Reporter influenza A viruses (IAVs) are valuable tools for studying virus fitness, screening antivirals, and assessing host-virus interactions. However, the compact and segmented nature of the IAV genome presents major challenges for engineering genetically stable reporter viruses with minimal fitness defects. To address this, we developed a replication-competent IAV incorporating a tandem split-GFP reporter strategy. Specifically, we appended seven tandem repeats of the GFP11 peptide (GFP11×7) to the C-terminus of the PB2 or PA polymerase subunit. Viruses either contained or lacked a self-cle

Wastewater-based surveillance (WBS) of multidrug-resistant bacteria could complement clinical data, serving as a population-level early warning tool. This study evaluated WBS as a pandemic preparedness tool, by selectively isolating and culturing methicillin-resistant Staphylococcus aureus (MRSA) with CHROMagar MRSA. Some 24-h composite wastewater samples (n = 80) were collected from ten treatment plants across Finland between February 2021 and January 2022. MRSA prevalence in wastewater samples was 27.5% (n = 22/80), showing seasonal and temporal variations. Phenotypic antimicrobial susceptib

BACKGROUND: The burden of the COVID-19 pandemic in terms of morbidity and mortality differentially affected populations. Between and within populations, behavior change was likewise heterogeneous. Factors influencing precautionary behavior adoption during COVID-19 have been associated with multidimensional aspects of risk perception; however, the influence of lived experiences during other recent outbreaks on behavior change during COVID-19 has been less studied.
METHODS: To consider how the direct disease experience ("near misses") and behavior change during the 2014-2016 Ebola virus disease

BACKGROUND: Diagnosing a coronavirus disease 2019 (COVID-19) infection with high specificity in chest computed tomography (CT) imaging is considered possible due to distinctive imaging features of COVID-19 pneumonia. Since other viral non-COVID pneumonia show mostly a different distribution pattern, it is reasonable to assume that the patterns observed caused by the novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) are a consequence of its genetically encoded molecular properties when interacting with the respiratory tissue. As more mutations of the initial SARS-CoV-2 wild-typ

BACKGROUND: Vaccines induce antigen-specific immunity, which provides long-lived protection from the target pathogen. Trials from areas with high incidence rates for infectious diseases indicated that the tuberculosis vaccine Bacillus Calmette-Guérin (BCG) induces in addition non-specific immunity against various pathogens and thereby reduces overall mortality more than would have been expected by just protecting from tuberculosis. Although recent trials produced conflicting results, it was suggested that BCG might protect from non-tuberculosis respiratory infections and could be used to bridg

INTRODUCTION: The COVID-19 pandemic has forced a transition from in-person to virtual learning, and educators must innovate and adapt to keep learners engaged. One way to achieve this is through gamification.1 The authors employed a novel approach to gamification of virtual learning which engaged not only learners' computers but also their mobile phones. Learners worked in teams communicating by text message to answer ABEM board-style questions and occupy sites on the virtual board.

We previously demonstrated that the combination of synthetic small-molecule Toll-like receptor 4 (TLR4) and TLR7 ligands is a potent adjuvant for recombinant influenza virus hemagglutinin, inducing rapid and sustained immunity that is protective against influenza viruses in homologous, heterologous, and heterosubtypic murine challenge models. Combining the TLR4 and TLR7 ligands balances Th1 and Th2-type immune responses for long-lived cellular and neutralizing humoral immunity against the viral hemagglutinin. Here, we demonstrate that the protective response induced in mice by this combined ad

UNLABELLED: Influenza virus infections are a major public health concern and cause significant morbidity and mortality worldwide. Current vaccines are effective but strain specific due to their focus on the immunodominant globular head domain of the hemagglutinin (HA). It has been hypothesized that sequential exposure of humans to hemagglutinins with divergent globular head domains but conserved stalk domains could refocus the immune response to broadly neutralizing epitopes in the stalk. Humans have preexisting immunity against H1 (group 1 hemagglutinin), and vaccination with H5 HA (also grou

UNLABELLED: Norovirus is the leading cause of acute gastroenteritis, responsible for 685 million cases and 200,000 deaths annually. Wastewater-based epidemiology (WBE) enables not only global surveillance of dominant strains but also the detection of cryptic and emerging viral variants. An important question, however, is how to link genomic diversity to clinical impact and phenotypic traits of the strains detected in wastewater. In the Global Urban Wastewater Project, phylogenetically distinct norovirus GI.3 and GII.12 lineages were detected in wastewater from Cameroon. In this study, we chara

BACKGROUND: The COVID-19 health crisis rapidly escalated into a broader social and economic emergency, affecting physical and mental health beyond the disease itself and placing a disproportionate burden on vulnerable populations. This study investigates the determinants of poor mental health related to COVID-19 among urban-poor homemakers residing in a social housing villa located on the outskirts of Santiago de Chile, a metropolitan city characterized by pronounced sociospatial segregation. METHODS: Capitalizing on the ongoing RUCAS cohort study, we introduced a COVID-19 module into the bian

AI is emerging as a promising tool for diagnosing COVID-19 based on chest CT scans. The aim of this study was the comparison of AI models for COVID-19 diagnosis. Therefore, we: (1) trained three distinct AI models for classifying COVID-19 and non-COVID-19 pneumonia (nCP) using a large, clinically relevant CT dataset, (2) evaluated the models' performance using an independent test set, and (3) compared the models both algorithmically and experimentally. In this multicenter multi-vendor study, we collected n=1591 chest CT scans of COVID-19 (n=762) and nCP (n=829) patients from China and Germany.

BACKGROUND: The COVID-19 pandemic exposed systemic vulnerabilities in public health infrastructure, underscoring the urgency for innovation in disease surveillance and emergency response. Artificial intelligence (AI) has emerged as a promising tool to enhance the accuracy, efficiency, and scalability of public health interventions. Yet, there remains a limited understanding of how AI has been applied in real-world infectious disease control and who is contributing to its development and implementation.
OBJECTIVE: This scoping review aimed to map current applications of AI in public health pra

The COVID-19 pandemic highlighted the importance of laboratory preparedness. Regular monitoring of diagnostic tools via external quality assessments (EQAs) is key to maintaining robust public health response service. We hereby conducted a third SARS-CoV-2 EQA assessing the diagnostic capabilities of European expert public health laboratories. A 10 samples panel containing Alpha (used in previous EQA), BA.4, BA.5, and BQ.1.18 variants along with human seasonal coronaviruses and negative controls was produced and validated. Participants were invited by the European Centre for Disease Prevention

Subtype H10 influenza A viruses (IAVs) have been recovered from domestic poultry and various aquatic bird species, and sporadic transmission of these IAVs from avian species to mammals (i.e., human, seal, and mink) are well documented. In 2015, we isolated four H10N7 viruses from gulls in Iceland. Genomic analyses showed four gene segments in the viruses were genetically associated with H10 IAVs that caused influenza outbreaks and deaths among European seals in 2014. Antigenic characterization suggested minimal antigenic variation among these H10N7 isolates and other archived H10 viruses recov

Infection with SARS-CoV-2 variants typically confers protective humoral immunity. Yet, the Omicron variant demonstrates a considerable capacity to evade immune responses in individuals previously infected with other SARS-CoV-2 strains. This has prompted investigation into Omicron's potential resistance to neutralizing antibodies elicited by natural infection or vaccination. Importantly, the molecular mechanisms underlying this cross-protection have been investigated; however, they are not yet fully elucidated. In this study, we investigated the neutralization capacity of convalescent sera from

UNLABELLED: A better understanding of viral factors that contribute to influenza A virus (IAV) airborne transmission is crucial for pandemic preparedness. A limited capacity for airborne transmission was recently observed in a human A(H9N2) virus isolate (A/Anhui-Lujiang/39/2018, AL/39) that possesses a leucine (L) residue at position HA1-226 (H3 numbering), indicative of human-like receptor binding potential. To evaluate the roles of the residue at this position in virus fitness and airborne transmission, a wild-type AL/39 (AL/39-wt) and a mutant virus (AL/39-HA1-L226Q) with a single substitu
