Investigating the Sudan virus outbreak in Uganda through the deployment of a mobile laboratory.

BACKGROUND: Uganda faced its sixth reported Ebola outbreak between September 2022 and January 2023, the fifth to be recorded as Sudan Virus Disease (SVD) in the country. In response to this, the Ugandan Ministry of Health (MoH), rapidly deployed a mobile laboratory from the National Health Laboratory and Diagnostics Services (NHLDS) within one week of the declared outbreak. Here we describe the deployment of the mobile laboratory to Mubende, the outbreak epicentre, as part of the national response. This provided (1) efficient diagnostics and characterization of Sudan virus cases to support nat

Developing global climate anomalies suggest potential disease risks for 2006-2007.

BACKGROUND: El Niño/Southern Oscillation (ENSO) related climate anomalies have been shown to have an impact on infectious disease outbreaks. The Climate Prediction Center of the National Oceanic and Atmospheric Administration (NOAA/CPC) has recently issued an unscheduled El Niño advisory, indicating that warmer than normal sea surface temperatures across the equatorial eastern Pacific may have pronounced impacts on global tropical precipitation patterns extending into the northern hemisphere particularly over North America. Building evidence of the links between ENSO driven climate anomalies a

Zoonotic risks of pathogens from dairy cattle and their milk-borne transmission.

Dairy products are major sources of high-quality protein and bioavailable nutrients and dairy production contributes to local, regional and national-level economies. Consumption of raw milk and raw milk products does, however, carry a zoonotic risk, as does direct contact with cattle by farm husbandry staff and other employees. This review will mainly focus on the latter, and deal with it from the standpoint of a well-developed dairy industry, using the example of the Netherlands. With regard to dairy cattle, the main bacterial pathogens are Salmonella spp., Listeria monocytogenes and Leptospi

TRIM69 Inhibits Vesicular Stomatitis Indiana Virus.

Vesicular stomatitis Indiana virus (VSIV), formerly known as vesicular stomatitis virus (VSV) Indiana (VSVIND), is a model virus that is exceptionally sensitive to the inhibitory action of interferons (IFNs). Interferons induce an antiviral state by stimulating the expression of hundreds of interferon-stimulated genes (ISGs). These ISGs can constrain viral replication, limit tissue tropism, reduce pathogenicity, and inhibit viral transmission. Since VSIV is used as a backbone for multiple oncolytic and vaccine strategies, understanding how ISGs restrict VSIV not only helps in understanding VSI

Knowledge mapping of risk mitigation measures against vector-borne diseases.

A comprehensive synthesis of current evidence on surveillance, prevention and control measures for 25 selected vector-borne diseases (VBDs) affecting animals in the EU is presented here. The assessment integrates evidence from systematic literature reviews, modelling studies, field investigations and expert judgement. The aim is mapping the availability and effectiveness of risk mitigation measures (RMMs), including surveillance, movement restrictions, vaccination, culling, medicinal treatments, on-farm biosecurity and vector control. Of the 25 VBDs considered, 13 are listed under the EU Anima

A Quadruple Gene-Deleted Live BoHV-1 Subunit RVFV Vaccine Vector Reactivates from Latency and Replicates in the TG Neurons of Calves but Is Not Transported to and Shed from Nasal Mucosa.

Bovine herpesvirus type 1 (BoHV-1) establishes lifelong latency in trigeminal ganglionic (TG) neurons following intranasal and ocular infection in cattle. Periodically, the latent virus reactivates in the TG due to stress and is transported anterogradely to nerve endings in the nasal epithelium, where the virus replicates and sheds. Consequently, BoHV-1 is transmitted to susceptible animals and maintained in the cattle population. Modified live BoHV-1 vaccine strains (BoHV-1 MLV) also have a similar latency reactivation. Therefore, they circulate and are maintained in cattle herds. Additionall

Socioeconomic risk markers of arthropod-borne virus (arbovirus) infections: a systematic literature review and meta-analysis.

INTRODUCTION: Arthropod-borne viruses (arboviruses) are of notable public health importance worldwide, owing to their potential to cause explosive outbreaks and induce debilitating and potentially life-threatening disease manifestations. This systematic review and meta-analysis aims to assess the relationship between markers of socioeconomic position (SEP) and infection due to arboviruses with mosquito vectors. METHODS: We conducted a systematic search on PubMed, Embase, and LILACS databases to identify studies published between 1980 and 2020 that measured the association of SEP markers with

Triazavirin might be the new hope to fight Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2).

Since the beginning of the outbreak, a large number of clinical trials have been registered worldwide, and thousands of drugs have been investigated to face new health emergency of highly contagious COVID-19 caused by the Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2). Drug repurposing, i.e., utilizing an approved drug for a different indication, offers a time- and cost-efficient alternative for making new (relevant) therapies available to physicians and patients. Considering given strategy, many approved and investigational antiviral compounds, alone or in various relevant combi

Schlafen 11 Restricts Flavivirus Replication.

Schlafen 11 (Slfn11) is an interferon-stimulated gene that controls the synthesis of proteins by regulating tRNA abundance. Likely through this mechanism, Slfn11 has previously been shown to impair human immunodeficiency virus type 1 (HIV-1) infection and the expression of codon-biased open reading frames. Because replication of positive-sense single-stranded RNA [(+)ssRNA] viruses requires the immediate translation of the incoming viral genome, whereas negative-sense single-stranded RNA [(-)ssRNA] viruses carry at infection an RNA replicase that makes multiple translation-competent copies of

Sirtuin Inhibitors Are Broadly Antiviral against Arboviruses.

Arthropod-borne viruses are diverse pathogens and are often associated with human disease. These viruses span multiple genera, including flaviviruses, alphaviruses, and bunyaviruses. In a high-throughput drug screen, we found that tenovin-1 was antiviral against the flaviviruses Zika virus and dengue virus. Tenovin-1 is a sirtuin inhibitor, and here we found that inhibition of sirtuins, but not inhibition of the related histone deacetylases, is potently antiviral against diverse arboviruses. Sirtuin inhibitors block infection of arboviruses in multiple human cell types. We found that sirtuin i

Elongin C Contributes to RNA Polymerase II Degradation by the Interferon Antagonist NSs of La Crosse Orthobunyavirus.

Mosquito-borne La Crosse virus (LACV; genus Orthobunyavirus, family Peribunyaviridae, order Bunyavirales) causes up to 100 annual cases of severe meningoencephalitis in children and young adults in the United States. A major virulence factor of LACV is the nonstructural protein NSs, which inhibits host cell mRNA synthesis to prevent the induction of antiviral type I interferons (IFN-α/β). To achieve this host transcriptional shutoff, LACV NSs drives the proteasomal degradation of RPB1, the large subunit of mammalian RNA polymerase II. Here, we show that NSs acts in a surprisingly rapid manner,

La Crosse virus, but not Jamestown Canyon virus, is dependent on the host translation termination factor eRF1 due to changes in the nonstructural protein NSm.

UNLABELLED: Bunyaviruses are a class of segmented, negative-sense RNA viruses that are transmitted by arthropod and rodent vectors. Members of the Orthobunyavirus genus and California Serogroup (CSG) circulate in the U.S., including La Crosse Virus (LACV), which is a leading cause of pediatric encephalitis. The host factors required for infection, particularly those that impact bunyavirus translation, are poorly understood. We found that host eukaryotic release factor 1 (eRF1) is important for viral infection; genetic depletion or pharmacological degradation of eRF1 inhibits LACV infection. eR

Mosquito dynamics and their drivers in peri-urban Antananarivo, Madagascar: insights from a longitudinal multi-host single-site survey.

BACKGROUND: Antananarivo, the capital city of Madagascar, is experiencing a steady increase in population growth. Due to the abundance of mosquito vectors in this locality, the population exposed to mosquito-borne diseases is therefore also increasing, as is the risk of epidemic episodes. The aim of the present study was to assess, in a resource-limited setting, the information on mosquito population dynamics and disease transmission risk that can be provided through a longitudinal entomological study carried out in a multi-host single site. METHODS: Mosquitoes were collected every 15 days ov

Distribution and relative densities of Aedes aegypti in Niger.

Its climate and environmental conditions put Niger at particularly high risk of the spread of Aedes mosquitoes, which can transmit arboviruses. In September 2016, the Republic of Niger reported its first outbreak of Rift Valley Fever (RVF) in the northern region of Tahoua, near the Mali border, particularly in the departments of Tchintabaraden, Tassara, and Abalak. The history of RVF has showed that epidemics and epizootics have a considerable socioeconomic impact in affected countries, such as Niger, whose populations are particularly concerned by livestock farming. Cross-sectional studies we

The production and evaluation of a standard diagnostic peste des petits ruminants (PPR) hyperimmune serum prepared from the Egyptian antigen (Egypt 87).

The aim of this study was to produce a specific hyperimmune serum for diagnosis of peste des petits ruminants (PPR). Based on good laboratory practices and standard operating procedures, we produced this reagent in goats using attenuated local strain of PPRV. The quality was assured to meet the internationally required levels of potency and sterility. The titer of the product was 1024 as evaluated by virus neutralization (VN) and agar gel immunodiffusion (AGID) tests. In its final form, it is a sterile disc of a lyophilized material with a pale yellow colour, placed into an evacuated, tightly

Climate-sensitive infectious diseases in transition: a scoping review of key diseases and their epidemiology, data applications and mechanism of outbreak response in Saudi Arabia.

BACKGROUND: Climate-sensitive infectious diseases (CSID), of both vector- and water-borne diseases present growing public health concerns in Saudi Arabia. By virtue of the absence of evidence-based vaccine for most of these diseases, adequate management should primeirly rely on timely and reliable data integration, robust disease surveillance, and temporal and spatial vector control strategies. OBJECTIVE: This scoping review aimed to examine the scale and scope of CSID in Saudi Arabia, with attempts to elicit all diseases' trends and patterns, the availability of associated epidemiological, e

Prioritization of zoonoses for multisectoral, One Health collaboration in Somalia, 2023.

BACKGROUND: The human population of Somalia is vulnerable to zoonoses due to a high reliance on animal husbandry. This disease risk is exacerbated by relatively low income (poverty) and weak state capacity for health service delivery in the country as well as climate extremes and geopolitical instability in the region. To address this threat to public health efficiently and effectively, it is essential that all sectors have a common understanding of the priority zoonotic diseases of greatest concern to the country. Linalool oxide: generalist plant based lure for mosquito disease vectors.

BACKGROUND: Lack of effective vaccines and therapeutics for important arboviral diseases such as Rift Valley fever (RVF) and dengue, necessitates continuous monitoring of vector populations for infections in them. Plant-based lures as surveillance tools has the potential of targeting mosquitoes of both sexes and females of varied physiological states; yet such lures are lacking for vectors of these diseases. Here, we present evidence of the effectiveness of linalool oxide (LO), a single plant-based lure previously developed for malaria vectors in trapping RVF vectors, Aedes mcintoshi and Aedes
