Proteases and protease inhibitors in taurocholate-induced acute pancreatitis in rats.

CONCLUSION: Taurocholate-induced acute pancreatitis (AP) in the rat mimics early necrotizing human pancreatitis. Protease activation and protease inhibitor consumption occur consistent with a two-stage development, and contact-phase activation is a possible primary event in this model. BACKGROUND: Proteases and protease inhibitors have been indicated to play an important role in both human and experimental acute pancreatitis, although little is known about them in rats. METHODS: Three percent sodium taurocholate was infused into the bilio-pancreatic duct to induce AP, and over 0-72 h we meas

The activities of amylase and a trypsin-like protease in the gut contents of germ-free and conventional chickens.

1. Proteolytic and amylase activities were assayed in the gut contents of germ-free and conventional chickens aged 3 and 14 d. 2. The tendency was for the proteolytic activity to be increased and the amylase activity to be decreased in conventional chicks. These differences were statistically significant in the caecum. 3. All of the additional proteolytic activity detectable in the caecum of conventional chicks was inhibited by soybean trypsin inhibitor. However, the increased activity was not detectable in the small intestine and was, therefore, not due to pancreatic trypsin. 4. Attempts to d

Nutritional significance of lectins and enzyme inhibitors from legumes.

Legumes have natural components, such as lectins, amylase, and trypsin inhibitors, that may adversely affect their nutritional properties. Much information has already been obtained on their antinutritional significance and how to inactivate them by proper processing. Chronic ingestion of residual levels is unlikely to pose risks to human health. On the other hand, the ability of these molecules to inhibit some enzymes such as trypsin, chymotrypsin, disaccharidases, and alpha-amylases, to selectively bind to glycoconjugates, and to enter the circulatory system may be a useful tool in nutrition

A starch-binding domain of α-amylase (AmyPG) disrupts the structure of raw starch.

Most raw starch-digesting enzymes possess at least one non-catalytic starch-binding domain (SBD), which enhances enzymatic hydrolysis of insoluble starch granules. Previous studies of SBD-starch interaction mainly focus on binding affinity for substrates, while the mechanism involved disruption of starch granules remains partially understood. Raw starch-digesting α-amylases AmyPG and AmyP were from Photobacterium gaetbulicola and an uncultured marine bacterium, respectively. Here, comparative studies on the two α-amylases and their SBDs (SBDPG and SBDAmyP) with high sequence identity were carr

Trypsin activity. A new marker of acute alcoholic pancreatitis.

A normal serum amylase level is found in up to 32% of patients with acute alcoholic pancreatitis. This underlines the need for more sensitive diagnostic tests in this frequent cause of pancreatitis. Animal and human studies have shown that chronic alcohol consumption leads to important modifications in trypsinogen metabolism. The present work has prospectively analyzed admission serum trypsin activity with a new biochemical test and usual markers such as amylase, lipase, and immunoreactive trypsin in 32 attacks of acute pancreatitis. Seventeen were due to alcohol and 15 to other causes, includ

Structural, quantitative, and functional characterization of N-glycans in porcine pancreatin extract, lipase, and α-amylase.

Porcine pancreatin extract (PPE), an enzyme mixture comprising porcine pancreatic lipase (PPL), porcine pancreatic α-amylase (PPA), and proteases, facilitates the digestion of fats, carbohydrates, and proteins and is widely used to treat pancreatic insufficiency. N-glycosylation is a critical post-translational modification influencing enzyme stability and activity. This study presents the N-glycan profiling of PPE, PPL, and PPA to evaluate their structural roles in enzymatic function. N-glycans were enzymatically released, procainamide-labeled, and analyzed using liquid chromatography-tandem

Nature-based interventions reduce physiological stress in children with chronic illnesses: evidence from salivary biomarkers.

INTRODUCTION: Contact with nature is increasingly recognized as an important determinant of public health, yet limited objective evidence exists regarding its benefits for children with chronic illnesses. This study aimed to evaluate the short-term effects of a forest-based intervention on stress-related salivary biomarkers in vulnerable pediatric populations, including cancer survivors and children with neurodevelopmental disorders. METHODS: A quasi-experimental design was used to assess 52 children aged 8-17 years, including both patients and their healthy siblings. Participants completed a

Early traumatic experiences impair the functioning of both components of the endogenous stress response system in adult people with eating disorders.

Childhood trauma is a non-specific risk factor for eating disorders (EDs). It has been suggested that this risk is exerted through trauma-induced long-lasting changes in the body stress response system. Therefore, we explored the activity of the hypothalamus-pituitary-adrenal axis and of the sympathetic nervous system in adult ED patients with or without a history of childhood trauma exposure. Salivary cortisol and alpha-amylase, a marker of the sympathetic nervous system activity, were measured at awakening and after 15, 30 and 60 min in 35 women with EDs. The Childhood Trauma Questionnaire (

Neuroendocrine Response to School Load in Prepubertal Children: Focus on Trait Anxiety.

At the time of school-age, the most frequent stress stimuli are related to school environment and educational process. Anxiety may play a big role in coping with stressful situations associated with school load. To approach this issue, we performed a real-life study at school during the classwork. The sample consisted of 36 healthy children aged 10 years, which were divided to low and high trait anxiety group based on the median value of the anxiety score. The investigations were carried out in the classroom during a stress condition (final exams) and non-stress condition (without any exam). I

Macroamylasemia induced by hydroxyethyl starch--confirmation by gel filtration analysis of serum and urine.

The concentration of alpha-amylase in blood rose significantly (Student's t-test) in seven normovolemic healthy subjects after they were given 30 g/m2 body surface area of hydroxyethyl starch 350/0.60 (HES 350/0.60, a new volemic colloid). Concomitant with elevated activity in blood, amylase clearance (Cam) was markedly reduced, even though clearance of creatinine (Ccr) was within normal limits. Similar findings were observed for one male subject given 500 ml (30 g) of HES 450/0.70. Sephadex G-100 gel elution profiles of serum recovered from one male subject after the infusion of HES 350/0.60

Exocrine and endocrine pancreatic function in rats treated with alpha-glucosidase inhibitor (acarbose).

Pancreatic exocrine and endocrine secretory dynamics were studied in the isolated perfused pancreata of rats fed a normal diet or a diet supplemented with the alpha-glucosidase inhibitor, acarbose (150 mg/100 g food). After 10 days, the body weight of acarbose-treated rats was slightly lower than that of the control rats despite a larger food intake. Pancreatic amylase levels were significantly decreased, trypsinogen levels were significantly increased, and lipase levels were unaltered in the treated group compared with the controls. Basal and caerulein-stimulated flow rates of pancreatic juic

Salivary amylase - The enzyme of unspecialized euryphagous animals.

INTRODUCTION: Alpha-amylase (EC 3.2.1.1) is the most abundant enzyme in the saliva of man and of several vertebrates. In humans, salivary amylase is mainly formed in the parotid gland; its activity is of high inter-individual and intra-individual variability. The physiological functions of α-amylase have not yet been explored completely. It is well known that the enzyme cleaves the α-(1,4)-glycosidic bonds of polysaccharides. Furthermore it plays an important role in initial bioadhesion in man, facilitating carbohydrate metabolism and bacterial adherence at the tooth surface and therewith cari

Activity of some hepatic enzymes in schistosomiasis and concomitant alteration of arylsulfatase B.

The levels of arylsulfatases A and B, alpha-amylase, aspartate transcarbamylase, and gamma-glutamyl transpeptidase were investigated during the infection of mice with schistosoma mansoni. This infection caused a significant (p < 0.001) increase in the activity of hepatic arylsulfatase B (ASB), aspartate transcarbamylases and gamma-glutamyl transpeptidase. A non-significant difference occurred for alpha-amylase (p < 0.3) and arylsulfatase A (p > 0.5) when compared to the control. The specific activity of hepatic ASB was progressively increased with the progression of the Schistosoma-infection.

Substance K and salivary secretion in the rat.

Intravenous injections of substance K (SK), a novel member of the family of tachykinins, evoked secretion from the three major salivary glands of the rat in the presence of muscarinic, alpha-adrenergic and beta-adrenergic receptor blockade; the submaxillary glands contributed most and the sublingual glands least to the total volume secreted. SK was less potent than substance P (SP) in evoking fluid and amylase secretion. However, the amylase concentration in parotid saliva evoked by SK was twice that evoked by SP, a finding which indicates that in the glands there are more than just one type o

Comparative analysis of pancreatic amylase activity in laboratory rodents.

Alpha-amylase is the main enzyme for starch digestion in the mammalian gastrointestinal tract. There are species differences in the enzymatic activity of pancreatic amylase that are related to the digestive strategy and natural diet of a species. This aspect is well investigated in pet and farm animals, while in common laboratory animal rodents, information is scarce. In the context of the 3R concept, detailed knowledge of the digestive physiology should be the basis of adequate nutrition, experimental planning and data interpretation. The present study aimed to obtain reference data on amylas

Synthesis of Some Novel Chalcone and 3,5-Disubstituted Pyrazoline Derivatives: Evaluation of Their α-Amylase and α-Glucosidase Inhibitory Activities In Vitro and In Silico.

This study reports the synthesis of some novel chalcone (C1-C3) and 3,5-disubstituted pyrazoline derivatives (P1-P3), structurally based on aminophenyl and trifluoromethylphenyl groups. Their potential as antidiabetic agents was evaluated through in vitro inhibition of α-amylase and α-glucosidase enzymes and supported by molecular docking studies. Among all the compounds, P2 showed potent dual inhibition, with IC50 values of 9.35 and 2.10 µM against α-amylase and α-glucosidase, respectively, comparable to or better than the standard inhibitor acarbose. Kinetic studies revealed that P2 acts via
