Unleashing antifungal power: Nanopriming with selenium boosts JA signaling, suppresses pathogen enzymes, and activates phytoalexins for enhanced Fusarium graminearum resistance in maize.

INTRODUCTION: Stalk rot caused by Fusarium graminearum (F. graminearum) poses a serious threat to maize production, further exacerbated by contamination with the mycotoxin deoxyninylenol (DON). Although selenium nanoparticle (SeNP) priming technology shows potential for enhancing crop resistance, the systematic regulatory mechanisms by which it reprograms plant defense responses against this pathogen remain unclear. OBJECTIVES: This study aimed to elucidate the molecular mechanisms through which SeNP priming regulates transcriptional and metabolic reprogramming in maize seedlings to resist F.

Enhancing Food Quality Through Radio Frequency Drying of In-Shell Oil Crops: A Critical Review of Mechanisms, Advantages, and Challenges.

The postharvest drying of in-shell oil crops is severely constrained by their shell-kernel structure, which creates stubborn heat and mass transfer resistance, leading to prolonged processing times, high energy consumption, and rapid quality deterioration, including lipid oxidation and color loss. Radio frequency (RF) drying offers an inside-out volumetric heating strategy that bypasses shell resistance and directly heats the kernel, thus reducing drying time and preventing quality deterioration. This review aimed to systematically clarify the mechanism of RF selective heating induced by diele

IRE1/FAM134B-mediated ER-phagy alleviates zearalenone-induced ER stress and developmental defects in porcine embryos.

Zearalenone (ZEN), a mycotoxin commonly found in animal feed, impairs the female reproductive function owing to its estrogen-mimicking ability. ZEN exposure induces endoplasmic reticulum (ER) stress, oxidative stress, and autophagy activation. However, the involvement of inositol-requiring enzyme 1 (IRE1)-mediated ER-phagy in ZEN-induced ER stress and embryonic competence during in vitro culture (IVC) remains unclear. Porcine embryos were cultured with 2.5, 5, and 10 μM ZEN for two days after in vitro fertilization (IVF) to assess blastocyst development. ZEN exposure caused a significant dose-

Alginate-encapsulated enzymes mitigate deoxynivalenol-induced hepatic mitochondrial dysfunction and glycolithocholic acid accumulation in piglets.

BACKGROUND: Deoxynivalenol (DON), a mycotoxin commonly found in grains and feed, poses a serious threat to animal and public health. This study aimed to investigate the underlying mechanisms of DON-induced liver damage in piglets and to elucidate the potential of alginate-encapsulated enzyme in mitigating hepatotoxicity in vivo. RESULTS: The results indicated that DON exposure induced growth retardation and hepatic mitochondrial damage in piglets, which were effectively rescued by DON-degrading enzymes (DDE) supplementation. Hepatic transcriptomic profiling revealed that DON induced downregul

Supplementation with Yellow Mealworm (Tenebrio molitor) Larvae Grown on Deoxynivalenol-Contaminated Substrate Improves Growth and Gut Integrity in Broilers.

Deoxynivalenol (DON), a Fusarium mycotoxin frequently detected in poultry feeds, impairs broiler health by damaging gut integrity, altering immune response, and compromising growth performance. Insect-derived proteins, such as those from yellow mealworm (Tenebrio molitor) larvae meal (YMW), are gaining interest as an alternate protein source for broiler chickens. Hence, this study evaluated whether defatted YMW produced from larvae reared on DON-contaminated substrates can be safely included in broiler diets. A total of 400 one-day-old Cobb 700 mixed-sex broilers were assigned to four dietary

Aflatoxin and Liver Cancer in China: The Evolving Research Landscape.

Aflatoxins, particularly aflatoxin B1 (AFB1), are among the most potent naturally occurring carcinogens and remain a major food-borne hazard in parts of Asia and Africa. China has generated a uniquely cohesive body of evidence connecting aflatoxin contamination to hepatocellular carcinoma (HCC), especially in settings where chronic hepatitis B virus (HBV) infection is highly prevalent and acts synergistically with aflatoxin exposure. Over five decades, field investigations and laboratory innovations-exemplified by long-term work in Qidong-have assembled a multi-layered causal chain spanning th

Signal-on photoelectrochemical aptasensor for ultrasensitive zearalenone detection via cation exchange reaction amplification in COF-367 NSs/CdS QDs/Ag2S heterojunction.

BACKGROUND: Zearalenone (ZEN) is a non-steroidal estrogenic mycotoxin closely related to our health. It is of great significance to detect ZEN quickly and accurately in agricultural production. This study presents a novel signal-on photoelectrochemical (PEC) aptasensor for ultrasensitive detection of ZEN by integrating COF-367 nanosheets (NSs), CdS quantum dots (QDs), and Ag2S heterojunctions with cation exchange reaction (CER)-based signal amplification. The detection of ZEN was based on signal enhancement of COF-367 NSs/CdS QDs/Ag2S heterojunction and CER without any biological assembly, sol

Impact of Low-Level Ergot Alkaloids and Endophyte Presence in Tall Fescue Grass on the Metabolome and Microbiome of Fall-Grazing Steers.

Fescue toxicosis (FT) is a mycotoxin-related disease caused by the ingestion of tall fescue, naturally infected with the ergot alkaloid (EA)-producing endophyte Epichloë coenophiala. Some grazing on endophyte-free (E-) or non-toxic (NT), commercial endophyte-infected pastures takes place in the US as well. Earlier, we found that grazing on toxic fescue with low levels of EAs during fall affects thermoregulation, behavior, and weight gain. Building on these findings, the current study aimed to investigate how the presence of low EA-producing E+ or NT endophytes can influence animal metabolome,

Alleviating effect of ellagic acid on ovarian reserve function injury induced by zearalenone and its mechanism.

PURPOSE: Zearalenone (ZEA), a widespread mycotoxin, harms reproductive health in humans and animals. Ellagic acid (EA), a plant-derived polyphenol, can reduce such damage. This study explores how EA helps repair ZEA's effects on goat ovarian granulosa cells (OGCs) and murine ovarian. METHODS: The detoxifying effects of EA on ZEA were examined both in vitro and in vivo. PRINCIPAL RESULTS: In Vitro Studies: EA ameliorated ZEA-induced oxidative stress, mitochondrial dysfunction, and apoptosis in OGCs. Specifically, EA downregulated the expression of endoplasmic reticulum stress (ERS) genes (ERN

Necroptosis contributes to the intestinal toxicity of deoxynivalenol and is mediated by methyltransferase SETDB1.

Deoxynivalenol (DON) is a secondary metabolite produced by fungi, which causes serious health issues worldwide due to its widespread presence in human and animal diets. Necroptosis is a newly proposed cell death mode and has been proposed as a potential mechanism of intestinal disease. This study aimed to investigate the role of necroptosis in intestinal damage caused by DON exposure. Piglets were fed diets with or without 4 mg/kg DON for 3 weeks or given a gavage of 2 mg/kg BW DON or sterile saline to investigate the effects of chronic or acute DON exposure on the gut, respectively. Phenethyl isothiocyanate as an anti-nutritional factor attenuates deoxynivalenol-induced IPEC-J2 cell injury through inhibiting ROS-mediated autophagy.

Deoxynivalenol (DON) is considered to be the most harmful mycotoxin that affects the intestinal health of animals and humans. Phenethyl isothiocyanate (PEITC) in feedstuff is an anti-nutritional factor and impairs nutrient digestion and absorption in the animal intestinal. In the current study, we aimed to explore the effects of PEITC on DON-induced apoptosis, intestinal tight junction disorder, and its potential molecular mechanism in the porcine jejunum epithelial cell line (IPEC-J2). Our results indicated that PEITC treatment markedly alleviated DON-induced cytotoxicity, decreasing the apop

Structural insights into bacterial resistance to cerulenin.

UNLABELLED: Cerulenin is a fungal toxin that inhibits both eukaryotic and prokaryotic ketoacyl-acyl carrier protein synthases or condensing enzymes. It has been used experimentally to treat cancer and obesity, and is a potent inhibitor of bacterial growth. Understanding the molecular mechanisms of resistance to cerulenin and similar compounds is thus highly relevant for human health. We have previously described a Bacillus subtilis cerulenin-resistant strain, expressing a point-mutated condensing enzyme FabF (FabF[I108F]) (i.e. FabF with isoleucine 108 substituted by phenylalanine). We now rep

Effect of salicylic acid on Fusarium graminearum, the major causal agent of fusarium head blight in wheat.

Salicylic acid (SA) is one of the key signal molecules in regulating plant resistance to diverse pathogens. In Arabidopsis thaliana, it is predominantly associated with resistance against biotrophic and hemibiotrophic pathogens, and triggering systemic acquired resistance. In contrast, the effect of SA on the defence efficiency of wheat against fusarium head blight (FHB) and its causal agent, Fusarium graminearum, is still poorly understood. Here we show that the F. graminearum mycelial growth and conidia germination were significantly inhibited, and eventually halted in the presence of increa

The nephrotoxin ochratoxin A induces key parameters of chronic interstitial nephropathy in renal proximal tubular cells.

Ochratoxin A (OTA) is a nephrotoxic and cancerogenic mycotoxin. There is epidemiological evidence that OTA exposition leads to cortical interstitial nephropathies in humans. However, virtually no data are available investigating the effect of OTA on renal cortical cells with respect to induction of nephropathy. Thus, we investigated whether OTA is able to induce changes of cellular properties potentially leading to interstitial nephropathy, using proximal tubular cell lines (OK, NRK-52E). OTA decreased cell number and cell protein time and dose dependently. Accordingly we investigated the effe

Impairment of cell cycle progression by aflatoxin B1 in human cell lines.

Aflatoxin B1 is a mycotoxin produced by Aspergillus flavus and Aspergillus parasiticum, which may be present as a food contaminant. It is known to cause acute toxic effects and act as a carcinogenic agent. The carcinogenic action has been related to its ability to form unstable adducts with DNA, which represent possible mutagenic sites. On the other hand, the primary cellular target responsible for its toxic action has not yet been clearly identified. Previous data suggested a possible correlation between cell proliferation and responsiveness to aflatoxin toxicity. These observations led us to
