Sensory neuro-immune interactions differ between irritable bowel syndrome subtypes.

OBJECTIVE: The gut is a major site of contact between immune and sensory systems and evidence suggests that patients with irritable bowel syndrome (IBS) have immune dysfunction. Here we show how this dysfunction differs between major IBS subgroups and how immunocytes communicate with sensory nerves. DESIGN: Peripheral blood mononuclear cell supernatants from 20 diarrhoea predominant IBS (D-IBS) patients, 15 constipation predominant IBS (C-IBS) patients and 36 healthy subjects were applied to mouse colonic sensory nerves and effects on mechanosensitivity assessed. Cytokine/chemokine concentrat

Neuropeptide regulation of feeding in dogs.

Norepinephrine and four families of neuropeptides, namely, neuropeptide Y (NPY), opioid peptides, galanin, and growth hormone-releasing factor (GRH), have been shown to stimulate feeding after central administration. Because these studies were mainly done on laboratory rats, the present study was designed to ascertain the central stimulators of feeding in dogs. We have shown that porcine and human pancreatic polypeptides (PPs), when administered into the third cerebral ventricle (intracerebroventricularly), increased food and water intake of satiated animals but that the COOH-terminal fragment

Intestinal ganglioneuromatosis: mucosal and transmural types. A clinicopathologic and immunohistochemical study of six cases.

Six cases of intestinal ganglioneuromatosis (GN) included in this study reveal the occurrence of two morphologic patterns. Transmural GN was characterized by neural hyperplasia in all layers of the bowel wall with predominant involvement of the myenteric plexus. It was found in three patients affected by multiple endocrine neoplasia IIb. Mucosal GN, having predominant involvement of the mucosa without concomitant hyperplasia of the myenteric plexus, was associated with von Recklinghausen's disease, adenocarcinoma of the colon, and multiple adenomas with megacolon in one case each. Clinicopatho

Opioids and breast cancer recurrence.

PURPOSE OF REVIEW: Breast cancer survival has improved motivating the need for better understanding of the sequelae of the disease and its treatments. Lab studies suggest opioids modify cancer cell growth but the association of opioids with cancer progression in humans is not clear. This review aims to summarize recent findings related to opioid use and breast cancer progression. RECENT FINDINGS: Opioid-sparing analgesia may be associated with better survival in cancer patients. In-vitro research suggests that treatment with μ-opioid receptor antagonists inhibits cancer proliferation, and sho

The effects of histamine injection on the histochemistry of the guinea pig parathyroid gland.

Histochemical studies on the activity of some neuropeptides (PGP, CT, NPY, SP and betaE) and enzymes (AP, AlP, SDH and MAO) were performed on guinea pig parathyroid glands after injections of histamine, histamine receptor blockers and muscarinic receptor blocker. Under conditions of histamine shock, the immunoreactivity of CT and the reactivity of SDH and MAO were found to decrease together with an increase in the activity of betaE and AP. The reactions for SP, NPY, PGP and AlP did not change in any of the groups.

Opioid binding profile of morphiceptin, Tyr-MIF-1 and dynorphin-related peptides in rat brain membranes.

Opioid properties of several morphiceptin- (Tyr-Pro-Phe-Pro-NH2), Tyr-MIF-1 (Tyr-Pro-Leu-Gly-NH2) and dynorphin-derivatives were characterized in rat brain in vitro receptor binding assay and in electrically stimulated longitudinal muscle strip preparation of guinea pig ileum. In the case of morphiceptin-related peptides, an excellent correlation was found between the [3H]-naloxone binding displacement data and the agonist potencies determined in the bioassay. The "turning point' was the C-terminal amidation in the tri- and tetrapeptide pairs in both series. Tyr-MIF-1 derivatives showed weak a

Opioid receptors on guinea-pig intestinal crypt epithelial cells.

1. Opioid peptides promote net intestinal absorption via two mechanisms: stimulation of Na+ and Cl- absorption and inhibition of Cl- secretion. Although these transport changes are predominantly mediated by submucosal neurones, it is currently unclear whether opioid peptides can regulate enterocyte function directly. We therefore tested the hypothesis that enterocytes have specific opioid receptors. 2. Villus and crypt jejunal epithelial cells were isolated by the distended sac method from anaesthetized guinea-pigs. Flow cytometry was used to resolve enterocytes from other cell types and to de

Enduring changes in brain and behavior produced by chronic amphetamine administration: a review and evaluation of animal models of amphetamine psychosis.

Some people who repeatedly use stimulant drugs, such as amphetamine (AMPH), develop an AMPH-induced psychosis that is similar to paranoid schizophrenia. There has been, therefore, considerable interest in characterizing the effects of chronic stimulant drug treatment on brain and behavior in non-human animals, and in developing an animal model of AMPH psychosis. A review of this literature shows that in non-human animals chronic AMPH treatment can produce at least two different syndromes, and both of these have been proposed as animal models of AMPH psychosis. The first syndrome is called 'AMP

The neural substrates for the motor-activating properties of psychostimulants: a review of recent findings.

Several different classes of pharmacological agents produce syndromes of behavioral activation in humans and infrahumans. While many of these agents, including direct and indirect sympathomimetics, methylxanthines, opiates and several neuropeptides have very distinct neurochemical profiles, it is not clear whether their behavioral stimulant action results from their action on a common neural substrate, or instead from their action on parallel but separate activation "circuits.' Using photocell measurements of motor activity in rats, it has been possible to demonstrate that some agents with ver

Neurochemical mechanism of action of drugs which modify feeding via the serotoninergic system.

The neurochemical mechanisms by which drugs acting on central serotoninergic system modify feeding were reviewed. Fenfluramine, a clinically effective appetite suppressant, releases serotonin from nerve terminals and inhibits its reuptake, and considerable evidence suggests that these effects mediate its anorectic activity. The D isomer of fenfluramine is particularly specific in affecting serotonin mechanisms and causing anorexia. Transmitters other than serotonin such as acetylcholine, catecholamines and GABA are also affected by systemic administration of fenfluramine, but some of these eff

Analgesia induced by local plantar injections of opiates in the formalin test in infant rats.

Morphine injected locally to the paw of an adult or an infant rat is analgesic. Opiates specific to micro and kappa opioid receptors, and less consistently to delta opioid receptors, given locally to the site of injury in adult animals are also analgesic in a variety of models of inflammatory pain. To determine which opioid receptor(s) are involved in local analgesia in the immature animal, agonists specific for micro, kappa, and delta opioid receptors were injected into the intraplantar pad in infant rats and the resultant nociceptive behavior and Fos expression assayed in the formalin test.

Receptors for opioid peptides in the guinea-pig ileum.

Cryptic receptor sites in the guinea-pig ileum preparation have been uncovered by the treatment of the preparation with the highly selective, irreversible mu opioid receptor antagonist, beta-funaltrexamine. These beta-funaltrexamine-insensitive sites appear to interact only with opioid peptides ([D-Ala2, D-Leu5]enkephalin, [D-Ala2, Met5]enkephalinamide, Tyr-D-Ser-Gly-Phe-Leu-Thr and [D-Ala2, MePhe4, Gly-ol5]enkephalin) but not with nonpeptide agonists. These new sites could not be protected by either mu-selective (morphiceptin and [D-Ala2, MePhe4, Gly-ol5]enkephalin) or delta-selective ([D-Ala

A novel bovine spinal cord endoprotease with high specificity for dynorphin B.

An endopeptidase that converts the opioid peptide dynorphin B (Tyr-Gly-Gly-Phe-Leu-Arg-aRg-Gln-Phe-Lys-Val-Val-Thr) to its bioactive fragment Leu-enkephalin-Arg6 was isolated from bovine spinal cord. The enzyme was purified about 230-fold from a concentrated spinal cord extract. Upon sodium dodecyl sulfate-polyacrylamide gel electrophoresis, it stained as a protein of Mr 55,000. The purified enzyme is optimally active at around pH7 and has essential thiol groups. It appears to be highly specific for dynorphin B (Km = 11 microM) but not for alpha-neoendorphin or dynorphin A, two other opioids i

Interactions of Tyr-MIF-1 at opiate receptor sites.

Binding of Tyr-MIF-1 (Tyr-Pro-Leu-Gly-NH2) to mu and delta opiate receptors was compared with other putative opiate antagonist peptides by displacement of iodinated ligands selective for mu (DAGO, FK33824, and morphiceptin) and delta (DPDPE) receptors. Tyr-MIF-1 and ACTH (1-24 and 1-39) inhibited binding of 125I-DAGO with IC50's of about 1 microM. FMRF-NH2 was about an order of magnitude weaker while CCK-8 and MIF-1 failed to inhibit 50% of binding at concentrations up to 100 microM. Morphiceptin, Tyr-MIF-1, and ACTH were less potent but more efficacious than DAGO, FK33824, morphine, or naloxo
