[Nicotinic acetylcholine receptors are possible therapeutic targets for schizophrenia].

The rate of smoking in patients with schizophrenia is higher than that in the general population. Nicotinic acetylcholine receptors (nAChR) are involved in the sensorimotor gating deficits in schizophrenia. We have revealed that nicotine ameliorates the disruption of the PPI, a model of sensorimotor gating, which is induced by apomorphine, a dopamine receptor agonist, but is not effective for the disruption of the PPI induced by phencyclidine, a glutamine NMDA receptor antagonist, in rats. Furthermore, the ameliorating effect of nicotine is antagonized by methyllycaconitine, a selective alpha7

Treatment of myasthenia gravis by preventing acetylcholine receptor modulation.

Myasthenia gravis (MG) is an autoimmune disease caused by antibodies mainly directed to the acetylcholine receptor (AChR) of the neuromuscular junction. Induction of antigenic modulation and complement activation by such autoantibodies leads to ultrastructural damage of the postsynaptic membrane and loss of AChR and associated proteins. Reduction of antigenic modulation by increasing the expression of the receptor-associated anchor protein, rapsyn, or by functionally monovalent competing IgG4 anti-AChR antibodies was shown to prevent MG disease activity. We propose that preventing antigenic mo

IgG1 antibodies to acetylcholine receptors in 'seronegative' myasthenia gravis.

Only around 80% of patients with generalized myasthenia gravis (MG) have serum antibodies to acetylcholine receptor [AChR; acetylcholine receptor antibody positive myasthenia gravis (AChR-MG)] by the radioimmunoprecipitation assay used worldwide. Antibodies to muscle specific kinase [MuSK; MuSK antibody positive myasthenia gravis (MuSK-MG)] make up a variable proportion of the remaining 20%. The patients with neither AChR nor MuSK antibodies are often called seronegative (seronegative MG, SNMG). There is accumulating evidence that SNMG patients are similar to AChR-MG in clinical features and t

Functional significance of non-neuronal acetylcholine in skin epithelia.

In recent years, the physiological role of non-neuronal acetylcholine (ACh) and its receptors (AChR) in epidermal physiology has been under intense investigation. However, little is known about the role of the non-neuronal cholinergic system in inflammatory skin diseases. We chose the clinically nicotine-dependent skin disease hidradenitis suppurativa (HS) as model to study the influence of long term nicotine ingestion on epidermal morphology and AChR expression. HS is a chronic inflammatory, disabling disease of unknown pathogenesis emerging from the pilosebaceous unit of the intertriginous a

Decalin-type acetylcholine-mimetic organophosphates as inhibitors of acetylcholinesterase.

The novel decalin-type N,O,P-heterocycles, P(3)-axially and P(3)-equatorially substituted cis- and trans-configurated 3-fluoro-2,4-dioxa-7-aza-, 2,4-dioxa-8-aza-, and 2,4-dioxa-9-aza-3-phosphabicyclo[4.4.0]decane 3-oxides, are configuratively fixed and conformationally constrained phosphorus analogues of acetylcholine. The compounds are suitable probes for the investigation of molecular interactions with acetylcholinesterase.

Acetylcholine: a novel regulator of airway smooth muscle remodelling?

Increased airway smooth muscle mass is a pathological feature that asthma and chronic obstructive pulmonary disease (COPD) have in common. This increase has gained renewed interest in view of recent developments showing that airway smooth muscle, instead of solely being a contractile partner, is capable of interacting dynamically with its environment, especially under inflammatory conditions. Airway smooth muscle cells are able to proliferate, to migrate, and to secrete chemokines, cytokines, extracellular matrix proteins and growth factors, and most importantly, to adapt to these functions by

Role for M5 muscarinic acetylcholine receptors in cocaine addiction.

Muscarinic cholinergic receptors of the M5 subtype are expressed by dopamine-containing neurons of the ventral tegmentum. These M5 receptors modulate the activity of midbrain dopaminergic neurons, which play an important role in mediating reinforcing properties of abused psychostimulants like cocaine. The potential role of M5 receptors in the reinforcing effects of cocaine was investigated using M5 receptor-deficient mice in a model of acute cocaine self-administration. The M5-deficient mice self-administered cocaine at a significantly lower rate than wild-type controls. In the conditioned pla

Inhibition of m3 muscarinic acetylcholine receptors by local anaesthetics.

1. Muscarinic m1 receptors are inhibited by local anaesthetics (LA) at nM concentrations. To elucidate in more detail the site(s) of LA interaction, we compared these findings with LA effects on m3 muscarinic receptors. 2. We expressed receptors in Xenopus oocytes. Using two-electrode voltage clamp, we measured the effects of lidocaine, QX314 (permanently charged) and benzocaine (permanently uncharged) on Ca(2+)-activated Cl(-)-currents (I(Cl(Ca))), elicited by acetyl-beta-methylcholine bromide (MCh). We also characterized the interaction of lidocaine with [(3)H]-quinuclydinyl benzylate ([(3)H

Doxepin affects acetylcholine induced cutaneous reactions in atopic eczema.

BACKGROUND: Atopic eczema (AE) is a chronic inflammatory skin disease with strong itching as the prominent symptom. The pathology of itch is still in discussion, but acetylcholine (ACH) seems to be a relevant pruritogenic mediator in AE. Since efficient benefit on pruritus and excoriations has been demonstrated with tricyclic agents, we investigated how the topical treatment with doxepin (5%, Boehringer Standard, Mannheim, Germany), a tricyclic compound with anticholinergic properties, may influence ACH induced itch and cutaneous sensations (erythema, wheal, axonreflex flare). METHODS: Eleven

Acetylcholine receptor antibodies in the Lambert-Eaton myasthenic syndrome.

Two patients were initially diagnosed with myasthenia gravis with elevated titers of acetylcholine receptor antibodies. Features including weakness that normalized with sustained contraction, areflexia, autonomic symptoms, and low-amplitude baseline compound muscle action potentials with abnormal increments following brief exercise and high-frequency repetitive stimulation, however, suggested that these patients had Lambert-Eaton myasthenic syndrome. One patient had antibodies directed against presynaptic calcium channels, confirming the diagnosis. The second patient was seronegative for these

Circulating antibodies against nicotinic acetylcholine receptors in chagasic patients.

Human and experimental Chagas' disease causes peripheral nervous system damage involving neuromuscular transmission alterations at the neuromuscular junction. Additionally, autoantibodies directed to peripheral nerves and sarcolemmal proteins of skeletal muscle have been described. In this work, we analyse the ability of serum immunoglobulin factors associated with human chagasic infection to bind the affinity-purified nicotinic acetylcholine receptor (nAChR) from electric organs of Discopyge tschudii and to identify the receptor subunits involved in the interaction. The frequency of serum ant

Responsiveness of brain muscarinic acetylcholine receptors in aged rats.

In the first part of the investigation the age-related changes in cortical, hippocampal and striatal cholinesterases (ChE), choline acetyltransferase (ChAT) and muscarinic acetylcholine receptors (mAChRs, measured as Bmax of 3H-QNB binding) were compared in male Wistar, Fischer 344 and Sprague-Dawley rats aged 3 and 24 months. Fischer rats exhibited 1.5 fold higher levels of ChAT and mAChRs than Wistar and Sprague-Dawley rats in the areas analyzed. The age-related decline of cortical ChAT and mAChRs was present in Fischer rats but not in those of the other two strains, i.e. was strain-dependen

Different types of blockade of crustacean acetylcholine-induced currents.

The voltage dependence, concentration dependence, and agonist dependence of blocking and unblocking produced by anticholinergic agents on the ionophoretically activated cholinergic currents of the lobster gastric mill 1 (g.m.1) muscle were examined. Although the ionophoretic technique provides only qualitative information as to blocking mechanisms it is useful in revealing slow components of the blocking action of some drugs. At least two qualitatively different types of voltage-dependent block of the crustacean cholinergic currents were observed. For pempidine, mecamylamine and decamethonium

Nicotinic acetylcholine receptor desensitization studied by [3H]perhydrohistrionicotoxin binding.

Desensitization of the nicotinic acetylcholine (ACh) receptor of Torpedo electric organs was studied by monitoring activated [3H]perhydrohistrionicotoxin binding to the channel sites of the receptor. Receptor desensitization was produced by incubating Torpedo membranes that had been pretreated with diisopropylfluorophosphate with ACh and was quenched by the addition of AChesterase. Desensitization caused concentration- and time-dependent decrease in the binding of [3H]perhydrohistrionicotoxin to carbamylcholine-activated receptors and an increase in the apparent affinity of carbamylcholine to

Reconstitution of acetylcholine receptor function using purified receptor protein.

Membrane preparations containing only the four acetylcholine receptor polypeptide subunits (40, 50, 60, and 65 x 10(3) daltons) were purified from Torpedo californica electroplax. The receptor protein was extracted from these membranes with 2% aqueous sodium cholate, and complete dissolution into discrete molecular species was confirmed by sedimentation analysis. The solubilized preparation reassociated with exogenous phospholipids when the detergent was removed by dialysis and formed spherically sealed vesicles 400--600 A in diameter. The reconstituted receptor preparations had nearly 90% of

Release of acetylcholine by chick embryo heart before innervation.

1. In chick embryo hearts, 3-day-old and not yet innervated, repetitive direct stimulation causes a transitory inhibition of the spontaneous rhythm.2. The degree of post-stimulation inhibition depends on the frequency and duration of the artificial stimulation and on the concentration of K and Ca ions in the extracellular solution.3. After treatment with atropine (10(-5) g/ml.) post-stimulation inhibition is no longer observed. The spontaneous rhythm is accelerated by atropine. The findings therefore suggest that an ACh-like substance is released from the non-innervated embryonic heart during

Citicoline in cardiovascular protection: mechanistic roles of choline and acetylcholine.

Citicoline (CDP-choline) is a rate-limiting precursor for phosphatidylcholine synthesis and acetylcholine production, and thus a unique metabolic hub linking membrane phospholipid homeostasis with cholinergic signaling. The biochemical rationale for the therapeutic potential of citicoline in heart disease is rooted in fundamental metabolic principles and conserved cytoprotective pathways. However, the cardiovascular evidence base for citicoline is not as extensive as its established neuroprotective profile. The cardioprotection is mainly mediated by the cholinergic system, including vagal effe

Binding mechanism and antagonism of the vesicular acetylcholine transporter VAChT.

The vesicular acetylcholine transporter (VAChT) has a pivotal role in packaging and transporting acetylcholine for exocytotic release, serving as a vital component of cholinergic neurotransmission. Dysregulation of its function can result in neurological disorders. It also serves as a target for developing radiotracers to quantify cholinergic neuron deficits in neurodegenerative conditions. Here we unveil the cryo-electron microscopy structures of human VAChT in its apo state, the substrate acetylcholine-bound state and the inhibitor vesamicol-bound state. These structures assume a lumen-facin
