Leveraging electronic health records to study pleiotropic effects on bipolar disorder and medical comorbidities.

Patients with bipolar disorder (BD) have a high prevalence of comorbid medical illness. However, the mechanisms underlying these comorbidities with BD are not well known. Certain genetic variants may have pleiotropic effects, increasing the risk of BD and other medical illnesses simultaneously. In this study, we evaluated the association of BD-susceptibility genetic variants with various medical conditions that tend to co-exist with BD, using electronic health records (EHR) data linked to genome-wide single-nucleotide polymorphism (SNP) data. Data from 7316 Caucasian subjects were used to test

Pleiotropic effect of the proton pump inhibitor esomeprazole leading to suppression of lung inflammation and fibrosis.

BACKGROUND: The beneficial outcome associated with the use of proton pump inhibitors (PPIs) in idiopathic pulmonary fibrosis (IPF) has been reported in retrospective studies. To date, no prospective study has been conducted to confirm these outcomes. In addition, the potential mechanism by which PPIs improve measures of lung function and/or transplant-free survival in IPF has not been elucidated. METHODS: Here, we used biochemical, cell biological and preclinical studies to evaluate regulation of markers associated with inflammation and fibrosis. In our in vitro studies, we exposed primary lu

Exploring the silent connection: unveiling the intricate relationship between gastroesophageal reflux disease and sleep apnea syndrome.

BACKGROUND: Gastroesophageal reflux disease (GERD) and Sleep Apnea Syndrome (SAS) are two prevalent medical conditions that significantly affect health and quality of life. GERD involves stomach content reflux into the esophagus, while SAS causes recurrent upper airway obstruction during sleep. Despite recent studies hinting at a link, the precise relationship and causality between GERD and SAS remain unclear. Our research uses bidirectional Mendelian randomization to explore this intricate relationship. Additionally, given SAS's high prevalence in cardiovascular patients (40-80%, as highlight

Bayesian-weighted Mendelian randomization reveals sleep apnea syndrome mediates the BMI-chronic pain link.

BACKGROUND: Body mass index (BMI) has a complex association with a variety of chronic pain conditions, which may be influenced by sleep apnea syndrome (SAS). However, the role of SAS within the causal pathway linking BMI to chronic pain remains unconfirmed. This study explored whether and to what extent SAS serves as a mediator between BMI and chronic pain using Bayesian-weighted Mendelian randomization (BWMR) techniques. METHODS: This study utilized genome-wide association study (GWAS) summary data for BMI (N=461,460) and SAS (N=476,853; based on ICD-10 code diagnostic registries) from the I

Mediation analysis of the causal pathway from gastroesophageal reflux disease to pancreatitis: A Mendelian randomization study.

Although gastroesophageal reflux disease (GERD) and pancreatitis are both common gastrointestinal disorders, few studies have investigated their potential causal relationship. Clarifying whether GERD contributes to the development of acute pancreatitis (AP) and chronic pancreatitis (CP), and the possible mediating role of cholelithiasis, may provide new insights into shared pathogenic mechanisms. A univariable Mendelian randomization (UVMR) analysis was performed to assess the causal effects of GERD on AP and CP. To improve statistical power, the results from the discovery and replication coho

Causal effects of psoriasis on breast cancer risk: The contribution of emotional distress explored by Mendelian randomization.

Psoriasis is a chronic inflammatory disease associated with an increased risk of various cancers, including breast cancer. However, the mechanisms underlying this association remain poorly understood. Negative emotional states, commonly experienced by individuals with psoriasis, may play a role in mediating this link. This study aims to investigate the causal effect of psoriasis on breast cancer risk and explore the potential mediating role of negative emotions using Mendelian randomization (MR) approaches. Genetic instruments for psoriasis and negative emotions were sourced from large-scale g

Investigating the metabolomic pathways in female reproductive endocrine disorders: a Mendelian randomization study.

INTRODUCTION: Reproductive endocrine disorders (RED), including polycystic ovary syndrome (PCOS), endometriosis (EMs), and female infertility (FI), significantly affect women's health globally, with varying prevalence across different regions. These conditions can be addressed through medication, surgical interventions, and lifestyle modifications. However, the limited understanding of RED's etiology and the substantial economic burden of its treatment highlight the importance of investigating its pathogenesis. Metabolites play a critical role in metabolic processes and are potentially linked

Integrating multi-omics and network toxicology to identify FIS1 as a key target of environmental pollutants in male infertility.

BACKGROUND: Male infertility (MI) impacts about one in seven couples globally, involving complex gene-environment interactions. Environmental pollutants may disrupt spermatogenesis via mitochondrial dysfunction, but key targets and mechanisms are unclear. Integrating multi-omics with computational toxicology offers a novel strategy to decipher these interactions. METHODS: We integrated blood transcriptomics from MI patients, GWAS summary statistics, and QTL data (methylation, expression, protein). MI-related mitochondrial genes were identified through differential expression analysis, followe

Mendelian randomization reveals probucol's preventive role in Behçet's disease via circulating metabolites.

Behçet's disease (BD) is a chronic, recurrent condition for which effective preventive medications are currently lacking. This disease often disrupts lipid metabolism, adversely affecting vascular endothelial function. Exploring preventive strategies, such as lipid-lowering medications, is crucial. Probucol, known for its lipid-lowering properties, emerges as a promising candidate. By inhibiting the ATP-binding cassette transporter A1 (ABCA1), probucol is hypothesized to regulate circulating metabolites, potentially reducing the risk of BD. This study employs Mendelian randomization (MR) to ev
