Perturbations of genes essential for Müllerian duct and Wölffian duct development in Mayer-Rokitansky-Küster-Hauser syndrome.

Mayer-Rokitansky-Küster-Hauser syndrome (MRKHS) is associated with congenital absence of the uterus, cervix, and the upper part of the vagina; it is a sex-limited trait. Disrupted development of the Müllerian ducts (MD)/Wölffian ducts (WD) through multifactorial mechanisms has been proposed to underlie MRKHS. In this study, exome sequencing (ES) was performed on a Chinese discovery cohort (442 affected subjects and 941 female control subjects) and a replication MRKHS cohort (150 affected subjects of mixed ethnicity from North America, South America, and Europe). Phenotypic follow-up of the fem

Integrated network toxicology, transcriptomic, and Mendelian randomization analyses explore potential molecular links between acrylamide exposure and clear cell renal cell carcinoma.

Acrylamide is a potential carcinogen widely encountered in occupational settings and heat-processed foods. However, the potential molecular relationships between acrylamide-associated targets and clear cell renal cell carcinoma remain poorly characterized. The toxicological characteristics of acrylamide were predicted using ProTox-3.0 and ADMETlab 3.0. Potential acrylamide-associated targets were identified using SwissTargetPrediction, the Similarity Ensemble Approach, and TargetNet and were intersected with clear cell renal cell carcinoma (ccRCC)-related genes obtained from GeneCards. Genetic predisposition to increased blood cholesterol and triglyceride lipid levels and risk of Alzheimer disease: a Mendelian randomization analysis.

BACKGROUND: Although altered lipid metabolism has been extensively implicated in the pathogenesis of Alzheimer disease (AD) through cell biological, epidemiological, and genetic studies, the molecular mechanisms linking cholesterol and AD pathology are still not well understood and contradictory results have been reported. We have used a Mendelian randomization approach to dissect the causal nature of the association between circulating lipid levels and late onset AD (LOAD) and test the hypothesis that genetically raised lipid levels increase the risk of LOAD. METHODS AND FINDINGS: We include

METTL21C is a potential pleiotropic gene for osteoporosis and sarcopenia acting through the modulation of the NF-κB signaling pathway.

Sarcopenia and osteoporosis are important public health problems that occur concurrently. A bivariate genome-wide association study (GWAS) identified METTL21c as a suggestive pleiotropic gene for both bone and muscle. The METTL21 family of proteins methylates chaperones involved in the etiology of both myopathy and inclusion body myositis with Paget's disease. To validate these GWAS results, Mettl21c mRNA expression was reduced with siRNA in a mouse myogenic C2C12 cell line and the mouse osteocyte-like cell line MLO-Y4. At day 3, as C2C12 myoblasts start to differentiate into myotubes, a signi

The causal association between inflammatory bowel disease and pancreatitis: A 2-sample Mendelian randomization study.

Clinical studies showed a relationship between inflammatory bowel disease (IBD) and pancreatitis. Nonetheless, the causal relationship between IBD and pancreatitis has yet to be established. We employed Mendelian randomization (MR) to determine potential causal associations between IBD and pancreatitis. Genome-wide association study data on IBD were acquired from the International Inflammatory Bowel Disease Genetics Consortium. Summary statistics for pancreatitis were from the FinnGen database and BioBank Japan. Inverse-variance weighting (IVW) was used to perform a 2-sample MR analysis. Heter

Genetically predicted plasma metabolites mediate the relation between inflammatory factors and Meniere's disease.

OBJECTIVE: Emerging evidence suggests inflammation contributes to Meniere's Disease (MD), a chronic vestibular disorder characterized by vertigo, hearing loss, and tinnitus. However, the causal role of Inflammatory Factors (IFs) and the mediating effects of Plasma Metabolites (PMs) in MD remain unclear. This study investigated bidirectional causality between IFs and MD and evaluated PMs as potential mediators. METHODS: Using Genome-Wide Association Study (GWAS) summary data, we performed bidirectional Mendelian Randomization (MR) analyses to assess causal links between 91 IFs, 1,400 PMs, and

Lung cancer and altered brain structure: A causal inference based on MR.

Lung cancer is one of the most prevalent malignancies globally and imposes a substantial burden on mortality and healthcare costs. Emerging clinical evidence suggests that lung cancer may influence brain morphology, particularly the cerebral cortex. This study employed Mendelian Randomization (MR) to investigate the potential causal relationship between lung cancer and cortical structural alterations. Single nucleotide polymorphisms associated with small cell lung cancer (SCLC) and non-SCLC (NSCLC) were instrumental variables derived from the FinnGen consortium. Summary-level data on brain cor

Bidirectional Mendelian randomization study of causal associations between white-matter structural connectivity and multiple sclerosis.

BACKGROUND: Multiple sclerosis (MS) is associated with widespread network disruption, but whether specific white-matter structural connectivity (WMSC) phenotypes contribute causally to MS susceptibility remains unclear. METHODS: We performed bidirectional two-sample Mendelian randomization (MR) using genome-wide association study (GWAS) summary statistics for 206 tractography-derived WMSC phenotypes (UK Biobank; N = 26,333) and MS susceptibility (IMSGC; N = 115,803 of European ancestry). Primary inference used inverse-variance weighted (IVW) MR under a multiplicative random-effects model, com

Use of Mendelian randomization to evaluate the effect of atrial fibrillation on cardiovascular diseases and cardiac death.

AIMS: Several observational studies indicated that atrial fibrillation might aggravate other cardiovascular diseases apart from ischaemic stroke. However, it remains to be determined whether these associations reveal independent causation. Using Mendelian randomization (MR), we systematically investigated how genetically predicted atrial fibrillation affected other cardiovascular diseases and cardiac death. METHODS AND RESULTS: Summary-level data for atrial fibrillation and other cardiovascular diseases were obtained from public genome-wide association study data. The random inverse-variance
