Follistatin levels and endocrine disorders: A two-sample Mendelian randomization study.

To investigate the causal relationship between follistatin (FST) levels and endocrine diseases such as polycystic ovary syndrome (PCOS), type 2 diabetes (T2DM), obesity, and osteoporosis (OP) using a 2-sample Mendelian randomization (MR) analysis. Instrumental variables closely associated with FST levels were obtained from large-scale genome-wide association study data in the IEU database. Summary-level data for 4 endocrine diseases were sourced from the latest version of the FinnGen database. Our primary method for MR analysis was the inverse-variance weighted (IVW) method, supplemented by th

Immune cell traits and causal relationships with cholecystitis: a mendelian randomization analysis.

Cholecystitis, characterized by inflammation of the gallbladder, is intricately linked to immune cells and the cytokines they produce. Despite this association, the specific contributions of immune cells to the onset and progression of cholecystitis remain to be fully understood. To delineate this relationship, we utilized the Mendelian randomization (MR) method to scrutinize the causal connections between 731 immune cell phenotypes and cholecystitis. By conducting MR analysis on 731 immune cell markers from public datasets, this study seeks to understand their potential impact on the risk of

Causal impact of gut microbiota and associated metabolites on pulmonary arterial hypertension: a bidirectional Mendelian randomization study.

BACKGROUND: Patients with pulmonary arterial hypertension (PAH) exhibit a distinct gut microbiota profile; however, the causal association between gut microbiota, associated metabolites, and PAH remains elusive. We aimed to investigate this causal association and to explore whether dietary patterns play a role in its regulation. METHODS: Summary statistics of gut microbiota, associated metabolites, diet, and PAH were obtained from genome-wide association studies. The inverse variance weighted method was primarily used to measure the causal effect, with sensitivity analyses using the weighted

Causal effect of body mass index and physical activity on the risk of joint sports injuries: Mendelian randomization analysis in the European population.

BACKGROUND: Observational studies can suggest potential associations between variables but cannot establish a causal effect on their own. This study explored the causal associations between body mass index (BMI), physical activity (PA), and joint sports injuries. METHODS: We conducted two-sample Mendelian randomization (MR) using publicly accessed genome-wide association studies (GWAS) datasets to investigate the causal effects of BMI and PA on joint sports injury risk. The inverse-variance weighted method was believed to be the primary MR analysis. Subsequently, sensitivity, pleiotropy, and

Reassessing the relationship between major depressive disorder and blood lipids: a comprehensive Mendelian randomisation study.

BACKGROUND: Numerous studies have consistently demonstrated that a considerable proportion of patients with major depressive disorder (MDD) frequently exhibit pronounced dyslipidaemia. However, the causal dynamics between MDD and dyslipidaemia remain elusive. AIMS: To comprehensively disentangle the genetic causality between MDD and various phenotypes of blood lipids, thereby facilitating the advancement of management strategies for these conditions. METHODS: We conducted a two-sample univariable Mendelian randomisation (MR) analysis using different models, including the inverse variance wei

Understanding Factors That Cause Benign Paroxysmal Positional Vertigo, Ménière Disease, and Vestibular Neuritis: A Two-Sample Mendelian Randomization Study.

OBJECTIVES: Vertigo is a prevalent clinical symptom, frequently associated with benign paroxysmal positional vertigo (BPPV), Ménière disease (MD), and vestibular neuritis (VN), which are three common peripheral vestibular disorders. However, there is a relative lack of research in epidemiology and etiology, with some existing studies presenting discrepancies in their conclusions. We conducted a two-sample Mendelian randomization (MR) analysis to explore potential risk and protective factors for these three peripheral vestibular disorders. DESIGN: Based on genome-wide association studies, we e

Body mass index and type 2 diabetes liability in osteomyelitis and cellulitis: A cross-biobank Mendelian randomization study.

Previous Mendelian randomization (MR) studies have linked obesity and type 2 diabetes (T2D) liability to osteomyelitis and skin or soft-tissue infection, but the reproducibility of these associations across independent outcome biobanks remains uncertain. We conducted cross-biobank univariable MR and multivariable MR analyses using European-ancestry genome-wide association study summary statistics. FinnGen Release 13 osteomyelitis (2519 cases and 471,012 controls) was designated as the primary clinical outcome, whereas cellulitis (10,223 cases and 468,418 controls) served as a comparative infec

The effects of coagulation factors on the risk of endometriosis: a Mendelian randomization study.

BACKGROUND: Endometriosis is recognized as a complex gynecological disorder that can cause severe pain and infertility, affecting 6-10% of all reproductive-aged women. Endometriosis is a condition in which endometrial tissue, which normally lines the inside of the uterus, deposits in other tissues. The etiology and pathogenesis of endometriosis remain ambiguous. Despite debates, it is generally agreed that endometriosis is a chronic inflammatory disease, and patients with endometriosis appear to be in a hypercoagulable state. The coagulation system plays important roles in hemostasis and infla

Tuber intake is independently associated with reduced risk of Hashimoto's thyroiditis: a community-based cross-sectional study.

BACKGROUND: Hashimoto's thyroiditis (HT) is the leading cause of autoimmune hypothyroidism, and dietary factors have been implicated in its pathogenesis through immune and metabolic pathways. We aimed to identify dietary and metabolic factors associated with HT risk in a large community cohort and to evaluate whether tuber-associated metabolites causally influence HT susceptibility using Mendelian randomization (MR). METHODS: Cross-sectional analysis of 7,878 community-dwelling adults (≥40 years) from the REACTION cohort, Dalian, China. HT was defined by thyroid autoantibody seropositivity (a

Causal association between remnant cholesterol level and risk of cardiovascular diseases: a bidirectional two sample mendelian randomization study.

Serum lipids have been associated with an increased risk of various cardiovascular diseases (CVDs) in several observational studies, but the causal inference between the remnant cholesterol (RC) levels and several CVDs risk has not been established. The purpose of this study was to investigate whether there is a causal relationship between RC levels and risk of developing CVDs by a bidirectional two-sample Mendelian randomization (TSMR) analysis. One TSMR analysis was performed using the publicly released large-scale genome-wide association study (GWAS) data. Inverse variance weighted (IVW) me

Causal associations between gut microbiota and inflammatory factors in neonatal jaundice: a Mendelian randomization study.

BACKGROUND: Neonatal jaundice is a common clinical condition, affecting approximately 60% of full-term infants and 80% of preterm infants. The majority of neonatal jaundice represents benign physiological hyperbilirubinemia that resolves spontaneously, yet a subset progresses to clinically significant hyperbilirubinemia (CSH), defined as elevated serum bilirubin requiring phototherapy or exchange transfusion to prevent permanent neurotoxic injury. The gut microbiota plays a crucial role in human health and disease, with inflammatory mediators frequently contributing to the body's response to i

Estimating causal effects of C-reactive protein on disease and health outcomes using multivariable Mendelian randomization adjusting for heritable confounding.

BACKGROUND: C-reactive protein (CRP) is a marker of inflammation associated with autoimmune, cardiovascular, and neuropsychiatric disorders. However, it remains unclear whether CRP causally affects these traits or if observed associations result from reverse causation or confounding. Mendelian randomization (MR) uses genetic variants as instrumental variables to estimate causal effects and avoid the biases present in observational studies. Prior MR studies have suggested causal effects of CRP on several traits, including low-density lipoprotein (LDL) cholesterol, schizophrenia (SCZ), and knee
