Continuous and Divergent Biotransformation of β-Ketoesters Into Enantiomerically Pure β-Hydroxyacids or Their Oligomers Through a Telescoped Multienzyme Cascade.

Efficient, selective, and sustainable synthetic platforms are increasingly important in modern chemical manufacturing, yet chemical flexibility often remains limited. In this study, we develop a modular biocatalytic system for the divergent and continuous transformation of ethyl acetoacetate (EAA) into either enantiomerically pure (R)-3-hydroxybutyric acid ((R)-3HBA) or oligomers of (R)-poly(3-hydroxybutyrate) (PHB). The cascade combines two telescoped packed-bed reactors containing an immobilized, self-sufficient Lactobacillus kefir ketoreductase (LkKRED) and commercial immobilized Candida an

Cerebral glucose and energy utilization during the evolution of hypoxic-ischemic brain damage in the immature rat.

The cerebral metabolic rate for glucose (CMRg1) and cerebral energy utilization (CEU) were assessed in immature rats during recovery from cerebral hypoxia-ischemia. CMRg1 was determined using a modification of the Sokoloff technique with 2-deoxy-[14C]glucose (2-DG) as the radioactive tracer. CEU was determined using the Lowry decapitation technique. Seven-day postnatal rats underwent unilateral common carotid artery ligation, followed 4 h thereafter by exposure to 8% oxygen at 37 degrees C for 3 h. At 1, 4, or 24 h of recovery, the rat pups underwent those procedures necessary for the measurem

Effect of excess and deficiency of vitamin A on the utilization of FFA by liver and skeletal muscle.

Fatty acid metabolism in liver and skeletal muscle has been studied in rats treated with high doses of vitamin A and in those made vitamin A-deficient. Ingestion of 30,000 IU of vitamin A for two days resulted in increased incorporation of palmitate-1-14C into triglycerides but not into phospholipids. Accumulation of hepatic triglycerides was observed in vitamin A-fed rats. Deficiency of vitamin A did not cause any change in the triglyceride or phospholipid content of the liver. The rate of hepatic fatty acid oxidation and ketogenesis was markedly increased in vitamin A-fed rats. The experimen

The influence of diabetic regulation on the diurnal variation in blood and the urinary excretion of ketone bodies. Studies in newly diagnosed diabetics.

A typical diurnal variation of blood ketone bodies (acetacetate+3-hydroxybutyrate) has been found in 23 newly diagnosed, untreated diabetics regardless of diabetic type. It was characterized by high morning concentrations, followed by a fall, and later on a rise in concentration during the evening and night. Satisfactory diabetic regulation, judged from glucose measurements, did not change this diabetic pattern, which differed fundamentally from the normal diurnal variation. In the regulated state the morning concentration of ketone bodies in blood was still abnormally high in the juvenile and

Multicomponent synthesis of novel imidazole-pyran derivatives: in silico and in vitro studies.

Herein, a three-component reaction was used for the synthesis of imidazole-pyran derivatives (4a-n) through the reaction between imidazol-5-carbaldehyde (1a-n), malononitrile (2), and methyl acetoacetate (3). The reaction process was simple, quick, proceeded without the need for any purification technique and used green solvents. The synthesized substances (4a-n) were evaluated for their potential anticancer effects on the MCF-7 (breast cancer), HT29 (colon cancer), and A2780cis (cisplatin-resistant ovarian cancer) cell lines, and a control normal cell line, CHO (Chinese hamster ovary). Requirement for oxidation of neuronal ketone bodies in aging and neurodegeneration.

Glucose is the brain's primary fuel, but the brain can also use alternative energy substrates, especially during development or starvation. Emerging evidence suggests ketone metabolism may help the brain adapt to energy stress in neurodegenerative diseases such as Alzheimer's disease, although its role in constitutive brain function in normal aging is poorly understood. Using iPSC-derived human neurons and adult-inducible, neuron-specific Bdh1 knockout mice, we show that ketone body metabolism is essential for maximum energy production, neuronal function, and mouse survival-even under normal n

Ketone metabolism at the interface of atherosclerosis and brain dysfunction.

Atherosclerosis and brain dysfunction converge through vascular inflammation, endothelial injury, and regional bioenergetic failure of the neurovascular unit. Ketone bodies, including acetoacetate and D-β-hydroxybutyrate, perform critical roles in metabolic signaling by serving as alternative oxidative substrates for ATP production and bioactive signaling molecules that modulate cellular pathways. In the arterial wall, ketones improve endothelial function, reduce inflammation, and shift macrophages away from proinflammatory states, as well as induce changes that are predicted to limit lesion g

Metabolomic analysis of larval stages of Onchocerca japonica (Spirurida: Onchocercidae), raised in black fly (Diptera: Simuliidae) vectors, by gas chromatography-tandem mass spectrometry.

To monitor and prevent the spread of zoonotic onchocerciasis, identification of the natural vectors (blood-sucking insects) of its causative agents, Onchocerca species, is crucial. To date, vector identification depends on the detection of infective third-stage larvae in insects by traditional dissection. We aimed to develop a novel, more efficient method for the discrimination of the four larval stages, i.e. microfilariae (Mf), first-stage larvae (L1), second-stage larvae (L2), and third-stage larvae (L3), of O. japonica by metabolomic analysis. Microfilariae of O. japonica, the causative age

Efficient fabrication of chitin-based films with high UV-blocking and fluorescence via Hantzsch reaction.

As the second most abundant polysaccharide on earth, chitin-based films are promising packaging and biomedical materials. However, the lack of special function (especially UV-blocking properties and fluorescence) usually restricts their further and high-value applications. Herein, high UV-blocking and fluorescence chitin-based films were fabricated via surface modification using acetoacetation and the Hantzsch reaction. The structure and properties of the films were characterized by NMR, FTIR, fluorescence spectrum, and tensile tests. The results showed that the benzene ring and 1,4-dihydropyr

Facile synthesis of functionalized quinolinones in a green reaction medium and their photophysical properties.

A facile and green chemical approach was successfully developed to construct functionalized quinolinones utilizing substituted alcohols, alkyl acetoacetate, and α-bromo ketones. Various quinolinones bearing either electron-rich or electron-deficient groups at different positions were synthesized in moderate to good yields under mild reaction conditions. The plausible mechanistic pathway for this transformation is supported by experimental evidence and control experiments. This simple approach for synthesizing quinolinones could open new avenues for discovering novel biological and pharmaceutic

A Novel Acetone Sensor for Body Fluids.

Ketones are well-known biomarkers of fat oxidation produced in the liver as a result of lipolysis. These biomarkers include acetoacetic acid and β-hydroxybutyric acid in the blood/urine and acetone in our breath and skin. Monitoring ketone production in the body is essential for people who use caloric intake deficit to reduce body weight or use ketogenic diets for wellness or therapeutic treatments. Current methods to monitor ketones include urine dipsticks, capillary blood monitors, and breath analyzers. However, these existing methods have certain disadvantages that preclude them from being

Unexplained Tachypneoa and Severe Metabolic Acidosis in a Three-Month-Old Child: A Rare Presentation of Beta-Ketothiolose Deficiency.

Beta-ketothiolase (mitochondrial acetoacetyl-CoA thiolase, T2) deficiency is a rare inborn error of metabolism that is characterized by impaired metabolism of ketones and isoleucine. The condition is inherited as an autosomal recessive disorder. Herein, we present a child with T2 deficiency from Mahaoya, Eastern Province, Sri Lanka. This three-month-old child presented with fever, difficulty in breathing, and irritability for one day and was subsequently found to have severe metabolic acidosis with positive ketone bodies in urine. His blood glucose was normal. Metabolic screening showed increa

Ketogenic Diet and Weight Loss: Is There an Effect on Energy Expenditure?

A dysregulation between energy intake (EI) and energy expenditure (EE), the two components of the energy balance equation, is one of the mechanisms responsible for the development of obesity. Conservation of energy equilibrium is deemed a dynamic process and alterations of one component (energy intake or energy expenditure) lead to biological and/or behavioral compensatory changes in the counterpart. The interplay between energy demand and caloric intake appears designed to guarantee an adequate fuel supply in variable life contexts. In the past decades, researchers focused their attention on

Catalytic and Multicomponent Reactions for Green Synthesis of Some Pyrazolone Compounds and Evaluation as Antimicrobial Agents.

A green synthetic approach and facile method was developed to produce pyrazole compounds (6a-d) by the reaction of ethyl acetoacetate (1), hydrazines (2a-d), and catalytic imidazole (3) in aqueous media. 4-Dicyanomethylene-2-pyrazoline-5-one derivatives (14a-d) were synthesized through the reaction of 2-pyrazoline-5-one derivatives (6a-d) with tetracyanoethylene (TCE) (7) by using catalytic imidazole (3) in an aqueous medium. Moreover, the 4-dicyanomethylene derivative (16) was obtained via treatment of 1-phenyl-3,5-pyrazolidinedione (15) with TCE (7). The spiropyrazoleoxirane derivatives (18

In Vivo Estimation of Ketogenesis Using Metabolic Flux Analysis-Technical Aspects and Model Interpretation.

Ketogenesis occurs in liver mitochondria where acetyl-CoA molecules, derived from lipid oxidation, are condensed into acetoacetate (AcAc) and reduced to β-hydroxybutyrate (BHB). During carbohydrate scarcity, these two ketones are released into circulation at high rates and used as oxidative fuels in peripheral tissues. Despite their physiological relevance and emerging roles in a variety of diseases, endogenous ketone production is rarely measured in vivo using tracer approaches. Accurate determination of this flux requires a two-pool model, simultaneous BHB and AcAc tracers, and special consi

Ketone bodies as a therapeutic for Alzheimer's disease.

An early feature of Alzheimer's disease (AD) is region-specific declines in brain glucose metabolism. Unlike other tissues in the body, the brain does not efficiently metabolize fats; hence the adult human brain relies almost exclusively on glucose as an energy substrate. Therefore, inhibition of glucose metabolism can have profound effects on brain function. The hypometabolism seen in AD has recently attracted attention as a possible target for intervention in the disease process. One promising approach is to supplement the normal glucose supply of the brain with ketone bodies (KB), which inc

β-hydroxybutyrate potentiates anti-tumor immunity by modulating cytotoxic CD8+ T cell responses.

Ketogenic diets (KDs) have been reported to influence tumor progression through metabolic and immunological modulation of the tumor microenvironment. β-hydroxybutyrate (βOHB), the predominant ketone body elevated by KD, functions not only as an energy substrate but also as a potent signaling metabolite. Despite its role in modulating the tumor microenvironment, the direct impact of βOHB on the function of CD8+ T cell, a key mediator of anti-tumor immunity, remains incompletely understood. Here, we demonstrate that βOHB suppresses tumor growth in multiple mouse tumor models by enhancing the acc
