GPR120: Mechanism of action, role and potential for medical applications.

G protein-coupled receptors (GPCRs) constitute a family of transmembrane proteins that mediate many cellular processes. GPR120/FFAR4, a receptor from this family that is activated by fatty acids, has received considerable attention recently. This paper presents a literature review concerning the role of GPR120 and its mechanism of action in animal and human studies as well as the potential use of GPR120 for the treatment of chronic diseases. Two electronic databases - Medline and Google Scholar - were searched for available studies addressing the review topic that were written in English and p

Potential Therapeutic Action of Adiponectin in Duchenne Muscular Dystrophy.

Adiponectin (ApN) is a hormone that exhibits anti-inflammatory effects on skeletal muscle exposed to acute and chronic inflammation. We have previously tested the implication of ApN in Duchenne muscular dystrophy (DMD) using mdx mice, a model of DMD, and by generating transgenic mdx mice overexpressing ApN. We showed that ApN can act as a preventive agent and delay disease progression by reducing muscle inflammation/injury and improving force/myogenesis. Herein, we took an opposite approach and crossed mdx mice with ApN knockout mice, to obtain mdx mice with ApN depletion. The aims were to tes

Potential therapeutic action of nitrite in sickle cell disease.

Sickle cell disease is caused by a mutant form of hemoglobin that polymerizes under hypoxic conditions, increasing rigidity, fragility, calcium influx-mediated dehydration, and adhesivity of red blood cells. Increased red cell fragility results in hemolysis, which reduces nitric oxide (NO) bioavailability, and induces platelet activation and inflammation leading to adhesion of circulating blood cells. Nitric Oxide inhibits adhesion and platelet activation. Nitrite has emerged as an attractive therapeutic agent that targets delivery of NO activity to areas of hypoxia through bioactivation by de

Melatonin potentiates the anticonvulsant action of phenobarbital in neonatal rats.

Phenobarbital is the most commonly utilized drug for neonatal seizures. However, questions regarding safety and efficacy of this drug make it particularly compelling to identify adjunct therapies that could boost therapeutic benefit. One potential adjunct therapy is melatonin. Melatonin is used clinically in neonatal and pediatric populations, and moreover, it exerts anticonvulsant actions in adult rats. However, it has not been previously evaluated for anticonvulsant effects in neonatal rats. Here, we tested the hypothesis that melatonin would exert anticonvulsant effects, either alone, or in

Predictions of the EC50 for action potential block for aliphatic solutes.

Experiments were conducted to test the hypothesis that aliphatic hydrocarbons bind to pockets/crevices of sodium (Na(+)) channels to cause action potential (AP) block. Aliphatic solutes exhibiting successively greater octanol/water partitition coefficients (K (ow)) were studied. Each solute blocked Na(+) channels. The 50% effective concentration (EC(50)) to block APs could be mathematically predicted as a function of the solute's properties. The solutes studied were methyl ethyl ketone (MEK), cyclohexanone, dichloromethane, chloroform and triethylamine (TriEA); the K (ow) increased from MEK to

Effect of calmodulin antagonists on the compound action potential of the cochlea.

This study aimed to evaluate the effect of calmodulin antagonists on the threshold of the compound action potential (CAP) and the functional recovery of the cochlea after transient ischemia. When trifluoperazine and W-7 were administered to albino guinea pigs with perilymphatic perfusion, these drugs did not significantly affect the CAP thresholds. Transient cochlear ischemia of 30-min duration was obtained via a skull base approach. Although trifluoperazine significantly ameliorated the post-ischemic CAP threshold shifts 4 h after the onset of reperfusion, 1 to 50 microM W-7 did not affect th

[Effect of histamine on the action potential of the maxillary nerve in rabbits].

The significant enhancement of action potential as recorded from rabbit maxillary nerve with topically applied 1.0 mmol/L histamine on the nasal mucosa was completely blocked by pretreatment with diphenhydramine (H1 antagonist) but not with cimetidine (H2 antagonist). This fact gives new support to the concept that histamine exerts its pathological effect on nasal mucosa at least partly via an afferent nervous pathway of the trigeminal nerve.

Naloxone potentiates epinephrine's pressor actions in endotoxemic rats.

Naloxone's pressor effects in shock may be mediated through antagonism of endogenous opioid inhibition of sympathoadrenal catecholaminergic systems. Since circulating catecholamine levels are not further elevated by naloxone in endotoxemic animals, it is possible that naloxone acts upon opiate receptors to enhance catecholamine actions at the receptor or postreceptor level. To investigate this hypothesis, we sought to determine whether naloxone treatment would augment the pressor actions of exogenous catecholamines (epinephrine) in normal and endotoxemic rats. Naloxone (3 mg/kg intravenous [i.

[Changes in the compound action potential in patients with acoustic neuroma].

Thirty-eight patients with proven acoustic neuroma were divided into three groups: 13 patients with normal threshold on pure tone audiogram, 15 patients with a hearing loss of 40 dB-80 dB (HL) in the low and middle frequencies, and 10 totally or almost deaf patients. Results of the first two groups are reported, using intra-tympanic electrodes with alternating clicks of 90 dB (nHL) intensity and a stimulus rate of 20/s. A total of 500 sweeps were averaged. Widening of the CAP complex by a mean of 1.6 ms, compared to 1.1 ms in normal hearing subjects, was the most striking finding. Summation po

Measurement of action potential thresholds in experimental endolymphatic hydrops.

Unilateral experimental endolymphatic hydrops was created by endolymphatic sac and duct obstruction in guinea pigs. Studies of action potential (AP) threshold were then performed and compared with histologic findings. Action potential thresholds were significantly increased in the hydrops ears. The close relationship between AP changes and hydrops was sufficient to allow the detection of hydrops by AP threshold measurements alone. The AP threshold changes were positively related to the degree of hydrops and postoperative interval in a manner consistent with hearing changes seen in Meniere's di

Potentiation by the injection vehicle of the teratological action of acetazolamide in rats.

The high alkalinity of the injection vehicle of certain parenteral solutions of acetazolamide produces necrosis of the skin upon sc injection. The possible modification of this effect on the teratogenicity of acetazolamide was examined. Acetazolamide in a vehicle of pH 10.5 produced 36.6% fetal malformations, in a vehicle of pH 8.7, 6.1%, and in neutral suspension, 11.8%. Adrenal medullectomy or phentolamine plus the high pH acetazolamide reduced the frequency to 23.2 and 24.4%, respectively. The teratogenicity of the low pH acetazolamide was increased by epinephrine to 64.2%. The frequency of

Cooperative activation of action potential Na+ ionophore by neurotoxins.

Four neurotoxins that activate the action potential Na+ ionophore of electrically excitable neuroblastoma cells interact with two distinct classes of sites, one specific for the alkaloids veratridine, batrachotoxin, and aconitine, and the second specific for scorpion toxin. Positive heterotropic cooperativity is observed between toxins bound at these two classes of sites. Tetrodotoxin is a noncompetitive inhibitor of activation by each of these toxins (KI = 4-8 nM). These results suggest the existence of three functionally separable components of the action potential Na+ ionophore: two regulat

EPCR in Wound Healing: Mechanisms of Action and Therapeutic Potential.

The endothelial protein C receptor (EPCR) is an important component of the protein C (PC) system, recognised for its diverse roles in blood coagulation, inflammation, and stem cell regulation. Wound healing is a complex physiological process that can be divided into four distinct but overlapping phases: haemostasis, inflammation, proliferation and remodelling. Recently, EPCR has emerged as a key regulator in wound repair and regeneration. During haemostasis, EPCR enhances the conversion of PC to its activated form (APC) to optimise local and systemic anticoagulation. In the inflammatory phase,

Mechanisms of action and therapeutic potential of PCSK9-regulating drugs.

CONTEXT: Proprotein convertase subtilisin/kexin type 9 (PCSK9) regulates plasma low-density lipoprotein cholesterol (LDL-C) metabolism and is a key target for cardiovascular therapies. It also plays roles in inflammation, cancer, and metabolic disorders, prompting interest in repurposing PCSK9-targeting drugs for non-lipid conditions. OBJECTIVE: This review comprehensively summarizes PCSK9-regulating medications, delves into their mechanisms of action, and explores their increasingly expanding therapeutic potential across multiple organ systems, such as the liver, immune system, small intesti

Simulation insights on the compound action potential in multifascicular nerves.

We developed an extended reciprocity theorem approach to model neuron signals which handles heterogeneous dielectric environments and arbitrary electrode shapes, and use it to study analytically the single fiber action potential. We then established a semi-analytic model that also uses hybrid electromagnetic-electrophysiological simulations to model evoked compound action potential (eCAP) signals from multi-fascicular nerves populated with heterogeneous fiber populations. For validation, model predictions were compared with cuff electrode recordings of activity induced by vagus nerve stimulati

Tetracyclines and bone: Unclear actions with potentially lasting effects.

Tetracyclines are a broad-spectrum class of antibiotics that have unclear actions with potentially lasting effects on bone metabolism. Initially isolated from Streptomyces, tetracycline proved to be an effective treatment for Gram +/- infections. The emergence of resistant bacterial strains commanded the development of later generation agents, including minocycline, doxycycline, tigecycline, sarecycline, omadacycline, and eravacycline. In 1957, it was realized that tetracyclines act as bone fluorochrome labels due to their high affinity for the bone mineral matrix. Over the course of the next

Glycolysis selectively shapes the presynaptic action potential waveform.

Mitochondria are major suppliers of cellular energy in neurons; however, utilization of energy from glycolysis vs. mitochondrial oxidative phosphorylation (OxPhos) in the presynaptic compartment during neurotransmission is largely unknown. Using presynaptic and postsynaptic recordings from the mouse calyx of Held, we examined the effect of acute selective pharmacological inhibition of glycolysis or mitochondrial OxPhos on multiple mechanisms regulating presynaptic function. Inhibition of glycolysis via glucose depletion and iodoacetic acid (1 mM) treatment, but not mitochondrial OxPhos, rapidl
