Tracking and localization of calmodulin in live cells.

The calcium signaling protein calmodulin (CaM) interacts with many target proteins inside the cell to regulate a wide range of biological signals. CaM's availability to propagate signals depends on its mobility, which may be regulated by interactions with multiple target proteins. We detected single molecules of CaM labeled with a fluorescent dye and injected into living HEK 293 cells, and we used high-speed, wide-field, single-molecule imaging to track single CaM molecules. Single-molecule trajectories were analyzed to characterize the motions of individual CaM molecules. Single-molecule loca

Gait behaviors as an objective surgical outcome in low back disorders: A systematic review.

BACKGROUND: Objective motor performance measures, especially gait assessment, could improve evaluation of low back disorder surgeries. However, no study has compared the relative effectiveness of gait parameters for assessing motor performance in low back disorders after surgery. The purpose of the current review was to determine the sensitive gait parameters that address physical improvements in each specific spinal disorder after surgical intervention. METHODS: Articles were searched with the following inclusion criteria: 1) population studied consisted of individuals with low back disorder

Physiology of the oesophageal transition zone in the presence of chronic bolus retention: studies using concurrent high resolution manometry and digital fluoroscopy.

Distinct contraction waves (CWs) exist above and below the transition zone (TZ) between the striated and smooth muscle oesophagus. We hypothesize that bolus transport is impaired in patients with abnormal spatio-temporal coordination and/or contractile pressure in the TZ. Concurrent high resolution manometry and digital fluoroscopy were performed in healthy subjects and patients with reflux oesophagitis; a condition associated with ineffective oesophageal contractility and clearance. A detailed analysis of space-time variations in bolus movement, intra-bolus and intra-luminal pressure was perf

Gait and posturographic parameters of children with achondroplasia - a long-term pilot study.

Walking and maintaining balance are essential for participating in daily life. For children with achondroplasia, reaching these milestones is with a delay and during growth, challenges persist for those skills. However, the impact of developmental changes on gait and balance in this population has not yet been investigated longitudinally. Therefore, this study aimed to examine the influence of growth on gait pattern and postural control in children with achondroplasia over a two-year period. 6 children with achondroplasia (whereas 5 are treated with vosoritide, a growth promoting peptide) and

Cuban history of CRF19 recombinant subtype of HIV-1.

CRF19 is a recombinant form of HIV-1 subtypes D, A1 and G, which was first sampled in Cuba in 1999, but was already present there in 1980s. CRF19 was reported almost uniquely in Cuba, where it accounts for ∼25% of new HIV-positive patients and causes rapid progression to AIDS (∼3 years). We analyzed a large data set comprising ∼350 pol and env sequences sampled in Cuba over the last 15 years and ∼350 from Los Alamos database. This data set contained both CRF19 (∼315), and A1, D and G sequences. We performed and combined analyses for the three A1, G and D regions, using fast maximum likelihood

Spatiotemporal distribution, abundance, and host interactions of two invasive vectors of arboviruses, Aedes albopictus and Aedes japonicus, in Pennsylvania, USA.

BACKGROUND: Aedes albopictus and Aedes japonicus, two invasive mosquito species in the United States, are implicated in the transmission of arboviruses. Studies have shown interactions of these two mosquito species with a variety of vertebrate hosts; however, regional differences exist and may influence their contribution to arbovirus transmission. METHODS: We investigated the distribution, abundance, host interactions, and West Nile virus infection prevalence of Ae. albopictus and Ae. japonicus by examining Pennsylvania mosquito and arbovirus surveillance data for the period between 2010 and

Transcriptome-wide transmission disequilibrium analysis identifies novel risk genes for autism spectrum disorder.

Recent advances in consortium-scale genome-wide association studies (GWAS) have highlighted the involvement of common genetic variants in autism spectrum disorder (ASD), but our understanding of their etiologic roles, especially the interplay with rare variants, is incomplete. In this work, we introduce an analytical framework to quantify the transmission disequilibrium of genetically regulated gene expression from parents to offspring. We applied this framework to conduct a transcriptome-wide association study (TWAS) on 7,805 ASD proband-parent trios, and replicated our findings using 35,740

Long-term cognitive deficits after traumatic brain injury associated with microglia activation.

Traumatic Brain Injury (TBI) is the most prevalent of all head injuries. Microglia play an essential role in homeostasis and diseases of the central nervous system. We hypothesize that microglia may play a beneficial or detrimental role in TBI depending on their state of activation and duration. In this study, we evaluated whether TBI results in a spatiotemporal change in microglia phenotype and whether it affects sensory-motor or learning and memory functions in male C57BL/6 mice. We used a panel of neurological and behavioral tests and a multi-color flow cytometry-based data analysis followe

Spatial and Temporal Analysis of the Stomach and Small-Intestinal Microbiota in Fasted Healthy Humans.

Although the microbiota in the proximal gastrointestinal (GI) tract have been implicated in health and disease, much about these microbes remains understudied compared to those in the distal GI tract. This study characterized the microbiota across multiple proximal GI sites over time in healthy individuals. As part of a study of the pharmacokinetics of oral mesalamine administration, healthy, fasted volunteers (n = 8; 10 observation periods total) were orally intubated with a four-lumen catheter with multiple aspiration ports. Samples were taken from stomach, duodenal, and multiple jejunal sit

Mild hyperthermia by MR-guided focused ultrasound in an ex vivo model of osteolytic bone tumour: optimization of the spatio-temporal control of the delivered temperature.

BACKGROUND: Magnetic resonance guided focused ultrasound was suggested for the induction of deep localized hyperthermia adjuvant to radiation- or chemotherapy. In this study we are aiming to validate an experimental model for the induction of uniform temperature elevation in osteolytic bone tumours, using the natural acoustic window provided by the cortical breakthrough. MATERIALS AND METHODS: Experiments were conducted on ex vivo lamb shank by mimicking osteolytic bone tumours. The cortical breakthrough was exploited to induce hyperthermia inside the medullar cavity by delivering acoustic en

Outcome measures of instrumented gait analysis in hereditary spastic paraplegia: a systematic review.

BACKGROUND: Hereditary spastic paraplegias (HSPs) comprise a group of genetic movement disorders characterized by progressive spasticity and weakness of the lower limbs leading to gait deficits. Instrumented gait measures are applied to quantify gait patterns in HSP objectively. However, there is no consensus on the most relevant HSP-specific digital outcome measures for future clinical studies. AIM: This systematic review aims to summarize outcome measures of instrumented gait analysis in HSP patients, focusing on both traditional motion capture (MOCAP) and inertial sensor systems. METHODS:

Spatial and phenotypic heterogeneity of resident and monocyte-derived macrophages during inflammatory exacerbations leading to pulmonary fibrosis.

INTRODUCTION: Genetic mutations in critical nodes of pulmonary epithelial function are linked to the pathogenesis of pulmonary fibrosis (PF) and other interstitial lung diseases. The slow progression of these pathologies is often intermitted and accelerated by acute exacerbations, complex non-resolving cycles of inflammation and parenchymal damage, resulting in lung function decline and death. Excess monocyte mobilization during the initial phase of an acute exacerbation, and their long-term persistence in the lung, is linked to poor disease outcome. METHODS: The present work leverages a clin

Spatio-temporal variability of near-surface air pollutants at four distinct geographical locations in Andhra Pradesh State of India.

India is highly vulnerable to air pollution in the recent decade, especially urban areas with rapidly growing urbanisation and industrialisation. Here, we present spatio-temporal variability of air pollutants at four distinct locations in Andhra Pradesh State of India. The mean concentrations of air pollutants were generally higher at Visakhapatnam site than Amaravati, Rajahmundry, and Tirumala sites. The mean concentration of particulate matter of diameter less than 2.5 μm (PM2.5) was higher at Visakhapatnam site (48.5 ± 27.3 μg/m3) by a factor of about 1.6 as compared to Tirumala site (29.5

Gait phenotypes in paediatric hereditary spastic paraplegia revealed by dynamic time warping analysis and random forests.

The Hereditary Spastic Paraplegias (HSP) are a group of heterogeneous disorders with a wide spectrum of underlying neural pathology, and hence HSP patients express a variety of gait abnormalities. Classification of these phenotypes may help in monitoring disease progression and personalizing therapies. This is currently managed by measuring values of some kinematic and spatio-temporal parameters at certain moments during the gait cycle, either in the doctor´s surgery room or after very precise measurements produced by instrumental gait analysis (IGA). These methods, however, do not provide inf

Digital Gait Measures Capture 1-Year Progression in Early-Stage Spinocerebellar Ataxia Type 2.

BACKGROUND: With disease-modifying drugs in reach for cerebellar ataxias, fine-grained digital health measures are highly warranted to complement clinical and patient-reported outcome measures in upcoming treatment trials and treatment monitoring. These measures need to demonstrate sensitivity to capture change, in particular in the early stages of the disease. OBJECTIVE: Our aim is to unravel gait measures sensitive to longitudinal change in the-particularly trial-relevant-early stage of spinocerebellar ataxia type 2 (SCA2). METHODS: We performed a multicenter longitudinal study with combin

Vertically resolved meteorological adjustments of aerosols and trace gases in Beijing, Taiyuan, and Hefei by using RF model.

Air pollution represents a complex phenomenon defined by the presence of various gases and particulate matter, leading to intricate spatio-temporal fluctuations. This study aims to enhance our understanding of how meteorological factors influence trace gases and aerosols, exacerbating air pollution in various geographical locations, specifically in Beijing's Fengtai (BJFT), Taiyuan City (SXTY), and Hefei's Science Island (HFDP). The study employs 2D-MAX-DOAS observations and utilizes the Random Forest (RF) model to decouple the influence of meteorological conditions from pollutant data. The ve

Influence of sagittal pelvic attitude on gait pattern in normally developed people and interactions with neurological pathologies: A pilot study.

BACKGROUND: Gait Analysis of healthy people, imitating pathological conditions while walking, has increased our understanding of biomechanical factors. The influence of the pelvis as a biomechanical constraint during gait is not specifically studied. How could mimicking a pelvic attitude influence the dynamic mechanical interaction of the body segments? We proposed an investigation of the pelvic attitude role on the gait pattern of typically developed people when they mimicked pelvic anteversion and posteroversion. MATERIALS AND METHODS: Seventeen healthy volunteers were enrolled in this stud

[Driving Mechanism of Carbon Emissions in China's Agriculture and the Economic Decoupling Effect].

Agriculture is a significant sector of carbon emissions in China. Clarifying its emission characteristics and driving mechanisms is of great significance for promoting the green and low-carbon transformation of agriculture. Based on the panel data of 31 provincial regions across the country from 2011 to 2022, this study uses the IPCC emission coefficient method to calculate the total amount and intensity of agricultural carbon emissions, revealing their spatio-temporal evolution characteristics. Research findings indicate that the overall carbon emissions from agriculture across the country ha
