Causal relationships between hundreds of plasma metabolites and PTSD: a bidirectional mendelian randomization study.

BACKGROUND: Recent studies have indicated a connection between plasma metabolites and Post-traumatic stress disorder (PTSD). Nevertheless, the precise causal relationship remains unclear. METHODS: We performed bidirectional Mendelian Randomization (MR) using two metabolite and two PTSD GWAS datasets to examine causal relationships between PTSD and 1009 plasma metabolites. Forward MR tested metabolite causally effects on PTSD, while reverse MR assessed PTSD causally effects on metabolites. Primary analysis employed the IVW method, supported by four supplementary methods. Four IVW results per d

Causal association of antibody-mediated immune responses to infectious disease agents with lung cancer: a Mendelian randomization study.

BACKGROUND: There is a causal relationship between many non-communicable diseases and other infectious diseases. However, the exact relationship between lung cancer and the immune response to antibodies associated with viral diseases is unclear. Compared with other traditional methods, Mendelian randomization (MR) studies reduce the influence of confounding factors, thus improving the accuracy of the results. This study used MR to investigate the relationship between lung cancer and its subtypes, and the antibody immune response. METHODS: A two-sample MR method was employed to examine the ass

Association of peripheral B cells and delirium: combined single-cell sequencing and Mendelian randomization analysis.

BACKGROUND: Delirium seriously affects the prognosis of patients and greatly reduces the ability to work and live. Peripheral inflammatory events may contribute to the development of delirium, the mechanism of which is still unclear. There is a lack of effective diagnostic and treatments for delirium in clinical practice. The study aims to investigate alterations in peripheral immune cell subsets under inflammatory stress and to explore causal associations with delirium. METHODS: Single-cell transcriptional sequencing data of human peripheral blood mononuclear cells (PBMC) before and after li

Associations between sleep-behavioral traits and healthspan: A one-sample Mendelian randomization study based on 388,909 participants of the UK-Biobank.

BACKGROUND: Although the association between sleep behavior and morbidity and mortality risk has been reported before, there is still uncertainty whether the observed associations are causal or confounding. Therefore, we investigated the causal relationships between sleep-behavioral traits and terminated healthspan risk using Mendelian randomization (MR). METHODS: We conducted a one-sample MR analysis to evaluate causality between six sleep-behavioral traits (sleep duration, chronotype/morningness, napping, sleeplessness/insomnia, and getting up from bed) and risk of healthspan termination am

Causal relationships between rheumatoid arthritis and neurodegenerative diseases: a two-sample univariable and multivariable Mendelian randomization study.

BACKGROUND: Observational research has highlighted a potential relationship between rheumatoid arthritis (RA) and neurodegenerative diseases (NDs). However, the confirmation of a causal connection is impeded by the inherent limitations of such studies, including vulnerability to confounding factors and the possibility of reverse causality. This study employs a two-sample Mendelian randomization (MR) approach to assess the causal impact of RA on three NDs, including Alzheimer's disease (AD), Parkinson's disease (PD), and amyotrophic lateral sclerosis (ALS). METHODS: We aggregated data from gen

Investigating genetic links between blood metabolites and preeclampsia.

BACKGROUND: Observational studies have revealed that metabolic disorders are closely related to the development of preeclampsia (PE). However, there is still a research gap on the causal role of metabolites in promoting or preventing PE. We aimed to systematically explore the causal association between circulating metabolites and PE. METHODS: Single nucleotide polymorphisms (SNPs) from genome-wide association study (GWAS) of 486 blood metabolites (7,824 participants) were extracted as instrumental variables (P < 1 × 10- 5), GWAS summary statistics for PE were obtained from FinnGen consortium

Mendelian randomization analysis and validation supports MEGF9 and MLLT11 as potential targets for the treatment of varicocele and male infertility.

OBJECTIVE: A growing body of research suggests a link between varicocele and male infertility (MI). However, current evidence is mainly based on retrospective studies, which are prone to interference from confounding factors and cannot establish causal relationships. Mendelian randomization (MR) studies on the causal relationship between varicocele and MI are very limited. Therefore, this study conducted a two-sample MR study to elucidate the causal effect between the two. METHODS: Download the data set GSE216907 from the GEO database, and use R software to screen differential genes in normal

Causal effects of specific gut microbiota on musculoskeletal diseases: a bidirectional two-sample Mendelian randomization study.

BACKGROUND: Recent observational studies and clinical trials demonstrated an association between gut microbiota and musculoskeletal (MSK) diseases. Nonetheless, whether the gut microbiota composition has a causal effect on the risk of MSK diseases remains unclear. METHODS: Based on large-scale genome-wide association studies (GWAS), we performed a two-sample Mendelian randomization (MR) analysis to investigate the causal relationship between gut microbiota and six MSK diseases, namely osteoporosis (OP), fracture, sarcopenia, low back pain (LBP), rheumatoid arthritis (RA), and ankylosing spond

Thyroid disease and breast cancer, benign breast neoplasm: a two-sample Mendelian randomization study.

BACKGROUND: Breast cancer (BC) is a prevalent and significant health issue and a major contributor to global cancer incidence, accounting for 31% of all reported cases in women. Benign breast neoplasm, as a benign tumor with a high incidence in women, may play an important role in the development of BC. Previous studies have shown that thyroid dysfunction and thyroid cancer (TC) can lead to the occurrence of many cancers. Therefore, we conduct Mendelian randomization (MR) analysis to explore the causality of thyroid dysfunctions, TC, and breast neoplasm. METHODS: The data of the analysis from

Causal effect of blood osteocalcin on the risk of Alzheimer's disease and the mediating role of energy metabolism.

Growing evidence suggests an association between osteocalcin (OCN), a peptide derived from bone and involved in regulating glucose and lipid metabolism, and the risk of Alzheimer's disease (AD). However, the causality of these associations and the underlying mechanisms remain uncertain. We utilized a Mendelian randomization (MR) approach to investigate the causal effects of blood OCN levels on AD and to assess the potential involvement of glucose and lipid metabolism. Independent instrumental variables strongly associated (P < 5E-08) with blood OCN levels were obtained from three independent g

Causal relationships between blood lipids and major psychiatric disorders: Univariable and multivariable mendelian randomization analysis.

BACKGROUND: Whether the positive associations of blood lipids with psychiatric disorders are causal is uncertain. We conducted this two-sample Mendelian randomization (MR) analysis to comprehensively investigate associations of blood lipids with psychiatric disorders. METHODS: Univariable and multivariable models were established for MR analyses. Inverse variance-weighted (IVW) MR was employed as the main approach; weighted median and MR-Egger were used as sensitivity analysis methods. The possibility of violating MR assumptions was evaluated utilizing several sensitivity analyses, including

Multi-omics study of ABCA1 in sepsis: Integrating mendelian randomisation, single-cell transcriptomics, in silico knockout and virtual screening.

BACKGROUND: Sepsis is a life-threatening organ dysfunction caused by a dysreg- ulated host response to infection, with the immune status dynamically evolving from an early hyperinflammatory phase to a late immunosuppressive phase. ABCA1 (ATP-binding cassette transporter A1) is involved in cholesterol efflux and inflammatory regulation, but its causal relationship with sepsis, cellular ori- gin, and potential for drug targeting remain unclear. This study is the first to integrate Mendelian randomisation, single-cell transcriptomics, in silico knock- out, and virtual screening to systematically
