns.

This study investigated the effects of phenolic acids with different functional groups (cinnamic acid: CIA, caffeic acid: CA, ferulic acid: FA) on corn starch (CS) digestibility by simulating dietary intake patterns (co-heating and non-co-heating) and their mechanism. Both treatments could reduce the digestibility of CS. Compared to the non-co-heating treatment, the resistant starch content of 10 % CA co-heating samples increased by 8.36 %. The co-heating case led to a decrease in the trough viscosity, peak viscosity, and final viscosity of CS. Phenolic acids reduced the short-range order of C

Immature leaves are the dominant volatile-sensing organs of maize.

Plants perceive herbivory-induced volatiles and respond to them by upregulating their defenses. To date, the organs responsible for volatile perception remain poorly described. Here, we show that responsiveness to the herbivory-induced green leaf volatile (Z)-3-hexenyl acetate (HAC) in terms of volatile emission, transcriptional regulation, and jasmonate defense hormone activation is largely constrained to younger maize leaves. Older leaves are much less sensitive to HAC. In a given leaf, responsiveness to HAC is high at immature developmental stages and drops off rapidly during maturation. Re

An unusual rectal foreign body: A sundried corn-stick.

Though common, the extraction of rectal foreign bodies is still a great challenge for surgeons. The foreign body's position can usually be verified by plain abdominal radiography. Given the potential of sexually transmitted disease, screening for HIV, hepatitis, and syphilis is warranted prior to intervention. The utilization and selection of surgical instruments need to be flexible, ingenious, and innovative.

Profiling of main metabolites in root exudates and mucilage collected from maize submitted to cadmium stress.

The aim of this study was to characterize qualitatively and quantitatively the composition of the main rhizodeposits emitted from maize (Zea mays) under Cd stress, in order to discuss their role in Cd availability and tolerance. Maize was grown for 6 weeks in sand at four Cd exposure levels (0, 10, 20, and 40 μM Cd in nutrient solution) and two types of rhizodeposits were collected at the end of cultivation period. Mucilage and other molecules adhering to rhizospheric sand were extracted with a buffer before root exudates were collected by diffusion into water. Total carbon, proteins, amino ac

Interference of three herbicides on iron acquisition in maize plants.

The use of herbicides to control weed species could lead to environmental threats due to their persistence and accumulation in the ecosystems and cultivated fields. Nonetheless, the effect of these compounds on plant mineral nutrition in crops has been barely investigated. This study aimed at ascertaining the effect of three herbicides (S-metolachlor, metribuzin and terbuthylazine) on the capacity of maize to acquire iron (Fe). Interferences on plant growth and reductions on the Fe contents were found in the plants treated. Furthermore, root cell viability and functionality losses were ascerta

Improvement of cerium of photosynthesis functions of maize under magnesium deficiency.

Rare earth elements can promote photosynthesis, but their mechanisms are still poorly understood under magnesium deficiency. The present study was designed to determine the role of cerium in magnesium-deficient maize plants. Maize was cultivated in Hoagland's solution added with cerium with and without adequate quantities of magnesium. Under magnesium-deficient conditions, cerium can prevents inhibition of synthesis of photosynthetic pigment, improves light energy absorption and conversion, oxygen evolution, and the activity of photo-phosphorelation and its coupling factor Ca(2+)-ATPase. Progress in breeding low phytate crops.

Populations that depend on grains and legumes as staple foods consume diets rich in phytic acid (myo-inositol-1,2,3,4,5,6-hexkisphosphate), the storage form of phosphorus in seeds. This compound binds tightly to important mineral nutrients such as iron and zinc, forming salts that are largely excreted. This phenomenon can contribute to mineral depletion and deficiency. As one approach to solving this and environmental problems associated with seed-derived dietary phytic acid, the U. S. Department of Agriculture and others have isolated cereal and legume low-phytic acid mutations and have used

KATANIN promotes cell elongation and division to generate proper cell numbers in maize organs.

Microtubule severing is essential for proper eukaryotic cell elongation and division. Here we show that the microtubule severing protein, KATANIN p60, is encoded by two genes in Zea mays L. (maize) called Discordia3a (Dcd3a) and Dcd3b. A semi-dominant mutant with short stature, poor fertility, and a clumped tassel was identified in Dcd3b called Clumped tassel1 (Clt1). Genetic enhancers that further reduced stature and fertility were identified in inbred lines and mapped to the Dcd3a locus, identifying several dcd3a alleles. Loss-of-function p60 allele combinations reduce microtubule severing,

Pseudomonas rhizozeae sp. nov., originated from rhizosphere soil of corn field.

A novel Pseudomonas species was isolated from the rhizosphere of Zea mays in Hungary. Phylogenetic analysis based on concatenated 16S rRNA, rpoD, and rpoB sequences placed the isolate within the Fluorescens superclade, while a proteome-based phylogenetic analysis elucidated the accurate phylogenetic position, a distinct lineage between the Corrugata and Jessenii clades. Whole-genome sequencing of the type strain SZMC 28357T (GenBank accession: JAVLSB000000000) revealed a 5.9 Mb genome with a G + C content of 61%. Comparative genomic analyses (ANI and dDDH) further confirmed that SZMC 28357T re

SbPL1CE8 from Segatella bryantii combines with SbGH28GH105 in a multi-enzyme cascade for pectic biomass utilization.

Pectinases are useful biocatalysts for pectic biomass processing and are extensively used in the food/feed, textile and papermaking industries. Two pectinase genes, a pectate lyase (SbPL1CE8) and a polygalacturonase (SbGH28GH105) were isolated from Segatella bryantii and functionally characterized. Recombinant rSbPL1CE8 was most active against polygalacturonic acid (PGA) and pectin with a 60 % degree of esterification, with kcat/Km values of 721.18 ± 64.77 and 327.02 ± 22.44 mL/s/mg, respectively. Truncated rSbPL1 acted as a mesophilic alkaline pectate lyase, which was highly resistant to inac

Ecological indicators and biological resources for hydrocarbon rhizoremediation in a protected area.

Spillage from oil refineries, pipelines, and service stations consistently leads to soil, food and groundwater contamination. Bacterial-assisted phytoremediation is a non-invasive and sustainable solution to eliminate or decrease the concentration of xenobiotic contaminants in the environment. In the present study, a protected area interested by a fuel discharge was considered to assess a bioremediation intervention. From the spill point, a plume of contamination flowed South-West into the aquifer, eventually reaching a wetland area. Soils, groundwaters and plants belonging to the species Scir

Sample Preparation for Quantitative Proteome and Phosphoproteome Profiling of Maize Tissues.

Zea mays (maize) is a globally important cereal crop and a key system for studying plant development and stress responses. Proteome profiling and phosphoproteome profiling provide direct, quantitative readouts of protein abundance and phosphorylation states, which offer insights into aspects of regulation and cellular function that transcript-level measurements alone cannot provide. Robust and reproducible methods are essential for generating accurate and biologically relevant data in proteomics studies. The complexity of plant tissues, however, poses challenges for developing reliable sample

Effects of Atmospheric CO2 Levels on the Susceptibility of Maize to Diverse Pathogens.

Rising atmospheric CO2 has significant implications for crop productivity and food security. Based on studies in C3 plants, elevated CO2 (eCO2) can shape plant-pathogen interactions, although the outcomes are often variable. The question of how eCO2 influences immunity and disease development in C4 plants, such as the globally important cereal crop maize (Zea mays L.), has not been systematically examined. We challenged maize plants grown under ambient CO2 (aCO2, 420 ppm) and eCO2 (550 ppm) with bacterial, viral, fungal, and oomycete pathogens. Plants grown in eCO2 were more susceptible to sug

Lipidomic and transcriptomic analyses reveal that the gene ZmLACS9 enhances salt stress tolerance by regulating chloroplast lipid metabolism and maintaining photosynthetic performance in maize.

Salt stress is a major environmental factor limiting crop growth and productivity. We found a long-chain acyl-CoA synthetase (LACS) gene ZmLACS9 that was associated with salt tolerance traits in maize (Zea mays). Mutation of ZmLACS9 impaired plant growth, while overexpression of ZmLACS9 improved plant growth vigour under salt stress conditions. Metabolome and transcriptome analyses indicated that ZmLACS9 may regulate the expression of lipid trafficking-related genes, thereby influencing the concentrations of galactolipids and phospholipid in the photosynthetic membrane under salt stress. A High-Throughput Absolute Quantification of Protein-Bound Tryptophan from Model and Crop Seeds.

This protocol describes a high-throughput absolute quantification protocol for the aromatic essential amino acid, tryptophan (Trp). This procedure consists of a milligram-scale alkaline hydrolysis followed by an absolute quantification step using a multiple reaction monitoring tandem mass spectrometric (LC-MS/MS) detection method. The approach facilitates the analysis of a few hundred samples per week by using a 96-well plate extraction setup. Importantly, the method uses only ∼4 mg of tissue per sample and uses the common alkaline hydrolysis protocol, followed by water extraction that include

Removal of nitrate from agricultural runoff in biochar electrode based biofilm reactor: Performance and enhancement mechanisms.

A biochar electrode based biofilm reactor was developed for advanced removal of nitrate from agricultural runoff. The corn-straw (Zea mays L.) biochar formed at 500 °C has an adsorption capacity of NO3--N up to 2.659 mg g-1. After 45-day start-up phase, the removal efficiency of nitrate reached 93.4% when impressed current was 20 mA, hydraulic retention time was 12 h and chemical oxygen demand/total nitrogen (C/N) ratio was 0.56 without additional carbon source. In comparison, neither electrochemical reduction alone nor microbial denitrification alone could obtain the ideal nitrate removal eff

Purple Corn Extract Improves Benign Prostatic Hyperplasia by Regulating Prostate Cell Proliferation and Apoptosis.

Purple corn (Zea mays L.), utilized as a natural pigment in food production and processing, has been used to treat obesity, cystitis, and urinary tract infections. However, no reports of its use for benign prostatic hyperplasia (BPH) exist. Purple corn extract (PCE) contains anthocyanins, particularly cyanidin-3-O-glucoside, which have various pharmacological characteristics. Therefore, this study sought to elucidate the ameliorative effect of PCE on BPH in dihydrotestosterone (DHT)-stimulated WPMY-1 cells and testosterone propionate (TP)-induced rats. Expression levels of the upregulated andr

Genome-wide association study of grain polyphenol concentrations in global sorghum [Sorghum bicolor (L.) Moench] germplasm.

Identifying natural variation of health-promoting compounds in staple crops and characterizing its genetic basis can help improve human nutrition through crop biofortification. Some varieties of sorghum, a staple cereal crop grown worldwide, have high concentrations of proanthocyanidins and 3-deoxyanthocyanidins, polyphenols with antioxidant and anti-inflammatory properties. We quantified total phenols, proanthocyanidins, and 3-deoxyanthocyanidins in a global sorghum diversity panel (n = 381) using near-infrared spectroscopy (NIRS), and characterized the patterns of variation with respect to g

High frequency of centromere inactivation resulting in stable dicentric chromosomes of maize.

Somatic chromosome spreads from maize (Zea mays L.) plants containing B-A translocation chromosomes undergoing the chromosome type breakage-fusion-bridge cycle were examined by FISH. The size and type of extra chromosomes varied among cells of the same individual. A collection of minichromosomes derived from the chromosome type breakage-fusion-bridge cycle was examined for the presence of stable dicentric chromosomes. Six of 23 chromosomes in the collection contained two regions with DNA sequences typical of centromeres. Functional analysis and immunolabeling of CENH3, the centromere-specific
